발생단계별 해마신경세포에서 eIF4E 및 eIF4EBP1의 표현 (Developmental Expression of Eukaryotic Initiation Factor 4E (eIF4E) and eIF4E-binding Protein 1 (eIF4EBP1) in Rat Hippocampal Neurons)

  • 신경세포의 가지돌기 내 단백질합성은 필요한 단백질을 실시간으로 제공할 수 있는 이점을 제공한다. 본 연구에서는 단백질합성인자 eIF4E와 그 억제 단백질인 eIF4EBP1의 발생단계별 표현을 배양한 해마신경세포를 면역 염색하여 조사하였다. eIF4E는 가지돌기에 점박이 모양으로 표현되었으며, 핵에는 표현되지 않았다. 그러나 eIF4EBP1는 가지돌기 뿐 아니라 발생초기(DIV 0.5)부터 핵에서 표현되었으며 성숙한 세포에서 핵에 더욱 뚜렷이 표현되었다. eIF4E 혹은 eIF4EBP1의 PSD95과의 colocalization은 $39.1{\pm}9.6%$$70.5{\pm}5.2%$ (DIV 7), $57.7{\pm}8.2%$$36.0{\pm}3.1%$ (DIV 10), $29.9{\pm}2.9%$$40.2{\pm}11.7%$ (DIV 20)이었다. eIF4E와 eIF4EBP1의 colocalizatin은 $18.5{\pm}2.6%$ (DIV 7), $11.1{\pm}3.9%$ (DIV 10), $38.6{\pm}5.6%$ (DIV 20)이었다. 이 결과는 eIF4E 및 eIF4EBP1의 많은 부분이 연접후에 위치하며, 발생초기에는 eIF4E가 활동적인 형태로 존재하지만, 성숙 신경세포에서는 eIF4EBP1과 결합하여 비활성적인 형태로 존재함을 의미한다.

  • 고려인삼의 묘를 GA, 2,4-D 및 B-9로 처리하여 암하에서 생육시켰다. 생장효율($E_1$ = St/Ro, $E_2$ = St/(Ro-Rt), $E_3$ = (Ro- Rt)/ Ro, 여기서 St는 지상부중, Ro와 Rt는 초기와 일정시간 후의 뿌리의 무게임)과 기타 관련된 요인 및 상호관계를 조사하였다. $E_1$$E_3$는 온도변화에 이차식 관계를 보인 반면 $E_2$는 부의 직선 상관을 보였다. $E_1$$E_2$보다 $E_3$에 더 의존하였다. $E_2$$E_3$값은 거의 같았다. $E_2$는 전에 보고된 값보다 커서 뿌리간 큰 차이가 있음을 나타낸다. GA는 고온역에서 $E_2$$E_3$값을 증대시켰으며 B-9은 모든 온도에서 $E_3$를 크게 감소시켰고고 저온역에서 $E_2$를 감소시켰다. 지상부중은 대부분의 생장조절제와 온도에서 기질량과 유의정상관을 보였으며 몇경우에는 호흡소모량과 유의정상관을 보였다. 호흡소모량은 저온역에서만 $E_1$$E_3$와는 유의정상관을 $E_2$와는 유의부상관을 보였다.

칡소와 비경흑색 한우의 Melanocortin Receptor 1 (MC1R) 유전자형 분석 (Analysis of Melanocortin Receptor 1 (MC1R) Genotype in Korean Brindle Cattle and Korean Cattle with Dark Muzzle)

  • 칡소와 비경흑색 한우의 MC1R 유전자의 유전자형를 조사하고 칡소의 모색 발현과 한우 비경색과의 관계를 규명하기 위하여 MC1R 유전자의 PCR-RFLP 분석을 수행하였다. 소에서 나타나는 6가지 유전자형($E^D/E^D,\;E^D/E^+,\;E^D/e,\;E^+/E^+,\;E^+$/e와 e/e) 중 칡소에서는 단지 2개의 유전자형 $E^+/E^+$$E^+$/e 만이 출현하였고, e/e 유전자형을 가지는 개체는 전혀 출현하지 않아 제주재래흑우에서와 같이 흑색 호반모가 발현되기 위해서는 기본적으로 $E^+$ allele이 필요한 것으로 사료되었다. 비경흑색 한우와 비경황색 한우에서는 모두 $E^+$/e 혹은 e/e 유전자형이 출현하였고 $E^+$와 e allele의 빈도는 각각 0.37, 0.63과 0.11, 0.89였다. 비록 $E^+$ allele의 빈도가 비경흑색 한우에서 비경황색 한우에서 보다 높았지만, $E^+$ allele과 비경 흑색과는 완전한 연관성은 없었다. 이러한 결과는 MC1R 유전자 분석이 칡소 뿐만 아니라 한우의 모색 고정에도 유용한 도구로 이용 가능함을 제시하고 있다.

  • C 형 간염 바이러스 (Hepatitis C Virus, HCV)는 두종류의 외피당단백질 $E1_{192-383}$$E2_{384-746}$를 갖고 있다. $E1_{192-383}$$E2_{384-740}$ 단백질은 glutathione S-transferae (GST) 융합단백질의 형태로 대장균에서 발현되지 않았으나, 이 단백질들의 C말단에 존재하는 소수성영역을 제거하였을 때 $GST-E1_{192-283}$ 융합단백질은 과량으로 가용성 형태로 발현되었고, $GST-E2_{384-649}$ 융합단백질은 비 가용성 형태로 발현되었다. 이 융합단백질들 각각은 HCV 양성환자의 혈청과 특이적으로 반응하였다. Thrombin으로 처리하여 얻은 정제된 $E1_{192-283}$ 단백질 및 융합형태의 $GST-E2_{384-649}$ 단백질 각각을 생쥐에 접종하였을 때 E1 및 E2 특이적인 항체가 생성되었다. 이 결과들은 $E1_{192-383}$$E2_{384-649}$ 융합단백질 C 말단에 존재하는 소수성영역이 이 단백질들의 발현량 및 가용성에 영향을 주며 $E1_{192-283}$ 단백질 영역내에 HCV 양성환자의 혈청과 특이적으로 반응하는 epitope (s)이 존재한다는 것을 제시해 주고 있다.

  • 톱다리개미허리노린재의 페로몬 성분인 (E)-2-hexenyl (E)-2-hexenoate (1) 과 (E)-2-hexenyl (Z)-3-hexenoate (2)의 합성에 있어 Steglich 에스테르화 반응의 최적 조건을 검토하였다. (E)-2-헥센산 또는 (Z)-3-헥센산에 대하여 1-1.5 당량의 디사이클로카보다이이미드 (DCC), 1.5-2.0 당량의 (E)-2-헥센놀과 0.1 당량의 4-디메틸아미노피리딘(DMAP)과 반응하였을 때, 각각 76-78% 와 87-91%의 수율로 1과 2를 얻었다.

  • This study is attempted to serve the fundamental theory for ‘The reorganization of Korean coastal and adjacent water fishery.’ On the Korean coastal and adjacent water fishery where one species stock is catched by multiple fisheries, traditional analysis is not suitable, as analyzing through adjusting the heterogeneous fishing effort among the fisheries to an unit having same fishing strength. Therefore, this study presents the ‘Multi-Variable Model’, adopting fishing effort from each fishery as independent variable, respectively, in order to analyze the quantitative fluctuation of fishery resource not with fishing strength but with amount of fishing effort, measured by the unit of each fishery. For the sake of simplication, this study assumes that one species is catched by two fishery, premise two assumption. 1) Every fishery has not the selectivity in fishing 2) The promotion of fishing efficiency is accomplished in the same speed. Resource equilibrium equation of each fishery is; $$CPUE_1=\frac{Y_1}{E_1}=a_1+ b_1\cdot E_1+c_1\cdot E_1$$ $$CPUE_1=\frac{Y_1}{E_1}=a_1+ b_1\cdot E_1+c_1\cdot E_1$$ Sustainable yield equation is; $$SY_1=a_1\cdot E_1+\cdot b_1E{_1}{^2}+c_1\cdot E_1\cdot E_1$$ $$SY_1=a_1\cdot E_1+b_1\cdot E_1\cdot E_1+c_1\cdot E{_1}{^2}$$ This study is rudimentary, hereafter, refinemental analysis will be supplemented.

Characterization of Virulence Function of Pseudomonas cichorii Avirulence Protein E1 (AvrE1) during Host Plant Infection

  • Pseudomonas cichorii secretes effectors that suppress defense mechanisms in host plants. However, the function of these effectors, including avirulence protein E1 (AvrE1), in the pathogenicity of P. cichorii, remains unexplored. In this study, to investigate the function of avrE1 in P. cichorii JBC1 (PcJBC1), we created an avrE1-deficient mutant (JBC1ΔavrE1) using CRISPR/Cas9. The disease severity caused by JBC1ΔavrE1 in tomato plants significantly decreased by reducing water soaking during early infection stage, as evidenced by the electrolyte leakage in infected leaves. The disease symptoms caused by JBC1ΔavrE1 in the cabbage midrib were light-brown spots compared to the dark-colored ones caused by PcJBC1, which indicates the role of AvrE1 in cell lysis. The avrE1-deficient mutant failed to elicit cell death in non-host tobacco plants. Disease severity and cell death caused by JBC1ΔavrE1 in host and non-host plants were restored through heterologous complementation with avrE1 from Pseudomonas syringae pv. tomato DC3000 (PstDC3000). Overall, our results indicate that avrE1 contributes to cell death during early infection, which consequently increases disease development in host plants. The roles of PcJBC1 AvrE1 in host cells remain to be elucidated.

칡소의 MC1R의 유전자형에 따른 교배 조합이 자손의 모색과 유전자형 변이에 미치는 영향 (Effects of Genotype Mutation and Coat Color Phenotype on the Offspring from Mating System of MC1R Genotype Patterns in Korean Brindle Cattle)

  • Bovine coat color is decided by the melanocortin receptor 1 (MC1R) genotype mutation and melanogenesis. Specially, in the various cattle breeds, dominant black coat color is expressed by dominant genotype of $E^D$, red or brown is expressed in the frame shift mutation of recessive homozygous e by base pair deletion and wild type of $E^+$ is expressed in various coat colors. However, not very well known about the effected of MC1R genotype mutation on the coat color through family lines in KBC. Therefore, this study were to investigate effect of MC1R genotype mutation on the coat color, and to suggest mating breed system in accordance with of MC1R genotype for increased on brindle coat color appearance. Parents (sire 2 heads and dam 3 heads) and offspring (total : 54 heads) from crossbreeding in KBC family line with the MC1R genotype and phenotype records were selected as experimental animals. The relationship between melanocortin 1 receptor (MC1R) genotypes expression verified by PCR-RFLP, and brindle coat color appearance to the family line of the cross mating breed from MC1R genotype pattern was determined. As a result, 4MC1R genetic variations, $E^+/E^+$ (sire 1), $E^+/e$ (sire 2 and dam 3), $E^+/e$ with 4 bands of 174, 207 and 328 bp (dam 1) and $E^+/e$ with 3 bands of 174, 207, 328 and 535 bp (dam 2) from parents (sire and dam) of KBC. However, 3 genetic variations, e/e (24%), $E^+/E^+$ (22%) and $E^+/e$ (56%) were identified in offspring. Also, brindle coat color expressrated was the e/e with the 0%, $E^+/E^+$ with 67% and $E^+/e$ with 77% from MC1R genotype in offspring on the cross mating of KBC. Furthermore, when the sire had $E^+/e$ genotype and the dam had $E^+/E^+$ with the 3 bands or $E^+/e$ genotype, and both had whole body-brindle coat color, 62% of the offspring had whole body-brindle coat color. Therefore, the seresults, the mating system from MC1R genotype patterns of the sires ($E^+/e$) and dams ($E^+/E^+$ with the 3 bands or $E^+/e$) with brindle coat color may have the highest whole body-brindle coat color expression in their offspring.

핸드스프링 몸접어 앞공중돌기동작의 운동학적 분석 (The Kinematic Analysis of Handspring Salto Forward Piked)

  • The purpose of this study is to compare and analyze the phase-by-phase elapsed time, the COG, the body joint angle changes and the angular velocities of each phase of Handspring Salto Forward Piked performed by 4 college gymnasts through 3D movement analysis program. 1. The average elapsed time for each phase was .13sec for Phase 1, .18sec for Phase 2, .4sec for Phase 3, and .3sec for Phase 5. The elapsed time for Phase 1 to Phase 3 handspring was .35sec on average and the elapsed time for Phase 4 to Phase 5 handspring salto forward piked was .7sec on average. And so it showed that the whole elapsed time was 1.44sec. 2. The average horizontal changes of COG were 93.2 cm at E1, 138. 5 cm at E2, 215.7 cm at E3, 369.2 cm at E4, 450.7 cm at E5, and 553.1 cm at E6. The average vertical changes of COG were 83.1 cm at E1, 71.3 cm at E2, 78.9 cm at E3, 93.7 cm at E4, 150.8 cm at E5, and 97.2 cm at E6. 3. The average shoulder joint angles at each phase were 131.6 deg at E1, 153.5 deg at E2, 135.4 deg at E3, 113.4 deg at E4, 39.6 deg at E5, and 67.5 deg at E6. And the average hip joint angles at each phase were 82.2 deg at E1, 60 deg at E2, 101.9 deg at E3, 161.2 deg at E4, 97.7 deg at E5, and 167 deg at E6. 4. The average shoulder joint angular velocities at each phase were 130.9deg/s E1, 73.1 deg/s at E2, -133.9 deg/s at E3, -194.4 deg/s at E4, 29.4 deg/s at E5, and -50.1 deg/s at E6. And the average hip joint angular velocities at each phase were -154.7 deg/s E1, -96.5 deg/s at E2, 495.9 deg/s at E3, 281.5 deg/s at E4, 90.3 deg/s at E5, and 181.7 deg/s at E6. The results shows that, as for the performance of handspring salto forward piked, it is important to move in short time and horizontally from the hop step to the point to place the hands on the floor and jump, and to stretch the hip joints as much as possible after the displacement of the hands and to keep the hip joints stretched and high in the vertical position at the takeoff. And it is also important to bend the shoulder joints and the hip joints fast and spin as much as possible after the takeoff, and to decrease the speed of spinning by bending he shoulder joints and the hip joints quickly after the highest point of COG and make a stable landing.

한국산 톱다리개미허리노린재 집합페로몬 성분의 조성과 배합비율별 유인력 (Composition of the Aggregation Pheromone Components of Korean Bean Bug and Attractiveness of Different Blends)

  • 톱다리개미허리노린재(Riptortus clavatus Thunberg) (Heteroptera: Alydidae) 수컷 성충은 (E)-2-hexenyl (Z)-3-hexenoate (E2HZ3H), (E)-2-hexenyl (E)-2-hexenoate (E2HE2H) 및 tetradecyl isobutyrate(TI)의 세 성분으로 조성되는 집합페로몬을 분비하고, 그 페로몬에 동종의 암수 성충과 약충을 유인되며, 난기생봉인 Ooencyrtus nezarae (Hymenoptera: Encyrtidae)는 이 페로몬을 기주탐색에 이용한다. 본 연구에서는 한국의 진주와 익산, 일본의 Tsuhba와 Kumamoto지역에서 채집한 톱다리개미허리노린재의 집합페로몬 성분의 조성비를 조사하고, 각 성분의 배합비율별 유인력을 진주와 Kumamoto에서 조사하였다. 진주와 익산계통의 집합페로몬 조성 비율(E2HZ3H:E2HE2H:TI)은 각각 1:1.4:0.2, 1:0.8:0.2였으며, Tsuhba와 Kumamoto계통은 각각 1:2.8:0.2, 1:1.5:0.1이었다. 야외 유인력 검정에서, 1:1:1 (E2HZ3H:E2HE2H:TI=16.7:16.7:16.7mg/고무격막) 또는 1:1:0.5 (E2HZ3H:E2HE2H:TI=20:20:10mg/고무격막)로 배합한 페로몬이 1:5:1 (E2HZ3H:E2HE2H:TI = 7.1:35.7:7.1mg/고무격막)로 배합한 것보다 유인력이 높았다.
