Comparison of Growth and Water Quality in Juvenile Japanese Eel, Anguilla Japonica Fed Commercial Extruded Pellet and Paste Type Diets (시판 뱀장어용 부상사료 및 반죽사료에 대한 치어기 뱀장어(Anguilla japonica) 성장 및 사육수질 비교)

  • Growth and water quality in Japanese eel, Anguilla japonica ($6.0{\pm}0.9g$) fed extruded and paste type diets were compared for 8 weeks. Fish in the duplicate tanks were fed with one of three experimental diets: two different extruded pellet diets and one paste type diet. Six FRP tanks were used with stocking density of 3.6 kg per tank (3 m3). Restricted feeds (ca. 2% of body weight) were served twice daily. Weight gains were checked at the end of every two-week interval, and water qualities in terms of NH4-N and NO2-N were determined daily one hour after feeding in the morning. Water temperature, dissolved oxygen, and pH were constantly kept within the optimum range, while fifty percent of total water volume was changed daily. Eels fed the extruded pellet diets showed significantly better (P<0.05) growth performance than did eels fed the paste type diet. The amount of feed waste from the paste type diet was significantly greater than those from the extruded pellet diets (P<0.05). Ammonia (NH4-N) waste was significantly higher from the paste diet than from the extruded pellet diets (P<0.05). These results show that extruded pellet diets are better than the paste type diet for growth and water quality management in eel culture.

Effects of antioxidant enzymes and bioaccumulation in eels (Anguilla japonica) by acute exposure of waterborne cadmium

  • This study was conducted to evaluate the acute effects of waterborne cadmium exposure on bioaccumulation and antioxidant enzymes in eels (Anguilla japonica) and to determine the median lethal concentration (LC50). Fish were exposed to different cadmium concentrations (0, 0.15, 0.30, 0.61, 1.83, 3.08, 3.67, 4.29, and 5.51 mg L-1) for 96 h. The LC50 of A. japonica to cadmium was 3.61 mg L-1. Cadmium accumulation generally increased in tissues with increasing waterborne cadmium concentrations. At ≥ 1.83 mg L-1 exposure, all tissues accumulated significant cadmium concentrations compared with the control group, in the order of kidney > liver > gill > spleen > muscle. Measurements of variation in actual cadmium concentrations showed that a reduction of the metal in experimental water was related to cadmium accumulation in tissues. As activity alteration of antioxidant enzymes for reactive oxygen species, superoxide dismutase and catalase activities increased at ≥ 0.61 mg L-1 significantly, glutathione peroxidase and glutathione S-transferase activities were not significantly changed. The results of this study suggest that acute exposure to waterborne cadmium is potentially fatal to A. japonica due to the metal's major accumulation in various tissues and the effect of antioxidant enzyme activity.

High-Voltage Liquid-Electrolyte Microbatteries Inspired from Electric Eels (전기뱀장어의 전기발생을 모사한 고전압 액체 전해질 미소전지)

  • We present high-voltage liquid-electrolyte microbatteries, inspired from the high-voltage generation mechanism of electric eels using serially connected multiple-cell arrays. In the microbatteries, we purge air into the electrolyte filled in a channel layer to isolate serially connected multiple cell arrays using three surface-tension valves (cell-front, outlet, and cell-end valves). Compared to the previous multi-cell stack or interconnection, present microbatteries provide a reduced multi-cell charging time. We have designed and characterized four different prototypes C1, C10, C20, and C40 having 1, 10, 20, and 40 cells, respectively. In the experimental study, the threshold pressures of cell-front, outlet, and cell-end valves were measured as $460{\pm}47$, $1,000{\pm}53$, and $2,800{\pm}170$ Pa, respectively. The average charging time for C40 was measured as $26.8{\pm}4.9$ seconds where the electrolyte and air flow-rates are 100 and $10{\mu}l/min$, respectively. Microbatteries showed the maximum voltage of 12 V (C40), the maximum power density of $110{\mu}W/cm^2$ (C40), and the maximum power capacity of $2.1{\mu}Ah/cm^2$ (C40). We also proposed a tapered-channel to remove the reaction gas from the cell chamber using a surface tension effect. The present microbatteries are applicable to high-voltage portable power devices.

The effect of the entrance size on the catch of trap for conger eel (스프링식 붕장어그물통발의 어획성능에 미치는 입구크기의 영향)

  • Recently, Fishermen have required to expand the circumference of trap entrance for conger eel because it was difficult to take the catch out as well as to put the baits into. A series of fishing experiments was carried out in coastal areas of Tongyoung, Korea in 2008 and 2009 using the traps to describe the effect of the entrance size on the catch. The experimental traps (five types) were used in filed experiments with four types circumference size traps (140, 180, 220 and 260mm) with mesh size 22mm and another type was used the same one usually using in filed with mesh size 35mm (750mm circumference). The experiment results were, the conger eels of total length 35mm more or so were caught 85, 93, 142 and 176 individual by the experiment traps with mesh size 22mm as increase circumference size from 140mm to 260mm, respectively. And the small conger eels of the total length below 35mm to be prohibited to catch by law were caught 145, 160, 288 and 304 individual according to increase the circumference size of trap, respectively. In addition, in case of the trap with mesh size 22mm, bycatches were 230 - 260 individuals and much more than bycatches of the trap with mesh size 35mm. In conclusion, when we expand the circumference size of trap according to fishermen's requirement, we should review not only economic of fishery but also increasement the bycatch of non-target fishes and small size fishes.

Variation in the Nanostructural Features of the nc-Si:H Thin Films with Substrate Temperature (수소화된 나노결정 실리콘 박막의 기판온도에 따른 나노구조 변화)

  • We investigated the nanostructural, chemical and optical properties of nc-Si:H films according to deposition conditions. Plasma enhanced chemical vapor deposition(PECVD) techniques were used to produce nc-Si:H thin films. The hydrogen dilution ratio in the precursors, [$SiH_4/H_2$], was fixed at 0.03; the substrate temperature was varied from room temperature to $600^{\circ}C$. By raising the substrates temperature up to $400^{\circ}C$, the nanocrystalite size was increased from ~2 to ~7 nm and the Si crystal volume fraction was varied from ~9 to ~45% to reach their maximum values. In high-resolution transmission electron microscopy(HRTEM) images, Si nanocrystallites were observed and the crystallite size appeared to correspond to the crystal size values obtained by X-ray diffraction(XRD) and Raman Spectroscopy. The intensity of high-resolution electron energy loss spectroscopy(EELS) peaks at ~99.9 eV(Si $L_{2,3}$ edge) was sensitively varied depending on the formation of Si nanocrystallites in the films. With increasing substrate temperatures, from room temperature to $600^{\circ}C$, the optical band gap of the nc-Si:H films was decreased from 2.4 to 1.9 eV, and the relative fraction of Si-H bonds in the films was increased from 19.9 to 32.9%. The variation in the nanostructural as well as chemical features of the films with substrate temperature appears to be well related to the results of the differential scanning calorimeter measurements, in which heat-absorption started at a substrate temperature of $180^{\circ}C$ and the maximum peak was observed at ${\sim}370^{\circ}C$.

  • The immunomodulatory effects of levamisole (LMS) were evaluated in leucocytes of eels in vitro. Proliferation of lymhocytes treated with T-cell mitogen (Con A or PHA) was markedly inhibited by LMS in a dose dependent manner. B cell mitogen (LPS), in contrast, slightly increased the proliferaion. On the other hand, production of MIF and MAF when treated with Con A was increased in a dose-dependent way. NK cell activities were somewhat increased when LMS was pretreated and this augmentation was due to an increase in binding capacity of effector-target cell, but not due to the target cell lytic activity of effector cells. Phagocytic activity, superoxide anion formation, hydrogen peroxide formation and lysozyme activity of leucocytes were enhanced by LMS in a dose related-manner. These results suggest that LMS might modulate the immmune responses by activation of cytokine production and by augmentation of leukocyte activity but not by increment of immunocompetent cell numbers.

  • The infection status of domestic farmed eels Anguilla japonica, Anguilla bicolor and Anguilla marmorata with Japanese eel endothelial cell-infecting virus (JEECV) and anguillid herpesvirus 1 (AnHV) was examined at the major eel farming areas in Korea. These viruses were detected in all areas examined, regardless of the eel species or age. Any farm with a history of viral infection in adult fish confirmed the infection to be transmitted to stocked fry within 3 to 5 months. It is proposed that both viruses are horizontally transmitted within a given farm. The primary symptoms and histopathological lesions produced by the two viral infections are similar, making it difficult to distinguish the two diseases through clinical symptoms. Both viruses displayed 100% detection in the gills, suggesting that the gills are an optimal tissue for JEECV and AnHV monitoring. This study concluded that JEECV and AnHV were prevalent on eel farms across the country and caused very high mortality when the two viruses co-infected fry. Additional studies, including experimental infections, are needed to clearly understand the pathogenicity of each virus and the risk of co-infection.

Evaluation of Fermented Blood Meal as a Functional Additive in the Diet of Japanese Eel Anguilla japonica (뱀장어(Anguilla japonica)용 사료 기능성 첨가제로써의 발효 혈분 이용성 평가)

  • The effects of dietary supplementation with fermented blood meal (FBM) in two forms were evaluated on the growth performance, hematological parameters, innate immune response, antioxidant capacity, digestive enzyme activity and intestinal histology of Japanese eel Anguilla japonica. A commercial powder feed was used as a basal diet (Con). Six other diets were prepared by supplementing 2, 4 and 6% FBM either in powder or liquid form to the Con diet (designated as P2, P4, P6, L2, L4 and L6, respectively). In total 420 eels (initial mean body weight: 55.1±1.71 g) were distributed into 21 tanks (450L) at a density of 20 eels per tank. Three replicate groups of fish were fed one of the seven diets twice daily for 12 weeks. No significant differences were observed in the growth performance and survival among all groups. Fish fed FBM-containing diets showed significantly improved hematological parameters, innate immunity and antioxidant capacity than those in the Con group. Fish fed FBM-containing diets showed significantly enhanced digestive enzyme activity, villus length and goblet cells than those in fish fed Con diet. These results indicate that both powder and liquid FBM could be a good functional feed additive to improve innate immunity and digestion in Japanese eel.

Vitamin K variations in conger eel (Conger myriaster) influenced by harvest time and size

  • This study analyzed the variation in vitamin K content in conger eel (Conger myriaster) caught off Tongyeong, Gyeongsangnam-do, Korea, focusing on the influence of size (large and small) and harvest period (monthly throughout 2021). We applied enzymatic extraction and HPLC-fluorescence methods for the analysis of vitamin K1 (phylloquinone) and K2 (menaquinone). The vitamin K content in conger eel varied significantly with size and harvest season (p<0.05). In large-sized samples, the phylloquinone content peaked in July (0.80±0.09 ㎍/100 g), while the highest menaquinone content was in May (0.79±0.11 ㎍/100 g). Conversely, in small-sized conger eels, the highest phylloquinone was found in December (1.94±0.15 ㎍/100 g), and the peak menaquinone level was in January (0.66±0.02 ㎍/100 g). The fat content was highest in July for large samples and in January for smaller ones. There was a positive correlation between fat and total vitamin K contents in conger eel (r=0.631, 0.667). Method validation and quality control measures ensured data reliability for vitamin K1 and K2 analyses. This study provides reliable information on the size and seasonal variations of vitamin K in conger eels, a staple in the Korean diet. This information is valuable for inclusion in Korea's national food nutrition database and for formulating future national health and nutrition policies.

On the Histopathological Changes and Methemoglobinemia to Nitrite Toxicity in the Culture Farms of Eel, Anguilla japonica (참장어(뱀장어) Anguilla japonica 양어장(養魚場)에서의 아질산(亞窒酸) 중독(中毒)에 의한 Methemoglobin증(症)과 병리조직학적(病理組織學的) 변성(變性))

  • Histopathological changes and methemoglobinemia of the eels (Anguilla japonica) reared at six culture farms in the vicinity of Y$\check{o}$su city were investigated under the conditions of $22.0{\sim}29^{\circ}C$, 4.33~7.33ppm of D.O., pH 6.1~8.2 and 0.03~10.1ppm of nitrite from December, 1989 to June, 1990. All of the eels showed no any abnormal state in action, color and blood, but the eels reared in pH 6.0 had planty of mucus and stickiness on their body surface. Methemoglobin levels were 1.03~9.86% of total erythrocyes in Dec. and 5.05~25.69% in June, and poikilocyte levels were 5.8~30.17% in Dec. and 8.67~74.45% in June. These results indicate the fact that the water management of the culture farms was worse in summer than in winter, even though not good in winter. Hypertrophy was found in the gill, hepatic and kidney tissue of the eels in all the culture farms. In March and June, Fish of all culture farms appeared the detachment of gill lamella epithelium cells, pyknosis and sinusoids of nucleus in the hepatic tissue and hemosiderosis in the kidney tissue. The trend of increase in methemoglobin levels appeared in the fish of culture farms with high nitrite concentration, which didn't appear equally in all the culture farms. However, in the culture farms of pH 6.0, methemoglobin and poikilocyte levels were increased independently of concentration of nitrite, and degeneration in the tissue of the gill, liver and kidney was also appeared.
