The present study was designed to demonstrate that ES cell lines efficiently could be isolated from explanted blastocysts of hybrid BCF1 mouse when grown on STO feeder layer derived from mouse fibroblasts in culture medium supplemented with leukemia inhibitory factor (LIF). The expanded blastocysts were attached to mitomycin C-inactivated STO feeder layer and were cultured for 4 days. Four days later the ICM was disaggregated by a short term trypsin treatment (0.25% trypsin / 0.04% EDT A for 2-3 min). The resulting cell suspension was seeded on a new STO feeder layer and covered with DMEM supplemented with 10% FCS, 0.1 mM nonessential amino acid, 0.1 mM sodium pyruvate, 0.1 mM mercaptoethanol and 1,000 U/ml LIF. Colonies of ES-like cells were observed after the second passage. These colonies were repeatedly passaged at approximately 5 day intervals. In this study, five ES-like celllines were isolated by directly explanting blastocysts, but three lines were lost after the 5th passage, possibly due to toxic effects of a new FCS batch. The characterization of developmental potential of isolated cell lines was performed with respect to in vitro differentiation and specific activity of alkaline phosphatase (AP). When cells were cultured in suspension, the aggregates of cell lines were capable of forming simple embryoid bodies (EB), and showed the capacity for forming cystic multilayer EBs. In addition, the cell lines were positive for AP staining, a biochemical marker characteristic of mouse ES cells.
Infection of Epstein-Barr virus(EBV) gives rise to a broad spectrum of clinical manifestations in children. Although renal involvement is rare, diverse renal manifestations are known from hematuria to acute renal failure. Secondary membranous nephropathy(MN) associated with systemic EBV infection is an uncommon renal pathology and only two cases have been reported. We are adding another case of MN associated with EBV infection in a child. An 8-year-old girl was admitted for renal biopsy. She had been followed up for microscopic hematuria and intermittent proteinuria for 5 months. There had been no specific findings in serology and radiology. Tonsil biopsy had been done due to exudative tonsillar hypertrophy and enlarged multiple cervical lymph nodes. And it showed EBV-associated lymphoproliferative findings. Serologic tests for EBV showed positive evidence of recent infection; viral capsid antigen(VCA) IgM was borderline positive, VCA IgG and early antigen IgG were positive, and EB nuclear antigen IgG was negative. In Situ Hybridization of tonsil for EBV mRNA was positive. Because her proteinuria and hematuria were aggravated at that time(protein 3 +, RBC >60/HPF), renal biopsy was done. Renal biopsy showed the findings of MN, characterized by thickened capillary walls with epimembranous spikes on light microscopy and subepithelial, mesangial and subendothelial electron dense deposits on electron microscopy. On immunofluorescence microscopy, IgG, C1q, kappa and lambda chains were positive. After steroid administration, proteinuria and hematuria resolved gradually within 6 months.

This research has a meaning to provide basic data for eco-friendly way of district unit plans and ecological landscape planning by evaluation of biotope preservation value at the level of district unit and designating land development of the site, the whole area of Sinseo-dong (Dong-gu, Daegu metropolitan city) for research site. The summary of analysis result is as follows. As a result of classification of biotope types on the research site, it is divided into 11 biotope groups such as a residential biotope group and 51 specific biotope types which is subordinate to the groups. As a result of the first value assessment on classified biotope types, there are 16 types of natural rivers which is full of vegetation as a I class. Also it is analysed as 9 types of IIclass, 14 of IIIclass, 8 of IVclass, and 4 of Vclass. In particular, in light of a wildlife habitat, EB, in case of broad-leaved tree of mixed forest assessed as a II class, was classified into Iclass which is one-step upgraded as a final class with the analysis as there is a structural characteristic (more than 71% of low density, 50 years of age-class). As a result of second assessment, it is analysed that there are 17 special sites (1a,1b) and 33 special sites (2a, 2b, 2c) respectively for preservation of species and biotope. Particularly, in case of the No. 27 space, it was assessed that it has the value of about medium (IIIclass) level, but its value was upgraded with the on-spot detailed investigation that most of Aristolochia contorta, designated as a rare plant by Ministry of Environment, is growing. It is regarded that the above-mentioned research result on evaluation of biotope preservation value is expected to provide very important basic materials for future district unit plans and smooth integration with landscape ecology plans and eco-friendly space development.
This study was conducted to examine whether the parthenogenetic mouse embryonic stem (P-mES) cells can differentiate into functional cardiomyocytes in vitro similar to (mES) cells. p-mES04 and IVF-derived mES03 cells were cultured by suspension culture for 4 days. The formed embryoid bodies (EBs) were treated with 0.75% dimethyl-sulfoxide (DMSO) for further 4 days (4-/4+), and then plated onto gelatin coated culture dish. The appearance of contracting cardiomyocytes from the P-mES04 and mES03 cells was examined for 30 days. The highest cumulative frequency was detected at days 13 (69.83%) and 22 (61.3%), respectively. By immunocytochemistry, beating P-mES04 cells were positively stained with muscle specific anti-sarcomeric a-actinin Ab and cardiac specific anti-cardiac troponin I Ab similar to contracted mES03 cells. When the expression of cardiac muscle-specific genes was analyzed by RT-PCR, beating P-mES04 cells were expressed cardiac specific L-type calcium channel, a1C, cardiac myosin heavy chain a, cardiac muscle heavy polypeptide $7{\beta}$, GATA binding protein 4 and atrial natriuretic factor, but not expressed skeletal muscle specific L-type calcium channel, a1S, which was similar to male adult heart cells and mES03-derived beating cardiomyocytes. The result demonstrates that the P-mES cells can be used as an alternative for the study on the characteristic analysis of in vitro cardiomyocyte differentiation from the ES cells.

In this paper, the performance of a multicarrier CDMA system using adaptive modulation and adaptive subchannel allocation scheme is analyzed in Rayleigh fading channel. The proposed adaptive modulator consists of modulation schemes using QPSK 16 QAM, 64 QAM and 256 QAM and constellations are pointed by Gray code. In addition, the threshold of the analysis is average E$\_$b//N$\_$o/ when the BER is 1 %. In the multicarrier system with adaptive subchannel allocation scheme, each DS waveform of user is transmitted over the K subchannels with the biggest fading among L subchannels. In case of the proposed system, total 4 subchannels are used and data are transmitted over 2 subchannels with the biggest fading, which results in the threshold of each channels is 5.2 dB, 9 dB, 13.2 dB and 8.4 dB, 12.2 dB, 16.3 dB. In the case of proposed system, the BER of 10$\^$-3/ is satisfied if average E$\_$b//N$\_$o/ is 8.1 dB. This is increased performance of 12.9 dB in comparison with conventional system. The BPS according to average channel is needed average E$\_$b//N$\_$o/ of about 15 dB in 7 bit. In the case of subchannel error, the BER of 10$\^$-3/ is 13.6 dB and is declined about 5.5 dB.
Cardiopulmonary bypass cannulas are usually characterized by the French number. However this de- scription provides only the external diameter of the cannula, which gives no information about the press- ure-flow characteristics of the cannula itself. A standardized system to describe the pressure-flow characteristics of a given cannula has recently been proposed and has been termed the M-number It is reported that the pressure-flow characteristics of a particular cannula can be determined from a novo- gram or chart, if the experimentally derived M-number of the cannula is known. In this regard, we conducted an investigation to analyze correlation between experimentally and clinical y derived M-numbers using three different sizes of pediatric aortic cannulas in fifty cardiac patients on cardiopulmonary bypass. The clinical and experimental M-numbers showed a strong correlation. The clinical M-numbers were typically 0.)5 to 0.55 greater than the experimental M-numbers. The clinical M-numbers also showed an inverse relationship to the temperature change of the patient, most probably due to an increase in blood viscosity from hypothermia. This inverse clinical M-numbersltemperature re- lationship was more marked in higher M-number cannulas. The clinical data obtained in this study suggest that the experimentally derived M-numbers correlated strongly with the clinical performance of the cannula with the significant influence of the temperature.
This study was performed to analyze quantitatively the number of living bacteria in forest soil samples collected from Mt. Keryong using improved direct viable count (DVC) and plate count (PC) methods. The number of living bacteria by DVC comprised 18~44% of the total direct count (TDC), whereas the number of living bacteria by PC was less than 1% of TDC. These results showed that viable but non-culturable (VBNC) bacteria existed in the soil with high percentages. Besides, DVC was proved to make it possible to make a quantitative detection of the VBNC bacteria. On the other hand, upon measuring the value from the conventional nutrient broth (NB) and $10^{-2}$ folded diluted nutrient broth (DNB), the values from the DNB showed 5 to 10 times higher than those from the conventional NB medium. These results indicate that oligotrophic bacterial groups, which could multiply in the low nutrient broth, abundantly exist in the soil ecosystem. It would also be possible to apply this kind of method to other substrate to make a quantitative detection of soil bacterial groups.
Efforts to evaluate water pollution using indicator microorganisms have been underway for decades, and driven by research on water purity control applications, water quality criteria are growing more and more strict. Furthermore, recent reports indicate that high concentrations of antibiotics are not absorbed, and are present in excrement from animals and humans dosed with unnecessarily high levels of antibiotics. This has emerged as very important issue from the standpoint of being an ecological and health hazard. In this study, water pollution was analyzed through physicochemical and microbiological means, and antibiotic resistance in indicator microorganisms was assessed. In physicochemical analysis, biochemical oxygen demand (BOD)$_5$ and chemical oxygen demand (COD)$_{Mn}$ evaluation showed that pollution by organisms was highest at the G1 location with a high human population, and the DP location which has many livestock-containing households. The indicator organism levels at the G1 location were: Total Coliforms (1205 colony forming units (CFU)/100 ml), Fecal Coliforms (270 CFU/100 ml), Escherichia coli (253 CFU/100 ml) and Fecal Streptococci (210 CFU/100 ml), while for the DP location levels were: Total Coliforms (1480 CFU/100 ml), Fecal Coliforms (438 CFU/100 ml), E. coli (560 CFU/100 ml), and Fecal Streptococci (348 CFU/100 ml). Levels of fecal indicator microorganisms such as Fecal Coliforms, E. coli and Fecal Streptococci were high at all locations in the fall (the period after the rainy season), and the yearly distribution was similar between these organisms. If the number of livestock-containing households was high, almost all strains of E. coli (as distinct from the other indicator organisms) showed resistance to antibiotics, with the degree of resistance varying between areas. E. coli strains from the OY area in particular, which has a high population density, showed strong resistance to AM10 and Va30. While strong antibiotic resistance was observed overall at the DP and OY locations, no resistance was observed at the EB location.

Microstructure and chemical analysis of sintered bricks containing recycled wastes were investigated by SEM and EDS. The recycled wastes for which substitute ceramic raw materials were EAF (electric arc furnace) dust, fly ash and stone ash. Yellowish and brownish regions on the surface and brownish and blackish regions in the inside of bricks were observed. Main component of yellowish region on the surface turned out to be Zn. No chemical difference between the black-core region and brownish matrix. Mullite crystallites of 1 fm size were distributed in the inside of bricks and enclosed by glass phases. It seems that alumine-silicate mixtures of kaolin and fly ash were transformed to mullite crystallites during the sintering. Relatively large pores ot several ten fm size were observed in the black-core region in the inside of bricks. The main components of the inside of brick were Al and Si. The minor components were C, Na, Mg, K, Ca, and Fe. Particularly, the precipitates of Fe-rich crystallites were observed in the amorphous matrix. These precipitates were formed due to the local reduction atmosphere in the inside of bricks. Zn-rich covers were found on the surface of bricks because Zn diffused from the inside of bricks to the surface under the reduction atmosphere.

This work was investigated the effects of the elevated $CO_2$ and Pb contamination on the growth of Pinus densiflora. Two-years pine trees were planted in Pb-contaminated soils (500 mg/kg-soil) and uncontaminated soils, and cultivated for 3 months in the growth chamber where $CO_2$ concentration was controlled at 380 or 760 ppmv. The growth of P. densiflora were comparatively analyzed in 4 kinds of soil samples (CA : $CO_2$ 380 ppmv + Pb 0 mg/kg, CB : $CO_2$ 380 ppmv + Pb 500 mg/kg, EA : $CO_2$ 760 PPmv + Pb 0 mg/kg, EB : $CO_2$ 760 ppmv + Pb 500 mg/kg). It was measured the growth changes of the p. densiflora caused by $CO_2$ concentration and Pb contamination. The growth of P. densiflora was remarkably inhibited in the Pb-contaminated soil, although the biomass and the root elongations were not significantly affected by the elevated $CO_2$. These results suggested that the growth of p. densiflora was sensitively influenced by Pb contamination rather than $CO_2$ concentration. Compared to the initial soil, total Pb concentration in the soil samples was decreased at 760 ppmv $CO_2$ as well as at 380 ppmv $CO_2$ after 3 months. The accumulation of Pb in the roots at 760 ppmv $CO_2$ was two-fold of that at 380 ppmv $CO_2$, indicating that Pb bioavailability in the root of p. densiflora might be affected by the elevated $CO_2$.
