Effects of various freezing containers for vitrification freezing on mouse oogenesis

  • Background: In the present study, various freezing containers were tested for mouse embryos of respective developmental stages; embryos were vitrified and then their survival rate and developmental rate were monitored. Mouse two cell, 8 cell, and blastula stage embryos underwent vitrification freezing-thawing and then their recovery rate, survival rate, development rate, and hatching rate were investigated. Methods: EM-grid, OPS, and cryo-loop were utilized for vitrification freezing-thawing of mouse embryos. Results: It was found that recovery rate and survival rate were higher in the group of cryo-loop compared to those of EM-grid (p < 0.05). Embryonic development rate, two cell embryos to blastocyst, as well as hatching rate were higher in the control group compared to the EM-grid group and OPS group (p < 0.05), yet no difference was noted between the control group and cryo-loop group. Development rate and hatching rate of eight cell morulae and blastocysts were all lower in the treatment groups than the control group whilst hatching rate of blastocysts was higher in the control group compared to the groups of EM-grid and OPS (p < 0.05); although the cryo-loop group was shown to be slightly higher than other groups, it was not statistically significant. Conclusions: In the study, we investigate effects of freezing containers on vitrified embryos of respective developmental stages; it was demonstrated that higher developmental rate was shown in more progressed (or developed) embryos with more blastomeres. There was however, no difference in embryonic development rate was shown amongst containers. Taken together, further additional studies are warranted with regards to 1) manipulation techniques of embryos for various vitrification freezing containers and 2) preventive measures against contamination via liquid nitrogen.

Analysis of Properties of Flake Sendust EM Wave Absorber according to Granularity (입도에 따른 Flake Sendust 전파 흡수체의 특성 분석)

  • In this paper, we analyzed the characteristics of the EM wave absorber which was fabricated by using flake sendust (soft metal magnetic powder). The flake sendust was made of 3 different granularity by attrition mill. First, we have fabricated 3 kind of EM wave absorbers using the flake sendust and CPE(Chlorinated Polyethylene) and measured the S-parameters of the EM wave absorber. The complex relative permittivity and permeability were calculated from the measured data and the variations according to a change of granularity were researched. As a result, it was confirmed that the EM wave absorber using flake sendust with the $140{\mu}m$ average granularity has outstanding absorption ability in high frequency range(C band) for the reduction of eddy current loss(increase of permeability) and the increase of space charge polarization(increase of permittivity).

Association of gastric cancer with cytochrome P450 2C19 single-nucleotide polymorphisms in Koreans

  • Cytochrome P450 2C19 (CYP2C19) is a clinically important enzyme involved in the metabolism of therapeutic drugs, including (S)-mephenytoin, omeprazole, proguanil, and diazepam. Individuals are characterized as either extensive metabolizers (EM) or poor metabolizers (PM) on the basis of CYP2C19 enzyme activity. The PM phenotype occurs in 2-5% of Caucasians, but in 18-23% of Asians. To clarify the association between CYP2C19 polymorphisms and gastric cancer in Koreans, we investigated CYP2C19 genotypes ($CYP2C19^*1,\;{^*2},\;and\;^*3$) in 109 patients with gastric cancer and 211 controls. Normal ($CYP2C19^*1$) and defective alleles were detected with polymerase chain reaction/restriction enzyme analysis. CYP2C19 has three hereditary genotypes: homozygous EM, with high enzymatic activity; heterozygous EM, with moderate enzymatic activity; and PM, with no enzyme activity. We found that CYP2C19 heterozygous EM is more closely associated with gastric cancer than is homozygous EM. Because the CYP2C19 genotype varies in Koreans, a genotyping test is desirable to prevent gastropathy recurrence in patients before their doses of omeprazole are reduced during maintenance therapy.

  • Ambient backscatter communication has a drawback in which the transmission power is limited because the data is transmitted using the ambient RF signal. In order to improve transmission efficiency between transceiver, a channel estimator capable of estimating channel state at a receiver is needed. In this paper, we consider the K-means algorithm to improve the performance of the channel estimator based on EM algorithm. The simulation uses MSE as a performance parameter to verify the performance of the proposed channel estimator. The initial value setting through K-means shows improved performance compared to the channel estimation method using the general EM algorithm.

  • Regression tree is a tree-structured solution in which a simple regression model is fitted to the data in each node made by recursive partitioning of predictor space. There have been many efforts to apply tree algorithms to various regression problems like logistic regression and quantile regression. Recently, algorithms have been expanded to the panel data analysis such as RE-EM algorithm by Sela and Simonoff (2012), and extension of GUIDE by Loh and Zheng (2013). The algorithms are briefly introduced and prediction accuracy of three methods are compared in this paper. In general, RE-EM shows good prediction accuracy with least MSE's in the simulation study. A RE-EM tree fitted to business survey index (BSI) panel data shows that sales BSI is the main factor which affects business entrepreneurs' economic sentiment. The economic sentiment BSI of non-manufacturing industries is higher than that of manufacturing ones among the relatively high sales group.

Evaluation of microbiological, cellular and risk factors associated with subclinical mastitis in female buffaloes

  • Objective: This study aimed to evaluate the microbiological and cellular milk profile for the diagnosis of subclinical mastitis in female buffaloes and to assess risk factors for predisposition of the disease. Methods: Analyses were carried out by standard plate count (SPC), identification of species and antibiotic resistance, somatic cell count (SCC), electrical electrical conductivity of milk (ECM), and lactoferrin content in milk. Teat cups were swabbed to evaluate risk factors, observing hyperkeratosis, milking vacuum pressure and cleanliness of the site. Hence, 30 female buffaloes were randomly selected (15 from a group in early lactation and 15 in late lactation). Results: The most common bacteria in the microbiological examination were Staphylococcus spp., Streptococcus spp. and Corynebacterium sp. In the antibiotic sensitivity test, 10 (58.82%) of the 17 antibiotics tested were sensitive to all isolates, and resistant bacteria were Streptococcus uberis, Streptococcus dysgalactiae, Streptococcus haemolyticus, and Escherichia coli. It was observed that positive samples in the microbiological examination showed total bacterial count between $9.10{\times}10^3$ to $6.94{\times}10^6$ colony forming units/mL, SCC between 42,000 to 4,320,000 cells/mL and ECM ranging from 1.85 to 7.40 mS/cm. It was also found that the teat cups had high microbial counts indicating poor hygiene, and even faults in the cleanliness of the animals' waiting room were observed. It is concluded that values of SCC above 537,000 cells/mL and ECM above 3.0 mS/mL are indications of mammary gland infection for this herd; however, the association of these values with a microbiological analysis is necessary to more accurately evaluate the health status of mammary glands with subclinical mastitis. Conclusion: Through phenotypic characterization of bacteria involved in the samples, the genera Staphylococcus spp., Streptococcus spp., and Corynebacterimum bovis were the most prevalent in this study. Faults in environment and equipment hygienization are factors that are directly associated with mastitis.

Analysis and probabilistic modeling of wind characteristics of an arch bridge using structural health monitoring data during typhoons

  • The accurate evaluation of wind characteristics and wind-induced structural responses during a typhoon is of significant importance for bridge design and safety assessment. This paper presents an expectation maximization (EM) algorithm-based angular-linear approach for probabilistic modeling of field-measured wind characteristics. The proposed method has been applied to model the wind speed and direction data during typhoons recorded by the structural health monitoring (SHM) system instrumented on the arch Jiubao Bridge located in Hangzhou, China. In the summer of 2015, three typhoons, i.e., Typhoon Chan-hom, Typhoon Soudelor and Typhoon Goni, made landfall in the east of China and then struck the Jiubao Bridge. By analyzing the wind monitoring data such as the wind speed and direction measured by three anemometers during typhoons, the wind characteristics during typhoons are derived, including the average wind speed and direction, turbulence intensity, gust factor, turbulence integral scale, and power spectral density (PSD). An EM algorithm-based angular-linear modeling approach is proposed for modeling the joint distribution of the wind speed and direction. For the marginal distribution of the wind speed, the finite mixture of two-parameter Weibull distribution is employed, and the finite mixture of von Mises distribution is used to represent the wind direction. The parameters of each distribution model are estimated by use of the EM algorithm, and the optimal model is determined by the values of $R^2$ statistic and the Akaike's information criterion (AIC). The results indicate that the stochastic properties of the wind field around the bridge site during typhoons are effectively characterized by the proposed EM algorithm-based angular-linear modeling approach. The formulated joint distribution of the wind speed and direction can serve as a solid foundation for the purpose of accurately evaluating the typhoon-induced fatigue damage of long-span bridges.

Comparative study on properties of commercial polyvinyl siloxane impression materials (시판 폴리비닐실록산 인상재의 물성에 관한 비교연구)

  • The purpose of this study was to compared with the properties of commercial polyvinyl siloxane impression materials on the Korean product and the other country one. The materials used in this study were 5 hydrophilic polvinyly siloxane impression materials of light body automixing type (Perfect-F, Handae Chemical, Korea; Examix, GC, Japan; Contrast, VOCO, Germany; Express, 3M, U.S.A; Extrude, Kerr, U.S.A.). Specimens of each impression material were fabricated from a mold with dimensions identical to that specified in ASTM D624-91 and were subjected to tensile lode at 500 mm/min until failure for tear strength. Properties of consistency, strain in compression, recovery from deformation, detail reproduction and linear dimensional change were tested according to the testing methods of the ISO specification no.4823(1992). From this study, the follow ing results were obtained 1. The tear strength values were high ET(3.4kN/m), EP(3.4kN/m), PF(3.0kN/m), CT(3.0kN/m), but the lowest was EM(2.2kN/m)(p<0.05). 2. In the consistency, CT(47 mm) had the highest value, followed by PF(42.1 mm), ET(41.2 mm), and EM(39.6 mm), EP(39.2 mm)(p<0.05). It means more flow that the consistency value is high. 3. The strain in compression values were high followed by EM(5.8%), PF(52%), CT(4.6%), ET(4.1%), EP(2.9%)(p<0.05). 4. In the recovery from deformation, ET(99.95%) and PF(99.90%) had the highest value, followed by EP(99.75%), EM(99.74%) without statistical signification, CT(99.64%) had the lowest value(p<0.05). 5. Detail reproduction reappeared to line $20{\mu}m$ all products. 6. The linear dimensional change was a little shrinked all products. and there were not statistical signification (p>0.05). All products were satisfied the ISO specification.

Electrical and VLF EM Responses for Conductive Dipping Dyke (맥상 황화광체에 대한 전기 및 전자탐사 반응)

  • Self-potential, VLF-EM and dipole-dipole resistivity methods have been widely used for exploration of conductive sulfide ore deposit, because of the convenience and low cost of field work and the reliability of their results. The geophysical responses for vein-type sulfide outcrop of Changkoom mine located in Bukwi-Myon, Jinan-Gun, Chollabuk-Do were investigated and compared with its drilling results. The geology around the survey area is composed of acidic volcanics and sediments of Yuchon Group. And sulfides bearing pyrite, pyrrotite, galena etc. are deposited in disseminated or vein type within acidic volcanics. Typical geophysical responses were detected from the above vein type ore body, respectively. From the shape and extent of S.P. anomaly, ore body is dipping westward and extending about 50 m. It is detected that the VLF EM response matching the outline of ore zone is considered as indication of dyke dipping westward. And also resistivity response indicating conductive dipping dyke is detected. From drilling results for outcrop and geophysical anomalies, the shape of ore body is vein type dipping about $70^{\circ}W$ and extending about 50 m.

  • We present a new approximate formulation of the integral equation (IE) method for models with variable background conductivity. This method overcomes the standard limitation of the conventional If method related to the use of a horizontally layered background only. The new approximate IE method still employs the Green's functions for a horizontally layered 1-D model. However, the new method allows us to use an inhomogeneous background with the IE method. The method was carefully tested for modeling the EM field for complex structures with a known variable background conductivity. It can find wide application in modeling EM data for multiple geological models with some common geoelectrical features, like a known inhomogeneous overburden, or salt dome structures.
