Association between PON1 Gene SNPs and Growth and Carcass Traits in Beef Cattle

  • Paraoxonase-1 (PON1), like lipoprotein lipase (LPL), plays a key role in the metabolism and physiology of mammalian growth. The objectives of this study were to estimate the allele and genotype frequencies at the PON1/EcoRV and PON1/AluI loci in three genetic groups of beef cattle and to determine associations between these polymorphisms and growth and carcass traits. Genotyping was performed on 30 Angus, 32 Hereford and 26 Simmental. The association analysis was carried out using the GLM procedure of SAS 9.1 and the least squares means of the genotypes were compared by the Tukey's test. Animals with AG genotype at the PON1/EcoRV locus had higher weight at the time of entry into the fattening corrals ($329.97{\pm}6.08kg$) and close to the time of slaughter ($577.56{\pm}8.32kg$) and net meat weight ($275.89{\pm}4.05kg$) and fitted tenderness ($3.10{\pm}0.19kg$) (p<0.05). Animals with AA genotype at the PON1/AluI locus had higher weight at the time of entry ($333.37{\pm}8.93kg$) and slaughter ($576.82{\pm}13.18kg$) and net meat weight ($275.49{\pm}6.43kg$) and average daily gain ($0.68{\pm}0.02kg/d$) (p<0.05). The meat color score was also significantly higher (p<0.05). Between genotypes and breeds, there were significant differences observed except for TBW, REMG, MBS, REA and MCS. As a metabolism gene, genotypes of the SNPs of PON1 gene might be reflecting BFT directly, such as $A_eA_eG_aG_a$ genotype in this experiment.

한국 청소년의 거주지역별 및 체격별 식사의 질 평가지수의 비교 (Comparison of Indices for Diet Quality Evaluation of Korean Adolescents by Residence Area and Body Size)

  • The purpose of this study was to assess several indices of diet quality based on nutrient, food and food group intake of Korean adolescents based on several indices on diet quality according to residence area and body size. Using the data from the 1998 National Health and Nutrition Survey, twenty-four-hour-dietary recalls of a total of 1,110 Korean adolescents aged 13 - 19 years (male 543, female 567) were analyzed for nutrient adequacy ratio (NAR) , index of nutritional quality (INQ) , the number of foods (Dietary Variety Score, DVS) and food group consumed (Dietary Diversity Score, DDS) . In doing that, it was attempted to apply only the minimum amount of solid foods of Kant's without inclusion of liquid foods because of the very limited variety in Korean foods. Based on weight length index, 13.1% of the subjects were categorized as obese, 14.2%, overweight, 44.4%, normal and 28.3%, underweight. Only vitamin B2 intake was higher in the obese group than in the underweight group. There was no meaningful difference in energy, protein and fat intakes according to the grade of the body size. In terms of residence area, intake of fat, niacin, vitamin $B_6$ and folic acid were lower in the rural areas than in the metropolitan city. Only vitamin E intake was higher in the rural areas. Mean value of NARs (MAR) and INQs (mINQ) was also higher in the metropolitan city than in the rural areas, but there was no significant difference of these two values according to body size of the subjects. Mean DVS was 21.02 for total subjects, and has no difference between male and female and between metropolitan city and other medium-small city. But, the rural areas showed the lowest DVS of 19.05. Mean DDS in which five is a maximum score was 3.3 with no significant difference by sex and by residence area in male subjects. However, in female subjects, DDS in the rural areas was the lowest. According to body size of the subjects, there was no meaningful difference in both scores of DVS and DDS. In conclusion, most indices of nutrient intake and food and food group intake were not significantly different by body size of the subjects, while most indices were significantly different by residence areas: higher in the metropolitan city than in the rural area.

Effect of Postmortem Metabolic Rate on Meat Color

  • The current study was conducted to evaluate carcass characteristics, objectives and sensory meat qualities of Hanwoo longissimus muscle as affected by ultimate pH. Twenty-four steers and bulls ($556{\pm}53$ kg and $0.63{\pm}0.32$cm for live weight and backfat thickness, respectively) were used. As there was a linear relationship (r = 0.77) between lean meat color and ultimate pH, cattles were thus segregated into normal $pH{\leq}5.8$, Normal, n = 13) and DFD (pH>5.8, n = 11) groups. Normal pH group had significantly (p<0.05) higher carcass weight, marbling score and backfat thickness than those for high pH group, while fat color and lean meat color were inverse. In principle component analysis for co-ordinates of DFD and normal meats, fat color, lean meat color, texture, time to pH 6.2 and pH at 24 h postmortem were associated with the positive range of the first factor (67.5%) while backfat thickness marbling score and temperature at 24 h were placed in negative values. The rate constant of decline in pH (pH k) did not differ between the two groups, implying that initial pH (i.e., pH at slaughtering) differed between two groups. Contour mapping of pH k between pH at 1 and 24 h postmortem indicated that high pH k was related to lower pHs at 1 and 24 h postmortem. This suggested that the high pH cattles (i.e., DFD cattle) resulted from their own potential. Although the time to reach pH 6.2 was significantly (p<0.05) shorter for normal meat (i.e., 3.2 h) than that for DFD one (i.e., 19.8 h), there were no significantly differences in both WB-shear force and sensory attributes. Given that the experimental animals were sampled from a similar group, which implies a similar myoglobin pigment content, the current data suggested that pre- and post-slaughter animal handling likely had a significant effect on ultimate pH and consequently meat color of Hanwoo longissimus muscle, and also small animals with lower marbling score and backfat thickness had a higher risk for DFD meat.

효소저항성 쌀전분의 첨가가 마들렌의 품질 및 텍스처 특성에 미치는 영향 (Effect of Addition of Enzyme-Resistant Rice RS3 on Quality and Textural Characteristics of Madeleine)

  • This study attempted to examine the application of retrograded starch (RS3) isolated from rice flour into Madeleine which is easy to make, supply enough energy and micro nutrients with adequate drinks, and prevent an adult disease. This could be a popular food to anyone regardless of age and gender who avoid rice and become high value-added, processed rice foods. For this, control Madeleine was made from wheat flour and an experimental one was made from 5 or 10% rice RS3 addition as well as wheat flour. Four different types of rice were produced from Premium Ho-Pyong Rice, that is, dry milled rice flour(RFD), soaked for 8 hours and milled, followed by air-dried rice flour(RFW), rice starch(RST), and retrograded rice starch or enzyme-resistant starch(RS3). The results found were as follows: Proximate compositions were decreased with soaking to make RFW, RST and RS3, compared to RFD. RS3 had the highest L, +a and ${\Delta}E$ with the lowest +b, changing it to a dark color, explaining the need for heat control during processing. At $80^{\circ}C$, the swelling power was shown in the order of RST>RFW>RFD>RS3 and the solubility of RS3 was the highest. There were significant differences in viscosities of peak, trough, cold, breakdown and total setback of all rice samples using RVA (p<0.001). Due to the pH of RS3, the Madeleine batter became acidic (p<.01) and expanded, resulting in more air cells and open texture. With an increasing RS3 level in Madeleine, several textural attributes among 'fresh' and 'stored at room temperature' Madeleine samples were significantly different by using Texture Analyzer. While the addition of RS3 in Madeleine did not significantly affect the sensory evaluation, indicating RS3 isolated from rice as a beneficial ingredient for processed rice products.

A Comparison of the In Vitro Inhibitory Effects of Thelephoric Acid and SKF-525A on Human Cytochrome P450 Activity

  • Thelephoric acid is an antioxidant produced by the hydrolysis of polyozellin, which is isolated from Polyozellus multiplex. In the present study, the inhibitory effects of polyozellin and thelephoric acid on 9 cytochrome P450 (CYP) family members (CYP1A2, CYP2A6, CYP2B6, CYP2C8, CYP2C9, CYP2C19, CYP2D6, CYP2E1, and CYP3A4) were examined in pooled human liver microsomes (HLMs) using a cocktail probe assay. Polyozellin exhibited weak inhibitory effects on the activities of all 9 CYPs examined, whereas thelephoric acid exhibited dose- and time-dependent inhibition of all 9 CYP isoforms ($IC_{50}$ values, $3.2-33.7{\mu}M$). Dixon plots of CYP inhibition indicated that thelephoric acid was a competitive inhibitor of CYP1A2 and CYP3A4. In contrast, thelephoric acid was a noncompetitive inhibitor of CYP2D6. Our findings indicate that thelephoric acid may be a novel, non-specific CYP inhibitor, suggesting that it could replace SKF-525A in inhibitory studies designed to investigate the effects of CYP enzymes on the metabolism of given compounds.

소의 뇌 Inositol triphosphate kinase와 Calmodulin-Affigel과의 친화도 (THE AFFINITY OF CALMODULIN-AFFIGEL FOR INOSITOL TRIPHOSPHATE KINASE FROM BOVINE BRAIN)

  • 세포막의 정보전달기전중 phosphoinositide system은 정보가 전달될때 phospholipase C 효소의 작용으로 phosphatidyl inositol bisphosphate로부터 inositol triphosphate($IP_3$)와 diacylglycerol이 생성되며 $IP_3$는 다시 $IP_3$kinase에 의해 inositol tetrakisphosphate($IP_4$)로 되어 이차전령 물로서 작용한다. 본 연구는 $IP_3$kinase효소가 $Ca^{2+}$와 calmodulin에 의해 활성화되는 성질을 이용하여 calmodulin을 정제하고 $IP_3$kinase효소와의 친화도를 비교 관찰하였다. Calmodulin정제는 phenyl-Sepharose resin을 이용하여 column chromatography를 시행하여 정제확인하였으며 분자량이 17,000임을 SDS-polyacrylamide gel 전기영동으로 확인하였다. 정제된 calmodulin을 affigel column에 결합시킨 gel에 소의 뇌로부터 분리한 $IP_3$kinase효소가 담긴 시료를 calmodulin-affigel column에 적용하여 결합 및 유출정도를 비교하였으며 $Ca^{2+}$이 든 buffer에서 친화도가 가장 컸으며 유출은 EGTA용액에서 일부 유출되었으며 calmodulin/$Ca^{2+}$이 든 buffer에선 강한 유출정도를 관찰하였다. 그러나 calmodulin/$Ca^{2+}$$IP_3$kinase효소의 활성을 증가시키며 calmodulin이 단백질이어서 정제면에서 효소와의 분리가 쉽지않아 여러 다른 detergent를 적용하였으나 0.2% chaps buffer에서 집중된 유출을 관찰하였다.

체외생산된 생쥐 배반포기배의 ICM과 Trophectoderm 세포수에 관한 연구 (ICM - Trophectoderm Cell Numbers of Mouse IVF/IVC Blastocysts)

  • 본 연구는 Polynucleotide-specific 형광물질을 이용한 Differential labelling 기법으로 체외수정 후 배양 4일째 생산된 B6CBA Fl 생쥐 배반포의 Total, ICM, Trophectoderm의 세포수를 조사함으로서 생쥐의 착상전 후기 배발달에 대한 기초 자료를 얻고자 실시하였다. 공시 배반포는 과배란처리에 의해 얻어진 난자를 $1{\times}10^6cells/ml$의 정자로 수정시키고, 95시간동안 M16배양액과 $37^{\circ}C$, 5% $CO_2$배양기 내에서 배양하여 배반포강의 확대와 투명대 두께의 감소를 기준으로 early, middle, expanded와 hatching으로 구분하였다. 본 연구에서 얻어진 결과는 다음과 같다. 1) 체외수정 후 95시간째 배반포 발달율은 86.7%였으며, early, middle, expanded와 hatching으로 16.3%, 18.9%, 10.5%, 40.9% 였다. 2) Bisbenzirnide를 이용한 배반포의 총 세포수는 early, middle, expanded, hatching 각각 $35.6{\pm}10.4$, $49.4{\pm}8.6$, $60.8{\pm}10.7$$62.7{\pm}13.9$를 얻었다. 3) Polynucleotide-specific형광물질을 이용한 Differential labelling으로 배반포 ICM과 Trophectoderm의 세포수를 early, middle, expanded, hatching으로 나누어 조사한 결과, ICM세포수는 각각 $9.6{\pm}3.0$, $13.6{\pm}3.9$, $16.0{\pm}3.3$, $19.5{\pm}4.6$개 이었고, Trophectoderm세포수는 $30.6{\pm}5.1$, $39.9{\pm}5.8$, $42.2{\pm}8.1$, $43.7{\pm}11.1$ 개로 나타나 ICM과 Trophectoderm 모두 동일하게 발달의 진행정도에 따라 세포수의 증가양상을 나타내었다. 또한, Bisbenzimide와 Differential labelling에서 얻어진 총세포수의 비교에서도 동일하게 발달의 진행정도에 따라 세포수의 증가를 나타내었으며 그와 동시에 세포수도 거의 유사하였다. 이러한 결과로 미루어 볼때, Differential labelling을 이용한 빠르고도 간편한 세포수 계산법은 착상전 후기 배발달을 고찰하는데 유용하며, 배양조건에 따른 Embryo의 Quality를 반영하는 Indicator로서 이용될 수 있다는 것을 시사한다.

제주지역 중학생의 총당류 섭취실태 연구 (A study of total sugar intake by middle school students in Jeju Province)

  • 본 연구는 제주지역 중학생 1,184명 (남학생 476명, 여학생 708명)에게 24시간 회상법으로 식사조사를 하여 총당류 섭취량을 추정하였으며 그 결과는 다음과 같다. 1. 조사대상자의 1일 총당류 섭취량은 전체 60.3 g, 남학생 50.5 g, 여학생 66.9 g으로서 여학생이 남학생보다 유의하게 높았다. 1일 총당류 섭취량 중 각 개별 당류가 차지하는 비중은 서당 (23.9 g) > 포도당 (12.9 g) > 과당 (12.3 g) > 유당 (6.0 g) > 맥아당 (3.3 g)의 순이었다. 2. 끼니별 1일 총당류 섭취량은 아침 6.5g, 점심 9.0g, 저녁 11.8 g, 간식 33.0 g으로 간식에서 섭취하는 양이 가장 많았다. 식품군별 1일 총당류 섭취량은 곡류 (23.0 g) > 우유류 (11.0 g) > 과일류 (7.3 g) > 당류 (6.1 g) > 채소류 (5.8 g)의 순으로 높았다. 음식종류별 1일 총당류 섭취량은 빵 과자류 (11.5 g) >면류 (10.2 g) > 유제품류 (7.5 g) > 빙과류 (6.2 g) > 음료류 (5.3 g) > 과일류 (4.5 g) > 튀김류 (3.2 g)의 순이었다. 3. 조사대상자의 1일 총당류 섭취량 60.3 g 중 천연당은 18.3 g (30.3%), 첨가당은 35.8 g (59.4%), 혼합당은 6.2 g (10.3%) 이었다. 총당류에서 얻는 열량비율이 20% 이상인 고당류섭취군은 20% 미만인 사람에 비해 탄수화물, 칼슘, 비타민 B1, B2, C의 섭취량은 유의하게 높은 반면 철, 아연, 니아신, 비타민 B6, 비타민 E 섭취량은 유의하게 낮았다. 단맛선호군은 단맛저선호군에 비해 열량, 탄수화물 및 총 당류섭취량이 유의하게 높았다. 이상의 결과를 종합해 보면 제주지역 중학교 2학년 학생들의 1일 평균 총당류 섭취량은 60.3 g이었고, 이중 첨가당으로 인한 섭취가 59.4%로 가장 많았으며, 간식으로부터 섭취되는 비율이 가장 높았다. 따라서 중학생의 총당류 섭취량을 감소시키기 위해서는 올바른 간식 식품선택에 대한 영양교육이 필요하다.

치수강(齒髓腔)과 치관외면(齒冠外面)과의 최단거리(最短距離)에 관(關)한 실측연구(實測硏究) (A STUDY ON MEASUREMENT OF MINIMAL DISTANCE BETWEEN PULP CHAMBER AND CORONAL SURFACE)

  • To determine the thickness of coronal hard structure the minimal distance between pupal surface and outer surface of crown was measured by means of Bowley gauge on extracted first molars. Upper(28 teeth) and lower(24 teeth) were carefully collected from 30-39 years of age and male. The teeth were split mesio-distally through central pit. On the split surface various part which are deeply related in cavity preparation were measured (schematic drawing). The results were as follows: A : Distance from mesio-cervical enamel to pulp chamber surface. upper $2.63{\pm}0.19$(mm) Lower $2.18{\pm}0.27$(um) B : Distance from mesial chamber ceiling to mesial surface upper $2.75{\pm}0.34$ Lower $2.62{\pm}0.31$ C : Distance from mesial chamber ceiling to occlusal surface upper $3.82{\pm}0.51$ Lower $3.49{\pm}0.50$ D : Distance from distal chamber ceiling to occlusal surface upper $4.28{\pm}0.69$ Lower $3.90{\pm}0.52$ E : Distance from distal chamber ceiling to distal surface upper $2.79{\pm}0.45$ Lower $2.41{\pm}0.40$ F : Distance from disto cervical enamel to pulp chamber surface upper $2.49{\pm}0.24$ Lower $2.39{\pm}0.25$.

비오톱 유형분류를 기반으로 한 경관평가 모형개발 및 적용 (A Development and Application of the Landscape Evaluation Model Based on the Biotope Classification)

  • 본 연구는 대규모 택지개발 예정지로 계획되어 있는 대구광역시 달성군 옥포면 논공읍 일대 약 $10km^2$를 대상으로 비오톱 유형분류를 기반으로 한 미 시각 측면에서의 경관가치 평가 모형을 설정해 보고, 이를 실사례지에 적용해 봄으로써 평가모형의 타당성을 검증해 보는데 가장 큰 의의를 두었다. 연구결과를 요약하면 아래와 같다. 비오톱 유형분류 결과, 평가대상이 되는 23개의 비오톱 유형과 이에 귀속되는 140개의 세부 비오톱 유형으로 분류되었다. 또한 평가지표 선정을 위한 설문조사는 1차, 2차, 3차 설문으로 나누어 수행하였다. 먼저 1차 설문분석은 우선 문헌연구 및 현장조사를 바탕으로 선정된 22개의 평가지표 항목들에 대한 중요도를 분석하는 단계이다. 다음으로 2차 설문분석은 요인분석을 실시, 내재된 상관관계를 이용하여 지표들을 재분류하였다. 3차 설문분석은 선정된 평가지표 항목의 가중치를 산정하는 단계로서, 평가지표 항목의 중요도 평균을 기준으로 합이 10이 되도록 표준화하는 방법을 사용하였으며, 2차 평가 시 각 지표항목의 점수에 가중치를 곱하여 최종 평가에 활용하였다. 이상과 같은 전문가 설문분석 결과, 최종 선정된 평가지표는 활력충전요소, 시각적 방해요소 등 총 19개 항목으로 조사되었다. 1차 평가 결과, 특별히 가치 있는 I등급은 식생이 풍부한 자연형 하천유형(D), 습지 및 늪지 유형(I) 등 총 7개 유형으로 나타났다. 또한 II등급으로는 식생이 풍부한 인공형 하천유형(E), 침엽수림 중심의 산림유형(Q) 등 총 4개 유형, III등급은 식생이 풍부한 자연형 실개천 및 도랑유형(F) 등 총 5개 유형으로 나타났다. 상대적으로 가치가 낮은 IV등급, V등급은 각각 식생이 빈약한 실개천 및 도랑유형(G) 등 총 2개 유형과 녹지가 빈약한 복합형 주거지역(B) 등 총 5개 유형으로 분석되었다. 2차 평가 결과, 경관 가치가 특별히 높은 비오톱 유형(1a, 1b)은 15개 공간으로 조사되었으며, 경관의 가치가 높은 유형(2a, 2b, 2c)은 총 28개 공간으로 분석되었다. 그러나 본 연구에서 제시된 경관평가 모형이 타 유사지역에서도 높은 적용성을 가지기에는, 본 연구에서 설정한 하나의 연구대상지만으로는 부족함이 있었던 바, 차후에는 다양한 사례지를 종합하고 표준화하여 객관성을 높여 나갈 필요가 있을 것으로 사료된다.
