This study was performed to observe the effects of dietary fat levels and sources on lipids contents and cellularities of liver, brain, and adipose tissue of early weaned rats. Male Sprague-Dawley rats were prematurely weaned from postnatal 17th day with the experimental diets differ in fat levels : low(5%), medium(10%), high(20%) and fat sources : butter, soybean oil, butter+ soybean oil. On the postnatal 29th day, contents of total lipid, triglyceride, cholesterol and phospholipid of serum, liver, brain and adipose tissue were determined, and DNA was determined to assess the cell growth. Rats early weaned fed high fat diet showed lower total lipid and triglyceride levels in serum and liver than those fed medium or low fat diet Rats early weaned fed high fat diet had adipocytes of fewer number, but larger size than those of rats fed low or medium fat diets. Rats early weaned fed soybean oil diet had more adipocytes thu those fed butter diet. Rats normally weaned to commercial chow diet showed lower total lipid, triglyceride, and cholesterol levels in serum and liver, had fewer adipocytes than all early weaned rats except for rats fed high fat-butter diet. These results suggest that high fat-butter diet is ideal weaning diet at early weaning.
This study was designed to examine how dietary protein levels affect organ growth and protein metabolism in early and normally weaned rats. Early and normally weaned rats separated fro the dam on the 15th and 121st day postpartum, respectively. were fed diets containing three levels of protein : low(10%) , normal (20%),and high(40%) . On the 35th day, the weight and DNA, RNA and protein contents in brain , liver, and kidney were determined to ascertain organ and cellular growth. Furthermore, serum total protein , albumin , $\alpha$-amino N and creatine and urinary urea N, and creatinine were determined in order to ascertain protein metabolism and renal functions. Dietary protein levels were not observed to significantly affect total DNA content, which may represent an index of cell number in the liver, brain and kidney. Fresh weight and protein/DNA ratio, which may represent indices of cell size, significantly increased in proportion to dietary protein in the kidney. As for the early weaned rats , the liver cell size significantly decreased. Dietary protein levels and weaning periods did not affect serum total protein and albumin . However, serum urea-N significantly increased in proportion to dietary protein levels whereas serum $\alpha$-amino N was decreased by early weaning . Nitrogen retention was lower in early weaned rats fed low or high levels of protein than in normally weaned rats. The results demonstrate that low or high levels of dietary protein have less desirable effects on protein metabolism in prematurely weaned rats.
This study was conducted to investigated the short-term effects of early weaning and protein intake on organ and cell growth, nitrogen metabolism and physiological functions of rats. Five groups of early weaned rats separated from the dam on the 15th day postpartum were each given one five diets consisting of either one of the three levels of casein-low(8%), -normal (16%), and -high(32%), or a normal level (16%) of isolated soy protein(ISP) or egg yolk protein, for 7 days. The normal weaned rats were fed maternal breast milk for three weeks from birth. On the 22nd day postpartum , all the rats were sacrificed . The weight gain of the early weaned rats, especially the ones fed high protein, was observed to be significantly lower than that of the normal weaned rats. By the 15th day, of early weaning and especially in the ISP-fed rats, the total DNA contents of liver and kidney, which may be said to represent an index of cell numbers, significantly decreased, but their fresh and dry weight and protein/DNA ratio, allegedly representing an index of cell size, significantly increased , not affecting the cell number and cell size of brain. There were no differences in total serum protein and albumin concentrations between early and normal weaned rats. In the early weaned rats observed , the serum urea N and $\alpha$-amino N concentrations significantly increased in high protein-fed rats, and decreased in low protein-fed rats. Another observation was that no significant difference was noticed as regards to serum GOT activity, total bilirubin, uric acid, and creatinine concentration, which may represent indices of liver and kidney functions, among rat groups, GPT activity was an exception . These results suggest that premature weaning and the quality and quantity of dietary protein significantly affect organ and cell growth and nitrogen metabolism but does not seriously affect physiological functions in the neonatal development of rats.

Melatonin, which is produced from tryptophan, exerts various biological functions, including the regulation of circadian rhythm, sedative agents, and antioxidant ability. Therefore, we conducted two experiments with early-weaned rats and pigs to investigate the antioxidant and sedative effects of melatonin. In the rat experiment, a total of 42 rats (21 days old) were used, and the antioxidant capacity was determined. Next, we used 120 early-weaned piglets (21 days old) to conduct a 5-week experiment to evaluate the reductive effect of melatonin on energy-wasting movement, including roaming and fight states. Dietary melatonin supplementation significantly improved growth in both rats and pigs compared to the control groups. Additionally, rats fed a melatonin-supplemented diet showed advanced antioxidant capacity with a decrease in hepatic malondialdehyde concentration compared to rats fed a basal diet. Moreover, dietary melatonin ingestion increased resting and feeding behaviors and reduced roaming and fight behaviors during Days 8-21 compared to the control diet group. Collectively, early weaned animals given dietary melatonin supplementation showed improved growth through upregulation of hepatic antioxidant capacity and minimization of energy-wasting behavior, including roaming and fight states, after pigs' social hierarchy establishment.
This study was conducted to investigate the effects of protein levels and casein/whey ratios on organ growth and protein metabolism in early weaned rats. Premature rats weaned by the 17th day were fed six semipurified synthetic, isocaloric and gel diets that contained three levels (low, medium and high) and two different combinations(casein/whey ; 80 : 20 or 20 : 80) of milk protein for 8 days. On the 25th day postpartum, frest weigth and DNA, RNA and milk protein contents in brain, liver, kidney and muscle were determined to ascertain organ and cellular growth. Futher, with a view to ascertain protein metabolism and renal functions, serum total protein, $\alpha$-amino N, urea N, and creatinine and creatinine and urinary urea N, creatinine and hydroxproline were determined. Total DNA contents of brain, liver and kidney, which may represent as an index of cell numbers in those organs were significantly decreased in the rats fed diets containing low level protein regardless of casein/whey ratio. However, as fat as the rats fed high protein diets were concerned, their fresh weight, protein contents and GFR of kidney were significantly increased. Furthermore, nitrogen components, $\alpha$-amino N, urea N and creatinie in serum and urine were also increassed. Another observation was that high casein/whey ratio significantly facilitated accumulation of porteins in muscle and kidney and urinary hydorxyproline excretion, not affecting the DNA content of those organs. This study showed that low(8%) or high(32%) contents of protein had less desirable effects either on protein metabolism or on organ cellular growth in prematurely weaned rats, whereas there were no effects on general growth and bone strength.
본 연구는 초산의 Wistar 계 rat를 이용하여 포유기간 중 제한포유시기와 이유시기를 조절하여 혈중 progesterone 수준을 검토하였다. 정상포유군 (NL)은 포유자의 수를 8 마리로 조절하였으며, 제한포유군 (RL)과 완전이유군 (W)은 RL0, RL5, RL10, RL15 및 RL20, 그리고 W0, W5, W10, W15 및 W20으로 각각 5 개 군으로 구분하여, 제한포유군은 각 개시일에 포유자의 수를 8마리에서 4마리로 조절하였으며, 완전이유군은 각 개시일에 포유자의 수를 완전히 이유시켰다. 1. 포유자의 발육상태는 제한포유군에서 정상포유군보다 성장이 유의적 (P<0.05)으로 증가하였고, 또한 분만직후 (RL0) 와 포유 5(RL5), 10(RL10), 15(RLl5) 일에 제한포유를 실시한 군에서는 시간이 경과함에 따라 정상포유군에 비해 유의적 (P<0.05) 으로 증가하였으나, 포유 20 일(RL20) 에 제한포유군에서는 정상포유군과 차이가 없었다 . 초산 rat 의 임신기간은 21.53$\pm$0.04일이었고 산자수는 13.75$\pm$0.07 마리였다. 2. 포유기간 중 모체의 성주기 변화는 정상포유군은 발정징후가 관찰되지 않았으나 분만직후 제한포유군은 20 일을 전후하여 불규칙적인 성주기가 관찰되었고, 분만직후 완전이유군은 10일을 전후하여 정상적인 성주기가 관찰되었다. 3. 정상포유군의 progesterone 농도는 포유 0 일에 33.16$\pm$2.64ng/$m\ell$에서 포유 10 일에 122.55$\pm$3.68ng/$m\ell$ 까지 증가하여 포유 20 일까지 높은 수준을 유지하다가 그 후 25 일에 97.30$\pm$3.21ng/$m\ell$ 까지 급속히 감소 (P<0.05) 하였다. 포유초기 (RL0, RL5)와 포유중기 (RL10)의 제한포유군은 제한포유실시 이후 5일 까지는 정상포유군과 같은 수준을 유지하다가 그 후 급격히 (P<0.05) 감소하였다. 결론적으로 흰쥐에 있어서 분만 후 포유자의 포유자극이 모체 난소의 기능에 중요하게 작용하는 것으로 사료된다.
본 연구는 Wistar계 rat를 이용하여 포유기간 중 제한포유시기와 이유시기를 조절하여, 혈중 IGF- I 수준변화에 대해 검토하였다. NL군 (NL)은 포유자의 수를 8 마리로 조절하였으며, RL군 (RL)과 W군 (W)은 RL0, RL0, RL10, RLl5 및 RL20, 그리고 W0, W5, WI0, W15 및 W20으로 각각 5개 군으로 구분하여, RL군은 각 개시일에 포유자의 수를 8마리에서 4마리로 조절하였으며, W군은 각 개시일에 포유자의 수를 완전히 이유시켜 다음과 같은 결과를 얻었다. 1. NL군의 혈중 IGF-I 농도는 분만직후 (Day 0) 750.59$\pm$3.52ng/$m\ell$ 을 나타내어 15일까지 일정한 수준을 유지하다가 그 후 포유 25일에 1,690.20$\pm$4.42ng/$m\ell$ 까지 증가되었다. 2. 포유초기 RL군 (RL0, RL5)의 IGF- I 농도는 포유 10일에 RL0군과 RL5군에서 각각 1,395.90$\pm$3.45 및 1,351.73$\pm$3.23ng/$m\ell$로서 NL 군의 745.96$\pm$2.24ng/$m\ell$ 보다 유의적 (P<0.05)으로 높게 나타났으나 포유 15일부터는 차이가 없었다. 3. 포유초기 W군의 IGF- I 농도는 이유 후 일주일 정도는 NL군에 비해 높은 수준 (P<0.05)을 나타내다가 10일경에 NL군과 같은 수준으로 감소하였다. 포유자극의 강약은 난소의 생리적 변화와 함께 혈중 IGF- I의 농도를 조절하는 것으로 사료된다.
The objective of this study was to determine the extent to which the compensatory nutrition regimen modulates lactation performance and milk protein gene expression in the first and second lactation cycles. Female rats(28 days of age) were assigned to 1)control ad libitum ; 2) stari-step compensatory nutrition(SSCN) regimen an alternating 3-2-3-4-week schedule beginning with an energy restriction diet(40% restriction) for 3 weeks followed by the control diet(ad libitum) for 2 weeks and then alternating another 3-4 week feeding regimen. The SSCN rats were received an overall 20% energy restriction(average from all stair-step periods) compared with the conventionally fed control group. Rats were bred during the first week of the second realimentation. All pups were weaned on day 21 of lactation. About 1 week after weaning all dams were mated for the second pregnancy. Mammary tissues were obtained from pregnant and lactating rats during the first and second lactation cycles. During these lactation cycles the SSCN group had a 11% increase in average lactation performance over that of control. The SSCN group had significantly increased levels of milk protein gene($\alpha$- and $\beta$-casein) expression in mammary tissues during the first lactation cycle compared with those of the control group. During the second lactation period the levels of milk protein gene expression in lactating mammary tissues of the SSCN group were also higher than those of the control group. These results suggest that the effects of compensatory growth imposed at an early age extend to the second lactation cycle with regard to increased lactation performance and milk protein gene expression.
간의 부분적 절제수술을 하지 않고 새로운 발암성 검색법을 찾고자, 화학물질에 민감한 신생암첫 Sprague-Dawley 랫드를 이용하여 diethylnitrosamine(DENA)으로 암을 유발시킨 후, 제1군에는 강력한 촉진제로서 2-acetylaminofluorene(2-AAF)을 사료하여 0.01%가 되게 섞어 투여하였고, 제2군은 약한 촉진제인 phenobarbital을 암수에 0.05% 농도로 녹여 토여하였으며, 제 3군은 대조군으로 DENA만을 1회 투여하였다. 그리고 발암성 평가는 glutathione S-transeferase placental form을 사용하여 검색하였다. 그 결과 체중에 대한 간의 무게비는 이유후 4주째 (7주)에 제2군이 제3군인 대조군에 비해 유의성 있게 (p<0.01)높았으며, 이유후 8주째(11주)에는 제1군과 제 2군이 대조군에 비해 각각 유의성 있게 (p<0.01, p<0.001)높았다. 그리고 이유후 4주(7주)째에 GST-P 양성병변을 이용한 전암병변의 면적을 비교해본바 제 1군과 제2군이 각각 유의성있게 (p<0.01, p<0.05)높게 나타났다. 이상의 결과에서 신생 랫드를 이용한 발암성 실험은 간의 부분적 절제수술을 하지 않고도 화학물질의 발암성을 좀더 이른 시기에 검색할 수 있음을 알 수 있었다. 그러므로 신생동물을 이용한 발암성 실험은 많은 화학물질들의 발암성을 검색하는데 매우 유용한 방법으로 사료된다.
