경기지역 목장수의 위생세균학적 분포 (Bacterial Sanitation of Farm Water in Kyunggi Province)

  • Hygienic condition of farm waters has an effect on the hygiene of dairy foods. So we examined Standard Plate Count, coliform, E. coli, heat-resistant bacteria, psychrophilic bacteria, Pseudomonas aeruginosa and enterococcus for the bacterial sanitation of 78 farm waters in Kyunggi Province. Of the 78 farm waters, the average number of psychrophilic bacteria was 750 $\pm$ 170/ml, SPC 440 $\pm$ 130/ml, Pseudomonas aeruginosa 130 $\pm$ 97/100/ml, Coliform 22 $\pm$ 17/ml, E. coli 10 $\pm$ 6/100/ml, Heatresistant bacteria 5 $\pm$ 1ml, and Enterococcus 2 $\pm$ 1/100/ml. The percent of over than 1000/ml in SPC and psychrophilic bacteria of 78 farm water was 11.5% and 23.1%, respectively. The rate of over than 10/ml in coliform and heat-resistant bacteria was 12.8% and 15.4%, respectively and the rate of over than 10/100 ml in E. coli, Pseudomonas aeruginosa and enterococcus was 8.9%, 33.3%, and 2.6%, respectively. Pseudomonas aeruginosa was no significant between any other indicator organisms, and psychrophilic bacteria was significant with only SPC but other indicator organisms were highly significant with each other.

소와 돼지 도체 표면에 대한 미생물 오염도 (Microbiological quality on surfaces of beef and pork carcasses in Seoul)

  • It was conducted to evaluate the microbiological quality on the surface of slaughtered beef and pork products in Seoul from January 2003 to December 2003. Two hundreds four beefs and 284 pork carcasses were surveyed on generic E coli, total bacterial count for microbiological quality, and Salmonella spp as food-borne pathogen. The prevalence of the excellent or good grade(10$^4$ CFU/$\textrm{cm}^2$ in SPC) of beef and pork carcasses were 99.7% and 97.9%, respectively. The frequency of beef carcasses with less than 10$^2$ CFU/$\textrm{cm}^2$ of E coli was 100%, while that of pork carcasses was 98.2%. Salmonella spp was not recovered from all of beef and pork carcasses.

상추와 생산환경의 미생물 안전성 평가 (Evaluation of Microbiological Safety of Lettuce and Cultivation Area)

  • 본 연구는 상추를 대상으로 재배~수확 후 단계의 미생물학적 안전성을 평가하고자 이천지역 상추농가와 수확 후 처리시설에서 위생지표세균(일반세균수, 대장균군,E. coli)과 병원성미생물(Escherichia coli O157:H7, Salmonella spp., Stphylococcus aureus, Listeria monocytogenes, Bacillus cereus)을 조사하였다. 상추 생산과정의 미생물 오염도 조사 결과, 대부분의 시료에서 일반세균수 수준은 4.0 log CFU/(ml, g, $100\;cm^2$, hand)이었다. 또한 수확 후 처리시설에서 사용되는 장갑에서 E. coli가 3.5 log CFU/hand로 비교적 높게 검출되었고, B. cereus는 토양과 저울에서 각각 3.5 log CFU/g, 2.6 log CFU/$100\;cm^2$로 타시료에 비해 높았다. 특히 정식 후 10일 째 상추의 일반세균수, 대장균군수 및 B. cereus의 오염수준이 정식 후 62일째 상추 보다 각각 l.19, 1.87, and 1.94 log CFU/g 정도 높은 것으로 나타났다. 한편 E. coli O157:H7, Salmonella spp, 및 L. monocytogenes은 검출되지 않았다. 본 연구의 결과를 통하여 상추의 유해 미생물 오염은 생산단계부터 발생되고 있음이 확인되었고 이러한 유해미생물의 오염을 사전에 관리하기 위해서는 생산에서부터 소비까지 위해요소를 관리하는 시스템의 도입이 필요하다.

일부 공중화장실 위생설비의 오염지표세균 조사 (The Load of Indicator Bacteria of Sanitary Ware in Public Restrooms)

  • Objective: The purpose of this study was to investigate the occurrence of indicator organisms in essential sanitary ware in public restrooms. Methods: Twelve public restrooms were randomly selected. Samples at three locations (toilet seat, toilet flush handle/lever, and faucet handle) in the restrooms were collected with moistened-sterile cotton swabs and applied to media in order to determine aerobic colony count (ACC), total coliforms, and Escherichia coli. Results: Most of the samples taken in summer showed a higher level of ACC than those taken in winter (p<0.05). Female restrooms showed higher ACC levels on faucet handles and male restrooms on toilet flush handles/levers (p<0.05). Overall, faucet handles contained the greatest level of ACC, followed by toilet seats, whereas the least load was found on toilet flush handles/levers. The ACC level of samples in the restrooms in public parks, subway stations, and educational institutions varied. Total coliforms were identified in about 20% of toilet seats and faucet handles in male restrooms and faucet handles in female restrooms in summer. These locations were also the sites of positive results of E. coli isolation. Conclusions: The public restrooms were significantly more contaminated in summer than in winter. Overall, the most contaminated locations in the restrooms were toilet seats in male restrooms, and faucet handles in female restrooms. Poor hygienic status was indicated by the positive results of total coliforms and E. coli on samples from some sites. Therefore, sanitary control of restrooms should be improved. These results should be confirmed in a larger study that includes more public restrooms.

  • Bioluminescence방법에 의하여 세균내에 존재하고 있는 ATP를 추출하여 그양을 정량분석하므로서 제육표면에 생존하고 있는 세균수를 신속하게 측정하는데 응용될 수 있는 가능성을 조사하기 위하여 본 실험을 실시하였으며, 그 결과는 1. Luciferin-luciferase에 작용하는 ATP농도가 $10^{-10}M$부터 $10^{-6}M$까지 증가함에 따라 발생된 빛의 양도 비례적으로 증가했다. 2 . 계육에서 분리동정된 부패균인 E. coli, H, alvei, P. putida, A. hydrophila에서 추출된 ATP 농도와 세균수와의 관계는 거의 일직선으로 나타난다. 3. E. coli에 있는 ATP양은 생존중 거의 변화하지 않는 것으로 나타났다. 4 계육의 저장기간동안 증식된 세균수에 따라 ATP 농도도 비례 적으로 증가되며 이때 상판계수(r)는 0.95로 나타났다. 5. 계육에 오염된 세균수와 ATP 농도와의 관계에서 볼 때 ATP농도가 $10^{-7}M$부근에서부터 부패가 시작된다고 사료된다. 6. 개발된 방법에 의하여 계육표면에 존재하는 세균의 ATP 농도를 측정하여 세균수를 간접적으로 신속히 ( 1시간내) 조사할 수 있다.

Microbial contamination including Vibrio cholerae in fishery auction markets in West Sea, South Korea

  • Background: The monitoring of pathogens of fishery auction markets is important to obtain safe fishery products regarding hygiene and sanitation. In this study, aerobic, coliform, Escherichia coli, and Vibrio cholerae were monitored in the fishery products and environmental samples obtained from fishery auction markets. Methods: The fishery products (flounder, octopus, skate, rock cod, sea bass, snail, monkfish, flatfish, comb pen shell, corb shell, conger eel, hairtail, croaker, and pilchard) were placed in filter bags, and the environmental samples (samples from the water tanks at the fishery auction markets, seawater from the fishery distribution vehicles, ice from wooden or plastic boxes, and surface samples from wooden and plastic boxes used for fish storage) were collected. Aerobic bacteria, E. coli, and coliform in the samples were enumerated on aerobic count plates and E. coli/coliform count plates, respectively. For V. cholerae O1 and V. cholerae non-O1 quantification, most probable number (MPN)-PCR analysis was performed. Results: Aerobic and coliform bacteria were detected in most samples, but E. coli was not detected. Wooden boxes were contaminated with high levels of aerobic and coliform bacteria in all seasons (spring, summer, and fall). During fall, V. cholerae non-O1 were detected in snails, hairtails, croakers, flatfishes, pilchards, plastic boxes, and water samples. Conclusions: These results indicate an increased prevalence of V. cholerae contamination in fishery products in fall, including food contact samples, which can be vehicles for cross-contamination.

소.돼지 도체표면의 미생물학적 고찰 (Survey on the microbiological quality of meat in Seoul)

  • This survey was conducted to evaluate the microbiological quality of raw beef and pork products from January to December in 1999. A total of 107 beef and 157 hog carcasses were collected from two abattoirs located in Seoul. The result showed that beef carcasses had an average bacterial loading around 139,000 bacteria/$\textrm{cm}^2$ of carcass surface, indicating a little bit higher count than the results reported in USA and Australian meat. However, overall hygienic status was found to be acceptable for all examined carcasses because 84.4% of product rated excellent, good or acceptable comparable to USA of 91.6% and Australia of 88%. The analysis of data on overnight-chilled to weekend-chilled carcasses indicated that the microbiological growth occurred in the chiller during the weekend chill with increases in total viable count from 130,000cfu/$\textrm{cm}^2$ to 400,000cfu/$\textrm{cm}^2$. Qualitative testing for escherichia coli, EC + MUG was used as a most probable number (MPN) method along with the petrifilm method. The average of MPN/$\textrm{cm}^2$ of E coli biotype 1 was 29MPN/$\textrm{cm}^2$ for beef carcasses and 1,100 MPN/$\textrm{cm}^2$ for hog carcasses, respectively. However, 41% of beef and 16.3% of hog carcasses were shown to be less than < 3 MPN/$\textrm{cm}^2$ in E coli biotype 1 examination. Although salmonella enteritis, S typhimurium and E coli O157:H7 were all negative, listeria monocytogenes was recovered from only one hog surface samples of the 89 carcasses tested.

  • Estrous and non-estrous rabbits were inoculated with E. coli or Streptococcus pyogenes, and the mixture of the two organisms, and bacterial count and histopathological studies of uterine horns were made to observe the effects of estrogen on the resistance of the uterus to bacterial infection. The results obtained were summarized as followings; 1. Four hours after inoculation of bactoria into uterine horn, the number of organisms was significantly lower in estrous rabbits than in non-estrous regardless of the kind of organisms inoculated. 2. The highest reduction rate of the organisms among the three bacterial inoculation groups was found in estrous rabbits inoculated with E. coli, and the lowest reduction rate was with Streptococcus pyogenes. 3. Histopathological changes of uterine horns induced five days after bacterial inoculation were observed. In estrous rabbits, a mild inflammatory reaction was found in Streptococcus pyogenes group, but a slight inflammatory reaction and only a negligible inflammatory reaction were observed in mixed bacteria group, and in E. coli group respectively. In non-estrous group, however, a marked inflammatory reaction was observed in Streptococcus pyogenes group, a moderate inflammatory reaction and a slight inflammatory reaction were observed in the mixed bacterial group and E. coli group, respectively.

  • 천연고분자인 chitin의 탈아세틸화물인 chitosan의 유리아미노기에 알킬 길이가 다른 알데히드를 이용하여 N-alkyl chitosan 유도체를 각각 합성한 다음, 이들을 methyl iodide로 4차화시킴으로서 4차 암모늄기가 도입된 새로운 수용성 chitosan 유도체를 합성하여, 도입된 알킬기의 길이에 따른 항균성을 그람양성균인 S. aureus(ATCC 6538P)와 그람 음성균인 E. coli(ATCC 14339)에 대해서 shake flask법을 이용하여 검토하였다. 그 결과 소수성이 증대될수록 항균력은 증가하였고, 특히 그람음성균인 E. coli에 비해 그람양성균인 S. aureus에 대한 항균력이 더욱 증대됨을 확인할 수 있었다. 이는 cytoplasmic membrane의 구조상의 차이에서 기인한 것으로 생각된다.

시판 즉석섭취 샌드위치류의 미생물학적 위해분석과 HACCP 적용 후 품질개선 효과 (Microbiological Hazard Analysis of Ready-to-eat Sandwiches and Quality Improvement Effect by Implementing HACCP)

  • This study was performed to analyze the microbiological hazards of ready-to eat sandwiches and to evaluate the effect of implementing a hazard analysis and critical control points (HACCP) for quality improvement of ready-to-eat sandwiches. In total, 174 samples were examined for the microbiological hazard analysis. About 37.4% of the samples had an aerobic plate count ${\geq}$6 log CFU/g, and the numbers of coliforms exceeded 3 log CFU/g in 82.2% of the samples. Additionally, Staphylococcus aureus, E. coli, and Salmonella spp. were detected in 10.3%, 4.6%, 1.1% of sandwiches, respectively. In contrast, E. coli O157:H7, and Listeria monocytogenes were not detected in any of the samples. The results of the microbiological verification after implementing a HACCP throughout the production process indicated that S. aureus, E. coli, Salmonella spp., E. coli O157:H7, and L. monocytogenes were not detected in any of the samples. Consequently, implementing a HACCP is effective for quality improvement of ready-to-eat sandwiches.
