Efficient and Cost-Reduced Glucoamylase Fed-Batch Production with Alternative Carbon Sources

  • Glucoamylase is an important industrial enzyme. Glucoamylase production by industrial Aspergillus niger strain featured with two major problems: (i) empirical substrate feeding methods deteriorating the fermentation performance; and (ii) the high raw materials cost limiting the economics of the glucoamylase product with delegated specification. In this study, we first proposed a novel three-stage varied-rate substrate feeding strategy for efficient glucoamylase production in a 5 L bioreactor using the standard feeding medium, by comparing the changing patterns of the important physiological parameters such as DO, OUR, RQ, etc., when using different substrate feeding strategies. With this strategy, the glucoamylase activity and productivity reached higher levels of 11,000 U/ml and 84.6 U/ml/h, respectively. The performance enhancement in this case was beneficial from the following results: DO and OUR could be controlled at the higher levels (30%, 43.83 mmol/l/h), while RQ was maintained at a stable/lower level of 0.60 simultaneously throughout the fed-batch phase. Based on this three-stage varied-rate substrate feeding strategy, we further evaluated the economics of using alternative carbon sources, attempting to reduce the raw materials cost. The results revealed that cornstarch hydrolysate could be considered as the best carbon source to replace the standard and expensive feeding medium. In this case, the production cost of the glucoamylase with delegated specification (5,000 U/ml) could be saved by more than 61% while the product quality be ensured simultaneously. The proposed strategy showed application potential in improving the economics of industrial glucoamylase production.

GTPase Activity Analysis of eRF3 in Euplotes octocarinatus

  • In eukaryotes, eRF3 participates in translation termination and belongs to the superfamily of GTPases. In this work, the dissociation constants for nucleosides bound to Euplotes octocarinatus eRF3 in the presence and absence of eRF1a were determined using fluorescence spectra methods. Furthermore, a GTP hydrolyzing assay of eRF3 was carried out using an HPLC method, and the kinetic parameters for GTP hydrolysis by eRF3 were determined. Consistent with data from humans, the results showed that eRF1a promoted the binding of GTP to eRF3 and the GTP hydrolyzing activity of eRF3. However, in contrast to the lack of GTP binding in the absence of eRF1 in human eRF3, the E. octocarinatus eRF3 was able to bind GTP by itself. The nucleotide binding affinity of the E. octocarinatus eRF3 also differed from the human data. A structure model and amino acid sequence alignment of potential G domains indicated that these differences may be due to valine 317 and glutamate 452 displacing the conserved glycine and lysine involved in GTP binding.

Cloning and Characterization of a PI-like MADS-Box Gene in Phalaenopsis Orchid

  • The highly evolved flowers of orchids have colorful sepals and fused columns that offer an opportunity to discover new genes involved in floral development in monocotyledon species. In this investigation, we cloned and characterized the homologous PISTALLATA-like (PI-like) gene PhPI15 ($\underline{Ph}alaenopsis$ $\underline{PI}$ STILLATA # $\underline{15}$), from the Phalaenopsis hybrid cultivar. The protein sequence encoded by PhPI15 contains a typical PI-motif. Its sequence also formed a subclade with other monocot PI-type genes in phylogenetic analysis. Southern analysis showed that PhPI15 was present in the Phalaenopsis orchid genome as a single copy. Furthermore, it was expressed in all the whorls of the Phalaenopsis flower, while no expression was detected in vegetative organs. The flowers of transgenic tobacco plants ectopically expressing PhPI15 showed male-sterile phenotypes. Thus, as a Class-B MADS-box gene, PhPI15 specifies floral organ identity in orchids.

Molecular Analyses of the Metallothionein Gene Family in Rice (Oryza sativa L.)

  • Metallothioneins are a group of low molecular mass and cysteine-rich metal-binding proteins, ubiquitously found in most living organisms. They play an important role in maintaining intracellular metal homeostasis, eliminating metal toxification and protecting against intracellular oxidative damages. Analysis of complete rice genome sequences revealed eleven genes encoding putative metallothionein (OsMT), indicating that OsMTs constitute a small gene family in rice. Expression profiling revealed that each member of the OsMT gene family differs not only in sequence but also in their tissue expression patterns, suggesting that these isoforms may have different functions they perform in specific tissues. On the basis of OsMT structural and phylogenetic analysis, the OsMT family was classified as two classes and class I was subdivided into four types. Additionally, in this paper we also present a complete overview of this family, describing the gene structure, genome localization, upstream regulatory element, and exon/intron organization of each member in order to provide valuable insight into this OsMT gene family.

Synthesis of Needle-like Aragonite from Limestone without Calcinations in the Presence of Magnesium Sulfate

  • Much attention has been paid to the processing of inorganic whisker, especially calcium carbonate whisker, which can be used as reinforcement materials of polymer composite due to its low price. Unfortunately, the present synthesis technique of calcium carbonate whisker starts from calcinations of limestone, which involves high energy consumption and furthermore is a highly environment polluting reaction. In this report, needle-like aragonite was synthesized with a reversible solution reaction from limestone without calcination. Optical microscopy, scanning electron microscopy (SEM) and X-ray diffraction (XRD) techniques were used to characterize the morphology and crystal structure of intermediates as well as that of the product, aragonite. GCC (grinding calcium carbonate) powder was dissolved in an aqueous solution of magnesium sulfate with reflux and air flush. EDTA titration was used to evaluate reaction rate of the dissolution. A kinetics equation of the dissolution reaction was constructed, which displayed second-order kinetics with respect to the concentration of magnesium sulfate. A rate constant of $0.0015\;l^{-3}{\cdot}mol^{-1}{\cdot}h^{-1}$ was obtained. The dissolution reaction gave fiber-like magnesium hydroxide sulfate and gypsum crystal. Then needle-like aragonite with a length of $9.13\;{\pm}\;1.02\;{\mu}m$ and an aspect ratio of $5.64\;{\pm}\;1.37$ was synthesized from the dissolution product with $CO_2$ bubbling at $70^{\circ}C$.

Reduction of the Retransmission Delay for Heterogeneous Devices in Dynamic Opportunistic Device-to-device Network

  • The dynamic opportunistic device-to-device (DO-D2D) network will frequently emerge in the fifth generation (5G) wireless communication due to high-density and fast-moving mobile devices. In order to improve the Quality of Experience (QoE) of users with different computing capacity devices in the DO-D2D network, in this paper, we focus on the study of how to reduce the packets retransmission delay and satisfy heterogeneous devices. To select as many devices as possible to transmit simultaneously without interference, the concurrent transmitters-selecting algorithm is firstly put forward. It jointly considers the number of packets successfully received by each device and the device's connectivity. Then, to satisfy different devices' demands while primarily ensuring the base-layer packets successfully received by all the devices, the layer-cooperation instantly decodable network coding is presented, which is used to select transmission packets combination for each transmitter. Simulation results illustrate that there is an appreciable retransmission delay gain especially in the poor channel quality network compared to the traditional base-station (BS) retransmission algorithm. In addition, our proposed algorithms perform well to satisfy the different demands of users with heterogeneous devices.

Development of a novel self-centering buckling-restrained brace with BFRP composite tendons

  • Buckling-restrained braces (BRBs) have excellent hysteretic behavior while buckling-restrained braced frames (BRBFs) are susceptible to residual lateral deformations. To address this drawback, a novel self-centering (SC) BRB with Basalt fiber reinforced polymer (BFRP) composite tendons is presented in this work. The configuration and mechanics of proposed BFRP-SC-BRBs are first discussed. Then an 1840-mm-long BFRP-SC-BRB specimen is fabricated and tested to verify its hysteric and self-centering performance. The tested specimen has an expected flag-shaped hysteresis character, showing a distinct self-centering tendency. During the test, the residual deformation of the specimen is only about 0.6 mm. The gap between anchorage plates and welding ends of bracing tubes performs as expected with the maximum opening value 6 mm when brace is in compression. The OpenSEES software is employed to conduct numerical analysis. Experiment results are used to validate the modeling methodology. Then the proposed numerical model is used to evaluate the influence of initial prestress, tendon diameter and core plate thickness on the performance of BFRP-SC-BRBs. Results show that both the increase of initial prestress and tendon diameters can obviously improve the self-centering effect of BFRP-SC-BRBs. With the increase of core plate thickness, the energy dissipation is improved while the residual deformation is generated when the core plate strength exceeds initial prestress force.

Research on Speed Estimation Method of Induction Motor based on Improved Fuzzy Kalman Filtering

  • An improved fuzzy Kalman filtering speed estimation scheme was proposed by means of measuring stator side voltage and current value based on vector control state equation of induction motor. The designed fuzzy adaptive controller conducted recursive online correction of measurement noise covariance matrix by monitoring the ratio of theory residuals and actual residuals to make it approach real noise level gradually, allowing the filter to perform optimal estimation to improve estimation accuracy of EKF. Meanwhile, co-simulation scheme based on MATLAB and Ansoft was proposed in order to improve simulation accuracy. Field-circuit coupling problems of induction motor under the action of vector control were solved and the parameter optimization accuracy was improved dramatically. The simulation and experimental results show that this algorithm has a strong ability to inhibit the random measurement noise. It is able to estimate motor speed accurately, and has superior static and dynamic characteristics.

Experimental research on the propagation of plastic hinge length for multi-scale reinforced concrete columns under cyclic loading

  • The plastic hinge lengths of beams and columns are a critical demand parameter in the nonlinear analysis of structures using the finite element method. The numerical model of a plastic hinge plays an important role in evaluating the response and damage of a structure to earthquakes or other loads causing the formation of plastic hinges. Previous research demonstrates that the plastic hinge length of reinforced concrete (RC) columns is closely related to section size, reinforcement ratio, reinforcement strength, concrete strength, axial compression ratio, and so on. However, because of the limitations of testing facilities, there is a lack of experimental data on columns with large section sizes and high axial compression ratios. In this work, we conducted a series of quasi-static tests for columns with large section sizes (up to 700 mm) and high axial compression ratios (up to 0.6) to explore the propagation of plastic hinge length during the whole loading process. The experimental results show that besides these parameters mentioned in previous work, the plastic hinge of RC columns is also affected by loading amplitude and size effect. Therefore, an approach toward considering the effect of these two parameters is discussed in this work.

Static behavior of stud shear connectors with initial damage in steel-UHPC composite bridges

  • For steel-concrete girders made composite using shear studs, initial damage on studs induced by weld defect, unexpected overloading, fatigue and others might degrade the service performance and even threaten the structural safety. This paper conducted a numerical study to investigate the static behavior of damaged stud shear connectors that were embedded in ultra high performance concrete (UHPC). Parameters included damage degree and damage location. The material nonlinear behavior was characterized by multi-linear stress-strain relationship and damage plasticity model. The results indicated that the shear strength was not sensitive to the damage degree when the damage occurred at 2/3d (d is the stud diameter) from the stud root. An increased stud area would be engaged in resisting shear force as the distance of damage location from stud root increased and the failure section becomes inclined, resulting in a less reduction in the shear strength and shear stiffness. The reduction factor was proposed to consider the degradation of the shear strength of the damaged stud. The reduction factor can be calculated using two approaches: a linear relationship and a square relationship with the damage degree corresponding to the shear strength dominated by the section area and the nominal diameter of the damaged stud. It was found that the proposed method is preferred to predict the shear strength of a stud with initial damage.
