Polygalacturonase를 검출하기 위한 종이 기반의 효소결합 면역반응 센서 제작 (Fabrication of a paper-based ELISA to detect polygalacturonase)

  • In this paper, we describe the fabrication of a paper-based enzyme-linked immunosorbent assay (ELISA) to detect polygalacturonase (PG), which is used as a biomarker to determine whether a plant is infected with a disease. The proposed paper-based ELISA can analyze the concentration of PG in a short time using a small sample compared to the traditional ELISA, which is generally performed using a well plate. To increase the resolution of the sensor, we optimized the dilution ratio of the HRP-conjugated goat anti-rabbit IgG antibody and the dilution ratio of the anti-PG and HRP-conjugated goat anti-rabbit IgG antibodies. Furthermore, for quantitative analysis of PG concentration, Delta RGB analysis was conducted to detect color changes in the sensing window displayed by the PG samples at various concentrations. Based on the experiment, the fabricated paper-based ELISA could measure at least 0.25 ㎍ of PG and the measurement range was 0.25-2 ㎍. Therefore, the paper-based ELISA for detecting PG is expected to be able to determine the presence or absence of disease in crops at the infection stage in the future.

간접 Latex 응집반응과 ELISA에 의한 중추신경계 질환 환자의 혈청 및 뇌척수액에서 Toxoplasmu gondii에 대한 항체 검출 (Detection of Antibodies in Serum and Cerebrospinal Fluid to Tonoplasma gondii by Indirect Latex Agglutination Test and Enzyme-Linked Immunosorbent Assay)

  • Toxoplasma증의 혈청학적 진단에 있어서 민감도를 증가시키기 위해 간접 latex 응집반응의 결과와 비교하면서 ELISA를 개발하였으며, 뇌척수액의 검사 시료로서의 가능성을 검토하였다. 아울러 중추신경계 질환환자로부 터기생충질환을 감별하기 위하여 1986년부터 1991년까지 전국 카 병원에서 채취한 혈청과 뇌척수액에 대하여 간접 latex 응집반응(ILA)과 ELISA를 실시하여 Toxoplasma 항체 보유 양상을 비교 검토하였다. 전체 2,016 건의 혈청에 대해 ILA를 실시하여 76건(3.8%)의 양성 (1:32이상의 titer)을 얻었다. 그러나 양성 혈청환자에서 채취한 뇌척수액에서는 낮은 titer의 반응은 있었으나 양성은 나타나지 않았다. 이들 양성 혈청의 양성 혈청 및 음성 혈청에 대하여 ELISA로 항체검사를 실시한바 ILA의 titer가 1 : 32인 군에서 통계적으로 유의한 차이를 나타내는 항체값을 얻었으며, 그 흡반도는 0.40이었다. 뇌척수액에 대한 ELISA로는 ILA의 1 : 64 titer군에서 통계 적으로 유의한 차이가 나타났고 그때의 흡광도 0.27을 양성 판단의 기준으로 사용하였다. ELISA에 의한 항체 검사상 전체 혈청에서 7.0%의 양성을 검출하여 ILA보다 약 2배 정도의 높은 민감도를 보였으며, 뇌척수액에서 는 5.6%의 양성률을 보여 ELISA는 뇌척수액에서의 항체 검출시 유용한 방법이라고 판단하였다. ILA에 비하여 ELISA는 약 2배 정도 높은 양성률을 내었고 양성률은 나이에 마라 40대 이후 급격한 증가를 보였으며, 여성보 다는 남성에서 약 2배 정도 양성률이 높게 나타났다. ELISA에 의한 뇌척수액의 항체 검사에서는 양성률의 성별 차이를 나타내지 않았다. 이상의 결과로 판단할 때, ELISA가 ILA보다 Toxoplasma 항체 검출의 민감도가 높았으며, 뇌척수액은 ELISA의 좋은 검사시료가 되며, 특히 중추신경계 Tocxoplnsma증의 진단에 있어 뇌척수액에 대한 항체 검사에서 ELISA가 유용하다고 판단하였다.

ELISA에 의한 유기인계 살충제 Acephate 잔류물 분석법 개발 (ELISA Development for the residue of the organophosphorus insecticide acephate)

  • 다클론항체를 이용하여 환경 시료중 유기인계 살충제 acephate를 분석할 수 있는 경합적 간접효소면역측정법(competitive indirect ELISA)을 개발하기 위하여 분석물과 화학구조가 유사하며 hexanoic acid를 지닌 3종의 hapten을 합성하였다. 이들 hapten을 active ester 방법으로 담체단백질인 keyhole limpet hemocyanin과 접합시켜 면역원으로 사용하있고, bovine serum albumin과 접합시켜 homologous 혹은 heterologous ELISA를 위한 코팅항원으로 사용하였다. 생산된 항체들과 코팅항원을 최종선발하여 acephate 잔류분석을 위한 ELISA를 위하여 최적화하였다. 단백질의 농도가 $1{\mu}g/mL$인 코팅항원 (hapten-3-BSA)과 다클론항체 #8377를 16000배로 희석한 heterologous ELISA에서 최적화된 acephate의 $IC_{50}$ 값은 110 ng/mL이었고, 분석범위와 최소검출한계는 각각 $10{\sim}1000$과 4 ng/mL 이었다. Acephate의 주분해산물이고 살충제로 사용되는 methamidophos을 위시한 몇몇 유기인계 살충제의 항체에 대한 교차반응은 모두 0.02%이하였다. 이와 같은 결과는 본 ELISA가 농산물 및 환경시료중의 acephate 잔류분석을 위하여 편리한 방법이 될 수 있음을 시사한다.

ELISA-LC/MS/MS 병행에 의한 식품 중 aflatoxins 분석 (Determination of Total Aflatoxins in Foods by Parallelism of ELISA and LC/MS/MS)

  • High performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS) have been widely used to quantify aflatoxins in food, but these methods are expensive, time-consuming, unsuitable for analysis of the routine screening of large sample numbers and require derivatization and high level techniques to perform. The objective of this study is to detect aflatoxins in a large number of foods by a high efficient analytical system of combined enzyme linked immunosorbent assay (ELISA) for screening and LC/MS/MS for confirmation. The samples spiked individually with aflatoxin $B_1$ (0.5 and 1.0 ng/g) and total aflatoxins (10 ng/g) were analyzed by ELISA and LC/MS/ MS, and the recoveries for ELISA and LC/MS/MS were 71.8~119.2% and 70.8~135.3%, respectively. A total of 378 samples (grains, nuts, soybean and fermented soybean foods, pepper and fermented pepper foods) were purchased from the six major cities in Korea and analyzed by ELISA-LC/MS/MS system. Twenty two (5.8%; peanut: 11, pistachio: 2, walnut: 6, almond: 1, pepper powder: 1, pepper paste: 1) out of 378 samples were screened as aflatoxin B1 positive by ELISA, but, 4 (1.1%; peanut: 2, pistachio:1, pepper powder: 1) out of the 22 samples screened were confirmed as aflatoxins positive at levels of 1.02~52.79 ng/g by LC/MS/MS. ELISA-LC/MS/MS system provides a more rapid, accurate and cost-effective method for the detection of aflatoxins in large number of samples.

  • We tested biomarker systems [enzyme-linked immunosorbent assay (ELISA) and immunochromatography assay (ICG) kits] for the screening of endocrine-disrupting chemicals in contaminated environments using antibodies resulting from $17{\beta}$-Estradiol-induced vitellogenin (Vtg) in the wild scorpion fish Sebastiscus marmoratus. Monoclonal antibodies of two clones (S28 and S15) were used as capture and tracer antibodies for ELISA and ICG assays. ELISA detected Vtg at levels greater than $0.1{\mu}g/mL$, while ICG detected Vtg at levels greater than $1{\mu}g/mL$. However, the ICG system was able to detect antibodies from $17{\beta}$-Estradiol-induced Vtg serum that had been diluted 1,000 times. Our results suggest that previously developed biomarker assays can be used as detection systems to detect known endocrine-disrupting chemicals in contaminated environments, and to measure their activity.

  • A competitive enzyme-linked immunosorbent assay(ELISA) for the detection of antibodies to Treponema pallidum(T.pallidum) was developed and evaluated. T.apllidum lysate was immobilized on the surface of microplate wells and horseradish peroxidase labeled human anti-T.pallidum lysate was immobilized on the surface of microplate wells and horseradish peroxidase labeled human anti-T.pallidum was prepared and used as a tracer. The performance of the competitive ELISA was evaluated by using different specimens. The competitive ELISA showed a sensitivity of 100% in a performance panel consisting of serum and plasma with anti-T.pallidum reactivity ranging from negative to strong positive by FTA-ABS test system and 120 plasma samples positive by TPHA. The specificity of the competitive ELISA was 100% in 1,200 plasma samples collected from healthy seronegative blood donors. These results suggest that the competitive ELISA provides an excellent assay method for the detection of antibodies to T.pallidum, and may be particularly useful for serological blood screening of syphilis.

  • Progesterone의 단크론성 항체를 생산, 이용하여 감도가 높으면서도 신속히 측정할 수 있는 ELISA 기법을 처음으로 개발코져 실시하였다. 단크론성 항체는 종래의 면역방법에 의해 획득한 항혈청에 비해 약 10배의 결합율을 보였고 titer 역시 높았다. Dot-blot 분석 결과 단크론성 항체는 IgM이었다. 경합반응은 2시간으로 충분하였고, progesterone 표준용액을 이용한 표준 곡선은 0~1000pg/well에서 거의 직선적이었다. Progesterone의 단크론성 항체를 이용한 ELISA는 임상적으로는 물론 연구용으로도 신속한 항체의 기능 측정에는 물론 각종 번식 관련의 지표로 충분히 활용될 수 있을 것으로 판단된다.

효소면역측정법에 의한 우유중의 Aflatoxin M$_{1}$ 분석

  • For a survey of the occurrence of aflatoxin M$_{1}$ (AFM$_{1}$) in domestic cow's milk, we developed an enzyme-linked immunosorbent assay (ELISA) system, and quantitated the toxin in cow's milk. In order to produce specific antibodies AFM, conjugated to bovine serum albumin (AFM$_{1}$-BSA) and Freund's adjuvant were immunized subcutaneously to rabbits. By use of the antiserum showing the highest titer and AFB$_{1}$-HRP conjugate, we established a competitive direct ELISA (cdELISA) for AFM$_{1}$, whose detection limit was 0.003 ppb. The cross-reactivities of the antiserum against aflatoxin M$_{1}$ M$_{2}$, B$_{1}$, B$_{2}$, G$_{1}$, G$_{2}$, B$_{2a}$, and G$_{2a}$, were 100, 29.9, 25.0, 2.7, 13.0, 0.65, 0, and 0%, respectively. When the cdELISA was applied to the cow's milk spiked with AFM$_{1}$ and followed by cleanup with C$_{18}$ cartridge, the mean recovery of the assay was 104% (mean of CV, 6.4%) in the final concentration of 0.01-1 ppb (10-1, 000 ppt). When cow's milk samples gathered from markets and farms were assayed by the cdELISA, the mean concentration and SD of AFM$_{1}$ was 80.4 $\pm$ 55.0 ppt (n=64; range, 5.6-280 ppt).

돼지 임신 진단을 위한 Heterologous ELISA 법 개발 (Development of Heterologous ELISA System for Diagnosis of Pregnancy in Swine)

  • Early diagnoses of pregnancy for animal such as swine and bovine is extremely important to increase income of a farmhouse and for the management of farm. For the development of immunoasaay system of pregnancy in swine, we report a competitive heterologous enzyme linked immunosorbent assay (ELISA) for the direct measurement of oestrone sulfate (E1S) in diluted urine using anti-E1G (glucuronide) monoclonal antibody which cross react with ElS. The principle of assay was based on the typical solid-phase competitive ELISA methods using E1G-HRP (horseradish peroxidase) as a tracer and E1S for standard. The method had a reasonable sensitivity for the detection of E1S with 0.15 ng/ml as a detection limit. The intra-assay and inter-assay precisions were raging coefficient of from 8.50~9.67% and 8.50~9.87%, respectively, which were quite acceptable. In a field trial with a group 37 sows (18 non-pregnancy and 19 pregnancy sows) after day 29~30 post service, the concentration of E1S were determined to be below 30 ng/ml in all non-pregnancy group and over 48 ng/ml in pregnancy group except one sample. The method described here, heterologous ELISA for the measurement of E1S in urine is good enough for monitoring the early pregnancy test of swine.

Developing Polyclonal Antibody-based Indirect-ELISA to Detect Anthracnose Inocula Prior to Soybean Sprout Rot

  • We developed a polyclonal antibody based-ELISA system to monitor inocula accurately and rapidly before onset of anthracnose on soybean sprouts. Titer of mouse antisera against conidia of Colletotrichum gloeosporioides, determined by indirect ELISA, was high enough to be detectable up to ${\times}$25,600 dilutions. Both PAb1 and PAb2 had the highest level of reactivity to Colletotrichum gloeosporioides. Absorbance readings exceeded 0.15. Sensitivity of PAb to C. gloeosporioides was precise enough to detect spore concentration as low as 500 conidia/well by indirect ELISA. Both antibodies are very sensitive and highly specific to the target pathogen Colletotrichum gloeosporioides, apparently discriminating other unrelated pathogen, or epiphytes. This kit fulfills the requirements far detecting inocula before infection and onset of anthracnose. Our ELISA system should also be feasible to detect C. acutatum (Mungbean sprouts rot) and G. cingulata (C. gleosporioides), (apple, pepper). It was remarkable that absorbance value was not reduced even after 4 consecutive washings (Fig.4), suggesting that antigenic determinants are on the surface of conidia. Antigenic determinant was characterized by heating and enzyme treatment: Both PAb1 and PAb2 bind to protein epitope that does not contain residue of amino acid, arginine, and Iysine, even though more work needs to be done.
