미성숙 감(Diospyros kaki Thunb. Sangju-Dungsi) 에탄올 추출물의 항알레르기 생리활성 연구 (Study on the Anti-allergic Effect using Immature Persimmon (Diospyros kaki Thunb. Sangju-Dungsi) Ethanol Extract)

  • This study was conducted to verify the availability of immature persimmon ethanol extract (IPEE) as a natural and functional ingredient in protecting inflammation and allergy of skin based on the mechanism. The major content analysis, antioxidant activities, anti-allergic activity, anti-inflammatory effect, and safety related to irritation of IPEE were evaluated. The gallic acid content per 10 mg/mL of IPEE was 0.522% (5.22 mg/g). The total polyphenol and flavonoid contents were 428.3 mg/g and 31.1 mg/g, respectively. In ABTS+ activity, DPPH ability and SOD-like activity, it showed a concentration-dependent increase, which indicated IPEE has excellent antioxidant activities. As for the anti-allergy test in RBL-2H3 cells, the IPEE showed a decrease in β-hexosaminidase secretion as the concentration increases, and IPEE tended to decrease IL-4 secretion in all RBL-2H3 cells compared to the IgE + HSA group. IPEE showed good anti-inflammatory effect in RAW 264.7 cells by decrease of NO production and inflammation cytokines (TNF-α and IL-6). Also IPEE showed non-irritant in BCOP assay. By the results of this study, the IPEE containing high tannins, had good antioxidant, anti-allergic, and anti-inflammatory effects, which indicated that the immature persimmon is considered to be a useful for the development of related functional ingredients.

Evaluation of pepper seedling growth according to the growing period and tray for automatic transplanting

  • The effective growth of pepper seedlings relies on the growing period and tray used, which both aim to minimize seedling damage during the transplantation process. Therefore, the objective of this study was to evaluate the effect of red and green pepper seedling growth in plug and cylindrical paper pot (CPP) trays with different growth periods of the seedlings. Two different seedling growth trays were used for the two varieties of pepper seedlings in the same growing media. The pepper seedling growth rates were investigated at 15, 30, and 45 days for each of variety. Important parameters, e.g., the plant height, fresh weight for the plant and root, number of leaves, leaf length and width, and leaf chlorophyll contents, were measured. During the experiment, the CPP tray maintained uniform seedling growth as compared to the plug tray. CPP trays ensured the height of the seedling, with these results 0.84 to 1.6 times higher than those of the plug tray for all conditions, indicating the good quality of the pepper seedlings. The shoot and root weights were found to be greater with more leaves in the CPP tray seedlings for 45 days compared to the seedlings grown in the plug tray, whereas the green pepper variety showed a greater leaf ratio than the red pepper seedlings. The analysis of pepper seedling growth presented in this study will guide the selection of suitable growth trays and seedling periods for farmers when they undertake automatic pepper transplantation in the field.

Impact of soft and stiff soil interlayers on the pile group dynamic response under lateral harmonic load

  • The interlayers, either softer or stiffer than the surrounding layers, are usually overlooked during field investigation due to the small thickness. They may be neglected through the analysis process for simplicity. However, they may significantly affect the dynamic behavior of the soil-foundation system. In this study, a series of 3D finite-element Direct-solution steady-state harmonic analyses were carried out using ABAQUS/CAE software to investigate the impacts of interlayers on the dynamic response of a cast in place pile group subjected to horizontal harmonic load. The experimental data of a 3×2 pile group testing was used to verify the numerical modeling. The effects of thickness, depth, and shear modulus of the interlayers on the dynamic response of the pile group are investigated. The simulations were conducted on both stiff and soft soils. It was found that the soft interlayers affect the frequency-amplitude curve of the system only in frequencies higher than 70% of the resonant frequency of the base soil. While, the effect of stiff interlayer in soft base soil started at frequency of 35% of the resonant frequency of the base soil. Also, it was observed that a shallow stiff interlayer increased the resonant amplitude by 11%, while a deep one only increased the resonant frequency by 7%. Moreover, a shallow soft interlayer increased the resonant frequency by 20% in soft base soils, whereas, it had an effect as low as 6% on resonant amplitude. Also, the results showed that deep soft interlayers increased the resonant amplitude by 17 to 20% in both soft and stiff base soils due to a reduction in lateral support of the piles. In the cases of deep thick, soft interlayers, the resonant frequency reduced significantly, i.e., 16 to 20%. It was found that the stiff interlayers were most effective on the amplitude and frequency of the pile group.

Efficacy of buccal piroxicam infiltration and inferior alveolar nerve block in patients with irreversible pulpitis: a prospective, double-blind, randomized clinical trial

  • Objectives: This randomized clinical trial aimed to assess the effectiveness of buccal infiltration with piroxicam on the anesthetic efficacy of inferior alveolar nerve block (IANB) with buccal infiltration in irreversible pulpitis, with pain assessed using the Heft-Parker visual analogue scale (HP-VAS). Materials and Methods: This study included 56 patients with irreversible pulpitis in mandibular molars, randomly distributed between 2 groups (n = 28). After evaluating the initial pain score with the HP-VAS, each patient received IANB followed by buccal infiltration of 2% lignocaine with adrenaline (1:80,000). Five minutes later, the patients in groups 1 and 2 were given buccal infiltration with 40 mg/2 mL of piroxicam or normal saline, respectively. An access opening procedure (AOP) was performed 15 minutes post-IANB once the individual showed signs of lip numbness as well as 2 negative responses to electric pulp testing. The HP-VAS was used to grade the patient's pain during caries removal (CR), AOP, and working length measurement (WLM). Successful anesthesia was identified either by the absence of pain or slight pain through CR, AOP, and WLM, with no requirement of a further anesthetic dose. A statistical analysis was done using the Shapiro-Wilk and Mann-Whitney U tests. Results: The piroxicam group presented a significantly lower (p < 0.05) mean pain score than the saline group during AOP. Conclusions: Buccal infiltration with piroxicam enhanced the efficacy of anesthesia with IANB and buccal infiltration with lignocaine in patients with irreversible pulpitis.

Shape and anatomical relationship of the mental foramen to the mandibular premolars in an Indian sub-population: a retrospective CBCT analysis

  • Objectives: This study assessed the shape and anatomical relationship of the mental foramen (MF) to mandibular posterior teeth in an Indian sub-population. Materials and Methods: In total, 475 existing cone-beam computed tomography records exhibiting 950 MFs and including the bilateral presence of mandibular premolars and first molars were assessed. Images were evaluated 3-dimensionally to ascertain the position, shape, and anatomical proximity of MFs to mandibular teeth. The position and shape of MFs were measured and calculated. The Pythagorean theorem was used to calculate the distance between the root apex of the mandibular teeth and the MF. Results: MFs exhibited a predominantly round shape (left: 67% and right: 65%) followed by oval (left: 30% and right: 31%) in both males and females and in different age groups. The root apices of mandibular second premolars (left: 71% and right: 62%) were closest to the MF, followed by distal to the first premolars and mesial to the second premolars. The mean vertical distance between the MF and the nearest tooth apex calculated on sagittal sections was 2.20 mm on the right side and 2.32 mm on the left side; no significant difference was found according to sex or age. The distance between the apices of the teeth and the MF was ≥ 4 mm (left; 4.09 ± 1.27 mm and right; 4.01 ± 1.15 mm). Conclusions: These findings highlight the need for clinicians to be aware of the location of the MF in treatment planning and while performing non-surgical and surgical endodontic procedures.

Comparison of the sealing ability of various bioceramic materials for endodontic surgery

  • Objectives: Endosequence Bioceramic Root Repair Material (BC-RRM) is used in endodontic microsurgery. It is available as a paste and a putty. However, no studies to date have examined the sealing ability of these forms alone or in combination as root-end filling materials. Hence, this study aimed to compare the sealing properties of these 2 forms of BC-RRM. Materials and Methods: Forty-two extracted upper anterior teeth were divided into 3 experimental groups, a positive and negative control. After the root canal treatment, the root ends were resected, retroprepared and retrofilled with either putty, paste + putty or mineral trioxide aggregate (MTA). The teeth were mounted in tubes so the apical 3 mm was submerged in Brain Heart Infusion (BHI) broth. The coronal portions of the canals were inoculated with Enterococcus faecalis and BHI broth and incubated for 30 days. The broth in the tubes was analyzed for colony forming units to check for leakage of bacteria from the canal. The teeth from the groups were sectioned and analyzed using scanning electron microscopy (SEM). The Kruskal-Wallis test and analysis of variance were used to analyze the data with a significance level p < 0.05. Results: The BC-RRM and MTA groups showed similar sealing ability. The positive control showed leakage in all samples. The SEM imaging showed the presence of bacteria in all experimental groups at the material-tooth interface. Conclusions: No significant differences were noted in the experimental groups, providing sufficient evidence that any combination could be effectively used during endodontic microsurgery.

A novel antimicrobial-containing nanocellulose scaffold for regenerative endodontics

  • Objectives: The aim of this study was to evaluate bacterial nanocellulose (BNC) membranes incorporated with antimicrobial agents regarding cytotoxicity in fibroblasts of the periodontal ligament (PDLF), antimicrobial activity, and inhibition of multispecies biofilm formation. Materials and Methods: The tested BNC membranes were BNC + 1% clindamycin (BNC/CLI); BNC + 0.12% chlorhexidine (BNC/CHX); BNC + nitric oxide (BNC/NO); and conventional BNC (BNC; control). After PDLF culture, the BNC membranes were positioned in the wells and maintained for 24 hours. Cell viability was then evaluated using the MTS calorimetric test. Antimicrobial activity against Enterococcus faecalis, Actinomyces naeslundii, and Streptococcus sanguinis (S. sanguinis) was evaluated using the agar diffusion test. To assess the antibiofilm activity, BNC membranes were exposed for 24 hours to the mixed culture. After sonicating the BNC membranes to remove the remaining biofilm and plating the suspension on agar, the number of colony-forming units (CFU)/mL was determined. Data were analyzed by 1-way analysis of variance and the Tukey, Kruskal-Wallis, and Dunn tests (α = 5%). Results: PDLF metabolic activity after contact with BNC/CHX, BNC/CLI, and BNC/NO was 35%, 61% and 97%, respectively, compared to BNC. BNC/NO showed biocompatibility similar to that of BNC (p = 0.78). BNC/CLI showed the largest inhibition halos, and was superior to the other BNC membranes against S. sanguinis (p < 0.05). The experimental BNC membranes inhibited biofilm formation, with about a 3-fold log CFU reduction compared to BNC (p < 0.05). Conclusions: BNC/NO showed excellent biocompatibility and inhibited multispecies biofilm formation, similarly to BNC/CLI and BNC/CHX.

Spectrophotometric evaluation of restorative composite shades and their match with a classical shade guide

  • Objectives: The aim of this study was to verify the match between 5 shades of composites from different manufacturers with a shade guide and among the systems using a portable spectrophotometer. Materials and Methods: Shade measurements were performed on specimens of Z350 XT (3M ESPE), Charisma Diamond (Heraeus Kulzer GmbH), Esthet X-HD (Dentsply Caulk), and Empress Direct (Ivoclar-Vivadent) for shades A1, A2, A3, B1, and C3 using a Vita Easyshade spectrophotometer (Vita Zahnfabrik) against a white background. Corresponding shades of Vitapan Classical (Vita Zahnfabrik) guide were measured likewise and shade variation (ΔE) was calculated based on International Commission on Illumination L*a*b* parameters. The ΔE of the composites in each shade was compared by one-way analysis of variance and Tukey's post hoc test (α = 0.05). Results: All composites presented ΔE > 3.7 compared with the shade guide. Variation in shades A3, B1, and C3 was significantly different for all composites. ΔE of Z350 XT was significantly lower for A1 than for the other shades, whereas ΔE of Z350 XT and Charisma Diamond were significantly lower for A2 than for the other shades. Conclusions: No composite shade matched with the shade guide. Equivalent shades of the restorative composite from different manufacturers may show clinically noticeable ΔE.

Postoperative pain after endodontic treatment of necrotic teeth with large intentional foraminal enlargement

  • Objectives: To evaluate postoperative pain after endodontic treatment of necrotic teeth using large intentional foraminal enlargement (LIFE). Materials and Methods: The sample included 60 asymptomatic necrotic teeth (with or without chronic apical periodontitis), and a periodontal probing depth of 3 mm, previously accessed and referred to perform endodontic treatment. After previous procedures, the position and approximate size of the apical foramen (AF) were determined by using an apex locator and K flexo-files, respectively. The chemomechanical preparation was performed with Profile 04 files 2 mm beyond the AF to achieve the LIFE, using 2.5 mL of 2.5% NaOCl at each file change. The filling was performed by Tagger's hybrid technique and EndoFill sealer. Phone calls were made to all the patients at 24, 48 and 72 hours after treatment, to classify postoperative pain. Statistical analysis was performed by different tests with a significance level of 5%. Results: Age, gender, periradicular status and tooth type did not influence postoperative pain (p > 0.05). Only 1 patient (1.66%) reported severe pain after 72 hours. Moderate pain was reported by 7, 4 and 3 patients after 24, 48 and 72 hours, respectively (p = 0.0001). However, paired analyses showed a statistically significant difference only between 24 and 72 hours (p = 0.04). Sealer extrusion did not influence the postoperative pain (p > 0.05). Conclusions: Acute or moderate postoperative pain was uncommon after endodontic treatment of necrotic teeth with LIFE.

KTX용 2경간 연속교량의 동적거동 및 공진감소 (Dynamic Behavior and Resonance Reduction of Two-Span Continuous Bridges for Korean Train eXpress)

  • 본 연구에서는 경부고속철도상의 대표적인 교량인 2경간 연속보 형태의 고속철도 교량에 대하여 동적거동을 해석하고 공진발생의 감소방안들을 제시하였다. 하중은 한국형 고속철도하중인 총길이 380.15 m의 TGV-K 열차(2P+18T)가 일정한 속도로 이동하는 것으로 하였다. 동적거동을 모사하는 지배방정식은 질량과 강성이 분포된 연속계에서 만들어진 편미분방정식을 이용하여 구하였으며 Duhamel 적분을 이용한 모달계수의 처리를 수반하는 모드중첩법을 이용하여 동적해석을 수행하였다. 열차의 주행속도에 관계없이 동적거동을 급격히 감소시킬 수 있는 지간장이 존재하였으며 지금까지 연구된 단순보 형태의 교량에서 분석된 결과와는 달랐다. 열차의 주행속도에 의존하여 동적응답이 급격히 증가되어 공진현상을 일으키는 경우에도 공진을 감소시킬 수 있는 여러 방안들을 제시하였다.
