Antibacterial activity of the water-soluble portion of Ulmus pzcmila L. extract against 10 bacterial species was studied by both cylinder plate dilution method and broth dilution test tube method. Inhibitory effect of the extract on the bacteria was also investigated by plotting bacterial survival at various concentration of the extract. The crude extract exhibited antibacterial activity against all of the tested bacterial species with exception of K pneurnoniae. The fractions of the extract prepared by CM Sephadex-C 50 ion exchange chromatography were also subjected to test the antibacterial activity, and the activity was studied after autoclaving for 20 minutes.
For the field application of dielectric barrier discharge plasma reactor, a multi-plasma reactor was investigated for the inactivation of microorganisms in sewage. We also considered the possibility of degradation of non-biodegradable matter ($UV_{254}$) and total organic carbon (TOC) in sewage. The multi-plasma reactor in this study was divided into high voltage neon transformers, gas supply unit and three plasma modules (consist of discharge, ground electrode and quartz dielectric tube). The experimental results showed that the inactivation of microorganisms with treated water type ranked in the following order: distilled water > synthetic sewage effluent >> real sewage effluent. The dissolved various components in the real sewage effluent highly influenced the performance of the inactivation of microorganisms. After continuous plasma treatment for 10 min at 180 V, residual microorganisms appeared below 2 log and $UV_{254}$ absorbance (showing a non-biodegradable substance in water) and TOC removal rate were 27.5% and 8.5%, respectively. Therefore, when the sewage effluent is treated with plasma, it can be expected the inactivation of microorganisms and additional improvement of water quality. It was observed that the $NH_4{^+}$-N and $PO{_4}^{3-}$-P concentrations of sewage was kept at the constant plasma discharging for 30 min. On the other hand, $NO_3{^-}$-N concentration was increased with proceeding of the plasma discharge.
An enantioselective lipase gene (esf) for the kinetic resolution of optically active (S)-flurbiprofen was cloned from the new strain Serratia marcescens ES-2. The esf gene was composed of a 1,845-bp open reading frame encoding 614 amino acid residues with a calculated molecular mass of 64,978 Da. The lipase expressed in E. coli was purified by a three-step procedure, and it showed preferential substrate specificity toward the medium-chain-length fatty acids. The esf gene encoding the enantioselective lipase was reintroduced into the parent strain S. marcescens ES-2 for secretory overexpression. The transformant S. marcescens BESF secreted up to 217kU/ml of the enantioselective lipase, about 54-fold more than the parent strain, after supplementing 3.0% Triton X-207. The kinetic resolution of (S)-flurbiprofen was carried out even at an extremely high (R,S)-flurbiprofen ethyl ester [(R,S)-FEE] concentration of 500 mM, 130 kU of the S. marcescens ES-2 lipase per mmol of (R,S)-FEE, and 1,000 mM of succinyl ${\beta}-cyclodextrin$ as the dispenser at $37^{\circ}C$ for 12h, achieving the high enantiomeric excess and conversion yield of 98% and 48%, respectively.

This study, for the first time, reports the functional expression of lipase B derived from the yeast Candida antarctica (CALB) in Corynebacterium strain using the Escherichia coli plasmid PK18. The CALB gene fragment encoding a 317-amino-acid protein was successfully obtained from the total RNA of C. antarctica. CALB was readily produced in the Corynebacterium strain without the use of induction methods described in previous studies. This demonstrated the extracellular production of CALB in the Corynebacterium strain. CALB produced in the Corynebacterium MB001 strain transformed with pEC-CALB recombinant plasmid exhibited maximum extracellular enzymatic activity and high substrate affinity. The optimal pH and temperature for the hydrolysis of 4-nitrophenyl laurate by CALB were 9.0 and 40℃, respectively. The enzyme was stable at pH 10.7 in the glycine-KOH buffer and functioned as an alkaline lipase. The CALB activity was inhibited in the presence of high concentration of Mg2+, which indicated that CALB is not a metalloenzyme. These properties are key for the industrial application of the enzyme.
A strain antagonistic to Fusarium solani, CHO104, was selected from approximately 100 microorganisms isolated from soil. Strain CHO104 was identified as Bacillus amyloliquefaciens and found to be Gram-positive based on the Biolog system and 16S rRNA methods. A culture broth of B. amyloliquefaciens CHO104 also exhibited antimicrobial activity against various microorganisms. As such, the EtOAc extract of the culture broth was isolated by various column chromatographic procedures and HPLC. The antimicrobial and antifungal substance was then characterized as macrolactin A $(C_{24}H_{34}O_5)$ using high-resolution EI-MS and NMR analyses, and found to be very effective in inhibiting the growth of Staphylococcus aureus, E. coli, and Botrytis cinerea, even when using a concentration of one-twentieth of the benzoic acid as the control compound.

A study was performed to examine the effects of reclaimed-water irrigation on microorganism con-centration in ponded-water of paddy rice plots. Several treatments were used and each one was triplicated to evaluate the change of indicator microorganisms (total coliforms (TC), fecal coliforms FC), and E. coli) concentrations in 2003 and 2004 growing seasons. Their concentrations increased significantly right after irrigation, but decreased about $45\%$ in 24 hours. It implies that agricultural activities such as plowing and fertilizing should be practiced one or two days after irrigation considering health-risks. Treatments with UV-disinfected water irrigation demonstrated significantly lower concentrations than others including control plots where natural surface water was irrigated. The monitoring results from actual paddy rice fields and experimental paddy plots showed that concentrations of indicator microorganisms ranged from $10^2\;to\;10^5$ MPN/100mL. A comprehensive assessment of existing agricultural practices and a thorough monitoring in the field as well as treatment-plots are recommended to make more realistic national guidelines more applicable. UV-disinfected water irrigation reduced microorganism concentrations in paddy fields down to below the concentration of conventional paddy rice culture, and is thought to be an effective and feasible measure fur agricultural reuse of secondary effluent.
Bacteria have evolved various types of resistance mechanism to toxic heavy metals, such as arsenic and antimony. An arsenical and antimonial resistant bacterium was isolated from a shallow creek draining a coal-mining area near Taebaek City, in Kangwon-Do, Korea. The isolated bacterium was identified and named as Pseudomonas sp. KM20 after biochemical and physiological studies were conducted. A plasmid was identified and its function was studied. Original cells harboring the plasmid were able to grow in the presence of 15 mM sodium arsenite, while the plasmid-cured (plasmidless) strain was sensitive to as little as 0.5 mM sodium arsenate. These results indicated that the plasmid of Pseudomonas sp. KM20 does indeed encode the arsenic resistance determinant. In growth experiments, prior exposure to 0.1 mM arsenate allowed immediate growth when they were challenged with 5 mM arsenate, 5 mM arsenite, or 0.1 mM antimonite. These results suggested that the arsenate, arsenite, and antimonite resistance determinants of Pseudomonas sp. KM20 plasmid were indeed inducible. When induced, plasmid-bearing resistance cells showed a decreased accumulation $of\;73^As$ and showed an enhanced efflux $of\;^73As$. These results suggested that plasmid encoded a transport system that extruded the toxic metalloids, resulting in the lowering of the intracellular concentration of toxic oxyanion. In a Southern blot study, hybridization with an E. coli R773 arsA-specific probe strongly suggested the absence of an arsA cistron in the plasmid-associated arsenical and antimonial resistance determinant of Pseudomonas sp. KM20.
Gene encoding cyclodextrin glycosyltransferase (CGTase), from thermotolerant Paenibacillus sp. T16 isolated from hot spring area in northern Thailand, was cloned and expressed in E. coli (JM109). The nucleotide sequences of both wild type and transformed CGTases consisted of 2139 bp open reading frame, 713 deduced amino acids residues with difference of 4 amino acid residues. The recombinant cells required 24 h culture time and a neutral pH for culture medium to produce compatible amount of CGTase compared to 72 h culture time and pH 10 for wild type. The recombinant and wild-type CGTases were purified by starch adsorption and phenyl sepharose column chromatography and characterized in parallel. Both enzymes showed molecular weight of 77 kDa and similar optimum pHs and temperatures with recombinant enzyme showing broader range. There were some significant difference in pH, temperature stability and kinetic parameters. The presence of high starch concentration resulted in higher thermostability in recombinant enzyme than the wild type. The recombinant enzyme was more stable at higher temperature and lower pH, with lower $K_m$ for coupling reaction using cellobiose and cyclodextrins as substrates.

A new treatment system for animal waste water has been developed as an alternative to the activated sludge process. It consists of two treatments; one is operated with 7 tanks, and the other is soil and plant cultivation bed. Aerobic microorganisms are added to the influent water in the tanks where the water is aerated so that the microbes utilize the pollutants, while sedimentation removes the indigestible solids. In the secondary treatment the water, which has already received a primary treatment, is filtered through soil where it also receives treatment by soil organisms. In addition there is transpiration of water and absorption of minerals by plants. In the primary treatment BOD, SS, coliforms (E. coli), TP and total bacteria were removed 79-99%, but COD and TN were removed only 58% and 36%, respectively. In the secondary treatment removal of nutrients proceeded further, and 93-99% of pollutants were removed. The treated waters met the quality standard of discharge water in Japan except for TN, which was in too great a concentration to meet discharge standards. This problem requires further study.
Total of 2,643 Korean soldiers were examined of their stool for parasitic infections by both cellophane thick smear and formalin-ether concentration techniques from August, 1983 to December, 1985. Out of them, 73.6% were free from any parasite, 22.6% were ova positive and 4.0% cyst positive. The ova positive rates by species were Ascaris lumbricoides 2.0%, Trichuris trichiura 13.0%, hookworm and Trichostrongylus orientalis 0.08% respectively, Clonorchis sinensis 7.6%, Metagonimus yokogawai 1.1%, Paragonimus westermani 0.08%, Echinostoma hortense 0.04%, Fibricola seoulensis 0.9%, Taenia sp. 0.3%, Hymenolepis nana 0.2% and H. diminuta 0.04%. Most of them (87.3%) were positive by a species, 11.9% by two and 0.8% by 3 species. The cyst positive rates were Entamoeba histolytica 0.6%, E. coli 1.4%, Endolimax nana 1.7%, Iodoamoeba butschlii 0.04% and Giardia lamblia 0.9%. Among the cyst positives, 89.5% were positive by a species, 7.6% by two and 0.5% by 3 species. The intestinal parasite infections among the Korean soldiers decreased distinctly compared with previous data.
