Background: Sulforaphane (SFN) is an isothiocyanate compound present in cruciferous vegetables. Although the anti-inflammatory effects of SFN have been reported, the precise mechanism related to the inflammatory genes is poorly understood. Objectives: This study examined the relationship between the anti-inflammatory effects of SFN and the differential gene expression pattern in SFN treated ob/ob mice. Methods: Nitric oxide (NO) level was measured using a Griess assay. The inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expression levels were analyzed by Western blot analysis. Pro-inflammatory cytokines (tumor necrosis factor [TNF]-α, interleukin [IL]-1β, and IL-6) were measured by enzyme-linked immunosorbent assay (ELISA). RNA sequencing analysis was performed to evaluate the differential gene expression in the liver of ob/ob mice. Results: The SFN treatment significantly attenuated the iNOS and COX-2 expression levels and inhibited NO, TNF-α, IL-1β, and IL-6 production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. RNA sequencing analysis showed that the expression levels of 28 genes related to inflammation were up-regulated (> 2-fold), and six genes were down-regulated (< 0.6-fold) in the control ob/ob mice compared to normal mice. In contrast, the gene expression levels were restored to the normal level by SFN. The protein-protein interaction (PPI) network showed that chemokine ligand (Cxcl14, Ccl1, Ccl3, Ccl4, Ccl17) and chemokine receptor (Ccr3, Cxcr1, Ccr10) were located in close proximity and formed a "functional cluster" in the middle of the network. Conclusions: The overall results suggest that SFN has a potent anti-inflammatory effect by normalizing the expression levels of the genes related to inflammation that were perturbed in ob/ob mice.
This study aimed to investigate changes in the quality of meat aged using Jeju scoria earthenware. Water-holding properties, pH, color, tenderization, fatty acid composition, and free amino acid characteristics of aged pork loin were evaluated to determine the effects of wet and dry (normal and Jeju Scoria) aging methods and aging time (0, 10, 20, 30, and 40 days) on meat quality. The aging methods altered pH and CIE L* after 10 days of aging. However, the aging method did not alter the pH and CIE L* of the aged pork loin after 10 days of aging. The shear force was significantly lower in the Scoria aging method than in the wet aging method after 10 and 20 days of aging. Both the normal and Scoria aging methods increased aging loss compared to the wet aging method, which could be attributed to longer air exposure times of the two non-packaged aging methods than in the packaged aging method. The scoria aging method had significantly higher contents and ratios of saturated (SFA) to mono-unsaturated fatty acids but lower ratios of poly-unsaturated fatty acids to SFA than the wet aging method. The content of most free amino acids significantly increased with aging time, especially those related to the sweet, and umami categories. In summary, this study suggests that the Scoria aging method provides positive aspects of eating quality, such as improvement of meat tenderness and taste, including umami, with minimum changes in the overall meat quality.
Objective : This study was conducted to investigate the muscle-improving and therapeutic effects of Boehmeria platanifolia (BP) in a mouse model of dexamethasone-induced muscle atrophy. Methods : Muscle atrophy was induced in C57BL/6 mice by intraperitoneal administration of dexamethasone for 12 days. BP extract was administered orally at doses of 100 mg/kg and 200 mg/kg for 19 days, starting 7 days before the intraperitoneal administration of dexamethasone. Mice were weighed during the experimental period, and muscle strength and muscle weight were measured at the end of the experiment. The gastrocnemius (GASTROC) muscles of mice were isolated and the cross-sectional area (CSA) of the muscle fibers was measured after H&E staining. Results : Dexamethasone-induced muscle atrophy mice had a decrease in body weight compared to normal mice, and BP-administrated mice did not show significant change in body weight compared with a control group. Muscle strength in mice with induced muscle atrophy was reduced compared to normal and significantly increased with BP administration and positive control. In addition, the weight of the quadriceps (QUAD) muscle and fiber size of the GASTROC muscle, which was reduced in sarcopenia-induced mice, was increased by BP. Conclusion : BP extract increased muscle strength, muscle weight, and muscle fiber size in dexamethasone-induced muscle atrophy mice. This suggests that the efficacy of BP extracts in improving muscle strength and preventing and treating sarcopenia may be beneficial for the development of potential therapeutic or functional products.

CD96 is a membrane-bound receptor discovered in humans in 1992 that is mainly present in natural killer cells and T cells derived from haematopoietic cells and performs immune functions. Based on the sequence of CD96 obtained from red seabream (Pagrus major), phylogenetic analysis with other species, infections of normal fish, Streptococcus iniae and red sea bream iridovirus (RSIV), and expression analysis was conducted using real-time polymerase chain reaction. Phylogenetic analysis showed the highest homology with Sparus aurata, and multiple sequence analysis confirmed the conservation of major domains between different fish species. Normal fish high expression results were confirmed in the head kidney, and spleen, which are the haematopoietic organs of the fish. High expression levels were confirmed in the gills, liver, spleen, and kidney on day three after RSIV infection. After S. iniae infection, high expression was confirmed in the gills and liver on day one, and high expression was confirmed in the spleen from 12 hours. These results show that PmCD96 functions as an immune gene in P. major and is considered a basic research case for CD96 in fish's hematopoietic organ immune system.

Background and Objectives Skin and soft tissue defects can be treated according to a range of strategies, such as local flap, skin graft, biological dressing, or free flap. On the other hand, free tissue transfer usually leaves a distinct scar with an inconsistency of color or hypertrophy. This problem is highlighted if the defect is located on the face, which could have devastating effects on a patient's psychosocial health. Materials and Methods The authors used an erbium : yttrium-aluminum-garnet (Er:YAG) laser to resurface the free flap skin and match the color with the surrounding facial skin. This study evaluated the effectiveness of laser skin resurfacing on the harmonious color matching of transferred flap. Patients who had undergone laser resurfacing on facial flap skin between January 2014 and December 2018 were reviewed retrospectively. An ablative 2,940-nm fractional Er:YAG laser treatment was delivered to the entire flap skin at 21 J/cm2 with the treatment end-point of pinpoint bleeding. Several months later, the clinical photographs were analyzed. The L*a*b* color co-ordinates of both the flap and surrounding normal skin were measured using Adobe Photoshop. The L*a*b* color difference (ΔE) for the scar and normal surrounding skin were calculated using the following equation: ${\Delta}E=\sqrt{({\Delta}L)^2+({\Delta}a)^2+({\Delta}b)^2}$ Results All five patients were satisfied with the more natural appearance of the flaps. The ΔE values decreased significantly from the pre-treatment mean value of 19.64 to the post-treatment mean value of 11.39 (Wilcoxon signed-rank test, p = 0.043). Conclusion Ablative laser resurfacing can improve the aesthetic outcome of free tissue transfer on the face.

This paper deals with the strongest static arches with the solid regular polygon cross-section. Both span length and volume of arch are always held constant regardless the shape functions of cross-sectional depth of regular polygon. The normal stresses acting on such arches are calculated when both static vertical and horizontal point loads are subjected. By using the calculating results of stresses, the optimal shapes of strongest static arches are obtained, under which the maximum normal stress become to be minimum. For determining the redundant of such indeterminate arches, the least work theorem is adopted. As the numerical results, the configurations, i.e. section ratios, of the strongest static arches are reported in tables and figures. The results of this study can be utilized in the field of the minimum weight design of the arch structures.
Septin4 belong to a family of polymerizing GTP-binding proteins that are required for many cellular functions, such as membrane compartmentalization, vesicular trafficking, mitosis, and cytoskeletal remodeling. Since, Septin4 is expressed specifically in the testis, we aimed to determine the association between Septin4 gene expression with sperm quality, DNA damage, and stress oxidative level in infertile patients. The present study included 60 semen samples that grouped into three groups: normozoospermia (n=20), asthenozoospermia (n=20), astheno-teratozoospermia (n=20). Initially, semen parameters were analyzed by using the World Health Organization protocol. The mRNA expression of Septin4 in sperm was examined using reverse transcription-polymerase chain reaction. Oxidative stress markers, i.e., total antioxidant capacity, superoxide dismutase, catalase, glutathione peroxidase, and malondialdehyde, were determined by ELISA kit. The current study showed a statistically significant highly positive correlation in Septin4 gene expression with sperm motility, normal morphology, viability, capacity, and sperm mitochondrial membrane potential (MMP). However, it showed significant negative correlation with sperm DNA fragmentation. Septin4 had a significant correlation with stress oxidative factor and antioxidant enzyme levels. In conclusion, Septin4 gene expression provides clinical useful information for the diagnosis of male infertility. It might be a marker for discrimination between fertile and infertile patients. To compare and analyze the improvement effects of white ginseng extract, red ginseng extract, and black ginseng extract on cognitive dysfunction and memory impairment caused by scopolamine in rats. In the cognitive behavioral test, the tendency of the SCOP+B group to overcome the escape time delay induced by scopolamine administration was observed, unlike the SCOP group. The frequency on plat form was significantly increased in the group treated with ginseng extracts compared to the SCOP group. As a result of measuring the duration time on goal quadrant, the time spent in the quadrant was significantly increased in the SCOP+B group compared to the SCOP group. In the hippocampus, the SCOP-treated group significantly decreased the activity of AChE compared to the normal group, but the ginseng extract-treated groups significantly increased it compared to the SCOP group. After sacrificing the rats after the behavioral test, the expression of PSD95 protein in the excised brain was significantly decreased in the SCOP group compared to normal, but it was observed that the SCOP+R and SCOP+B groups were significantly increased compared to the SCOP group. CREB1 protein expression was significantly increased in the SCOP+R group, and the expression of Cdk5 was significantly increased in the SCOP+B group. Ginseng extracts significantly restored the memory damaged by scopolamine suggesting that red ginseng increased the expression of CREB1 and PSD95 proteins, and black ginseng increased the protein expression of Cdk5 and PSD95 to induce memory recovery.
This study was carried out to study the changing patterns in muscle activities in Angle's II and Class III malocclusion patients following activator therapy, To study the activities of anterior temporal(T.A.), masseter(M.M.), posterior temporal(T.P.), and anterior belly of digastric(D.A.) muscles, surface electrodes were placed on 15 Angle's Class II and 17 Angle's Class III malocclusion patients and following conclusions were obtained after electromyographic recordings were taken at prior to the activator therapy and at some time between 6 -12 month after the start of activator therapy. 1. Class II and Class III malocclusion groups, when compared to normal occlusiongroup, showed no significant differences during resting and swallowing. But significant differences were observed in masseter and posterior temporal muscles during clenching. 2. Most of the muscles studied showed a certain pattern, that is those groups whose pre-treatment E.M.G. were lower than the normal value showed significant increase after activator therapy, whereas those groups whose pre-treatment E.M.G. were higher than the normal value showed significant decrease after activator therapy. This pattern was observed in all of the muscle groups studied except T.A.(swallowing), T.P.(clenching) and D.A.(swallowing, clenching) in Angle's Class III malocclusion group and in Angle's Class II malocclusion group, above tendency were observed only in T.A.(swallowing), M.M.(resting, swallowing) and T.P.(clenching). This pattern was less obvious during clenching. 3. When E.M.G. activity after activator therapy between Angle's Class II and Class III malocclusion group was compared, Class III malocclusion group showed more increase during resting, and Class II malocclusion group showed more increase during swallowing and clenching excepting M.M. and D.A. respectively.
Acanthopanax sessiliflorus (A. sessiliflorus) has been known as a traditional medicine having anti-stress, antioxidative and platelet aggregation inhibitory effects. This study was undertaken to investigate the functional properties of water extracts from four parts of A. sessiliflorus. Root, stem, leaf and fruit extracts from A. sessiliflorus were prepared with hot water ($80^{\circ}C$). The contents of functional substances, eleutheroside B and E, polyphenol, antioxidative activity, nitrite scavenging ability and anti-cancer activity of the extracts were determined. The contents of eleutheroside E in stem, root and fruit extracts were 542.50 ${\mu}$g/g, 343.35 ${\mu}$g/g and 30.78 ${\mu}$g/g, respectively. A large part of eleutheroside B was found in fruit (372.01 ${\mu}$g/g) and root (289.33 ${\mu}$g/g) extracts. Root and stem extracts contained 227.21 mg/100g and 131.22 mg/100g of polyphenols, respectively. Antioxidative activities (electron donating ability) of stem and root extracts were 79.87% and 77.27%, respectively. It appears that the antioxidative activities were related to polyphenol contents of the extracts. Most extracts showed 76-81.5% of nitrite scavenging ability at pH 1.2. It reveals that water extract from parts of A. sessiliflorus can inhibit formation of nitrosoamine in food. Effects of the extracts on the growth of normal and cancer cell lines were investigated. Extracts showed no cytotoxicity to normal dendritic cell line (DC2.4). Especially, the root extract promoted the growth of normal cell line. Root and stem extracts had 20-23% of inhibitory effect against stomach cancer cell line (SNU-719) and liver cancer cell line (Hep3B). These result indicated that the extracts from A. sessiliflorus can be used as functional food materials with antioxidative activity and nitrite scavenging ability to eliminate nitrosoamine in food.
