Web-Based Database and Viewer of East Asian Copy Number Variations

  • We have discovered copy number variations (CNVs) in 3,578 Korean individuals with the Affymetrix Genome-Wide SNP array 5.0, and 4,003 copy number variation regions (CNVRs) were defined in a previous study. To explore the details of the variants easily in related studies, we built a database, cataloging the CNVs and related information. This system helps researchers browsing these variants with gene and structure variant annotations. Users can easily find specific regions with search options and verify them from system-integrated genome browsers with annotations.

  • The concepts of Yeongwi(營衛) were made from "Hwangjenaegyeong(黃帝內經)" the most ancient and important book in East Asian Traditional Medicine, that have been used basic theories for the purpose of explaining physiology and pathology of The Oriental Medicine. In this article, through comparing with concepts of Yeongwi(營衛) in Onbyeonghak(溫病學) of Qing[淸] dynasty, some problems were investigated, through what process of thinking the unique pattern identification[辨證] of Wigiyeonghyeol(衛氣營血) had been made although there could be another inductive way, and what is the difference of concepts of Yeongwi(營衛) between "Hwangjenaegyeong(黃帝內經)" and Onbyeonghak(溫病學).

노루궁뎅이 버섯 자실체의 비극성 분획에서 분리된 성분들 (Chemical Constituents from Non-polar Fraction of the Fruiting Bodies of Hericium erinaceum)

  • Lion's Mane Mushroom, Hericium erinaceum, is a traditional edible mushroom widely used in culinary applications. It has been also used as a medicine in East Asian countries due to its various biological activities. Chemical investigation of fruiting bodies of this mushroom afforded many aromatic compounds, which were mostly isolated from polar fraction of its extracts. Herein we tried to investigate non-polar compounds from the extracts of this mushroom. $CHCl_3$-soluble fraction of the extracts was subjected to chemical investigation, which resulted in isolation of four compounds. Their chemical structures were elucidated as ircicerebroside (1), cortenuamide A (2), 1-D-arabinitol-monolinoleate (3), and cinnamic acid (4) on the basis of spectroscopic data. To the best of our knowledge, this is the first report of compounds 1, 2, and 4 from Hericium erinaceum.

Genome-wide single-nucleotide polymorphism data and mitochondrial hypervariable region 1 nucleotide sequence reveal the origin of the Akhal-Teke horse

  • Objective: The study investigated the origin of the Akhal-Teke horse using genome-wide single-nucleotide polymorphism (SNP) data and mitochondrial hypervariable region 1 (HVR-1) nucleotide sequences Methods: Genome-wide SNP data from 22 breeds (481 horses) and mitochondrial HVR-1 sequences from 24 breeds (544 sequences) worldwide to examine the origin of the Akhal-Teke horse. The data were analyzed using principal component analysis, linkage disequilibrium analysis, neighbor-joining dendrograms, and ancestry inference to determine the population relationships, ancestral source, genetic structure, and relationships with other varieties. Results: A close genetic relationship between the Akhal-Teke horse and horses from the Middle East was found. Analysis of mitochondrial HVR-1 sequences showed that there were no shared haplotypes between the Akhal-Teke and Tarpan horses, and the mitochondrial data indicated that the Akhal-Teke horse has not historically expanded its group. Ancestral inference suggested that Arabian and Caspian horses were the likely ancestors of the Akhal-Teke horse. Conclusion: The Akhal-Teke horse originated in the Middle East.

최근 북한 고려 의학에 반영된 『동의보감』 연구 (A Study in the Influence of The Treasured Mirror of Eastern Medicine (Donguibogam) upon the Koryo Medicine in North Korea)

  • Objectives: This article aims to clarify the understanding and publication status of both The Treasured Mirror of Eastern Medicine (Donguibogam) and some of its recent research progress made in North Korea, as well as their influences upon the academic of Koryo medicine in North Korea. Methods: Analysis was carried out based on various reference books, dictionaries and Korean translation of Donguibogam (1964 Ver.) published by North Korea, as well as the North Korean quarterly medical journal, Koryo Medicine. Results: The academic of Koryo medicine in North Korea regards Donguibogam, a Korean medical classic as it was written by native Korean physicians and it became an important ground material for basic research and clinical study. Various terminologies used in the original version of Donguibogam were defined as unscientific and superstitious by North Korean academics, which were then subsequently removed from the Korean translation of Donguibogam (1964 Ver.) published by North Korea. Therefore, this version cannot be seen as a complete edition of Donguibogam. Conclusions: The Donguibogam is known as one of the 'three major books of Koryo medicine' in North Korea. As a treasure of East-Asian traditional medical classic, Donguibogam is expected to become the medium for a closer research collaboration between the North and South Koreas including China in the future.

조선의 주요 국가간행의학서의 편제구성과 질병분류인식에 대한 개설적 연구 (A Study on the Contents of State-sponsored Medical Texts Published in the Joseon Dynasty Korea)

  • In many periods of Korean history, state-sponsored medical books played a crucial role in terms of distributing medical knowledge as well as systemizing medical information. This study uses comparative analysis to examine the tables on contents of state-sponsored medical publications in Korea. These tables of contents reveal the placement and categorization of medical knowledge, which implicates the ways in which diseases were classified. Historically, Korean medicine has been influenced by Chinese medicine, and at the same time, it has made steady efforts to localize Chinese medicine. This paper argues that Korean medicine adopted the Chinese styles of categorizing medical knowledge in the middle of 15th century for the first time and shows the tendency to Koreanize medical knowledge through the early 17th century. In the 18th century the Complete Records of Medicine (醫部全錄) shows the trace of referring the style of Korean medical book, the Treasured Mirror of Eastern Medicine (東醫寶鑑) in terms of categorizing medical knowledge.

Functional characterization of Clonorchis sinensis choline transporter

  • Clonorchis sinensis is commonly found in East Asian countries. Clonorchiasis is prevalent in these countries and can lead to various clinical symptoms. In this study, we used overlap extension polymerase chain reaction (PCR) and the Xenopus laevis oocyte expression system to isolate a cDNA encoding the choline transporter of C. sinensis (CsChT). We subsequently characterized recombinant CsChT. Expression of CsChT in X. laevis oocytes enabled efficient transport of radiolabeled choline, with no detectable uptake of arginine, α-ketoglutarate, p-aminohippurate, taurocholate, and estrone sulfate. Influx and efflux experiments showed that CsChT-mediated choline uptake was time- and sodium-dependent, with no exchange properties. Concentration-dependent analyses of revealed saturable kinetics consistent with the Michaelis-Menten equation, while nonlinear regression analyses revealed a Km value of 8.3 µM and a Vmax of 61.0 pmol/oocyte/h. These findings contribute to widen our understanding of CsChT transport properties and the cascade of choline metabolisms within C. sinensis.

Modulation of L-Arginine-Arginase Metabolic Pathway Enzymes: Immunocytochemistry and mRNA Expression in Peripheral Blood and Tissue Levels in Head and Neck Squamous Cell Carcinomas in North East India

  • Background: Arginine may play important roles in tumor progression by providing ornithine for polyamine biosynthesis, required for cell growth. The aim of this work was to determine the expression of arginine metabolic pathway enzymes in head and neck squamous cell carcinoma (HNSCC) in northeast India. Materials and Methods: The expressions of arginase isoforms (ARG1 and ARG2), ornithine aminotransferase (OAT) and ornithine decarboxylase (ODC) were examined in fifty paired HNSCC and adjacent non-tumor tissues by immunohistochemistry. Immunocytochemistry, semiquantitative reverse transcription sq-PCR and quantitative real-time qPCR were used to assess protein and mRNA expressions in peripheral blood of fifty HNSCC patients and hundred controls. Results: ARG1 and ODC protein and mRNA were strongly expressed in peripheral blood from HNSCC patients. No ARG2 expression was observed. In vivo, expression of ARG1, ARG2 and ODC was significantly higher in tumor than in non-tumor tissues. Most tumors expressed low levels of OAT, with no difference in tissues or blood, compared to controls. The absolute extent of maximal ARG1 upregulation with qPCR showed 6.23 fold increase in HNSCC. Conclusions: These findings strongly suggest that in HNSCCs, the ARG1 pathway is stimulated leading to the formation of polyamines as indicated by higher ODC expression, which promote tumor growth.

MicroRNAs May Serve as Emerging Molecular Biomarkers for Diagnosis and Prognostic Assessment or as Targets for Therapy in Gastric Cancer

  • Gastric cancer (GC) is one of the most common cancers, with high incidences in East Asia countries. Most GC patients have been reported with low early diagnosis rate and show extremely poor prognosis. Therefore, it is necessary to develop novel and more sensitive biomarkers to improve early diagnosis and therapy in order to provide longer survival and better quality of life for gastric cancer patients. MicroRNAs (miRNAs) play crucial roles in GC development and progression. miRNAs have emerged as a novel molecular biomarker for cancer diagnosis, prognosis and therapy with surprising stability in tissues, serum or other body fluids. This review summarizes major advances in our current knowledge about potential miRNA biomarkers for GC that have been reported in the past two years.
