A series of in vitro incubation studies with washed rumen bacteria were conducted to determine the influence of incubation time and concentrations of peptides, alanine, ammonia nitrogen and carbohydrate on the rate of peptide disappearance and on bacterial growth. Disappearance rate of tri-alanine (ala3) under various conditions was between 30.6 and $58.2mg\;hr^-$ per gram bacterial dry matter. Ala3 was removed from the incubation medium in an almost linear fashion as incubation time and ala3 concentration was increased. Washed rumen bacteria utilized ala3 faster than di-l-alanine (ala2) at all concentrations. Adding 9mM carbohydrate significantly increased ala3 disappearance, but level of ammonia nitrogen had no influence on ala3 disappearance. The presence of alanine in the medium significantly lowered ala3 utilization by rumen bacteria. Bacterial dry matter and nitrogen growth yield were not influenced by alanine and peptides when incubation medium already contained a sufficient level of ammonia nitrogen. Increased ammonia nitrogen in the presence of ala3 did not stimulate bacterial growth. Carbohydrate significantly increased bacterial dry matter and nitrogen growth as expected. Results indicate that the rate of peptide utilization by rumen bacteria may be altered by type and concentration of peptides, and energy supply, and this may be mediated through changes in numbers and type of bacteria.

A three year's field experiment was carried out on newly reclaimed tidal saline soils to evaluate the salt tolerance and growht characteristics, and their relationship to dry matter production and nutrient quality of main selected pasture species. Nine temperate grasses (14 varieties) and two forage crops (sorghum and pearl millet) were grown under different mulching treatments with medium sand and red-yellow soils (fine loamy materials of Typic Hapludults) from 1986 to 1988. Tall wheatgrass, tall fescue, reed canarygrass and alfalfa showed a good tolerance to soil salinity, especially tall wheatgrass (cv. Alkar) produced 19.6 ton/ha dry matter yield annualy under mulching treatment with medium sand depth in lcm. Pearl millet (cv. Gahi-3) was also evaluated as a salt tolerable forage species. Under salt stress in newly reclaimed tidal lands, plant showed a decrease in the assimirable leaf area (LA) as well as specific leaf area (SP. LA) and a low leaf weight ratio(LWR), and it resulted in a low concentration of crude protein and low digestible dry matter contents. Absorption of macro and micro elements in the plant on tidal lands was increased markedly.

This trial evaluated the effects of dietary supplementation with monensin sodium on dry matter intake, metabolic parameters and milk yield and milk composition of dairy Saanen goats in the periparturient period. Twelve Saanen pregnant dairy goats were assigned to a treatment and a control group. Saanen goats were fed monensin as its 10% sodium salt in the amount of 33 mg/kg of total dry matter intake during 30 d before parturition till d 42 in milk. Blood samples were collected from each goat at d 30, 15 and 7 before expected kidding time and also in d 1, 7, 13, 19, 21, 28, 35 and 42 postpartum. The serum concentrations of ${\beta}$-Hydroxybutyrate (BHBA), non-esterifed fatty acid (NEFA), calcium, magnesium, inorganic phosphorus, cholesterol, triglyceride, urea, total protein, albumin and glucose and aspartate aminotransferase (AST) activity were determined. The BHBA concentration significantly decreased in goats, which received monensin in comparison to controls in the postpartum period (p = 0.049). The concentration of sodium (Na) was significantly influenced by monensin treatment in the postpartum period (p = 0.048). In addition, the difference in dry matter intake was highly significant between the two groups during the pre-partum period. Controls had more dry matter intake (DMI) than treatment goats (p = 0.0001). Mean 3.5% fat corrected milk production was not influenced by monensin treatment. However, milk fat percentage was significantly decreased in monensin treated goats (p = 0.0017).

Winter green manure crops including legume increase grain yield of subsequent crop and substitute N fertilizer requirement with organic-No Hairy vetch grows vigorously and can provide N-rich green manure for corn with its soil incorporation after wintering. But, grain yield of corn as succeeding crop would be reduced if its planting time is delayed until late spring. This experiment was carried out to find the proper incorporation time of hairy vetch green manure and planting time of subsequent corn in cropping system with winter hairy vetch(green manure)-summer corn. Hairy vetch was incorporated into soil at a ten-day interval between April 10 and May 10 and corn was planted at 5 days after each hairy vetch incorporation. Soil nitrate concentration on April 10 and 20 in hairy vetch plot was slightly lower than that at winter fallow. Above-ground dry matter and organic-N of hairy vetch increased linearly with delayed hairy vetch incorporation time from April 10 to May 10. Average dry matter and organic-N produced by hairy vetch were 5.7 ton/ha and 248 kgN/ha, respectively. Corn growth and yield decreased as delayed corn planting time after May in spite of increasing dry matter and N-yield of hairy vetch. Nitrogen concentration of corn grain, stalk and whole plant at harvest were the highest in May 5 planting, but total N-uptake of May 5 planting were not different from that of April 25 planting because of lower grain yield. It was concluded that the proper incorporation time of hairy vetch and corn planting time were April 20 and April 25, respectively, because grain yield was the highest and corn could use hairy vetch-N effectively to produce dry matter.
Three wethers fitted with silastic catheters for collection of pancreatic juice, and cannulas located in the abomasum and the duodenum were used to investigate the effects of different hay and energy intake on pancreatic exocrine secretion. The wethers were fed Italian ryegrass hay or alfalfa hay at maintenance energy requirement and alfalfa hay ad libitum. High energy intake from alfalfa significantly increased abomasal flow of dry matter and both the concentration and daily secretion of ${\alpha}-amylase $. The high energy intake also tended to increase daily secretion of lipase, trypsin and chymotrypsin through the large volume of pancreatic juice. Compared with Italian ryegrass hay, alfalfa hay at the maintenance decreased abomasal dry matter flow, but increased concentration of ${\alpha}-amylase $ in the pancreatic juice, and tended to increase daily secretion of ${\alpha}-amylase $. The secretion of the other enzymes was not different between the two hays at maintenance intake. These results suggest that the kind of hay could change the concentration of ${\alpha}-amylase $ in the pancreatic juice, and that the intake level of alfalfa hay affects the ${\alpha}-amylase $ concentration and the juice volume secreted from the pancreas.

Sorghum cv. Pioneer 93 1, sorghum-sudangrass hybrid cv. Sioux and maize plant cv. Blizzard were assayed for toxic concentrations of nitrate-nitrogen ($NO_3$-N) and their relationship to morphological characteristics and environmental temperature in a field and phytotron trial. In the phytotron, sorghum and maize plants ranging from emergence to heading stage, were grown under different day/night temperatures of 30125, 25/20,28/18 and 1818 degree C. Nitrate-nitrogen in sorghum and maize plants was accumulated mainly in stems. Therefore nitrate concentration in the young plants was increased as development of stalks advanced and was highest at the stage of 3-4 leaves, when the plants had a leaf weight ratio 0.78-0.80 g/g plant weight. However, nitrate concentrations of the plant decreased as morphological development progressed, especially from the stage of growing point differentiation. Correlation coefficients showed a positive correlation of nitrate concentration with leaf weight ratio, leaf area ratio and specific leaf area, while plant height, dry matter percentage and absolute growth rate showed a negative association with TEX>$NO_3$-N ($P{\le}0.1$%). Cyanogenic glycosides, total nitrogen and crude protein were close associated with nitrate accumulation, and positively significant ($P{\le}0.1$%). High temperature over 30/25^{\circ}C.$ for 3 weeks increased N-uptake and dry matter accumulation, but reduced nitrate concentration. Under cold temperature below 18/8^{\circ}C.$ concentration of nitrate-N was increased in spite of its limited nitrogen uptake and plant growth.
The field observations on a seasonal characteristic of organic matter and its origin in the surface sediment were carried out at 35 stations in Deukryang bay, southern coast of Korean Peninsula from May 1995 to February 1996. The analytical parameters were mud temperature, ignition loss(IL), chemical oxygen demand(COD), pheopigment, sulfide and water content. The origin and seasonal dynamics of organic matter in Deukryang Bay were analyzed by COD/IL, COD/sulfide ratio and principal component analysis(PCA). As a results of the mud temperature fluctuated between 2.1$^{\circ}C$ with the lowest mean 4.6$^{\circ}C$ in winter and 27.6$^{\circ}C$ with the highest mean 25.5$^{\circ}C$ in summer. The range of ignition loss(IL) was from 3.1% in autumn to 21.5% in winter. Chemical oxygen demand(COD) showed the highest mean value of 8.45 mg/g dry in spring within the range of 2.90∼18.21 mg/g dry, while it showed the lowest value of 4.33 mg/g dry in autumn within the range of 0.67∼10.37 mg/g dry. Pheopigments showed the highest mean value of 9.04 $\mu\textrm{g}$/g dry in autumn within the range of 1.36∼20.44 $\mu\textrm{g}$/g dry, while it did the lowest mean value of 2.20 $\mu\textrm{g}$/g dry in summer within the range of 0.33∼11.36 $\mu\textrm{g}$/g dry. The range of total sulfide (H$_2$S) was from no detect(ND) to 3.30 mg/g dry in spring. And water content showed the annual mean value of 43.6% within the range of 23.6∼54.9%. The source of organic matter by COD/IL and COD/sulfide ratio in Deukryang Bay had been producted by primary producer in sea water areas except the areas effected by small stream, domestic and animal wastes. And the analytical results of PCA was able to be divided into three different regions. The former was characterized by the shallow depth and authigenic organic matter from phytoplankton in northwest area and northeastern inner bay, the secondary was done by deeper depth and allochthonous one from lands in southeast area and eastern entrance of bay, and the latter was done by authigenic one from the farm of seaweeds such as, sea cabbage, sea mustard etc in western entrance of bay. But a study on the relationship between sulfide and COD concentration in the northeastern inner bay which was characterized by the water stagnation will to take much more studying including major constituents of organic matter in the future.

The potential of the spot urine sampling technique as an alternative to performing a total urine collection to predict the microbial nitrogen supply was evaluated in crossbred bulls. In a completely randomized design, 20 growing crossbred bulls were assigned four levels of feed intake (120, 100, 80 and 60% of voluntary dry matter intake) on diets comprised of wheat straw and concentrate mixture (50:50). After three months of experimental feeding, a metabolism trial was conducted for ten days, during which spot urine collections were performed every 6 h post feeding on days 9 and 10. The daily urinary excretion of allantoin (A) and purine derivatives (PD) decreased with the reduction in feed intake while creatinine (C) excretion remained similar in animals fed at different levels. The microbial nitrogen (MN) supply calculated from the PD excreted in total urine (35.08 to 72.08 g/d) was higher at increased levels of feed intake. PD concentration in spot urine samples had poor correlation with feed intake except at 12 h post feeding. A/C ratio and PD/C ratio in spot urine samples remained similar irrespective of sampling time and significantly (p<0.01) correlated with daily urinary PD excretion, digestible organic matter intake and dry matter (DM) intake. However, no significant differences were evident in these ratios among animals fed at levels 120, 100 and 80% of voluntary dry matter intake (VDMI) at different times post feeding. These results suggests that the spot urine sampling technique to predict the microbial protein supply is not suitable for detecting small differences in MN supply and hence, estimation of PD excreted in total urine (mmol/d) is necessary to assess precisely the MN supply in crossbred bulls.

The effect of substituting potato by-products based silage (PBS) for concentrates on ruminal fermentation, methane production and nitrogen utilization in Holstein steers was evaluated. Three growing Holstein steers ($490{\pm}19kg$, initial body weight) were used in a $3{\times}3$ Latin square experiment with three experimental diets in which PBS was included at (1) 0, (2) 19 and (3) 27%, on a dry matter basis, replacing concentrates and wheat bran. Increasing replacement levels of PBS slightly increased dry matter (DM), organic matter (OM), NDF and ADF intakes by the steers (p<0.05). Inclusion of PBS at 19% increased crude protein (CP) digestibility of the experimental diets compared with the control (p<0.05). Substitution with PBS increased ADF digestibility and nitrogen retention (p<0.05), but did not affect energy retention. Energy loss as methane ranged between 5.0 and 6.1% of the total gross energy intake. There were no significant differences in carbon dioxide and methane production among all PBS levels, while daily methane production numerically increased with PBS inclusion. Substituting PBS for concentrates did not significantly affect ruminal pH and ammonia N concentration. Total VFA concentration, VFA molar proportions and blood metabolites were also unaffected by PBS replacement. These results suggest that substitution of PBS up to 27% of diet dry matter did not significantly increase methane production and was equal or superior to concentrates in ADF digestibility and nitrogen retention for growing steers.

A two year's field experiment was conducted on newly reclaimed saline tidal lands to measure the effects of sand mulching on salinity of the soils and their relationship to crop performance. Hybrid sorghum cv. Pioneer 931 was grown under different mulching treatments using of medium sand and red earth (Fine loamy, Typic Hapludults). Salinity in the root zone was decreased markedly under soil mulching using of medium sand, especially during the dry season, and it caused a great increase in the root growth and R/T ratio. Seasonal values of electrical conductivity at sand mulching were 6.6 in April and 1.6 mmhos in August, but it was still high with a concentration of 12.7 (April) and 3.8 mmhos (August) in untreated check plot. Sand mulching increased plant growth and the rate of dry matter accumulation. However, treatment of red earth additionally over sand mulching produced lower dry matter yield than those of soil mulching using of medium sand only. Under salt stress sorghum plant showed a decrease in the leaf weight ratio (LWR) and it resulted in a low concentration of crude protein of the plant. Sand mulching enhanced leaf weight ratio and rate of protein synthesis.
