치과용 인상재에서의 클로르헥시딘과 에센셜 오일의 항균성능에 대한 상승효과 (Synergy Effect of Chlorhexidine and Essential Oils on Antimicrobial Activity in Dental Impression Materials)

  • There is growing concern about cross infection among the patients to patients, patients to staffs, and tools to patients in healthcare facilities, especially in dentistry. In this study, the most widely used dental impression materials were prepared and the synergy effect of Chlorhexidine and essential oil on antimicrobial activity was examined in the impression materials. Chlorhexidine concentration of 0.1 wt% and 0.5 wt% showed no antimicrobial activity on Escherichia coli (E. coli) and Candida albicans. At 1.0 wt% Chlorhexidine, 0% of E. coli and 34.7% of Candida albicans were survived. Bergamot (Essential oil) concentration of 0.5 wt% and 1.0 wt% showed no antimicrobial activity on E. coli. At 2.0 wt% Bergamot oil, 71.9% of E. coli were survived. Tea tree oil (Essential oil) of 0.5 wt% showed no antimicrobial activity on E. coli. At 1.0 wt% Tea tree oil, 11.2% of E. coli was survived. At 2.0 wt% Tea tree oil, no E. coli was survived. However, no E. coli was survived at the concentration of 0.8 wt% Bergamot with 0.3 wt% Chlorhexidine. At the concentration of 0.8 wt% Tea Tree oil with 0.3 wt% Chlorhexidine, 1.3% of E. coli were survived. The experimental results showed that the synergy effects between Chlorhexidine and essential oils on antimicrobial activity were prominent.

Production of Poly(3-hydroxybutyrate) [P(3HB)] with High P(3HB) Content by Recombinant Escherichia coli Harboring the Alcaligenes latus P(3HB) Biosynthesis Genes and the E. coli ftsZ Gene

  • Filamentation-suppressed recombinant Escherichia coli strain harboring the Alcaligenes latus polyhydroxyalkanoate (PHA) biosynthesis genes and the E. coli ftsZ gene was constructed and cultivated for the production of poly(3-hydroxybutyrate) [P(3HB)] with high concentration and high content. By the pH-stat fed-batch culture of this recombinant E. coli strain XL1-Blue(pJC5), the final cell concentration and P(3HB) concentration obtained in 44.25h were 172.2g cell dry weight/l and 141.9g P(3HB)/l, respectively, resulting in productivity of 3.21g P(3HB)/l-h. More importantly, the P(3HB) content obtained was 82.4 wt %, which was significantly higher than that obtained with the recombinant E. coli harboring only the PHA biosynthesis genes.

저온저장중 Allspice(Pimenta dioica L.)에 의한 Escherichia coli 0157:H7과 Staphylococcus aureus 196E의 생존억제 (Inhibition of Escherichia coli 0157:H7 and Staphylococcus aureus 196E by allspice (Pimenta dioica L.) during cold storage)

  • 향신료인 allspice(Pimenta dioica L.)를 액체배지에 첨가하여 2종류의 식중독세균(Escherichia coli 0157:H7과 Staphylococcus aureus 196E)의 증식과 저온저장중 생존에 미치는 항균효과를 조사하였다 저농도(0~2%, w/v)의 allspice를 함유한 tryptic soy broth(TSB)에 E. coli와 S. aureus를 $10^{5}$~$10^{6}$ cells/$m\ell$가 되게 접종하여 35$^{\circ}C$에서의 증식과 냉장(5$^{\circ}C$)및 냉동(-2$0^{\circ}C$)저장중 생존억제효과를 생균수의 변화로서 조사하였다. 35$^{\circ}C$에서의 E. coli 증식은 1%이내의 향신료 농도에서는 증식이 저해되지 않았으나, 2%의 allspice 존재하에서는 긴 유도기를 거친 후에 증식이 시작되었다. 35$^{\circ}C$에서의 S. aureus의 증식은 0.1%와 0.3%의 allspice 존재하에서는 긴 유도기를 거친 후에 증식이 시작되었으나 0.5~2%에서는 세균의 증식이 억제되어 사멸하였다. 5$^{\circ}C$에서 냉장한 경우에는 E. coli와 S aureus 모두 0.3%의 allspice에 의해 생존이 크게 억제되었으며 0.5%이상의 농도에서는 저장말기에 사멸하였다. -2$0^{\circ}C$에 동결저장하였을 때 E. coli는 저장기간이 길어질수록, 첨가한 향신료의 농도가 높을수록 생존 억제효과가 증대되었다. 동결저장중 S. aureus의 생균수는 저장초기에 급격히 감소한 후 거의 일정수준을 유지하였으며 첨가한 향신료의 농도에 따른 생균수의 차이는 나타나지 않았다. 동결저장에서 0.1%의 향신료를 첨가했을 때 E. coli와 S. aureus는 대조구의 생균수에 비해 1/100이하로 생균수가 감소하여 allspice는 우수한 항균효과를 나타내었다.다.

  • The cellabiase (${\beta}$-glucosidase) gene in a Cellulomonas sp. CS1-1 was cloned into E. coli HB101 using the vector plasmid pBR322, and the expression of the gene in E. coli studied. The chromosomal DNA of the cellulomonas was digested by seveal restriction enzymes, each of which has only one cleaving site in plasmid pBR322. The recombinant plasmid, pSB2, created with Sal I frament, was expressed for the cellobiase gene in E. coli. The recombiant plasmid was estimated to contain 6.4 Kb foreign DNA at the Sal I site of plasmid pBR322 and the inserted DNA was mapped by single and double digestion with several enzymes. E. coli HB101(pSB2) has slowly grown in a mineral liquid medium containing cellobiose as a sole carbon source. The cellobiase activity in the transformed E. coli was 132 units per liter, which is equivalent to one twenty fifth of that in doner strain Cellulomonas sp. CS1-1. The transforned cell with plasmid containing cellulase gene grow well in the LB mediuns. The synthesis of cellobiase in the strain, E. coli HB101 (pSB2), was inhibited by glucose and at high concentration of cellobiose, and induced by cellobiose at low concentration.

마늘 물추출물의 Escherichia coli O157:H7, Salmonella typhimurium, Staphylococcus aureus에 대한 항균활성 (Antibacterial Activity of Aqueous Garlic Extract Against Escherichia coli O157:H7, Salmonella typhimurium and Staphylococcus aureus)

  • 본 연구는 마늘의 물추출물(AGE)의 E. coli O157:H7, S. typhimurium, 그리고 S. aureus에 대한 항균효과를 조사하였다. AGE의 E. coli O157:H7, S. typhimurium, and S. aureus에 대한 최소억제농도(MIC)는 각각 24, 48 그리고 24 mg/mL이었으며, 최소살균농도(MBC)는 모든 균에 대하여 96 mg/mL이었다. AGE 24 mg/mL을 E. coli O157:H7에 투여하고 배양 24시간 후에, 균의 증식이 무처리 대조군과 비교하여 유의성 있게 감소하였다(p < 0.01). 그러나 AGE 24 mg/mL을 S. aureus에 투여하고 배양 24시간 후에, 대조군과 비교하여 유의성 있는 균의 증식억제 효과가 나타나지 않았으나, AGE 96 mg/mL을 투여한 경우에서는 배양 24시간 후에 대조군과 비교하여 유의성 있는 균의 증식억제가 나타났다(p < 0.01). AGE 48 (p < 0.05)과 96 mg/mL (p < 0.001)을 S. typhimurim에 투여하고 24시간 배양 후에, 대조군과 비교하여 통계적으로 유의성 있는 균의 증식억제 효과가 나타났다. 본 연구의 결과로부터, 마늘의 물추출물은 항생제와 화학적 식품보존제의 대체제로서 사용이 가능할 것으로 사료된다.

Phloretin Protects Macrophages from E. coli-Induced Inflammation through the TLR4 Signaling Pathway

  • Macrophages are the cells of the first-line defense system, which protect the body from foreign invaders such as bacteria. However, Gram-negative bacteria have always been the major challenge for macrophages due to the presence of lipopolysaccharides on their outer cell membrane. In the present study, we evaluated the effect of phloretin, a flavonoid commonly found in apple, on the protection of macrophages from Escherichia coli infection. RAW 264.7 cells infected with standard E. coli, or virulent E. coli K1 strain were treated with phloretin in a dose-dependent manner to examine its efficacy in protection of macrophages. Our results revealed that phloretin treatment reduced the production of nitric oxide (NO) and generation of reactive oxygen species along with reducing the secretion of proinflammatory cytokines induced by the E. coli and E. coli K1 strains in a concentration-dependent manner. Additionally, treatment of phloretin downregulated the expression of E. coli-induced major inflammatory markers i.e. cyclooxygenase-2 (COX-2) and hemeoxygenase-1 (HO-1), in a concentration dependent manner. Moreover, the TLR4-mediated NF-κB pathway was activated in E. coli-infected macrophages but was potentially downregulated by phloretin at the transcriptional and translational levels. Collectively, our data suggest that phloretin treatment protects macrophages from infection of virulent E. coli K1 strain by downregulating the TLR4-mediated signaling pathway and inhibiting NO and cytokine production, eventually protecting macrophages from E. coli-induced inflammation.

대장균의 실관투석배양 (Hollow Fiber Dialysis Culture of E. coli)

  • 설관막 장치를 이용하여 대장균의 투석배양을 수 행했다. 투석액으로 배지 내의 초산을 제거함으로써 초산의 저해효과를 경감할 수 있었다. 초산 생성속도는 포도당과 용존산소농도에 대단히 민감하였고, 따라서 막을 통한 포도당의 투과속도는 산소공급속도와 균형을 유지해야 했다. 막을 통해 포도당이 천 천히 공급될 때, 대장균의 비성장속도는 포도당 투 과속도에 좌우되었고 초산의 생성은 억제되였다.

Linear Correlation between Online Capacitance and Offline Biomass Measurement up to High Cell Densities in Escherichia coli Fermentations in a Pilot-Scale Pressurized Bioreactor

  • To yield high concentrations of protein expressed by genetically modified Escherichia coli, it is important that the bacterial strains are cultivated to high cell density in industrial bioprocesses. Since the expressed target protein is mostly accumulated inside the E. coli cells, the cellular product formation can be directly correlated to the bacterial biomass concentration. The typical way to determine this concentration is to sample offline. Such manual sampling, however, wastes time and is not efficient for acquiring direct feedback to control a fedbatch fermentation. An E. coli K12-derived strain was cultivated to high cell density in a pressurized stirred bioreactor on a pilot scale, by detecting biomass concentration online using a capacitance probe. This E. coli strain was grown in pure minimal medium using two carbon sources (glucose and glycerol). By applying exponential feeding profiles corresponding to a constant specific growth rate, the E. coli culture grew under carbon-limited conditions to minimize overflow metabolites. A high linearity was found between capacitance and biomass concentration, whereby up to 85 g/L dry cell weight was measured. To validate the viability of the culture, the oxygen transfer rate (OTR) was determined online, yielding maximum values of 0.69 mol/l/h and 0.98mol/l/h by using glucose and glycerol as carbon sources, respectively. Consequently, online monitoring of biomass using a capacitance probe provides direct and fast information about the viable E. coli biomass generated under aerobic fermentation conditions at elevated headspace pressures.

  • There is an increasing incidence in health problems related to environmental issues that originate from inadequate treatment of sewage. This has compelled scientists to engage in innovative technologies to achieve a effective disinfection process. Electrolysis has emerged as one of the more feasible alternatives to conventional disinfection process. The objectives of the present paper were to investigate the effect of chemical characteristics on oxidant formation and Escherichia coli (E. coli) disinfection in synthetic sewage effluents. The influence of parameters such as COD, SS, T-N and T-P were investigated using laboratory scale batch reactor. The results showed that the higher COD, T-N and T-P concentration, the lower N, N-Dimethyl-4-nitrosoaniline (RNO, indicator of the generation of OH radical) degradation and E. coli disinfection was observed. The order of effect of RNO degradation and E. coli disinfection was T-P > COD > T-N > SS. When 4 parameter of water quality were worked simultaneously, oxidants formation and disinfection was decreased with increase of the concentration of sewage. To increase of the disinfection performance, the increase of disinfection time or electric power was need.

하구퇴적토 환경에서 E.coli의 부착에 영향을 주는 물리화학적 요인 (Physicochemical factors affecting the adsorption of E. coli in estuarine sediments)

  • The higher bacterial numbers on clay than on sand were caused by different environmental factors. Such factors affecting the adsorption of E. coli ATCC 11775 in the sediment as follows; optimal pH range for the adsorption of E. coli ATCC 11775 was pH 7.5-pH 9.5. E. coli ATCC 11775 were shown maxima in the salinity of 18.$75%_{o}$ on sand type sediment and $12.5%_{o}$ on clay type sediment. Bacteria attached better to clay typed sediment than to sand typed sediment when organic substance was eliminated. Beef extract of 0.5%-1% concentration was found to promote the attachment of E. coli ATCC 11775 effectively. Peptone of 0.5% was enganced the attachment on the clay, and peptone of 1.3%-5%, on the sand. E. coli ATCC 11775 was found to adsorb onto benthonite with the highest efficiency and on celite with the lowest efficiency. Efficiency of adsorption by inorganic ions was shown due to higher values of ion. Adsorption was achieved in the order of $Al^{3+}, Ca^{2+}, Na^{+}$.
