Multiplex RT-PCR Assay for Detection of Common Fusion Transcripts in Acute Lymphoblastic Leukemia and Chronic Myeloid Leukemia Cases

  • Background: Acute lymphoblastic leukemia (ALL) is a heterogeneous disease which requires a risk-stratified approach for appropriate treatment. Specific chromosomal translocations within leukemic blasts are important prognostic factors that allow identification of relevant subgroups. In this study, we developed a multiplex RT-PCR assay for detection of the 4 most frequent translocations in ALL (BCR-ABL, TEL-AML1, MLL-AF4, and E2A-PBX1). Materials and Methods: A total of 214 diagnosed ALL samples from both adult and pediatric ALL and 14 cases of CML patients (154 bone marrow and 74 peripheral blood samples) were assessed for specific chromosomal translocations by cytogenetic and multiplex RT-PCR assays. Results: The results showed that 46 cases of ALL and CML (20.2%) contained the fusion transcripts. Within the positive ALL patients, the most prevalent cryptic translocation observed was mBCR-ABL (p190) at 8.41%. In addition, other genetic rearrangements detected by the multiplex PCR were 4.21% TEL-AML1 and 2.34% E2A-PBX1, whereas MLL-AF4 exhibited negative results in all tested samples. Moreover, MBCR-ABL was detected in all 14 CML samples. In 16 samples of normal karyotype ALL (n=9), ALL with no cytogentic result (n=4) and CML with no Philadelphia chromosome (n=3), fusion transcripts were detected. Conclusions: Multiplex RT-PCR provides a rapid, simple and highly sensitive method to detect fusion transcripts for prognostic and risk stratification of ALL and CML patients.

Radixin Knockdown by RNA Interference Suppresses Human Glioblastoma Cell Growth in Vitro and in Vivo

  • Radixin, a member of the ERM (ezrin-radixin-moesin) family, plays important roles in cell motility, invasion and tumor progression. It is expressed in a variety of normal and neoplastic cells, including many types of epithelial and lymphoid examples. However, its function in glioblastomas remains elusive. Thus, in this study, radixin gene expression was first examined in the glioblastoma cells, then suppressed with a lentivirus-mediated short-hairpin RNA (shRNA) method.We found that there were high levels of radixin expression in glioblastoma U251cells. Radixin shRNA caused down-regulation of radixin gene expression and when radixin-silenced cells were implanted into nude mice, tumor growth was significantly inhibited as compared to blank control cells or nonsense shRNA cells. In addition, microvessel density in the tumors was significantly reduced. Thrombospondin-1 (TSP-1) and E-cadherin were up-regulated in radixin- suppressed glioblastoma U251 cells. In contrast, MMP9 was down-regulated. Taken together, our findings suggest that radixin is involved in GBM cell migration and invasion, and implicate TSP-1, E-cadherin and MMP9 as metastasis-inducing factors.

An Application of the Localized Finite Element Method to Two-dimensional Free Surface Wave Problems (2차원 자유표면파 문제에서의 국소 유한요소법의 응용)

  • The numerical calculation for solving boundary-value problem related to potential flows with a free surface is carried out by application of the localized finite element method. Only forced motion of 2-D body in infinitely deep fluid is considered, although this schemes is equally applicable to any first order time-harmonic problems of similar nature. The infinite domain of the fluid is separated into the inner flow field and the outer flow field with common inter-surface boundary. The finite element method is applied to obtain the solution in the inner flow field and the Green functions are utilized to represent the solution in the outer flow field. At the inter-surface boundary, the continuity of the value of potential and the normal derivative of the potential(i.e. matching condition) is conserved. The present method has better computational efficiency than the previous LFEM and the integral equation method of Frank. This enhanced computational efficiency is presumably due to the fact that the present method gives a symmetric coefficient matrix and requires less computational time in calculating the influence coefficient matrix of Green function than the integral equation method. And the irregular frequency desen't exist because the uniqueness of the solution is assured by the such that the exact free surface condition is satisfied on the boundary of the localized finite element region(i.e. inner region). As an example of the above method, the hydrodynamic forces for the circular cylinder and the rectangular cylinders are calculated. In the computed results, the small number of singularity distribution segments($3{\sim}6$) give good result relative to Ursell's and Vugts'.

Histopathological Studies on the Effect of Korean Safflower (Carthamus tinctorious L.) Seed Powder on Cadmium Poisoning in Mice (홍화씨분말이 카드뮴 중독에 미치는 병리조직학적 관찰)

  • This study was undertaken to find out the effect of Korean safflower seed powder on histopathological changes of cadmium toxicity in mice. Fifty BALB/c mice were divided into a control group(A) and four experimental groups(B, C, D, E) : group A received tap water and basal diet, group B received tap water and diet supplemented with 3% Korean safflower seed powder alone, group C received basal diet and 300 $\mu\textrm{g}$/g of cadmium, group D and E received basal diet supplemented with 3% and 10% Korean safflower seed powder and 300$\mu\textrm{g}$/g of cadmium respectively. Cadmium dissolved in tap water was used, and the Korean safflower seed powder were mixed with feed. All mice were dissected on the 56th day. Histopathological changes in liver, kidney, lung, cortical osseous tissue of femoral shaft, bone trabecular of femur, and epiphyseal cartilage plate of femur were observed. Group B showed no significant changes compared with the control group. But group C showed the unclearness of specific cells in liver, the loss of architecture and focal necrosis of hepatocyte, the glomerular swelling, degeneration and necrosis of convoluted tubules, desquamation and vacuolization of the greater part of the renal tubular epithelium, the marked congestion and thickness of the wall of alveolus in lung, slightly thinning of the cortical osseous tissue in femoral shaft, reduction of cancellous bone volume and marked narrowness of bone trabecular, marked thinning of epiphyseal cartilage plate and irregular arrangement of columnar structure of cartilage cells. On the other hand, Korean safflower seed powder-treated group showed a little convalescent changes and maintained their normal architectures in liner, kidney, lung, cortical osseous tissue of femoral shaft, bone trabecular of femur and epiphyseal cartilage plate of femur.

Micro Total Hip Replacement in Two Dogs with Legg-Calvé-Perthes Disease

  • A Maltese (case 1) and a Pomeranian (case 2) presented with acute right and left hind limb weight-bearing lameness. On physical examination, they exhibited severe pain and crepitus on the coxofemoral joint of affected hind limb. In addition, decreased thigh girth measurements were noted compared with the opposite leg. Radiological exam revealed necrotic areas in the femoral head of affected hind limb. These dogs were diagnosed with Legg-$Calv{\acute{e}}$-Perthes disease (LCPD). The dogs underwent micro total hip replacement (THR). After surgery, at 3 years (case 1) and 7 months (case 2), both dogs recovered normal activity. The thigh girth and lameness scores were apparently improved in the affected limbs of both dogs. No complications of prosthesis implants, such as loosening, were noted. The clinical outcomes of these cases indicate that dogs with LCPD can be successfully treated with micro THR and have a good prognosis immediately after surgery.

Correlation of Overexpression of Nestin with Expression of Epithelial-Mesenchymal Transition-Related Proteins in Gastric Adenocarcinoma

  • Background: Nestin is associated with neoplastic transformation. However, the mechanisms by which nestin contributes regarding invasion and malignancy of gastric adenocarcinoma (GAC) remain unknown. Recent studies have shown that the epithelial-mesenchymal transition (EMT) is important in invasion and migration of cancer cells. In the present study, we aimed to investigate the expression of nestin and its correlation with EMT-related proteins in GAC. Materials and Methods: The expression of nestin and EMT-related proteins was examined in GAC specimens and cell lines by immunohistochemistry and Western blotting. Clinicopathological features and survival outcomes were retrospectively analyzed. Results: Positive nestin immunostaining was most obviously detected in the cytoplasm, nucleus or both cytoplasm and nucleus of tumor cells in 19.2% (24/125) of GAC tissues, which was significantly higher than that in normal gastric mucosa tissues (1.7%, 1/60) (p=0.001). Nestin expression was closely related to several clinicopathological factors and EMT-related proteins (E-cadherin, vimentin and Snail) and displayed a poor prognosis. Interestingly, simultaneous cytoplasmic and nuclear nestin expression correlated with EMT-related proteins (E-cadherin, vimentin and Snail) (p<0.05) and lymph node metastasis (p=0.041) and a shorter survival time (p<0.05), but this was not the case with cytoplasmic or nuclear nestin expression. Conclusions: Nestin, particularly expression in both cytoplasm and nucleus, might be involved in regulating EMT and malignant progression in GAC, with potential as an unfavorable indicator in tumor diagnosis and a target for clinical therapy.

Remote monitoring of light environment using web-camera for protected chrysanthemum production (웹 카메라를 이용한 시설 내 국화생산 광 환경 원격 모니터링)

  • Increase of national family income improved demand of high-quality and year-round horticultural products including chrysanthemum. To meet these demand, farmers have introduced protected facilities, such as greenhouses, of which environmental conditions could be monitored and controlled. Environment management up to three weeks after transplanting is critical for chrysanthemum quality. Artificial lighting and light-blocking screen are especially important for long-day (day period > 13 hours) and short-day (night period > 13 hours) treatments. In this study, a web-camera was installed, and the image was obtained and transmitted to mobile phones to monitor the status of 3-wavelength(RGB) lighting environments. RGB pixel values were used to determine malfunctioning of the lighting lamps, and leaking out and incoming illumination status during short-day and long-day treatment periods. Normal lighting lamps provided RGB pixel values of 240~255. During long-day treatment period, G pixel values were useful to detect abnormal lighting conditions (e.g., leaking). During short-day treatment period, R pixel values were useful to determine incoming light (e.g., sun-light). Results of this study would provide useful information for remote monitoring of light conditions for protected chrysanthemum production under artificial lights.

Effect of Ulmi radicis Cortex Extract on Systemic and Local Anaphaylaxis in Rats (유근피(楡根皮)가 전신적(全身的) 및 국소적(局所的) 아나필락시스에 미치는 효과(效果))

  • Ulmi radicis cortex is a herb medicine which has been used for the treatment of such allergic disease as urticaria, allergic rhinitis and athma. To assess the contribution of an aqueous extract of Ulmi radicis cortex(URC) in systemic anaphylaxis, we used compound 48/80 as a fatal anaphylaxis inducer in rats. URC inhibited anaphylactic shock 100% with a dose of 1.0 mg/g body weight (BW) 1 hr before injection of compound 48/80. URC significantly inhibited serum histamine levels induced by compound 48/80. URC (1.0 mg/g BW) also inhibited to 79.1% passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. URC dose-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. Moreover, URC had a significant inhibitory effect on anti-DNP IgE-induced histamine release or tumor necrosis $factor-{\alpha}$ production from RPMC. The level of cAMP in RPMC, when URC was added, significantly increased compared with that of normal control. These results indicate that URC may possess strong antianaphylactic effect.

The Effects of Lonicerae Flos, Forsythiae Fluctus and Hwangryunhaedok Decoction Pharmacopuncture on Atopic Dermatitis in NC/Nga Mice (금은화, 연교 및 황련해독탕 약침이 아토피 피부염 유발 NC/Nga 생쥐에 미치는 영향)

  • Objectives : Atopic dermatitis is a chronic inflammatory skin disease characterized by pruritic and erythematous skin lesions. The purpose of this study was to investigate the suppressive effects of Lonicerae Flos, Forsythiae Fluctus and Hwangryunhaedok Decoction Pharmacopuncture on the development of atopic dermatitis-like skin lesions in NC/Nga Mice. Methods : The Atopic Dermatitis was induced by biostir AD on the mice's back skin. Experimental groups were divided into three including LFP(Lonicerae Flos Pharmacopuncture, EtOH extract), FFP(Forsythiae Fluctus Pharmacopuncture, EtOH extract) and HHP(Hwangryunhaedok Decoction Pharmacopuncture, Hydrodistillation extract). Every second day, the mice of three groups were treated with $0.1m{\ell}$ of pharmacopuncture using a syringe at right and left acupoints ($BL_{13}$), alternatively. On the control group, normal saline was used instead of pharmacopuncture. Subsequently optical observation with a handscope, a clinical skin score, Tissue(general/immune) mast cell, Serum IgE level, Serum histamine level, and Serum lymphokine(IL-2, IL-4, $IFN-{\gamma}{\gamma}$) were measured. Results : FFP and HHP decreased the clinical skin score, the total cell number of mast cells, and the Serum total IgE level and Serum histamine level. In Serum lymphokine levels, all groups were decreased to the IL-4 level, LFP and FFP were increased to the IL-2 level, and LFP was increased to the $IFN-{\gamma}$ level. Conclusions : From the above results, Forsythiae Fluctus Pharmacopuncture (EtOH extract) and Hwangryunhaedok Decoction Pharmacopuncture (Hydrodistillation extract) exerted anti-allergic and anti-inflammatory effects, suggesting a promising agent for improving atopic dermatitis related symptoms.

Design and Implementation of a Browser for Educational PDA Contents (교육용 PDA 컨텐츠 브라우저의 설계 및 구현)

  • Recently various electronic books (I-Book) based on PDA (personal digital assistance) that we can easily use anytime and anywhere have been developed. Volume and weight of the E-Book is much less than that of traditional books. In that reason, it is easy to carry and serve us with contents by diverse functions such as searching, bookmark, dictionary, and playing of color image, sound or moving picture. On account of these advantages, many products connected with I-Book have been emerged in the market. However a product connected with educational contents is scarce, because it requires not only normal function but also additional functions such as a problem solving. Therefore it is actually necessary to develop a browser and an editor for educational contents. In this paper, we express educational contents by XML and define structure of document with XML schema. Then, we design and implement an editor and a browser that can manage educational contents on PDA.
