Cryopreservation of Human Multi-Pronuclear (PN) Zygote by Ultra-Rapid Freezing (인간 다-전핵기 (>2PN) 수정란의 초급속 동결에 관한 연구)

  • The objective of this study was to test whether the developmental capacity of human multi-pronuclear (PN) zygotes after ultra-rapid freezing using EM grid can be maintained. For this experiment, multi-PN zygotes which produced in human IVF program were used as an alternatives of normal 2PN zygotes, and they were separated into 3PN or $\geq4PN$ zygotes to compare their in vitro development and cryoinjury according to PN number. As freezing solution, EFS30 which consisted of 30% ethylene glycol, 18% bcoll, 0.5 M sucrose and 10% FBS added D-PBS was used. The result obtained in this experiment was summarized as follows; When the multi..PN zygotes were ultrarapidly frozen and thawed, the high mean percentages (85.5%) were survived. Also when the cleavage rates between control and freezing group were compared with PN number, there were not significantly different in each group (3PN; 81.3% & 85.4% and $\geq4PN$; 90.0% & 95.7%). When the in vitro development rates after thawing were examined, freezing 3PN group (22.0%) was not differed to control 3PN group (38.5%), although the development result of freezing $\geq4PN$ group (45%) was significantly lower than that of control $\geq4PN$ group (44.4%) (p<0.05). These results demonstrate that developmental capacity of human zygote can be obtained by ultra-rapid freezing method using EM grid and EFS30.

Extensions of X-means with Efficient Learning the Number of Clusters (X-means 확장을 통한 효율적인 집단 개수의 결정)

  • K-means is one of the simplest unsupervised learning algorithms that solve the clustering problem. However K-means suffers the basic shortcoming: the number of clusters k has to be known in advance. In this paper, we propose extensions of X-means, which can estimate the number of clusters using Bayesian information criterion(BIC). We introduce two different versions of algorithm: modified X-means(MX-means) and generalized X-means(GX-means), which employ one full covariance matrix for one cluster and so can estimate the number of clusters efficiently without severe over-fitting which X-means suffers due to its spherical cluster assumption. The algorithms start with one cluster and try to split a cluster iteratively to maximize the BIC score. The former uses K-means algorithm to find a set of optimal clusters with current k, which makes it simple and fast. However it generates wrongly estimated centers when the clusters are overlapped. The latter uses EM algorithm to estimate the parameters and generates more stable clusters even when the clusters are overlapped. Experiments with synthetic data show that the purposed methods can provide a robust estimate of the number of clusters and cluster parameters compared to other existing top-down algorithms.

Development of Effective Cryopreservation Method for Mouse Oocytes (생쥐 난자의 효율적인 냉동보존 방법 확립을 위한 연구)

  • Objective: The purpose of this study was to evaluate the efficacy and effect of various cryopreservation method on the survival and the cytoskeletal stability of metaphase II mouse oocyte. Methods: Mouse ovulated oocytes were collected and cryopreserved by a modified slow-freezing method with 1.5 M 1, 2-propanediol (PrOH)+0.1 M sucrose or by vitrification using cryo loop and EM grid with 40% ethylene glycol+0.6 M sucrose. Four hours after thawing, intact oocytes were fixed and stained with fluorescein isothiocyanate (FITC)-conjugated monoclonal anti-$\beta$-tubulin antibody to visualize spindle and propidium iodide (PI) to visualize chromosome. Spindle morphology was classified as follows: normal (barrel-shaped), slightly and absolute abnormal (multipolar or absent). Results: Survival rate of the frozen-thawed oocytes in vitrification group was significantly higher than that of slow-freezing group (62.7% vs. 24.4%, p<0.01). Vitrification with cryo loop showed significantly higher survival rate than that with EM grid (67.7% vs. 53.5%, p<0.05). On the other hand, proportion of normal spindle and chromosome configurations of the frozen-thawed oocytes between two vitrification group was not significantly different. Conclusion: For mouse ovulated oocytes, vitrification with cryo loop may be a preferable procedure compared to slow-freezing method. Further study should be needed to investigate developmental competency of frozen-thawed mouse oocytes.

An Improved Method for EM Radioautographic Techniques using Cork (EM Radioautographic Techniques에 관(關)한 연구(硏究) - Cork 방법(方法) -)

  • Electron microscope radioautography introduced by Liquier-Milward (1956) is now used routinely in many laboratories. Most of the technical difficulties in specimen preparation have been overcome. This method is modified from loop method for improvement of EM radioautographic techniques. The advantages of this method are: 1. the use of single specimens on small corks and of a large wire loop, allows the experimenter to avoid the blemishes in the membrane; 2. the surfactant dioctyl sodium sulphosuccinate is added to diluted ILford L4, thus greatly prolonging the period of time over which good emulsion layers can be made; 3. corks can be handled in perspex holder which allows about 20 specimens to be developed simultaneously. The steps of the method comprise: 1. Cut ribbons of ultrathin sections of silver interference colour 2. Pick them up on formvar-coated 200 mesh grids 3. Prestaining of tissues 4. Coat the specimens with a thin layer of carbon by evaporation (30-60A) 5. Mount the specimens on corks (about 1cm apical diameter) using double-sided scotch tape 6. Emulsion coating; a. Take a 250m1 beaker, place it on the pan of a sliding weight balance and weigh it. Add 10 grams extra to the beam. Add pieces of ILford L4 emulsion to the beaker until the balance is swinging freely. Add the 20ml of distilled water that was previously measured out. b. Surfactant dioctyl sodium sulphosuccinate is added to diluted ILford L4. 7. Prepare a series of membranes of gelled emulsion with the wire loop and apply one to each cork-borne specimen. 8. Put the specimens away to expose by pushing the corks into short length of PVC tubing, each tube having a small hole in the side 9. Place the tubes in small boxes together with silica gel. 10. Exposure 11. Developer - Kodak Microdol X for 3 minutes 12. Fixer - A perspex holder can be manufactured which allows 20 specimens to be developed simultaneously. 12. Fixer - 30% sodium thiosulfate for 10 minutes 13. Examination with Siemens Elmiskop 1A electron microscope

  • SAR calculation method following the Mobi-Kids study protocol is analyzed and evaluation method of cumulative RF dose from mobile phones which have been used by a subject of case and control groups is proposed. An SAR database is built by calculating SAR distributions in 4 head models at different ages for representative phone models with the same conducted power. To obtain SAR distribution in a subject's head for a specific commercial phone which had/have been used by him/her, an SAR correction factor using SAR compliance test results is determined. Cumulative dose is calculated by considering mobile phone characteristics and use pattern such as call time and laterality(right and left).

Evaluation of physicochemical and textural properties of chicken breast sausages containing various combinations of salt and sodium tripolyphosphate

  • This study was to investigate the effect of salt alone or in combination with phosphate on physicochemical and textural properties, and chemical interactions of low-fat model sausages. pH, color, expressible moisture (EM), cooking loss (CL), proximate analysis, textural profile analysis and low-vacuum scanning electron microscopy were performed. As salt content increased, color tended to decrease, as did EM and CL parameters, indicating that the ability to retain moisture was improved with increased salt levels (p < 0.05). In addition, textural hardness, gumminess and chewiness all increased with increasing salt (p < 0.05). Sausages with 0.3% salt showed the lowest cohesiveness compared to those with salt levels higher than 0.3% (p < 0.05). Addition of sodium tripolyphosphate (STPP) increased pH of sausages. Increasing salt and STPP did not affect lightness (p > 0.05), but did increase redness and yellowness (p < 0.05). The moisture content was higher when the salt and STPP contents were increased (p < 0.05), but no differences in the fat and protein contents (%) were observed (p > 0.05). EM and CL tended to decrease with increasing salt and STPP. In textural properties, the combination of 1.8% salt and 0.3% STPP was the best among other treatment (p < 0.05). Surface microstructure showed a flat and dense structure with increasing salt and STPP. Since the addition of salt and phosphate improved the functionality, textural and physicochemical properties of meat products in this study, meat products will need to be developed in line with consumer's preference.

Effects of Neuroticism on Situation Awareness, Eye Movement, and Driving Performance (운전자의 신경증 수준에 따른 상황인식, 안구운동 및 운전수행에서의 차이)

  • In this study, the effects of driver's neuroticism on situation awareness(SA), eye movement(EM), and driving performance were examined. In Experiment 1, in which static driving scenes were presented, it was found that the drivers of higher neuroticism showed shorter percentage of dwell time and less percentage of fixation counts for the peripheral area than those of lower neuroticism, although SA scores yielded no difference between the two neuroticism groups. In Experiment 2, differences in the drivers' driving performances as well as SA and EM as a function of neuroticism were compared using driving simulation. The results of Experiment 2 showed the followings. First, contrasting to Experiment 1, the high neuroticism group showed lower SA score than low neuroticism group. Second, the pattern of EM was similar to those of Experiment 1. Third, high neuroticism group deviated more from center of road, and drove slower than low neuroticism group.

  • Titania nanomaterials are widely used as cosmetics and dyes, however the impacts on human health are uncertain, We investigated the biodistribution of inhaled titania nanoparticles in rats, Methods Eight weeks-old SD rats were intubated and inhaled with 3 mg titania nanoparticles, twice a week, for 2 weeks, After inhalation, the rats were sacrificed and tissues or heart, lung. intestine, brain, and liver were obtained, We investigated the tissues with optical microscope (OM), transmission electron microscope (EM), scanning EM, And to analyze titania concentration of each tissue, we lysed the tissues with radioimmunoprecipitation assay (RlPA) lysis buffer or acid. Results Granulation tissues in lung were confirmed on the optical microscope, however the other organs had no abnormalities in OM images, In EM images, the rats which inhaled titania nanoparticles showed calcium deposition at heart, brain, and intestine, Titania concentration in lung was increased on the inhaled rat sacrificed I month after last exposure. Conclusion Inhaled titania nanoparticles is thought to be deposited and make inflammatory reaction in lung, and the deposition was not efficiently cleared over a month. However inhaled titania nanoparticles may rarely pass through the alveolus-blood barrier and distribute to other organs of the bod.

  • In this paper we consider an application of the method of electromagnetic (EM) migration to the interpretation of a typical marine controlled-source (MCSEM) survey consisting of a set of sea-bottom receivers and a moving electrical bipole transmitter. Three-dimensional interpretation of MCSEM data is a very challenging problem because of the enormous number of computations required in the case of the multi-transmitter and multi-receiver data acquisition systems used in these surveys. At the same time, we demonstrate that the MCSEM surveys with their dense system of transmitters and receivers are extremely well suited for application of the migration method. In order to speed up the computation of the migration field, we apply a fast form of integral equation (IE) solution based on the multigrid quasi-linear (MGQL) approximation which we have developed. The principles of migration imaging formulated in this paper are tested on a typical model of a sea-bottom petroleum reservoir.

  • For GPR applications, accurate analysis of the scatterered field is necessary to identify the unknown target. Dyadic Green's Function of the multilayered medium is developed and applied to analyse of underground conduting object. We used method of moment(MOM) with dyadic Green's function, and Discrete Complex Image Method(DCIM).
