산풍고삼환(散風苦蔘丸)이 알레르기성 접촉피부염에 미치는 영향 (The Effects of Sanpunggosamhwan on the Allergic Contact Dermatitis)

  • Objectives: To study the effectiveness of Sanpunggosamhwan(SGH) against Allergic Contact Dermatitis(ACD), the contact hypersensitivity assay, change of cutaneous shape, anti-allergic effect, anti-inflammatory effect, and the effect on skin barrier were observed. Methods: The 200g rats were divided into three groups of 15 rats. The first group is the Normal group which was applied Acetone olive oil only. The second group is the ACD group which has intentionally activated Allergic Contact Dermatitis by DNCB. The third group is the SGH group which was given medication of Sanpunggosamhwan extract after the induction of Allergic Contact Dermatitis by DNCB. Each group of rats was observed after 24, 48 and 72 hours. Results: I. With the result of contact hypersensitivity assay, at 24hours SGH group showed appreciably less ear swelling than ACD group. 2. Regarding general change of skin, SGH group showed less hyperplasia of epidermis, less damage of epidermis than ACD group. 3. Regarding the number of WBC, ACD group showed significantly less than normal and SGH group at 72 hours. 4. Regarding the number of RBC in blood, ACD and SGH group showed significantly more RBC than normal group at 24, 48, 72 hours. 5. Regarding the ratio of neutrophil in WBC, ACD and SGH group showed significantly high percentage than normal group at 24, 48 hours. 6. Regarding the ratio of lymphocyte in WBC, ACD and SGH group showed significantly high percentage than normal group at 48 hours. 7. Regarding the erythema, SGH group showed significantly more erythema than normal and ACD group at 48 hours. 8. Regarding the melanin, SGH group showed significantly less melanin than normal group at 24 hours.9. Regarding the skin hydration, SGH group showed significantly high value than and ACD group at 72 hours. 10. Regarding the skin pH, ACD group showed significantly high value than normal and SGH group af 24 hours. 11. Regarding the number of Total IgE, ACD and SGH group showed more Total IgE than normal. g개up at 24 hours. 12. At Electro microscope-morphologic changes of skin, the damage of epithelium was decreased and regeneration power of skin was increased in the SGH group. Conclusions: The Sanpunggosamhwan extract administration was effective on the mitigation of skin damage in rats with allergic contact dermatitis.

가미강활산(加味羌活散)이 집먼지 진드기 추출물로 유도된 NC/Nga mouse의 아토피 피부염에 미치는 영향 (Effect of Kami-kanghwalsan (KKHS) on Atopic Dermatitis-like Skin Lesions Induced in NC/Nga Mouse by Mite Antigen Stimulation)

  • Objectives The purpose of this study is to investigate the effect of KKHS on atopic dermatitis in an in-vivo experiment using an NC/Nga atopic dermatitis mouse, which has histological and clinical similarities to this condition in humans. Methods To investigate the effect of KKHS on atopic dermatitis (AD), we evaluated atopic dermatitis-like skin lesions by clinical skin index and analyzed immunological parameters in peripheral blood mononuclear cells(PBMCs), splenocytes, draining lymph node(DLN) and performed skin histology in ears and dorsal skin of atopic dermatitis of NC/Nga mouse in vivo. Results In vivo, clinical skin severity score was significantly lower in the KKHS group than in the control group. IgE, IL-6, TNF-${\alpha}$, IgM, IgG2a and IgG2b levels in serum decreased remarkably in the KKHS group than in the control group, and the level of IFN-${\gamma}$ production which is secreted from Th1 cell was increased by KKHS. After this experiment we analyzed immunological cells ($CD3^+$, $CD19^+$, $CD4^+$, $CD8^+$, $CD3^+CD69^+$, $CD4^+CD25^+$ and $CD49b^+$) by flow cytometry. It results that the total absolute number of $CD3^+$, $CD19^+$, $CD4^+$ and $CD8^+$ cells were recovered as much as normal state, and the level of $CD3^+CD69^+$ in isolated DLN and PBMCs were significantly decreased, and total absolute number of $Gr-1^+$, $CD11b^+$ and $CD3^+$ in dorsal skin of NC/Nga mouse were decreased by KKHS. We analyzed ear, DLN, and neck-back skin after biopsy and dyeing by hematoxyline/eosin(H&E), toluidine staining (mast cells marker). KKHS were very effective to the histological symptoms which are in dermal and epidermal thickening, hyperkeratosis and inflammatory cell infiltration. Ear thickness was significantly decreased compared with the control group and the size of inflammatory lymphocytes cells (ILC) and plasma cells (PC) in DLN were also decreased. Conclusions KKHS on atopic dermatitis in an in-vivo experiment using an NC/Nga atopic dermatitis mouse was very effectiveness to the atopy dermatitis treatment.

A Functional SNP in the MDM2 Promoter Mediates E2F1 Affinity to Modulate Cyclin D1 Expression in Tumor Cell Proliferation

  • Background: The MDM2 oncogene, a negative regulator of p53, has a functional polymorphism in the promoter region (SNP309) that is associated with multiple kinds of cancers including non-melanoma skin cancer. SNP309 has been shown to associate with accelerated tumor formation by increasing the affinity of the transcriptional activator Sp1. It remains unknown whether there are other factors involved in the regulation of MDM2 transcription through a trans-regulatory mechanism. Methods: In this study, SNP309 was verified to be associated with overexpression of MDM2 in tumor cells. Bioinformatics predicts that the T to G substitution at SNP309 generates a stronger E2F1 binding site, which was confirmed by ChIP and luciferase assays. Results: E2F1 knockdown downregulates the expression of MDM2, which confirms that E2F1 is a functional upstream regulator. Furthermore, tumor cells with the GG genotype exhibited a higher proliferation rate than TT, correlating with cyclin D1 expression. E2F1 depletion significantly inhibits the proliferation capacity and downregulates cyclin D1 expression, especially in GG genotype skin fibroblasts. Notably, E2F1 siRNA effects could be rescued by cyclin D1 overexpression. Conclusion: Taken together, a novel modulator E2F1 was identified as regulating MDM2 expression dependent on SNP309 and further mediates cyclin D1 expression and tumor cell proliferation. E2F1 might act as an important factor for SNP309 serving as a rate-limiting event in carcinogenesis.

  • This paper presents a fuzzy-based method for classification skin color object in a complex background under varying illumination. Parameters of fuzzy rule base are generated using a genetic algorithm(GA). The color model is used in the YCbCr color space. We propose a unique fuzzy system in order to accommodate varying background color and illumination condition. This fuzzy system approach to skin color classification is discussed along with an overview of YCbCr color space.

  • Preliminary pile load tests for the design of large diameter drilled shaft were performed on two of reduced scale(D=1370mm) test piles. The maximum loads of 2350 tonf in each direction were applied using bi-directional hydraulic jacks(Osterberg Cell) at toe. Neither of the test piles yielded in terms of skin friction and end bearing. Comparisons of the test results with several methods that estimate pile capacity show that the method of Horvath and Kenney(1979) for skin friction and Zhang and Einstein(1998) for end bearing were most appropriate for the site. The test results were directly applied to pile design in case RQD of skin and toe was larger than that of the test pile. It is desirable, therefore, to consider not only unconfined compression strength but also rock mass properties(i.e. TCR, RQD) for skin friction and end bearing evaluation in the future.

Hyperspectral Fluorescence Imaging for Mouse Skin Tumor Detection

  • This paper presents a hyperspectral imaging technique based on laser-induced fluorescence for non-invasive detection of tumorous tissue on mouse skin. Hyperspectral imaging sensors collect image data in a number of narrow, adjacent spectral bands. Such high-resolution measurement of spectral information reveals contiguous emission spectra at each image pixel useful for the characterization of constituent materials. The hyperspectral image data used in this study are fluorescence images of mouse skin consisting of 21 spectral bands in the visible spectrum of the wavelengths ranging from 440 nm to 640 nm. Fluorescence signal is measured with the use of laser excitation at 337 nm. An acousto-optic tunable filter (AOTF) is used to capture images at 10 nm intervals. All spectral band images are spatially registered with the reference band image at 490 nm to obtain exact pixel correspondences by compensating the spatial offsets caused by the refraction differences in AOTF at different wavelengths during the image capture procedure. The unique fluorescence spectral signatures demonstrate a good separation to differentiate malignant tumors from normal tissues for rapid detection of skin cancers without biopsy.

Improvement of skin barrier function using lipid mixture

  • Dry skin is caused mainly by the perturbation of stratum corneum lipids which affected by ageing, change of season, excess use of surfactant and the effect of disease like atopic dermatitis and psoriasis. Intercellular lipid structures in stratum corneum are responsible for the barrier function of mammalian skin. The major lipd classes that can be extracted from stratum corneum are ceramides, cholesterol and fatty acid, which make up approximately 50, 25, 10 percent of the stratum corneum lipid mass, respectively. Small amount of cholesterol sulfate, phospholipids, glycosylceramide and cholesterol esters are also present. Recent studies have shown that application of one or two these lipids to the perturbed skin delays barrier recovery; only equimolar mixtures allow normal recovery. We observed that barrier recovery rate was improved in hairless mouse by topical application of single neutral lipids (ceramide, free fatty acid, cholesterol) and lipid mixtures. Whereas the application of single lipid didn’t allows a significant enhancement comparing with normal barrier repair, the equimolar mixtures of 3 components(including synthetic pseudoceramide PC104) improved barrier repair, as assessed by the transepidermal water loss. At clinical study to the volunteers aged over sixty, skin dryness recuperated by the increase of moisture(capacitance) and the reduction of scaling. Utilization of physiologic lipid mixture containing natural ceramides or synthetic pseudoceramide could lead to new forms of topical therapy for the dryness and dermatoses(e.g., psoriasis, atopic dermatitis and irritant dermatitis).

A numerical method for improving the reliability of knee translation measurement in skin marker-based motion analysis

  • In skin-marker based motion analysis, knee translation measurement is highly dependent on a pre-selected reference point (functional center) on each segment determined by the location of anatomical landmarks. However, the placement of skin markers on palpable anatomical landmarks (i.e., femoral epicondyles) has limited reproducibility. Thus, it produces large variances in knee translation measurement among different subjects, as well as across studies. In order improve the repeatability of knee translation measurement, in this study an optimization method was introduced, by which the femoral functional center was numerically determined. At that point the knee anteroposterior translation during the stance phase of walking was minimized. This new method was tested on 30 healthy subjects during walking in gait lab with motion capture system. Using this new method, the impact of skin marker position (at anatomical landmarks) on the knee translation measurement has been minimized. In addition, the ranges of anteroposterior knee translations during stance phase were significantly (p<0.001) smaller than those measured by conventional method which relies on a pre-selected functional center ($11.1{\pm}3.5mm$ vs. $19.9{\pm}5.5mm$). The results of anteroposterior translation using this new method were very close to a previously reported knee translation (12.4 mm) from dual fluoroscopic imaging technique. Moreover, this new method increased the reproducibility of knee translation measurement by 50%.

Ovalbumin으로 유도한 아토피성 피부염의 마우스 종별 차이에 관한 예비연구 (Ovalbumin Induces Atopic Dermatitis-like Skin Lesions in Different Species of mice: pilot study)

  • Objectives: Atopic dermatitis (AD) is an easily recurrent inflammatory skin disease. Since AD has complex pathology, people have been investigating it on different aspects with various experimental models. One of them is in vivo model which has spontaneously developed AD-like skin lesions by various inducers. Methods: In this study, two kinds of mouse species were applied in the experiment; BALB/c and C57BL/6 mice. We compared features among the animal species making AD mouse model with protein allergen, ovalbumin. AD-like skin lesions were induced by ovalbumin on two kinds of scheme and were evaluated with the histological results and size of spleen which is a critical immunological organ. Also, we measured the level of immunoglobulin E in serum. In addition, we investigated the results of ovalbumin induced-AD-like skin lesions along with obesity. Results and Conclusion: We evaluated weight of organs and thickness of epidermis. These results suggest the possibility of the appropriate in vivo experimental model for AD or the comorbidity with obesity.
