We seasonally examined marine macroalgal community structures in the subtidal zones at Gapado Island, Jeju, Korea, from February 2013 to November 2019. A total of 164 macroalgal species were identified, including 15 green, 40 brown, and 109 red algae. Species richness ranged from 54-106 species, with the maximum observed in 2013 and the minimum observed in 2015. Cladophora wrightiana var. minor, Ecklonia cava, Amphiroa anceps, Corallina aberrans, Corallina crassisima, Synarthrophyton chejuense, Sonderophycus capensis and Plocamium telfairiae occurred in all seasons. The average annual seaweed biomass was 1,258.22 g wet wt./m2, ranging from 917.51 g/m2 in 2017 to 1,551.95 g/m2 in 2014. E. cava was the most dominant species, accounting for 46.57% of the total seaweed biomass. The subdominant species were P. telfairiae and A. anceps, comprising 8.64% (108.75 g/m2) and 6.65% (83.61 g/m2) of the total biomass, respectively. The vertical distribution of subtidal seaweeds were represented by E. cava and P. telfairiae at 5-20 m, A. anceps at 10-20 m, C. aberrans and C. crassisima at 5 m and C. wrightiana var. minor at 10 m. Cluster analysis revealed three distinct groups: group A (2014-2018), group B (2019) and group C (2013), indicating significant differences in the annual seaweed community.
In the course of study for the development of functional food ingredients from seaweeds having useful biological activities, the in-vitro antioxidant and nitrite scavenging activities of the methanol extracts prepared from 35 different seaweeds (17 phaeophyta, 11 rhodophyta and 7 chlorophyta) were determined. At $500\;{\mu}g/ml$ concentration of the methanol extracts, Ecklonia cava, Ecklonia stolonifera, Eisenia bicyclis (Kjellman) Setchell, Ishige foliacea, I. okamurai, Sargassum confusum, S. fulvellum, S. yamade and Zostera marina showed 60% more DPPH and ABTS scavenging activity. The exceptions were found in I. okamurai and Z. marina, which showed 45% and 53% ABTS scavenging activity, respectively. The correlation coefficient between DPPH and ABTS scavenging activities was 0.855, suggesting that the 9 different seaweeds extracts could scavenge anion and cation radicals concurrently. In reducing power, only E.cava, E.stolonifera and E.bicyclis showed above 0.88 ($Abs_{700}$). In a while, in nitrite scavenging activity at $100\;{\mu}g/ml$ concentration of the methanol extracts, 19 different seaweeds extracts including I. foliacea, I. okamurai, S. confusum, S. fulvellum, and S. yamade showed 60% more activity. Calculation of $IC_{50}s$ of DPPH, ABTS and nitrite scavenging activities of 9 different seaweeds extracts further showed that I. foliacea and S. yamade, besides E.cava, E.stolonifera and E.bicyclis, have strong antioxidant and nitrite scavenging activity. These results suggest that the selected 9 different seaweeds could be developed as functional food ingredients and I. foliacea and S. yamade have potential as novel natural sources of antioxidant and nitrite scavenger.
To analyze nucleotide sequence encoding internal transcribed spacer (ITS) regions specific to the Laminariaceae family, genomic DNA was isolated from six brown algae species distributed along the east coast of Korea. These included three species from the Laminariaceae family (Agarum clathratum Dumortier, Costaria costata [C. Agardh] Saunders, and Saccharina japonica Areschoug) and two species from the Alariaceae family (Undaria pinnatifida [Harvey] Suringer and Ecklonia cava Kjellman), both in the order Laminariales, and one species from the family Sargassaceae in the order Fucales (Sargassum serratifolium). Based on a sequence analysis of ITS-1 and ITS-2 for A. clathratum, C. costata, and E. cava, oligonucleotides were designed from the regions that showed sequence conservation in Laminariaceae. Following polymerase chain reaction using three sets of primers, amplification of ITS-1 and ITS-2 was detected in reactions using genomic DNA isolated from the species belonging to Laminariaceae, but not from the species belonging to the other families. The results indicate that this method can be used for the detection and identification of Laminariaceae species.

Marine algal flora and community structure were investigated seasonally at three sites in subtidal zone of Wangdol-cho on the east coast of Korea from May 2012 to August 2013. A total of 122 species were collected and identified, including 12 green algae, 29 brown algae, and 81 red algae. Of these, 38 species were found throughout the survey period. Average seaweed biomass was 472.19-1,198.77 g wet wt. $m^{-2}$ in spring, 68.99-631.14 g wet wt. $m^{-2}$ in summer, 200.91-401.20 g wet wt. $m^{-2}$ in autumn, and 53.61-922.32 g wet wt. $m^{-2}$ in winter. The vertical distribution based on biomass were Grateloupia lanceolata, Acrosorium venulosum, Lomentaria catenata - Undaria pinnatifida, Dictyopteris pacifica, Sargassum horneri, Ecklonia cava - Desmarestia viridis, E. cava, S. horneri from upper to lower subtidal zone. The flora was classified into six functional groups: filamentous form (34.43%), coarsely branched form (25.41%), sheet form (24.59%), thick leather form (9.71%), crustose form (5.74%) and jointed calcareous form (0.82%). The C/P, R/P and (R+C)/P values reflecting the flora characteristics were 0.41, 2.79 and 3.21, respectively. Also, diversity index (H') and dominance index (DI) indicate that the algal community and environmental condition of Wangdol-cho is stable. We recommend that Wangdol-cho subtidal zone should be more protected from human activities such as turbulence and eutrophication in order to maintain species diversity and abundance of algae.

Ecklonia cava Kjellman, which has recently gained popularity due to the spread of farming techniques, is expected to be developed in various varieties in the future. There exist increased needs for research on the basis of natural populations and inter-regional morphological variations. We compared the morphology of the aquaculture and natural populations from 16 coastal areas in Korea. The 18 traits found suitable for distinguishing varieties were selected from 14 measurement traits and 4 ratios related to the main morphology and characteristics of primary blade, secondary blade, and stipe. In the cluster analysis, Janggil (E4) and Sorok (S7) showed significant differences from those of the same coastal region. Two groups, including Suyou (Q6, Q8, and Q10) which was the second year of farming, of the rest of the populations from East sea and southern coast were distinguished. Three populations of Jeju were divided into a regional group. In the principal component analysis (PCA), a large number of populations from East sea and Southern coast appeared in the center with aquaculture populations. PC1 and PC2 associated with traits of secondary blade index, stipe length and diameter, stipe length/primary blade length, primary blade length and width, secondary blade number, secondary blade length and width, divided E4, S7 and populations of Jeju region. As a result, the 18 characters of this study were found to be useful as criteria for discrimination of populations with significant differences in each coastal region, and these populations were expected to be candidates for new varieties.

The seasonal and vertical biomass of marine forest seaweeds were examined to select a suitable species at 12 sites of the South Sea in Korea between 2018 and 2019. The Habitat Suitability Index (HSI) was also calculated in terms of biomass for six species (three kelp and three Sargassum species). A total of 16 marine forest-forming species including four kelp and 12 Sargassum species were observed at the 12 sites. The average annual seaweed biomass by season and depth ranged from 843.73-2,925.85 g wet wt. m-2 at the eastern South Sea and from 343.87-4,580.10 g wet wt. m-2 at the western South Sea. In the kelp species, the Ecklonia cava biomass was predominant, followed by E. stolonifera. The macroalgal species with the greatest biomass was Sargassum macrocarpum, followed by S. horneri. The HSI values of E. stolonifera were between 0.76-1.0 at eight sites and those of E. cava were 0.58-0.92 at four sites, indicating that E. stolonifera was more suitable than E. cava. In the HSI values of the Sargassum species, S. horneri ranged between 0.84-1.0 at all 12 sites and the S. macrocarpum values were between 0.68-0.99. The results indicate that E. cava and S. macrocarpum were the most suitable for the marine forest construction in terms of the seaweed biomass, and E. stolonifera and S. horneri in terms of the HSI values. Thus, we suggest that seaweed biomass and HSI values should be considered when choosing suitable forest-forming species.

Neurodegenerative disorders in the elderly are characterized by gradual loss of memory and cognitive function. Oxidative stress caused by reactive oxygen species is associated with progressive neuronal cell damage and death in Alzheimer's disease, one of the most common neurodegenerative disorders. An edible brown seaweed, Ecklonia cava, contains a variety of biologically active compounds such as phlorotannins. In this study, we comparatively evaluated the total phenolic content, antioxidant capacity, and neuroprotective effects of the phlorotannin-rich extract from E. cava (PEEC). The total phenolic content of PEEC and dieckol was 810.8 mg gallic acid equivalents (GAE)/g and 996.6 mg GAE/g, respectively. Antioxidant capacity of PEEC was 1,233.8 mg vitamin C equivalents (VCE)/g and 392.1 mg VCE/g determined using ABTS and DPPH assays, respectively, while those of dieckol were 2,238.4 mg VCE/g and 817.7 mg VCE/g. High-performance liquid chromatography results revealed 48.08 ± 0.67 mg dieckol/g of PEEC. PEEC had neuroprotective effects in pheochromocytoma (PC-12) and human neuroblastoma (SH-SY5Y) cells against H2O2- and AAPH-induced oxidative damage, partly due to reduced intracellular oxidative stress. PEEC treatment inhibited acetylcholinesterase and butyrylcholinesterase in a dose-dependent manner. Taken together, these findings suggest that PEEC is a good source of antioxidants and neuroprotective materials.

This research was done to verify the effect of Ecklonia cava water extracts (ECWE) on inhibition of allergic reactions using ovalbumin (OVA)-immunized food allergy mouse model. For in vitro test, $10\sim100{\mu}g$/mL of ECWE and OVA were added to splenocytes obtained from OVA-immunized mice. The significant reduction of IgE antibody level in culture supernatants of splenocytes was shown in ECWE adding group at all tested concentrations. In addition, ECWE decreased IL-4 and IFN-$\gamma$ levels in supernatants of splenocytes. To confirm the effect of ECWE in in vivo test, ECWE was injected to peritoneal cavity of OVA-immunized mice. Subsequently, IgE level was measured in serum and cultured supernatants of splenocytes. As a result, the injection of ECWE (5 and 10 mg/kg BW) significantly attenuated the secretion of IgE antibody in both serum and splenocytes. In conclusion, the present study indicates that ECWE could suppress in a food allergy mouse model through the inhibition of IgE secretion.

This study was conducted to investigate the antioxidant and acetylcholinesterase (AChE) inhibitory activities of extracts from various seaweed. The extracts of $Sargassum$$thunbergii$ (91.3%), $Polysiphonia$$morrowii$ (90.7%), $Ecklonia$$cava$ (89.9%), and $Artemisia$$fukudo$ (85.9%) showed over 80% high radical scavenging activities at the final concentration of 40 ${\mu}g$/mL. The $Artemisia$$fukudo$ extract showed the highest inhibition activity of 30.2% on AChE at the final concentration of 10 ${\mu}g$/mL. The extract of $Porphyra$$tenera$, $Costaria$$costata$, $Monostroma$$nitidum$, $Ecklonia$$cava$, and $Agarum$$clathratum$ against AChE at a concentration of 10 ${\mu}g$/mL exhibited inhibition of 26.6%, 25.3%, 23.4%, 21.7%, 20.4% and 19.9%, respectively. The bioautography results showed that the mixtures of structurally diverse compounds were thought to affect AChE inhibitory activity. These results suggest that extracts from seaweed with their high quality components may be effective in the prevention of Alzheimer's disease and may be used to develop various functional food products.
