Ex vivo Boosted Immune Cell Therapy for Canine Hepatic Disease

  • A 12-year-old male American Cocker Spaniel was diagnosed with a type of chronic hepatits (CH) called cholangioheaptits. Routine supportive medication was administered to the patient, and ex vivo boosted immune cell (EBI-C) therapy was used for the treatment. A histopathologic examination of the liver 19 months later revealed that the cholangiohepatitis had progressed to cholangiocarcinoma. The medication and immune cell therapy was maintained. Two months after the new diagnosis, the patient's state worsened, and the dog died 635 days after the first visit. EBI-C therapy is a type of immunotherapy, where immune cells are isolated from the patient's peripheral blood mononuclear cells, expanded ex vivo, and then infused into the patient intravenously every two weeks. EBI-Cs (mean: 2.78 × 108 cells) were obtained 38 times and infused every two weeks. Most EBI-C were T-lymphocytes (99.24% of total EBI cells). T-lymphocytes produce large interferon (IFN)-γ, and IFN-γ inhibits liver fibrosis in dogs with CH. Moreover, in bile duct cancer, an increase in T-lymphocytes correlates with decreasing tumor invasion and metastasis. Thus, we propose that EBI-C therapy is applicable as a new supportive therapy for canine liver disease if other treatments like drug medication, surgery, or radiation are unavailable.

Influence of oxidative atmosphere of the electron beam irradiation on cyclization of PAN-based fibers

  • In order to study the impact of atmosphere during electron beam irradiation (EBI) of polyacrylonitrile (PAN) precursor fibers, the latter were stabilized by EBI in both air and oxygen atmospheres. Gel-fraction determination indicated that EBI-stabilization under an oxygen atmosphere leads to an enhanced cyclization in the PAN fibers. In the Fourier-transform infrared spectroscopy analysis, the PAN fibers stabilized by EBI under an oxygen atmosphere exhibited a greater decrease in the peak intensity at 2244 cm−1 (C≡N vibration) and a greater increase in the peak intensity at 1628 cm−1 (C=N absorption) than the corresponding PAN fibers stabilized under an air atmosphere. From the X-ray diffraction analysis it was found that oxygen uptake in PAN fibers leads to an increase in the amorphous region, produced by cyclization.

  • Polymerizations of higher $\alpha$-olefins were carried out in toluene by using highly isospecific catalyst, rac-(EBI)M(NMe$_2$)$_2$ (EBI=1,2-ethylenebis-(1-indenyl); M=Zr(rac-1); M=Hf(rac-2)) In the presence of Al(i-Bu)$_3$/[Ph$_3$C][B($C_{6}F_{5}$)$_4$]. The polymerization of high $\alpha$-olefin showed high activity and similar polymerization behavior. The polymerization activity was affected by both monomer size and lateral size of polymer chain. The conversion of monomer to polymer decreases with the increased lateral size in the order of 1-pentene>1-hexene>1-octene>1-decene. The same dependences of melting behavior and intrinsic viscosity of polyolefin on lateral size were observed according to the results obtained by differential scanning calorimetry and intrinsic viscosity. All poly($\alpha$-olefin)s showed very high isotacticity (triad) and the isotacticity increases in the order of poly(1-pentene)$^1H$ NMR and Raman spectra analysis showed that chain transfer to cocatalyst, which generates saturated methyl groups, Is a main chain termination. The $\beta$-hydride eliminations, which generate unsaturated vinylidene, tri-substituted, and vinylene end group. are found to be minor chain terminations.

Copolymerizations of Ethylene with 1-Hexene over ansa-Metallocene Diamide Complexes

  • We have performed copolymerizations of ethylene with 1-hexene using various ansa-metallocene compounds in the presence of the non-coordinative [CPh$_3$][B(C$\_$6/F$\_$5/)$_4$ion pair as a cocatalyst. The metallocenes chosen for this study are isospecific metallocene diamide compounds, rac-(EBI)Zr(NMe$_2$)$_2$ [1, EBI = ethylene-l ,2-bis(1-indenyl)], rac-(EBI)Hf(NMe$_2$)$_2$ (2), rac-(EBI)Zr(NC$_4$H$\_$8/)$_2$ (3), and rac-(CH$_3$)$_3$Si(1-C$\_$5/H$_2$-2-CH$_3$-4-$\^$t/C$_4$H$\_$9/)2 Zr(NMe$_2$)$_2$ (4), and syndiospecific metallocene dimethyl compounds, ethylidene(cyclopentadienyl)(9-fluorenyl) ZrMe$_2$ [5, Et(Flu)(Cp )ZrMe$_2$] and isopropylidence (cyclopentadienyl)(9-fluorenyl)ZrMe$_2$ [6, iPr(Flu)(Cp)ZrMe$_2$]. The copolymerization rate decreased in the order 4 >1-3>2 >5>6. The reactivity of I -hexene decreased in the order 2 >6>1- 3-5> 4. We characterized the microstructure of the resulting poly(ethylene-co-l-hexene) by $\^$l3/C NMR spectroscopy and investigated various other properties of the copolymers in detail.

Selection of Fungicides for the Control of Soybean Black Root Rot Caused by Calonectria ilicicola (콩 검은뿌리썩음병 방제를 위한 살균제 선발)

  • Fungicidal screening was performed to control soybean black root rot caused by Calonectria ilicicola through in vitro and greenhouse assays. In in vitro assay, 25 fungicides were assessed by an agar dilution method and a 96-well microtiter plate method. While protective fungicides including dithianon, dichlofluanid, mancozeb, and captan showed a very low activity against the mycelial growth C. ilicicola SC03-15 in the agar dilution method, they displayed potent inhibitory activity against spore germination in a 96-well microtiter plate method with $EC_{50}$ values of 4.65, 0.61, 4.64, and $0.29{\mu}g\;mL^{-1}$, respectively. Ergosterol biosynthesis-inhibiting (EBI) fungicides showed different antifungal activity against mycelial growth and spore germination according to molecules. Difenconazole displayed higher antifungal activity against spore germination rather than mycelial growth, and prochloraz inhibited potently both mycelial growth and spore germination with EC50 values less than $1.8{\mu}g\;ml^{-1}$. In contrast, the other EBI fungicides inhibited more highly mycelial growth than spore germination. Carbendazim+diethofencarb and dazomet also inhibited both mycelial growth and spore germination of C. ilicicola SC03-15 at very low concentrations. In greenhouse assay, carbendazim+diethofencarb effectively controlled a soybean black root rot by drenching 2 days before or after inoculation. In addition, tebuconazole showed potent curative activity against soybean black root rot.

Nonlinear response of r.c. framed buildings retrofitted by different base-isolation systems under horizontal and vertical components of near-fault earthquakes

  • Near-fault ground motions are characterized by high values of the ratio between the peak of vertical and horizontal ground accelerations, which can significantly affect the nonlinear response of a base-isolated structure. To check the effectiveness of different base-isolation systems for retrofitting a r.c. framed structure located in a near-fault area, a numerical investigation is carried out analyzing the nonlinear dynamic response of the fixed-base and isolated structures. For this purpose, a six-storey r.c. framed building is supposed to be retrofitted by insertion of an isolation system at the base for attaining performance levels imposed by current Italian code in a high-risk seismic zone. In particular, elastomeric (e.g., high-damping-laminated-rubber bearings, HDLRBs) and friction (e.g., steel-PTFE sliding bearings, SBs, or friction pendulum bearings, FPBs) isolators are considered, with reference to three cases of base isolation: HDLRBs acting alone (i.e., EBI structures); in-parallel combination of HDLRBs and SBs (i.e., EFBI structures); FPBs acting alone (i.e., FPBI structures). Different values of the stiffness ratio, defined as the ratio between the vertical and horizontal stiffnesses of the HDLRBs, sliding ratio, defined as the global sliding force divided by the maximum sliding force of the SBs, and in-plan distribution of friction coefficient for the FPs are investigated. The EBI, EFBI and FPBI base-isolation systems are designed assuming the same values of the fundamental vibration period and equivalent viscous damping ratio. The nonlinear dynamic analysis is carried out with reference to near-fault earthquakes, selected and scaled on the design hypotheses adopted for the test structures.

Response to Fungicides of Colletotrichum Species Isolated from Infected Tissues of Several Crops (다양한 작물로부터 분리한 탄저병균(Colletotrichum spp.)의 살균제에 대한 반응)

  • This study has been conducted to investigate the responses of various isolates of four Colletotrichum species such as C. gloeosporioides, C acutatum, C. coccodes, and C. dematium isolated from infected tissues of several crops to fungicides such as carbendazim, carbendazim+diethofencarb, four protective fungicides, and three ergosterol biosynthesis-inhibiting (EBI) fungicides. All the isolates of C. acutatum showed $EC_{50}$ values in a range of 0.001-3.040 ${\mu}g/ml$ against carbandazim, a benzimidazole fungicide. As for the response to carbendazim, the isolates of C. gloeosporioides obtained from pepper, apple, and strawberry were clearly divided into two groups, resistant or sensitive isolates. All the resistant isolates showed $EC_{50}$ values above 1000 ${\mu}g/ml$, whereas the sensitive isolates had lower $EC_{50}$ values than 0.550 ${\mu}g/ml$. The isolates of C. gloeosporioides exhibited a negative cross resistance between carbendazim and diethofencarb (a N-phenylcarbamate fungicide), but isolates of C. acutatum did not. Toward carbendazim, C. coccodes and C. dematium isolates showed a similar response to C. acutatum isolates and the sensitive isolates of C. gioeosporioides, respectively. As for response of Protective fungicides, all the isolates of C. acutatum showed a more resistant reaction than all the isolates of C. gloeosporioides. However, there was no difference among 4 species of Colletotrichum against EBI fungicides.

A Facile Pretreatment Method for Rice Straw using Electron Beam Irradiation and 4-methylmorpholine-N-oxide Solution (전자선 조사와 4-메틸모포린-N-옥사이드 용액을 이용한 볏짚의 전처리 방법)

  • In this study, a facile two-step pretreatment method was investigated for producing fermentable sugars. Rice straw was pretreated using electron beam irradiation (EBI) and 4-methylmorpholine-N-oxide (NMMO) prior to enzymatic hydrolysis. In the first stage, the EBI on the rice straw was carried out at various doses (100, 300, 500 kGy) and then, irradiated rice straw was stirred with NMMO solution at 120°C for 1 h for the second stage. The pretreated rice straw was hydrolyzed by cellulase 1.5 L (70 FPU/ml) and Novozyme-188 (40 CbU/ml) at 50°C for 24, 48, and 72 h. A sugar yield of 83.8% was obtained from the pretreated rice straw after 72 h of enzymatic hydrolysis. Also, FTIR and XRD results indicate that the pretreatment of the rice straw was effective due to the synergic effects of the two-step pretreatment. In conclusion, rice straw might be a potential substrate for bioethanol production by yeast fermentation.

Environmentally-Friendly Pretreatment of Rice Straw by an Electron Beam Irradiation (전자선 조사를 이용한 볏짚의 친환경 전처리 공정)

  • The autoclaving assisted by an irradiation pretreatment method was developed without toxic chemicals to produce fermentable sugars for their conversion to bioethanol. In the first step, electron beam irradiation (EBI) of rice straw was performed at various doses. The electron beam-irradiated rice straw was then autoclaved with DI water at $120^{\circ}C$ for 1 h. A total sugar yield of 81% was obtained from 300 kGy electron beam-irradiated rice straw after 72 h of enzymatic hydrolysis by Cellulase 1.5L (70 FPU/mL) and Novozyme-188 (40 CbU/mL). Also, the removal of hemicellulose and lignin was 32.0% and 32.5%, respectively. This result indicates that the environmentally-friendly pretreatment method of rice straw by an electron beam irradiation could be applied for bioethanol production in plant.

Pretreatment of Kenaf Core by Combined Electron Beam Irradiation and Water Steam for Enhanced Hydrolysis (향상된 가수분해율을 얻기 위한 전자선 조사와 물찜의 복합 전처리공정을 이용한 케냐프 코어 전처리)

  • We have investigated the combined pretreatment of electron beam irradiation (EBI) and water steam as a kenaf core pretreatment process. After each sample was exposed to electron beam dose ranging from 50 to 1,000 kGy, the irradiated sample was treated by water steam using an autoclave for 5-h at $120^{\circ}C$. The pretreated samples were characterized using FTIR-ATR and XRD. FTIR spectra and XRD analysis of nonpretreated and pretreated samples confirm that crystallinity changes were observed before and after the pretreatment. The crystallinity index (CrI) was increased from 50.6% for nonpretreated sample 55.0% for 500 kGy exposed sample. And then, we analyzed sugar yield that is the amount of produced mono-saccharides in pretreated sample by enzymatic hydrolysis; an enzyme activity rate was 70 FPU/mL and 40 CBU/mL, and the loading time was 24, 48 and 72-h. The highest sugar yield was 83.9% at 500 kGy after 72-h for enzymatic hydrolysis. The sugar yield of enzymatic hydrolysis for pretreatment samples was increased as doses are subsequently changed to 100, 200 and 300 kGy, allowing to give 50.8%, 58.6% and 67.9%, respectively.
