Ex vivo Boosted Immune Cell Therapy for Canine Hepatic Disease

  • A 12-year-old male American Cocker Spaniel was diagnosed with a type of chronic hepatits (CH) called cholangioheaptits. Routine supportive medication was administered to the patient, and ex vivo boosted immune cell (EBI-C) therapy was used for the treatment. A histopathologic examination of the liver 19 months later revealed that the cholangiohepatitis had progressed to cholangiocarcinoma. The medication and immune cell therapy was maintained. Two months after the new diagnosis, the patient's state worsened, and the dog died 635 days after the first visit. EBI-C therapy is a type of immunotherapy, where immune cells are isolated from the patient's peripheral blood mononuclear cells, expanded ex vivo, and then infused into the patient intravenously every two weeks. EBI-Cs (mean: 2.78 × 108 cells) were obtained 38 times and infused every two weeks. Most EBI-C were T-lymphocytes (99.24% of total EBI cells). T-lymphocytes produce large interferon (IFN)-γ, and IFN-γ inhibits liver fibrosis in dogs with CH. Moreover, in bile duct cancer, an increase in T-lymphocytes correlates with decreasing tumor invasion and metastasis. Thus, we propose that EBI-C therapy is applicable as a new supportive therapy for canine liver disease if other treatments like drug medication, surgery, or radiation are unavailable.

  • Pulps were separated from wood chips using an Electron beam irradiation (EBI) treatment without a NaOH-AQ (anthraquinone) treatment for cooking. The methods were based on a hot water treatment after EBI and two-step bleaching processes. Chemical compositions and FT-IR spectroscopy demonstrated that the content of lignin and hemicellulose in the bleached wood pulps treated with various EBI dose decreased with an increase of EBI doses. Specifically, the lignin in the bleached with pulps treated at 600 kGy of EBI dose was almost completely removed. Moreover, TGA analysis showed that a thermal stability increased with increasing the content of cellulose but the lignin decomposed slowly over the wide region.

Influence of oxidative atmosphere of the electron beam irradiation on cyclization of PAN-based fibers

  • In order to study the impact of atmosphere during electron beam irradiation (EBI) of polyacrylonitrile (PAN) precursor fibers, the latter were stabilized by EBI in both air and oxygen atmospheres. Gel-fraction determination indicated that EBI-stabilization under an oxygen atmosphere leads to an enhanced cyclization in the PAN fibers. In the Fourier-transform infrared spectroscopy analysis, the PAN fibers stabilized by EBI under an oxygen atmosphere exhibited a greater decrease in the peak intensity at 2244 cm−1 (C≡N vibration) and a greater increase in the peak intensity at 1628 cm−1 (C=N absorption) than the corresponding PAN fibers stabilized under an air atmosphere. From the X-ray diffraction analysis it was found that oxygen uptake in PAN fibers leads to an increase in the amorphous region, produced by cyclization.

국내 유통 농산물 중 EBI계 농약 모니터링과 위해도 평가 (Monitoring of Ergosterol Biosynthesis Inhibitor (EBI) Pesticide Residues in Commercial Agricultural Products and Risk Assessment)

  • 우리 국민의 주식이 되는 쌀(백미)과 콩류, 감귤류, 열대과 일류, 엽채류, 엽경채류, 근채류, 과채류, 버섯류에 속하는 농산물(989건)에 대해 8종의 EBI계 농약의 잔류 여부를 분석하였다. 그 결과 8건의 검체에서 농약이 검출되었고 농약 수는 5종이 검출되었다. 기준 미설정 농약은 2종이었으며, 곡류 134건 중 4건 검출로 3.0%, 채소류 471건 중 4건 검출로 0.8%의 검출율을 나타내었다. 그 외 콩류(48건), 견과종실류(72건), 과실류(197건), 버섯류(67건)에서는 전혀 검출되지 않았으며 부적합율은 0.1%로 나타났다. 국내 미등록농약인 paclobutrazole이 근대에서 잠정허용기준을 초과하는 것으로 확인되었다. 그리고 EBI계의 검출율은 0.8%로 매우 낮았으며 검출된 농약의 경우도 기준규격 이하로 안전한 수준이었다.

IL-35 Over-expression is Associated with Genesis of Gastric Cancer

  • Overexpression of interleukin (IL)-35 has been found in a variety of malignancies, but the expression status in gastric cancer has yet to be elucidated clearly. In the present study, positive expression of EBI3 and p35 was 63.3% and 70.0% of cases, respectively. EBI3 expression was strongly related with larger tumor size and invasion depth (P<0.05). Similarly, expression of p35 was also correlated with larger tumor size (P<0.05). These results indicate that IL-35 might be involved in growth of gastric cancer. Interestingly, EBI3 and p35 expressions were positive correlated with Ki-67 expression. Moreover, EBI3 immunoreactivity was associated with Bcl-2 staining. Our data suggest IL-35 is correlated with genesis of gastric cancer by regulating growth and apoptosis.

보호용 살균제와 ergosterol 생합성 저해 살균제에 대한 고추 탄저병균의 약제 반응 (Response of Colletotrichum sup. Causing Red Pepper Anthracnose to Protective and Ergosterol Biosynthesis-inhibiting Fungicides)

  • 전국의 고추 재배지에서 1999년과 2002년에 채집한 병든 고추로부터 각각 130개와 258개의 고추 탄저병균을 분리한 후, 4종의 보호용 살균제와 3종의 ergosterol생합성 저해 살균제에 대한 균사 생장 저해 정도를 측정함으로서 살균제들에 대한 저항성 모니터링을 실시하였다. 1999년에 분리한 균주는 세 가지 보호용 살균제, 즉 dithianon, chlorothalonil 및 propineb에 대해서는 2002년의 균주보다 더 저항성을 보였다. 이에 반하여 다른 보호용 살균제인 iminoctadine에 대해서는 오히려 2002년의 균주가 더 저항성인 것으로 나타났다. 세 개의 ergosterol 생합성 저해(EBI) 살균제, 즉 tebuconazole, hexaconazole 및 prochloraz에 대해서는 2002년의 균주가 1999년의 균주보다 더 저항성이 컸으며, 2002년도의 균주들의 $EC_{50}$ 값은 1999년 균주들보다 1.2배 내지 4.4배가 높았다 위의 살균제들에 대한 탄저병균의 반응을 보면, 채집한 지역 간에 저항성을 차이가 크게 나타나고 있었다. 단기간내에 충북에서 분리한 균주들은 채집한 시기가 늦어질수록, 고추의 생육 초기에 주로 많이 사용되는 보호용 살균제들에 대한 $EC_{50}$값은 감소하였다. 그러나 ergosterol 생합성 저해 살균제에 대한 균주들의 반응은 정해진 생육 기간 중에는 변화가 거의 없었다.

  • 이소특이성 촉매인 rac-(EBI)M($NMe_2$)$_2$ [EBI=1,2-ethylenebis-(1-indenyl); M=Zr (rac-1), M=Hf(rac-2)와 공촉매계로서 Al(iBu)$_3$/[Ph$_3$C][B($C_{6}F_{5}$)$_4$]를 이용하여 고차 $\alpha$-올레핀의 중합특성을 조사하였다. 고차 $\alpha$-올레핀의 중합은 높은 활성을 나타냈으며, rac-1과 rac-2의 두 촉매계는 유사한 중합거동을 나타냈다. 중합 활성은 단량체의 크기와 중합된 고분자 가지의 길이에 의해 영향을 받았다. 단량체의 전환율은 1-pentene>1-hexene>1-octene>1-decene의 순서로 감소하였다. 또한 합성된 폴리($\alpha$-올레핀)의 고유점도값과 시차주사열분석기에 의한 용융거동도 유사한 경향을 나타냈다. 폴리 ($\alpha$-올레핀)은 매우 높은 이소탁틱도 트리에드를 나타냈으며 poly(1-pentene)$^1H$ NMR과 Raman 스펙트럼을 이용한 고분자 사슬의 말단기 분석 결과로부터, 주로 일어나는 사슬정지반응은 말단에 포화된 메틸기를 생성하는 공촉매로의 전이이며, 소수 반응으로 비닐리덴, 삼차치환 및 비닐렌 이중결합을 생성하는 $\beta$-수소제거 반응이 일어남을 알 수 있었다.

Copolymerizations of Ethylene with 1-Hexene over ansa-Metallocene Diamide Complexes

  • We have performed copolymerizations of ethylene with 1-hexene using various ansa-metallocene compounds in the presence of the non-coordinative [CPh$_3$][B(C$\_$6/F$\_$5/)$_4$ion pair as a cocatalyst. The metallocenes chosen for this study are isospecific metallocene diamide compounds, rac-(EBI)Zr(NMe$_2$)$_2$ [1, EBI = ethylene-l ,2-bis(1-indenyl)], rac-(EBI)Hf(NMe$_2$)$_2$ (2), rac-(EBI)Zr(NC$_4$H$\_$8/)$_2$ (3), and rac-(CH$_3$)$_3$Si(1-C$\_$5/H$_2$-2-CH$_3$-4-$\^$t/C$_4$H$\_$9/)2 Zr(NMe$_2$)$_2$ (4), and syndiospecific metallocene dimethyl compounds, ethylidene(cyclopentadienyl)(9-fluorenyl) ZrMe$_2$ [5, Et(Flu)(Cp )ZrMe$_2$] and isopropylidence (cyclopentadienyl)(9-fluorenyl)ZrMe$_2$ [6, iPr(Flu)(Cp)ZrMe$_2$]. The copolymerization rate decreased in the order 4 >1-3>2 >5>6. The reactivity of I -hexene decreased in the order 2 >6>1- 3-5> 4. We characterized the microstructure of the resulting poly(ethylene-co-l-hexene) by $\^$l3/C NMR spectroscopy and investigated various other properties of the copolymers in detail.

Capability for Change at Community Health Centers Serving Asian Pacific Islanders: An Exploratory Study of a Cancer Screening Evidence-based Intervention

  • Background: Understanding and enhancing change capabilities, including Practice Adaptive Reserve (PAR), of Community Health Centers (CHCs) may mitigate cancer-related health disparities. Materials and Methods: Using stratified random sampling, we recruited 232 staff from seven CHCs serving Asian Pacific Islander communities to complete a self-administered survey. We performed multilevel regression analyses to examine PAR composite scores by CHC, position type, and number of years worked at their clinic. Results: The mean PAR score was 0.7 (s.d. 0.14). Higher scores were associated with a greater perceived likelihood that clinic staff would participate in an evidence-based intervention (EBI). Constructs such as communication, clinic flow, sensemaking, change valence, and resource availability were positively associated with EBI implementation or trended toward significance. Conclusions: PAR scores are positively associated with perceived likelihood of clinic staff participation in cancer screening EBI. Future research is needed to determine PAR levels most conducive to implementing change and to developing interventions that enhance Adaptive Reserve.

Facile preparation of self-assembled wool-based graphene hydrogels by electron beam irradiation

  • Three dimensional self-assembled graphene hydrogels were easily fabricated by electron beam irradiation (EBI) using an aqueous solution of wool/poly(vinyl alcohol) and graphene oxide (GO). After exposure to various levels of EBI radiation, the highly porous, self-assembled, wool-based graphene hydrogels were characterized using scanning electron microscopy and Fourier-transform infrared spectroscopy; to determine the gel fraction, degree of swelling, gel strength, kinetics-of-swelling analyses and removal of hexavalent chromium (Cr(VI)) from the aqueous solution. X-ray diffraction results confirmed that EBI played a significantly important role in reducing GO to graphene. The adsorption equilibrium of Cr(VI) was reached within 80 min and the adsorption capacity was dramatically increased as the acidity of the initial solution was decreased from pH 5 to 2. Changes in ionic strength did not exert much effect on the adsorption behavior.
