해조단백질의 추출에 관한 연구 1. 수용성 단백질의 추출 (STUDIES ON THE EXTRACTION OF SEAWEED PROTEINS 1. Extraction of Water Soluble Proteins)

  • 분포량이 많고 식용되고 있는 중요 해조의 효율적인 이용과 영양학적인 의의를 구명하기 위한 기초자료를 얻기 위하여 동근돌김(Porphyra suborciculata). 잎파래(Enteromorpha linza). 톳(Hizikia jusiforme). 모자반(Sargassum fulvllum), 미역(Undaria pinnatifida), 청각(Codim fragile), 셀만모자반(Sargassum kjellmanianum), 구멍갈파래(Ulva pertusa)등 8종을 선정하여, 이들 해조의 불용성 단백질 추출에 영향을 주는 제요소 즉 조체처리별, 추출온도와 시간별, 시료일추출용매화, pH의 영향등을 검토하고 또한 추출액중의 단백질 심전조건에 대하여 검토하였다. 1. 식용 해조의 조단백질 함량은 둥근돌김(Porphyra suborbiculat) $24.14\%$, 미역(Undaria pinnatifida) $15.7\%$, 톳(Hizikia jusiforme) $7.66\%$, 모자반(Sargasum fulvellum) $12.61\%$, 셀만모자반(Sargassum kjellmanianum) $14.08\%$, 잎파래(Enteromorpha linza) $10.86\%$, 구멍갈파래(Ulva Pertusa) $16.60\%$, 청각(Codium fragile)$9.89\%$이었다. 2. 시료의 처리조건에 따른 추출효과는 dry icemethanol 한제 중에서 동결시켜, 해사를 첨가하여 마쇄한 시료가 수용성 단백질 추출성적이 월등하게 좋아, 막지사발에서 마쇄한 $1.5\~2.5$배의 추출성적을 보였다. 3. 시료일추출용매비는 점질물이 적은 구멍갈파래, 잎파래는 1:20(w/v)에서, 점질물량이 많은 청각,미역, 톳, 모자반, 둥근돌김 등은 1:30(w/v)에서, 셀만모자반은 1:40(w/v)일 때 추출성적이 좋았다. 4. 추출시간은 구멍갈파래, 잎파래는 1시간, 둥근돌김, 톳, 미역, 모자반, 셀만모자반은 2시간, 청각은 3시간 정도에서 가장 효과적이었다. 5. 추출온도는 톳이 $40^{\circ}C, 둥근돌김, 청각, 잎파래 미역, 모자반, 셀만모자반 $50\%$에서, 구멍갈파래는 $60^{\circ}C$에서 최고의 추출성적을 보였다. 6. pH의 영향은 각 시료마다 대동소이하여 $pH9\~12$에서 가장 좋은 결과를 얻었다. 7. 추출 최적조건에서의 수용성 단백질 추출율은 1차 추출에서 전조단백질에 대하여 평균 $63\%$, 2차 유출에서 평균 $8.6\%$ 합계 $74\%$ 정도의 수용성 단백질 추출이 가능했으며, 1차에 대한 2차의 추출비율이 7.5:1이었다. 8. 단백질 침전분리 조작으로는 Barnstein method, methanol 처리법, TCA 처리법, 가열처리법 순으로 침전효과가 있었다.

ICE-POP 2018 기간 드롭존데 자료를 활용한 강설 구름의 열역학적 특성 (Thermodynamic Characteristics of Snowfall Clouds using Dropsonde Data During ICE-POP 2018)

  • The aircraft observation campaign was performed to investigate thermodynamic conditions of snowfall cloud over the East Sea of Korean peninsula from 2 February to 16 March 2018. During this period, four snowfall events occurred in the Yeongdong region and three cases were analyzed using dropsonde data. Snowfall cases were associated with the passage of southern low-pressure (maritime warm air mass) and expansion of northern high-pressure (continental polar air mass). Case 1 and Case 2a were related to low-pressure systems, and Case 2b and Case 3 were connected with high-pressure systems, respectively. And their thermodynamic properties and horizontal distribution of snowfall cloud were differed according to the influence of the synoptic condition. In Case 1 and Case 2a, atmospheric layers between sea surface and 350 hPa contained moisture more than 15 mm of TPW with multiple inversion layers detected by dropsonde data, while the vertical atmosphere of Case 2b and Case 3 were dry as TPW 5 mm or less with a single inversion inversion layer around 750~850 hPa. However, the vertical distributions of equivalent potential temperature (θe) were similar as moist-adiabatically neutral condition regardless of the case. But, their values below 900 hPa were about 10 K higher in Case 1 and Case 2a (285~290 K) than in Case 2b and Case 3 (275~280 K). The difference in these values is related to the characteristics of the incoming air mass and the location of the snowfall cloud.

실내동결시스템을 이용한 노상토의 동상 특성 (The Frost Heaving Characteristics of Subgrade Soils Using Laboratory Freezing System)

  • 겨울에는 시베리아기단의 영향으로 한랭 건조한 대륙성 고기압의 영향을 받아 춥고 건조하여 1월 평균기온이 $-6{\sim}-7^{\circ}C$의 영하의 온도로 낮아져 지반동결시 수분이동으로 동상현상이 발생하여 도로의 불균형 동결팽창을 초래하며, 결국 포장체를 파손시킨다. 동상발생시 토립자는 모관력에 의해 지하수를 흡수하여 아이스렌즈를 형성하며, 이 모관 흡수력은 토립자의 크기에 영향을 받는다. 도로는 다양한 재료와 단면으로 구성된 구조물이기 때문에 환경성과 재료 물성뿐만 아니라 포장체 각 층의 구조적 적정성 또는 지지력을 파악하는 것이 무엇보다 중요하다. 현재 기존 동상방지층 설계법에 따르면, 동상방지층은 포장체의 구조적 적정성과는 무관하게 온도 조건에 따른 동결깊이에 따라 일률적으로 결정되고 있다. 이러한 동결깊이를 포장구조설계에 적용함으로써 포장의 과다설계 우려가 있다. 따라서 본 논문에서는 도로 동상방지층의 효용성 검증 및 설치기준 확립을 위해 실내동결시스템을 활용하여 도로 노상토의 동상 특성에 대한 민감성을 판별, 도로건설 현장 노상토에 대한 역학적 특성과 실내 동결 시험을 수행하였으며, 외부 동결온도의 지속 조건에 대한 시료의 온도변화, 동상팽창압, 동상팽창량, 부동수분 등의 결과값을 통하여 동결 과정에 따른 지반공학적 특성을 평가하였다.

1623~1800년 서울지역의 기상기후 환경 -'승정원일기'를 토대로- (Weather and Climatic Environment of Seoul Area in South Korea during 1623~1800, Reconstructed from 'The Daily Records of Royal Secretariat of Joseon Dynasty(承政院日記)')

  • 본 연구에서는 승정원일기에 기록된 매일의 날씨와 기상현상을 토대로 기상기후 정보를 자료화하고 조선왕조실록 등에 기록된 기상 및 천변재이에 관한 정보를 추가 조사하여, 지금까지 연구되지 않은 1623~1800년 기간 서울지역의 기상과 기후 환경을 파악하였다. 1500~1760년은 소빙기에 해당하여 조사기간 서울지역에서는 대부분의 여름이 서늘하였으며 대체로 기상현상의 변화가 심하고 한랭 건조했던 것으로 파악된다. 특히 날씨와 기상현상 및 기후변화가 1650년, 1710년, 1770년을 전후로 현저하게 나타났다. 비와 눈의 일수는 1640년대에 큰 변화를 나타내기 시작해 1650년대~1710년대에 크게 감소하였고, 1710년대 이후 급격히 증가하였다. 계절적으로 여름의 비 일수는 1710년대 말 이후에 급격히 증가하였고, 눈의 일수는 1770년대 중반 이후 크게 감소하였다. 우박 일수는 1720년대 말에 크게 증가해 1760년대까지 계속되었다. 흐린 날의 일수는 1710년대를 전후로 여름에는 크게 감소하나 겨울에는 다소 증가하였다. 안개일수는 1770년경 이후에 현대의 평년값보다 적은 일수를 나타내기 시작하였다. 이들 시기는 상대적으로 강화된 한랭화와 건조화의 경향을 따르며, 역사기후학적 비교자료 및 지구과학 생물학적 자료의 연구 결과와도 대체로 일치한다.

남극 엽층리 퇴적물로부터 규조 종을 이용한 고기후 변화 연구 (Diatom Succession Representing the Paleoclimatic Change from Laminated Sediments around Antarctica)

  • 남극 브랜스필드 해협의 동부분지로부터 획득한 GC08-EB01 코어퇴적물의 고해상 엽층리로부터 규조를 이용하여 고기후변화를 연구하였다. 규조 정량 분석의 결과 규조 개체수 농도는 $0.4-4.2{\times}10^8g^{-1}$로 매우 풍부하게 산출되었으며, 엽층리는 생물기원의 규조가 풍부한 층과 육성기원 퇴적물이 풍부한 층으로 구분된다. 규조 엽층리는 Corethron crilophilum, Eucampia antarctica, Fragilariopsis curta, F. kerguelensis, Odontella weissflogii, Proboscia inermis, R. styliformis, Thalassiosira antarctica와 Chaetoceros resting spores 등의 번성이 특징적이며, 육원성 엽층리에서는 규조 군집이 혼합되어 나타나는 것이 특징이다. 홀로세 후기 동안 Fragilariopsis curta+F. cylindrus)/Thalassiosira antarctica의 산출 수직 분포비가 급격히 증가하는 구간은 해빙의 확장을 지시하며, 연구 코어 GC08-EB01 코어퇴적물이 퇴적되는 동안 적어도 5번의 한랭기가 있었음을 알려준다.

Experimental infection of Anopheles sinensis with Korean isolates of Plasmodium vivax

  • The objectives of the present study were to (1) determine the susceptibility of Anopheles sinensis to Korean isolates of Plasmodium vivax, (2) establish a method to collect large quantities of P. vivax sporozoites for use as antigen in seroepidemiological studies, and (3) investigate the characteristics of Korean isolates of P. vivax sporozoites. Females of Anopheles sinensis were collected at non-epidemic area, Seokwha-ri, Cheongwon-gun and Chungcheongbuk-do using tent-trap methods coupled with dry ice. The females were artificially infected with gameiocytes of P. vivax using blood obtained from P vivax malaria patients. Individual mosquitoes were infected using either a parafilm-covered glass feeding apparatus or were allowed to feed on naturally infected volunteers. Mosquitoes were sacrificed between 16 and 18 days post-feeding and an enzyme-linked immunosorbent assay (ELISA) was used to detect sporozoites. Four (33.4%) of 12 mosquitoes, which were fed on naturally infected volunteers directly, were positive for sporozoites. In cases, the mosquitoes allowed to feed on whole blood which were extract from three different patients with heparin treated vacuutainers using a parafilm-covered glass apparatus. Two of 55 (3.6%) were positive which blood sample was maintained at room temperature for 8 hours, 1 of 68 (1.5%) was positive which blood was maintained at $4^{\circ}C$ for 24 hours and 1 of 47 (2.3%) was positive at 4$^{\circ}C$ for 48 hours. The mean number of sporozoites was estimated about 818 (n=8; range of 648-1,056) based on optical density values of ELISA.

개심술에 있어서 GIK 의 심근 보호효과에 대하여 (Effect of GIK Solution for Myocardial Protection)

  • Although anoxic cardiac arrest produces a dry, quiet field, the ability of the myocardium to withstand the anoxic insult is uncertain. The current growth of interest in the use of various cardioplegic solutions ` has resulted in the development of a number of different solutions. In this study, 51 consecutive cases of elective open heart surgery with the aid of extracorporeal circulation were reviewed retrospectively to compare two methods of myocardial preservation. All of these open heart operations had been performed, using hemodilution principle under the moderate hypothermia at the Department of Thoracic and Cardiovascular Surgery, College of Medicine, Kyungpook National University from December, 1975 to July, 1979. In the 31 consecutive cases that form the anoxic arrest group, the operations were done with intermittent aortic cross-clamping and topical cardiac hypothermia. The heart was cooled topically by cold normal saline, which was converted to ice slush before application to the pericardial sac. Twenty of 51 consecutive cases were assigned to the cold cardioplegic method [the cardioplegic group], in which two kinds of cold cardioplegic solutions [Young solution and G IK solution] were infused into the aortic root proximal to the aortic cross clamp for myocardial preservation. Mean total aortic cross clamp times were 43 minutes in the anoxic arrest group and 67 minutes in the cardioplegic group. In the post-operative period, spontaneous regular heart beatings were recovered in 80 percent of the cardioplegic group as opposed to 25.7 percent of the anoxic arrest group. Ventricular fibrillation requiring DC shock was seen in 32.3 percent of the anoxic arrest group and 10 percent of the cardioplegic group. In the cardioplegic group, mean CPK-MB was one positive value on the first post-operative day, and mean LDH 1 was elevated to 51 0 units/ml on the 2nd post-operative day. These results indicate that protecting the myocardium with cold cardioplegia is superior to use of the anoxic cardiac arrest.

The Role of Somatostatin in Nociceptive Processing of the Spinal Cord in Anesthetized Cats

  • Somatostatin (SOM) is one of the major neuropeptides in dorsal root ganglion cells, but its role in spinal nociceptive process has not been well known. In present study we aimed to investigate the effect of SOM on the response of dorsal horn neurons to the various types of peripheral nociceptive stimuli in anesthetized cats. Using carbon-filament microelectrode, the single cell activities of wide dynamic range neurons were recorded from the lumbosacral enlargement after noxious mechanical (squeeze), thermal (radiant heat lamp) and cold (dry ice) stimulation to the receptive field. Sciatic nerve was stimulated electrically to evoke $A\;{\delta}-$ and C-nociceptive responses. SOM analogue, octreotide $(10\;{\mu}g/kg),$ was applied intravenously and the results were compared with those of morphine (2 mg/kg, MOR). Systemic SOM decreased the cellular responses to the noxious heat and the mechanical stimulation, but increased those to the cold stimulation. In the responses to the electric stimuli of sciatic nerve, $A\;{\delta}-nociceptive$ response was increased by SOM, while C-nociceptive response was decreased. On the other hand, MOR inhibited the dorsal horn cell responses to all the noxious stimuli. From the above results, it is concluded that SOM suppresses the transmission of nociceptive heat and mechanical stimuli, especially via C-fiber, while it facilitates those of nociceptive cold stimuli via $A\;{\delta}-fiber$.

An ESR Study of Amino Acid and Protein Free Radicals in Solution Part Ⅴ. an ESR Study of Gamma-Irradiated Lysozyme in Frozen Aqueous Solutions

  • An electron spin resonance study has been made on lysozyme in. frozen aqueous solutions irradiated with $_{60}Co$ r-rays in air at $77^{circ}K.$ Water resonances are dominant when the concentration and the temperature are both below 20% and $130^{circ}K$ respectively. More solute radicals are produced in the solution of higher concentration. Majority of the solute radicals results from direct hit of the radiation. The same types of radicals are induced at $77^{circ}K$ whether the substances are irradiated in the dry state or in frozen aqueous solution. Based on these results, it is assumed that the number of ESR centers produced by the secondary intermolecular radical reacions and stabilized in aqueous solutions may depend on the concentration of the solution, and the presence of water may facilitate the secondary radical reactions occuring in the solute molecules after heat treatment. Majority of the solute radicals above around $193^{circ}K$ are believed to react with oxygen to form peroxytype radicals. However, when the solution is subiected to heat-treatment at $265^{circ}K$ after irradiation at $195^{circ}K$ the peroxy-type resonance was not observed, suggestin that an appreciable amount of oxygen is condensed into the ice, at $77^{circ}K.$ in addition to the oxygen that has already been dissolved in solution and react with solute free radicals during the process of heat-treatment. When the solution contains $H_2O_2$, no water resonance but $HO_2$, type resonance was observed probably indicating that the radiation-induced OH radicals are trapped in $H_2O_2$ aggregates and react readily with $H_2O_2$ molecules to poroduce $HO_2$ type radicals even at $77^{circ}K.$.

Performance of Imported Sweet Corn Hybrids in Korea

  • The performance of 7 sugary (su) and 12 shrunken-2 (sh2) sweet com hybrids which are commercially grown in the United States was tested in Korea. The 100-seed weight of su hybrids (16.5-23.6 g) was much heavier compared to that of sh2 hybrids (10.9-17.5 g). The germination rate of su and sh2 hybrids at $25^{\circ}C$ ranged 93.3-100% and 86.7-98.9%, the emergence rate of su and sh2 hybrids in cold test ranged 78.9-97.8% and 62.2-97.8%, and field emergence rate of su and sh2 hybrids ranged 74.4-100.0% and 79.9-98.2%, respectively. In su hybrids, there was a significantly positive correlation between germination rate at $25^{\circ}C$ and emergence rate in cold test or early growth. In contrast, in sh2 hybrids seed weight was positively correlated with early plant growth, while not with the germination rate at $25^{\circ}C$ or emergence rate in cold test and field. Most sh2 hybrids produced larger and more marketable ears compared to su hybrids although there were significant differences among the hybrids in the same genotype. At harvest (24 days after pollination) soluble solids content of su hybrids (24.3-27.1 Brix %) was much higher than that of sh2 hybrids (13.8-18.0 Brix %), while total sugars of sh2 hybrids (21.4-28.6% on the dry weight basis) was much higher compared to su hybrids (2.4-15.9%). Considering germination and emergence rates, marketable ear production, and total sugar content, 'GCB 70' and 'Sweet Satin' in su hybrids and 'Ice Queen', 'Aspen', 'Sweet Magic', 'Bandit', 'Xtrasweet 82', 'Aspen', and 'Cambella 90' in sh2 hybrids performed better than other hybrids.
