Methods : Male rats weighing $195.21{\pm}5.85g$ fed high fat diet for 8 weeks and 40 rats(above 400 g) were divided into 4 groups. Each groups were divided into a control group and 3 experimental groups. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats Basal diet and administered an extract of Cornus walteri Wanger(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of proinflammatory cytokines, anti-oxidative activity and gene expression. Results : 1. Concentration of plasma free fatty acid, LDL-cholesterol showed a tendency to decrease in Cornus walteri Wanger ext. groups. Concentration of plasma triglyceride, total cholesterol showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. HDL-cholesterol showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group. 2. Concentration of liver total cholesterol showed a tendence to decrease in Cornus walteri Wanger ext. groups. Concentration of triglyceride liver showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. 3. Concentration of plasma and liver TBARS showed a tendence to decrease in Cornus walteri Wanger ext. groups. The values of GSH-Px, SOD and CAT activity showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group than that of control group. 4. The values of plasma AST and ALT activity showed no significantly different in all treatment groups. 5. Concentration of plasma $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ showed a tendency to decrease in the Cornus walteri Wanger ext. groups. However the concentration of IL-10 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly increment than that of control group. Concentration of liver $IL-1{\beta}$, $TNF-{\alpha}$ and IL-10 showed no significantly difference in all treatment groups. However concentration of IL-6 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly decrement than that of control group. 6. In the analysis of RT-PCR, gene expression of $TNF-{\alpha}$, Apo-B, Apo-E and leptin in the Cornus walteri Wanger ext. groups showed a lower expression than that of control group. 7. The ratio of $TNF-{\alpha}$, Apo-E and leptin expression per $\beta$-actin expression in the 200 and 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. The ratio of Apo-B expression per $\beta$-actin expression in the 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. Conclusions : According to above results, in lowering lipid effect, antioxidative activity and antiinflammatory effect, the Cornus walteri Wanger ext. gives positive effect.

  • We have investigated the EPR signal properties in 12 components of two mobile phones (LCD, OLED) using electron paramagnetic resonance (EPR) spectrometer in this study.EPR measurements were performed at normal atmospheric conditions using Bruker EXEXSYS-II E500 spectrometer with X-band bridge, and samples were irradiated by $^{137}Cs$ gamma-ray source. To identify the presence of radiation-induced signal (RIS), the EPR spectra of each sample were measured unirradiated and irradiated at 50 Gy. Then, dose-response curve and signal intensity variating by time after irradiation were measured. As a result, the signal intensity increased after irradiation in all samples except the USIM plastic and IC chip. Among the samples, cover glass(CG), lens, light guide plate(LGP) and diffusion sheet have shown fine linearity ($R^2$ > 0.99). Especially, the LGP had ideal characteristics for dosimetry because there were no signal in 0 Gy and high rate of increase in RIS. However, this sample showed weakness in fading. Signal intensity of LGP and Diffusion Sheet decreased by 50% within 72 hours after irradiation, while signals of Cover Glass and Lens were stably preserved during the short period of time. In order to apply rapidly EPR dosimetry using mobile phone components in large-scale radiation accidents, further studies on signal differences for same components of the different mobile phone, fading, pretreatment of samples and processing of background signal are needed. However, it will be possible to do dosimetry by dose-additive method or comparative method using unirradiated same product in small-scale accident.

  • Parameters of mating system and pollen flow of a Pinus densiflora population in Buan, South Korea, were estimated using seven nuclear microsatellite markers. The expected heterozygosity ($H_e$) was 0.614 in mother trees and 0.624 in seeds. Fixation index (F) was 0.018 and 0.087 in each generation. There was no significant genetic difference between the generations (P > 0.05). From MLTR, the outcrossing rate ($t_m$), the biparental inbreeding ($t_m-t_s$), and the correlation of paternity ($r_p$) were 0.967, 0.057, and 0.012, respectively. tm was larger but $t_m-t_s$ and $r_p$ were smaller than those of allozyme markers in Pinus densiflora. These values were similar to those of microsatellite markers in other pine species. The optimal pollen dispersal model from TwoGener was the normal dispersal model with the effective density of 220 trees/ha and its level of genetic differentiation in pollen pool structure (${\Phi}_{ft}$) was 0.021. The average radial distance of pollen flow (${\delta}$) was calculated as 11.42 m, but no correlation between the pairwise-${\Phi}_{ft}$ and the geographical distance among mother trees was at Mantel test (r = -0.141, P > 0.05). Although the effective pollen dispersal in the population seems to be restricted, the amount of genetic variation might be maintained in each generation without a loss of genetic diversity. It might be because the genetic diversity in pollen pool was high but the genetic difference between pollen donors was small under the complete random mating condition in the Pinus densiflora population in Buan.

  • In soybeans, responses of high temperature according to shift of sowing dates during the growing season was explored using the crop model, CROPGRO-soybean. In addition, it analyzed impact on change of sowing dates affects yield potential of soybean under future climate scenario (2041-2070). In Jeonju and Miryang during 1981-2010, if sowing at 15 or ten days ahead from 10 June, namely in shorten of the sowing day (i.e. when sown on 25 or 30 May), the yield potential reduced. However, the yield potential increased when sown 5 June. In the case of delay of sowing day (i.e. when sown on 15 or 20 June), reduction of yield potential in the average -5% was higher than increase in the average +2%. In particular, the relative changes for shorten of the sowing day or delay of the sowing day do not be shown in normal years which high temperatures did not abnormally occur during the growing season from 2003 to 2010 except when sown on 25 May. In abnormal years which high temperatures occurred during the critical period, especially R5 to R7, shorten of the sowing day affected to the increase of yield potential in Miryang, while the yield potential decreased in Jeonju except when sown on 5 June. However, delay of the sowing day influenced on the reduction of yield potential both in two sites. In future climate scenario of Representative Concentration Pathway (RCP) 8.5 during from 2041 to 2070, the increase and decrease of yield potential for shorten of the sowing day were +10/-9% for RCP 8.5 of Jeonju, and +14/-9% for RCP 8.5 of Miryang, respectively. Additionally, it showed +10/-17% for RCP 8.5 in Jeonju, and +10/-29% for RCP 8.5 in Miryang, respectively in the increase and decrease of yield potential for delay of the sowing day.

  • This paper described the extraction/purification of $\beta$-carotene from recombinant E.coli and evaluation of anti-wrinkle activity of purified $\beta$-carotene. No significant differences in extraction yields were observed when hexane or isobutyl acetate was used. However, extraction from wet-cell cake resulted in 2-fold higher amount of $\beta$-carotene than that from dry cells. Disruption of 5 g-wet cells by ultrasonic homogenizer, acetone dehydration, extraction with isobutyl acetate resulted in 36 mg of $\beta$-carotene corresponding to 61.2% of recovery. The formation and separation of $\beta$-carotene crystal improved the purity. 633 mg of $\beta$-carotene crystal with 93% purity was obtained from 223 g/L of wet-cell cake harvested from 2.5-L fed-batch culture broth. The cultures of normal human primary fibroblast were performed to investigate the effect of $\beta$-carotene on cytotoxicity as MTT assay and anti-wrinkle activity as collagen synthesis assays. $1.7{\mu}M$ of $\beta$-carotene was found to be optimal concentration at which 1.4-fold higher amount of collagen was synthesized than that in absence of $\beta$-carotene. This indicates that highly purified $\beta$-carotene can be obtained from recombinant E.coli by applying simple method with less toxic solvent and can be used in functional cosmetics as anti-wrinkle agent.

  • Purpose: The triple therapy with proton pump inhibitor (PPI) has been recognized as the treatment of choice in Helicobacter pylori (H. pylori) infection in adults. However, the effect of triple therapy with omeprazole, amoxicillin and clarithromycin (OAC) on eradication of H. pylori infection in children has not been established yet. This study was performed to evaluate the efficacy of OAC triple therapy and to compare the effect of one-week with two-week therapy on H. pylori eradication. Methods: From July 1998 to July 2000, 34 children with upper gastrointestinal symptoms, who underwent upper gastrointestinal endoscopy with biopsy at entry and 4 or more weeks after therapy, were enrolled in this study. H. pylori infection was assessed by CLO test and histologic examination (Hematoxylin-Eosin stain or Alcian yellow stain) with biopsy specimens. The regimen consisted of omeprazole (0.7 mg/kg/day), amoxicillin (50 mg/kg/day), and clarithromycin (25 mg/kg/day) for 1 week (n=21) or 2 weeks (n=13). Eradication of H. pylori was determined after the termination of treatment by the CLO test and histologic examination. Results: One-week treatment group consisted of 21 children (11 male, 10 female) with a mean age of $9.5{\pm}3.0$ years. Two-week group consisted of 13 children (4 male, 9 female) with a mean age of $9.9{\pm}4.0$ years. The endoscopic diagnoses included nodular gastritis in 19 cases, superficial gastritis in 7 cases, gastric ulcer in 4 cases, purpuric duodenitis in 2 cases, and normal in 2 cases. H. pylori was eradicated in 28 of total 34 children (82.4%). In 1-week group, H. pylori was eradicated in 17 of 21 children (81%). In 2-week group, H. pylori was eradicated in 11 of 13 children (84.6%). In remaining 6 cases in whom H. pylori had not been eradicated with OAC regimen, H. pylori infection persisted despite of the treatment with additional drugs such as colloidal bismuth subcitrate ($Denol^{(R)}$) and metronidazole. Conclusion: In this study, eradication rate of H. pylori with OAC regimen was 82.4%, and the triple therapy would be highly effective as primary treatment. However, there was no significant difference in the eradication rate between the 1-week and 2-week treatment group (P=0.785).

  • Litsea populifolia, a plant species of the Lauraceae family, is widely distributed in the tropical and subtropical areas of Asia. The phylogenetic relationships and botanical characteristics of L. populifolia have been reported; however, its anti-oxidative and anti-cancer activities remain unclear. In this study, we evaluated the anti-oxidative and anti-cancer effects of ethanol extracts of L. populifolia (EELP) together with the molecular mechanism of its anti-cancer activity in human lung adenocarcinoma A549 cells. EELP showed significant anti-oxidative effects with a 50% inhibitory concentration at $11.71{\mu}g/ml$, which was measured by the 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay. EELP exhibited cytotoxic activity and induced cell cycle arrest at the G1 phase in A549 cells in a dose-dependent manner, whereas EELP did not have the cytotoxic effect on the normal human lung cell line IMR90. Treatment with EELP also resulted in a decreased expression of G1/S transition-related molecules-including cyclin-dependent kinase (CDK) 2, CDK6, cyclin D1, and cyclin E-both for the transcription and translation levels. EELP-induced G1 arrest was associated with the phosphorylation of checkpoint kinase 2 (CHK2), p53, cell division cycle 25 homolog A (CDC25A), and the reduction of CDC25A expression in A549 cells. Collectively, these results suggest that EELP may exert an anti-cancer effect by cell cycle arrest at the G1 phase through both p53-dependent and p53-independent (ATM/CHK2/CDC25A/CDK2) pathways in A549 cells.

  • Prostate cancer (PCa) is one of the most metastatic tumor. Although hormone therapy or surgical castration is mostly conducted to treat PCa, it has a lot of side effects. Recently, many researchers have been exploring the tumor microenvironment to remedy these circumstances. Immune cells, especially macrophages, are an important composition of the tumor microenvironment. Under normal conditions, macrophages exhibit mild tumoricidal activity against tumors. However, once activated by interferon gamma or lipopolysaccharides, macrophages can kill cancer cells directly or indirectly by secreting cytokines and chemokines. In this study, murine macrophage RAW 264.7 cells were treated with Phellinus linteus extract. To analyze their pro-inflammatory phenotype, we were used several assays such as a real-time polymerase chain reaction, an enzyme-linked immunosorbent and nitric oxide assay. Prostate cancer cells were treated with the RAW 264.7-conditioned media, which was identified as a pro-inflammatory nature, for 48 h, and the expression of epithelial-mesenchymal transition (EMT)-related genes was determined. Not only N-cadherin, Snail, Twist, Slug, and Cadherin 11, which are mechenchymal-related proteins, were decrease, but epithelial marker of E-cadherin was increased. In addition, the mRNA level of vimentin, ccl2, and vegfa were decreased, as the EMT is closely related to the migration and invasion of cancer cells. In conclusion, the RAW 264.7-conditioned media stimulated with P. linteus extract inhibited migration and invasion and regulated the EMT pathway in human prostate cancer cells.

  • It is likely that the heading would occur early when air temperature increases. In 2022, however, the heading date was delayed unusually, e.g., by 3 to 5 days although temperature during the vegetative growth stage was higher than normal years. The objective of this study was to identify the cause of such event analyzing weather variables including average temperature, sunshine hours, and day-length for each growth stage. The observation data were collected for medium-late maturing varieties, which has been grown at crop yield experiment sites including Daegu, Andong, and Yesan. The difference in heading date was compared between growing seasons in 2021 and 2022 because crop management options, e.g., the cultivars and cultivation methods, were identical at those sites during the study period. It appeared that the heading date was delayed due to the difference in temperature responsiveness under a given day-length condition The effect of the temperature increase on the heading date differed between the periods during which when the day-length was more than 14.3 hours before and after the summer-solstice.. The effect of the temperature decrease during the period from which the day-length decreased to less than 14.3 hours to the heading date was relatively greater. This merits further studies to examine the response of rice to the temperature change under different day-length and sunshine duration in terms of heading.

  • To assess the industrial possibility of mixed-extracts containing Hovenia dulcis Thunberg and 12 different botanical ingredients, a protective effect was confirmed in the chronic ethanol-induced the liver, brain, and blood injury in mouse. Blood glucose levels of the normal control group(NG) and ethanol administration group(EG) were respectively 119.43mg/dL and 305.25mg/dL, and the mixed-extracts administration group(100, 200mg/kg body weight + 25% ethanol 5g/kg body weight respectively; ME100 & ME200) were decreased to 272.76mg/dL and 234.60mg/dL. Blood ethanol contents were decreased in ME100 and ME200(3.85mg/dL, 3.08mg/dL) compared to EG(4.08mg/dL), and blood acetaldehyde contents were also decreased in ME(15.76mg/dL, 15.16mg/dL) compared to EG(18.72mg/dL). The contents of hepatotoxic indicators such as glutamine pyruvic transaminase(GPT) and glutamic oxaloacetic transaminase (GOT), nephrotoxic indicators such as blood urea nitrogen(BUN), and creatine(CRE), and total cholestero(TCHO), and triglyceride(TG) in mouse blood serum were significantly decreased in the ME compared to EG. The acetylcholinesterase(AChE) activity of ME(109.00% and 108.47%, respectively) in mouse brain tissues was decreased in ME compared to EG(116.10%). Finally, ME was remarkable in vivo antioxidant activities in the mouse liver and brain tissues by superoxide dismutase(SOD), oxidized glutathione(GSH)/total GSH ratio and the malondialdehyde (MDA) assay. Therefore, the mixed-extracts was considered to be effective a high value food with protective effect against chronic ethanol traetment-induced cytotoxicity in liver and brain tissues.
