The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

Six brands of encapsulated fishoil products and five fishes were analyzed for their contents of eicosapentaenoic acid (EPA 20 :5 n-3) , docosahexaenoic acid(DHA 22: 6 n-3) and tocopherols. In both of the fish oil products and the fishes, major fatty acids were palmitic acid, oleic acid, eicosapentaeoic acid and docosahexaenoic acid and fatty acid compositions were also similar pattern each other. EPA showed variable amounts from 19.2 to 50.3% in the oil products whereas DHA were 13.2% to 28.3% inthe fishes. Tocopherols were studied in relation to the oxidative stability of fish products no relation was observed. However the amount of tocopherols in fish oil proudcts were higher than that of fishes. Contents of EPA, DHA and tocopherols in encapsulated fishoil products oil products were variable comparing with manufactures' claimed contents.
This study designed to compare the hypolipidemic e(feats of n6 linoleic acid (LA), n3 w-linolenic acid(LL) and n3 eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) In rats fed high fat (40% Cal) diet. Male Sprague-Dawley rats fed experimental diets for 6 weeks, which were different only in fatty acid composition. The dietary fats were beef tallow (BT) as a source of saturated fatty acid (SFA), corn oil(CO) for n6 LA, perilla oil (PO) for n3 a-LL and fish oil (FO) for n3 EPA+DHA. Plasma total cholesterol (T-chol) level was increased by n6 LA but decreased by n3 LL and n3 EPA+DHA and most effectively reduced by n3 EPA+DHA. Plasma triglyceride(TG ) level was reduced by n6 LA, but lipogenesis in liver was not affected by n6 LA. However, plasma TG level was lowered by n3 LL and EPA+DHA. Both lipogenic enzyme activity and liver TG level were also decreased by n3 PUFA. PO and FO groups were significantly higher in the relative Proportions of C20:5 and C22:6 of plasma and liver and lower in those of C20:4/C20:5 ratio. Overall, the lipid-lowering effect was in the order of n3 EPA+DHA >n3 LL > n6 LA and fish oil and perilla oil rich in n3 PUFA may have important nutritional applications in the prevention and treatment of hyperlipidemia.
To compare the hypolipidemic effects of n6 and n3 PUFA at different fat levels, male Sprague Dawley rats were fed either low fat (LF, 10% Cal) or high fat (HF, 40% Cal) diet which was different only in fatty acid composition for 6 weeks. Dietary fats were beef tallow, corn oil, perilla oil, and fish oil concentrate as a source of saturated fatty acid, n6 linoleic acid(LA). n3 ${\alpha}-linolenic$ acid(LL) and n3 eicosapentaenoic acid(EPA)+docosahexaenoic acid(DHA), respectively. VLDL fraction was separated by ultracentrifugation and chemical composition was determined by thin layer chromatography. Plasma cholesterol level was increased by n6 LA but decreased by n3 LL and n3 EPA in LF and HF diets, and the hypocholesterolemic effect of n3 EPA was most significant in HF diet. HDL-Chol level was raised by n6 LA in LF and HF diets, but significantly reduced by n3 EPA in HF. Plasma TG level was reduced by n6 LA n3 LL and EPA in LF and HF with the reduction of lipogenic enzyme activity only by n3 PUFAs. The proportion of TG in VLDL fraction was significantly lowered by n3 EPA in LF and HF. The proportion of apo-B in VLDL fraction was not changed in LF, but was significantly decreased in HF by n3 EPA. Therefore, the hypotriglyceridemic effect of n3 PUFA could be from the reduced lipogenesis in liver and resulted in the depressed secretion of TG as VLDL in LF and HF with significant lower production of apoB in HF diet.
Chum salmon (Oncorhynchus keta) meat oil contained high amounts of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) compared to oils extracted from other tissues of the fish. EPA and DHA occupied more than 25% of the total fatty acids in chum salmon meat oil. The main lipid classes in the meat oil were triacylglycerides and phospholipids. The major fatty acids of the molecular species composition of phosphatidylcholine isolated from the meat oil were DHA and EPA. DHA and EPA were the major molecular species in the phosphatidylcholine of chum salmon meat oil, representing 44% and 17%, respectively.

HDL-Chol level was raised by n6 LA, but there was no significant change in HDL-Chol levels by n3 LL and was lowered by n3 EPA. Plasma TG level was reduced by n6 LA, but lipogenesis in liver was not affected by n6 LA. However, plasma TG level was lowered by n3 LL and EPA. Both lipogenic enzyme activity and liver TC level were also decreased by n3 PUFA. The relative proportions of TG in VLDL was significantly lowered by n3 EPA. but the proportions of Apo B in VLDL was not changed by n3 EPA. The effects of sardine oil(high in eicosapentaenoic acid : EPA) and tuna oil(high in docosahexaenoic acid : DHA, also high in EPA) on fatty acid composition of brain and learning ability were evaluated in male weanling Sprague-Dawley rats and compared with the effects of corn oil and beef tallow. Animals assigned by randomized block design to one of the four experimental diet groups containing dietary lipid at 15%(w/w) level were given ad libitum for 7 weeks. Food intake and body weight gain of the fish oil groups were significantly lower than those of the corn oil and beef tallow groups. However, brain weights of the groups were not significantly different. In the brain fatty acid composition, the corn oil group showed high concentrations of n-6 fatty acids, the fish oil groups of n-3 fatty acids, and the beef tallow group of saturated fatty acids. Brain EPA and DHA contents of the fish oil groups showed significantly higher than the other groups while the brain ratio of saturated/monounsaturated/polyunsaturated fatty acid was controlled in a narrow range. In a maze test, the fish oil groups appeared to arrive at the goal faster than the corn oil and beef tallow groups. It explained that EPA in diets might efficiently convert to DHA resulting in DHA accumulation in brain tissue and might increase the learning performance as DHA did.
The objectives of this study were to investigate the effects of dietary fish oil inclusion on the eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) contents and organoleptic characteristics of breast meat in mule ducks. Three hundred mule ducks at four weeks of age were randomly assigned to three dietary treatments with five replicate pens in each. One replicate pen had ten males and females each with a total of 100 ducks in each treatment. The diet in the three treatments contained 0, 1.5, and 3.0% fish oil, respectively. Body weights at 4, 6, 8, and 10 weeks of age, and feed efficiency at 4 to 6, 6 to 8, and 8 to 10 weeks of age were recorded. At 10 weeks of age, one male and one female from each replicate were sacrificed for oxidative stability of breast meat and the sacrificed males were employed for the analysis of fatty acids in breast meat and skin. Sensory evaluation of breast meat was also performed. A level of 3.0% fish oil in the diet significantly deteriorated feed efficiency and body weight gain. Dietary fish oil inclusion had a trend of increasing abdominal fat deposition and decreasing the flavor of breast meat. The EPA and DHA contents in the breast meat were higher than those in the breast skin irrespective of oil sources. The EPA and DHA contents in breast meat and breast skin was significantly increased in the 3.0% fish oil group. Although EPA and DHA were not efficiently deposited in the duck meat through dietary fish oil inclusion, this method can still provide a partial supplementation of EPA and DHA.
Fatty acid contents of 59 food items which are major fat sources in Korean diet were analyzed. The contents of EPA and DHA in fish were 0.02-2.66g and 0.02-3.01g per 100g edible portion, respectively. Items with high amount of EPA and DHA were canned Sardine, Mackerel, Mackrel pike, Atka-fish, Hair tail, Conger eel and Herring. But white fish such as Croaker, Gindaro, Flounder and frozen Alaskan pollack contained less than 0.1g of EPA and DHA per 100g edible portion. Other sources of n-3 fatty acid were perilla oil, rapeseed oil, soybean oil and walnuts all of which contained relatively high amounts of linolenic acid.
In order to determine the position and the content of fatty acids sttached to glycerides and the migration degree of fatty acids in the migration reaction, fish oil was hydroyzed with lipolase-100T which was derived from Aspergillus oryzae. The content of fatty acids in the glyceride mixture was analyzed and compared with that of fish oil. The amounts of fatty acid in a 2-position and the migration degree of the fatty acid in 2,3-DG (diglyceride) and 2-MG (monolyceride) were carefully calculated. The results showed that approximately 95% (w/w) of DHA (docosahexanoic acid) and 65% of EPA(eicosapentaenoic acid) were attached to the 2-position of glycerides in fish oil. Approximately 87% (w/w) of DHA and 75% of EPA remained in 2,3-DG, and 88% of DHA and 65% of EPA in 2-MG were not involved in the migration reaction.
