Effects of Ato-tang on DNCB-induced atopic dermatitis in Mice (아토피피부염 동물 병태 모델에서 아토탕의 개선효과)

  • Objectives : The aim of this study is to investigate anti-atopic dermatitis effect using ato-tang.Methods : Ato-tang was external treatment to NC/Nga mice for 4 weeks, where atopic dermatitis was induced by DNCB at 1% and 0.4% for 3 weeks. Atopic dermatitis index score was measured using eye observation and picture evaluation. The histopathological change of dorsal skin was observed by hematoxylin and eosin (H&E) staining. Cytokines including IL-4, IL-5 and IL-13 were measured by Luminex or reverse transcriptase polymerase chain reaction (RT-PCR) analysis and immunoglobulin E (IgE) was measured by ELISA reader.Results : The dorsal skin of Ato-tang group showed decrease in erythema, pruritus, dry skin, edema, excoriation, erosion and lichenification level through naked eye observations. Immunoglobulin cell infiltration and the thickness of epidermis were significantly decreased in the dorsal skin compared to control. Production of Th2 cytokines (IL-4, IL-5, IL-13) and IgE level in serum were all significantly decreased, in comparison with control. In addition, mRNA expression level of Th2 cytokines (IL-4, IL-5, IL-13) in spleen was decreased, in comparison with control.Conclusion : The results indicated that external treatment of ato was improved skin barrier function in the symptoms of atopic dermatitis disease. Also, atopic dermatitis factors where cytokine as well as immunoglobulin E in serum and mRNA expression were decreased, respectively, in comparison with control. Therefore, we suggest that ato could be effectively used as a external therapeutic drug based on atopic dermatitis factors.

Novel Sporichthyaceae Bacterium Strain K-07 Skin Barrier, Moisturizing and Anti-inflammatory Activity (신규 Sporichthyaceae Bacterium Strain K-07 배양액의 피부장벽, 보습 및 항염증 활성)

  • The human skin is an ecosystem that provides habitat to various microorganisms. These comprise the skin microbiome and provide numerous benefits in addition to maintaining a symbiotic relation with the host. Various metabolites generated by the skin microbiome exert beneficial effects such as strengthening the skin barrier, and anti-aging and anti-inflammatory functions. In this study, we isolated a novel bacterium, designated Sporichthyacae strain K-07, from the human skin. Analysis of 16S rRNA gene sequences showed that the newly found bacterium shares 93.4% homology with the genus Sporichthya, thus corroborating the discovery of a novel genus. We further analyzed the effect of the novel strain in vitro, by treating HaCaT cells with bacterial metabolite products. Treatment resulted in changes in the mRNA expression levels of filaggrin, claudin1, claudin4, SMase, CERS3, HAS3, aquaporin3, IL-6, TNF-${\alpha}$, TSLP, and TARC. Specifically, the levels of filaggrin, claudin1, claudin4, SMase, CERS3, HAS3, and aquaporin3 were higher in strain K-07 metabolite product-treated cells than in control cells. These results showed that metabolite products of the novel strain K-07 enhanced the skin barrier and exert anti-inflammatory effects. Therefore, these metabolite products could be potentially used for treatment of skin conditions.

  • In this study, we investigated an inhibitory effect of AST cream on atopic dermatitis (AD) using a 2,4-dinitrochlorobenzene-induced AD murine model. Topical treatment with AST cream ameliorated the severity of AD-like lesional skin through decreases in infiltration of inflammatory cells and time of scratching behaviors. Also, AST cream reduced histamine and IgE levels in serum. The protein levels of IL-4 and IL-6 in AD-like lesional skin were suppressed by AST cream. These findings suggest that AST cream would be an alternative therapeutic agent for AD-like skin diseases.

Effect of Kami-chungsimyeunjatang on atopic dermatitis-like skin lesions induced in NC/Nga mice by mite antigen stimulation (가미청심연자탕(加味淸心蓮子場)이 NC/Nga mice의 아토피양(樣) 피부염에 미치는 영향)

  • Objectives : The purpose of this study is to examine of the effect of Kami-chungsimyeunjatang(KCSYJT) medicine on the atopy eruption control. Methods : The expression of IgE, IL-4, IL-6, $TNF-{\alpha}$, IgG2b, IgM, IgG2a and IgG1 level in serum, and $IFN-{\gamma}$ production by KCSYJT were analyzed. CD3e+/CD69+, CD4+/CD25+, B220+/IgE+ and B220+/CD23+ positive cells by flow cytometry in splenocytes were assayed and the revelation of CD3e+/CD69+, CD4+/CD8+ and CD4+/CD25+ marker in PBMC, spleen and DLN were observed. The outturn of IL-4, eotaxin 2, CCR3, TARC mRNA in splenocytes werw observed. We also analyzed NC/Nga mice's ear, DLN and neck-back skin after biopy and dye by H&E, and toluidine staining (mast cells marker) method, measured about epidermis and dermis part in comparison with control group. Results : NC/Nga mice suffered from dermatitis very similar to human AD with IgE hyperproduction. Specially, result that measure IgE content in serum on 8 weeks, 12 weeks, 16 weeks decreased remarkably than control group. After experiment end, result that observe revelation CD3e+/CD69+, CD4+/CD8+ and CD4+/CD25+ marker in PBMC, spleen and DLN establishment observed recover as normal with political background. And decreased than result control group which measure IL-4, IL-6, $TNF-{\alpha}$, IgG2b, IgM, IgG2a, IgG1 level in serum, and $IFN-{\gamma}$ production secreted in Th1 cell displayed increase by KCSYJT medicines. Ear thickness decrease than control group in result that observe effect that get in ear of a NC/Nga mouse. Course inflammation immunocyte etc.. permeated of result that effect that KCSYJT medicines get to NC/Nga mouse's skin establishment analyzes ear, DLN and neck-back skin after biopy, and dye by H&E, and toluidine staining (mast cells marker) method decreased about epidermis. and inflammation of dermis part remarkably than control group. Immunohistochemical examination of the skin lesion showed decrease by KCSYJT medicines on numbers of mast cells (CCR3) and CD4+ T cells containing IL-4 necessary for IgE. Conclusions : Th1 cell and Th2 cell was observed to be shift by secretion amount of IL-4 and $IFN-{\gamma}$ by KCSYJT medicines. Therefore, the KCSYJT medicine turned out to be useful in allergy autoimmune disease.

Simultaneous Determination of Prostaglandin E1 and Prostaglandin E1 Ethyl Ester in Hairless Mouse Skin Homogenate by High-Performance Liquid Chromatography

  • A rapid and specific high-performance liquid chromatographic method was developed and validated for the simultaneous determination of prostaglandin $E_{1}\;(PGE_{1})$ and prostaglandin $E_{1}$ ethyl ester $(PGE_{1}-EE)$ in hairless mouse skin homogenate. The sample treatment procedure involved deproteination and precipitation by acetonitrile. $PGE_{1}$ and $PGE_{1}-EE$ in supernatant were separated in a reversed-phase C18 column without being interfered by other components present in hairless mouse skin homogenate. 9-Anthracenecarboxylic acid was used as an internal standard. The retention times of $PGE_{1}$, 9-anthracenecarboxylic acid and $PGE_{1}-EE$ were, 4.5, 9.5 and 18.0 min, respectively. The assay showed linearity from 1 to $40\;{\mu}g/ml$ for both $PGE_{1}$ and $PGE_{1}-EE$. Precision expressed as RSD ranged from 2.3 to 14.1 % for $PGE_{1}$ and 1.6 to 11.0% for $PGE_{1}-EE$. Accuracy ranged from 100.5 to 119.6 % for $PGE_{1}$ and from 98.0 to 103.7% for $PGE_{1}-EE$. This method was employed successfully to follow the time course of concentrations of $PGE_{1}$ and $PGE_{1}-EE$ in hairless mouse skin homogenate for stability study.

IL-4 and $IFN-{\gamma}'$ gene revelation amount displayed marked decrease than control group in result that observe effect that get in skin of a NC/Nga mice's skin establishment analyzes neck-back skin after biposy, and dye by H&E method decreased about epidermis and inflammation of dermis part remarkable than control group. Conclusions : These results suggest that Th1 cell and Th2 cell observe to be shifted by secretion amount of IL-4 and $IFN-{\gamma}$ by KDKEJ extract could know that KDKEJ extract can use usefully in allergy autoimmune disease.

  • The inhibitors of prostaglandin $E_2\;(PGE_2)$ production and cyclooxygenase-2 (COX-2) activity have been considered as potential anti-inflammatory agents. In this study, we evaluated 9 compounds isolated from 5 Korean phytomedicinal plants (Spirea prunifolia, Paeonia suffruticosa, Salvia miltiorrhiza, Scutellaria baicalensis, and Artemisia capillaris) for the inhibition of $PGE_2$production and COX-2 expession in lipopolysaccharide (LPS)-stimulated human macrophages U937 cells. As a result, several compound such as prunioside A, penta-O-galloyl-beta-D-glucose, tanshinone IIA, baicalin, baicalein, wogonin, scopolatin, scoparone and decursinol showed potent inhibition of $PGE_2$production (50-70% inhibition at the test concentration of $10\;{\mu}M$). In addition, these compounds were also considered as potential inhibitors of COX-2 activity (45-73% inhibition at the test concentration of $10\;{\mu}M$). These active compound mediating COX-2 inhibitory activities are warranted for further elucidation of active principles for development of anti-inflammatory agents and these properties may contribute to the anti-atopic dermatitis activity.

Essential Oil of Thujopsis dolobrata Suppresses Atopic Dermatitis-Like Skin Lesions in NC/Nga Mice

  • We examined the effects of essential oil from Thujopsis dolobrata Sieb. et Zucc. var. hondai Makino (EOTD) (Cupressaceae) on atopic dermatitis (AD)-like skin lesions in NC/Nga mice. Treatment with EOTD twice daily for two weeks ameliorate AD-like skin lesions induced by DNCB (2,4 dinitrochlorobenzene), and clinical scores were reduced to 7.29, 7.07, and 4.5 points in the groups treated with 1.5%, 3.0%, and 6.0% extract (p<0.01) respectively, from the 15.0 score obtained using vehicle. EOTD inhibited the infiltration of mast cells into the AD-like skin lesion in NC/Nga mice (p<0.01) and also reduced serum histamine and IgE levels (p<0.05). Furthermore, it dose-dependently inhibited the release of beta-hexosaminidase from rat basophilic leukemia RBL 2H3 cells. These results indicate that EOTD reduces AD-like skin lesions by inhibiting the production of IgE and histamine, and, thus, IgE-mediated degranulation.

Effect of SPZZC, a Composition of Herb Extracts, on Atopic Dermatitis in BALB/c and NC/Nga Mouse (BALB/c 및 NC/Nga 마우스의 아토피성 피부질환모델에서 생약조성물 SPZZC의 치료효능)

  • The aim of this study was to evaluate if a composition of herb extracts, PLX-PLS was effective to treat atopic dermatitis (AD) in mice. SPZZC is a composition of herb extracts containing the roots of Scopolia parviflora and Paeonia lactiflora, the herb of Zizania caudiflora, the fruit of Ziziphus jujuba and the leaf of Chinese arborvitae. AD in BALB/c mouse was induced by patching ovoalbumin on the backside, while it in NC/Nga mouse was induced by repeated application of 1-chloro 2,4-dinitrobenzene (DNCB). Mice were topically treated with SPZZC or Domohorn ointment on the backside for 2 weeks (BALB/c) or 1 week (NC/Nga). Scratching behavior, clinical skin severity and the levels of WBC, neutrophil, eosinophil and total serum IgE were measured. After AD induction, scores of scratching behavior and clinical skin severity and the levels of WBC, neutrophil, eosinophil and total serum IgE were increased. Treatment with SPZZC significantly decreased scores of scratching behavior and clinical skin severity in a dose dependent manner in NC/Nga and BALB/c mice. Treatment with SPZZC 2% significantly decreased also the levels of WBC, neutrophil, eosinophil and total serum IgE. Especially, treatment of SPZZC 2% reduced more rapidly score of clinical skin severity than clobetasol cream. These results suggest that the SPZZC may be an alternative substance for the management of AD.

Acceleration of heat shock-induced collagen breakdown in human dermal fibroblasts with knockdown of NF-E2-related factor 2

  • Heat shock increases skin temperature during sun exposure and some evidence indicates that it may be involved in skin aging. The antioxidant response mediated by the transcription factor NF-E2-related factor 2 (Nrf2) is a critically important cellular defense mechanism that serves to limit skin aging. We investigated the effects of heat shock on collagenase expression when the antioxidant defense system was downregulated by knockdown of Nrf2. GSH and collagenases were analyzed, and the expression of inducible Nrf2, HO-1, and NQO1 was measured. HS68 cells were transfected with small interfering RNA against Nrf2. Heat shock induced the downregulation of Nrf2 in both the cytosol and nucleus and reduced the expression of HO-1, GSH, and NQO1. In addition, heat-exposed Nrf2-knockdown cells showed significantly increased levels of collagenase protein and decreased levels of procollagen. Our data suggest that Nrf2 plays an important role in protection against heat shock-induced collagen breakdown in skin. [BMB Reports 2015; 48(8): 467-472]
