Effects of light, methyl jasmonate(MJ), 6-benzylaminopurine(BA), thidiazouron(TDZ), and 2,4-dichlorophenoxyacetic acid(2,4-D) on the contents of ecdysteroids (${\beta}$-ecdysone+polypodine B) in Polypodium vulgare L. and Achyranthes japonica Nakai were studied. When the plants of P. vulgare were cultured under light control, the ecdysteroids contents in both leaves and rhizomes decreased with increasing light intensity. The ecdysteroids contents in A. japonica were also lower when cultured under light than under dark. Among the tested plant growth regulators, MJ, BA, and TDZ increased the ecdysteroids contents in both P. vulgare and A. japonica.

One of the biggest problems encountered during the last phase of silkworm rearing is non-uniform maturation of the silkworms especially during cooler months. Phytoecdysteroid (20-hydroxy ecdysone) was extracted in large-scale from a plant belongs to Caryophyllaceae and fed to silkworm larvae to test the effect of phytoecdysteroid. About 80% of the silkworms were ready for mounting by 18 hrs after treatment (when the treatment is done for uniform spinning), whereas in control batch only 37% worms were ready for mounting by the same time.
The prothoracicotropic hormone (PTTH) produced by the neurosecretory cells in insects is involved in molting and metamorphosis by activating the prothoracic frins) glands to secrete ecdysone (or related ecdvsteroidsl. In the present study, the small PTTH-like gene was isolated by screening of CDNA library using the bombvxin (corresponding to small PTTH in Bombvx moril gene probe in Drosophilo melonogaster. It showed 50-6096 sequence homology to bombyxin gene. The expression patterns of this gene showed developmental stage- and tissue-dependent manners. The mRNA was detected only in the late third instar larval-prepupa which is stases showing the highest hormonal activity to secrete ecdysteroids, and detected in the brain pan of the Isle third instar lanrae.
The herbs of Lamium takesimense Nakai (Lamiaceae) is used to treat spasmodic and inflammatory disease. The four polar compounds, ecdysterone, isoacteoside, rutin and lamiuside C, were isolated and identified from the BuOH fraction of the L. takesimense MeOH extract. HPLC quantification was performed on a Capcell Pak C18 column ($5{\mu}m$, $4.6mm{\times}250mm$) with a gradient elution of $H_2O$ and 0.05% acetic acid in MeOH. The HPLC method was validated in terms of linearity, sensitivity, stability, precision, and accuracy. The quantitative level in plant material was determined as the following order: lamiuside C (4, 3.75 mg/g dry weight) > ecdysterone (1, 1.93 mg/g) > isoacteoside (2, 1.32 mg/g) > rutin (3, 0.97 mg/g).
Adult diapause in insects is characterized by suppression of reproductive development. It is induced by environmental cues such as photoperiod, temperature, food availability, and other conditions Diapause-inducing environment is recognized and analyzed by the brain of the insects. The interpreted information is conveyed via endocrine system to target tissues such as ovaries, fat body, and other tissues. From this signal hierarchy of a brain-endocrine-target tissue axis, several factors are involved to express a diapause trait in a quantitative mode, even though the insects show a binomial phenotye between being in diapause or not. Recent works estimated that the number of the factors is relatively small by a series of crossing trials between high and low diapause lines. Heritability of the diapause is quite high (ca. 70%) in some species. Epistasis, sex-linkage, pleiotropism, and other nongenetic components also affect diapause inheritance. Most physiological studies have been focused on control mechanisms of the juvenile hormone (JH) synthesis in corpora allata (CA) because JH level in hemolymph of teneral adults is critical to decide a later developmental mode. Allatostatin, an antagonizer of JH synthesis, has been believed to be a potent brain message to CA for adult diapause induction.
These studies were carried out to investigate the effects of nonsteroidal ecdysone agonist, tebufenozide on embryonic and postembryonic development of sycamore lace bug, Corythucha ciliata. Tebufenozide exhibited ovicidal activity($LC_{50}=4.0$ ppm). Susceptibility of each instar nymphs to tebufenozide was nearly same($LC_{50}=2.3{\sim}6.0$ ppm). The tebufenozide treatment in the final instar affected the emergence, longevity and fecundity of surving adults significantly. Tebufenozide applied at high concentration (10 ppm) to younger adults delayed prioviposition period, and decreased adult longevity, the number of eggs laid per female and hatchability. The treatment also inhibited the ovarian development of the female adults.

Athyrium acutipinnulum, called as Ulleungdo ladyfern (Seom-go-sa-ri), is a native plant of South Korea. A. acutipinnulum has been consumed as foods and also traditionally used for the treatment of epilepsy, gonorrhea and nerve disorder. The methanolic extract and EtOAc soluble fraction of A. acutipinnulum showed the antioxidant activity. Fractionation using various chromatographic techniques resulted in the isolation of 13 compounds. The structures were elucidated on the basis of spectroscopic methods as seven phenolic compounds, methyl 2-hydroxy-3-phenylpropanoate (1), protocatechualdehyde (2), caffeic acid (3), trans-p-coumaric acid (4), (-)-4-E-caffeoyl-L-threonic acid (5), 5-O-caffeoyl shikimic acid (6) and 5-O-caffeoyl quinic acid (7), three flavonoids, quercetin 3-O-β-glucoside (8), naringenin-7-O-β-glucoside (9) and sutchenoside A (10), two steroids, ponasterone A (11) and ecdysone (12) and a coumarin, esculetin (13). Among them, compounds 5 and 10 were first reported from Athyrium spp and compounds 2, 5, 6 and 7 showed the antioxidant activity.
Recent studies provide some evidence that the HtrA2 protein is intimately associated with the pathogenesis of neurodegenerative disorders and that endoplasmic reticulum (ER) quality control and ER stress-associated cell death play critical roles in neuronal cell death. However, little is known about the intimate relationship between HtrA2 and ER stress-associated cellular responses. In the present study, we have demonstrated that the HtrA2 protein level was gradually and significantly increased by up to to-fold in the mitochondria under tunicamycin (Tm)-induced ER stress, which eventually promoted cell death through the release of HtrA2 into the cytoplasm. Using an ecdysone-inducible mammalian expression system, we demonstrate that the extent of cell death in 293-HtrA2 cells was approximately 20 times higher under Tm-induced ER stress, indicating that the increase in the HtrA2 protein level in the mitochondria itself is necessary but not sufficient for the promotion of cell death. Taken together, these results suggest that HtrA2 may serve as a mediator of ER stress-induced apoptosis and ER-mitochondrial cross-talk in some cellular processes.
This study was carried out to investigate the effective extraction condition for increase of phytoecdysteroids from fronds of Matteuccia struthiopteris (FMS) and Osmunda japonica (FOJ). Lyophilizated fronds were mixed with three different solvents (MeOH, 80% EtOH or water) and then extraction was carried out by using six different methods, such as, immersion (room temp.), heating (60$^{\circ}C$), stirring (200 rpm) for 6 h, or sonication in 42 kHz ultrasonic bath for 15, 30 and 45 minutes. Contents of 20-hydroxyecdysone (20E) and ponasterone A (PonA) were measured by using HPLC after purification of the extracts by $C_{18}$ cartridge. Altogether, our results indicate that the extraction using sonication with MeOH as a solvent (for 30 minutes) was the most effective condition for 20E and PonA from both MFS and FOJ. Resulting contents of 20E from FMS and FOJ were 66.76 and 104.48 ${\mu}g{\cdot}g^{-1}$ and PonA were 53.43 and 43.82 ${\mu}g{\cdot}g^{-1}$, respectively.
