GWAS of Salt Tolerance and Drought Tolerance in Korean Wheat Core Collection

  • Abiotic stress is a major problem in global agriculture as it negatively affects crop growth, yield, and quality. Wheat (Triticum aestivum) is the world's second-highest-producing food resource, so the importance of mitigating damage caused by abiotic stress has been emerging. In this study, we performed GWAS to search for SNPs associated with salt tolerance and drought tolerance. NaCl (200 mM) treatment was performed at the seedling stage using 613 wheat varieties in Korean wheat core collection. Root length, root surface area, root average diameter, and root volume were measured. Drought stress was applied at the seedling stage, and the above phenotypes were measured. GW AS was performed for each phenotype data using the MLM, MLMM, and FarmCPU models. The best salt-tolerant wheat varieties were 'MK2402', 'Gyeongnam Geochang-1985-3698', and 'Milyang 13', showing superior root growth. The significant SNP AX-94704125 (BA00756838) were identified in all models. The genes closely located to the significant SNP were searched within ± 250 kb of the corresponding SNP. A total of 11 genes were identified within the region. NB-ARC involved in the defense response, FKSI involved in cell wall biosynthesis, and putative BP Ml involved in abiotic stress responses were discovered in the 11 genes. The best drought-tolerant wheat varieties were 'PI 534284', 'Moro of Sind', and 'CM92354-33M-0Y-0M-6Y-0B-0BGD', showing superior root growth. This study discovered SNPs associated with salt tolerance in Korean wheat core collection through GWAS. GWAS of drought tolerance is now proceeding, and the GWAS results will be represented on a poster. The SNPs identified by GWAS can be useful for studying molecular mechanisms of salt tolerance and drought tolerance in wheat.

Drought Tolerance in Italian Ryegrass is Associated with Genetic Divergence, Water Relation, Photosynthetic Efficiency and Oxidative Stress Responses

  • Drought stress is a condition that occurs frequently in the field, it reduces of the agricultural yield of field crops. The aim of the study was to screen drought-adapted genotype of Italian rye grass. The experiments were conducted between the two Italian ryegrass (Lolium multiflorum L.) cultivars viz. Hwasan (H) and Kowinearly (KE). The plants were exposed to drought for 14 days. The results suggest that the morphological traits and biomass yield of KE significantly affected by drought stress-induced oxidative stress as the hydrogen peroxide (H2O2) level was induced, while these parameters were unchanged or less affected in H. Furthermore, the cultivar H showed better adaptation by maintaining several physiological parameter including photosystem-II (Fv/Fm), water use efficiency (WUE) and relative water content (RWC%) level in response to drought stress. These results indicate that the cultivar H shows improved drought tolerance by generic variation, improving photosynthetic efficiency and reducing oxidative stress damages under drought stress. These findings can be useful to the breeder and farmer for improving drought tolerance in Italian rye grass through breeding programs.

Overexpresssion of the OsbZIP66 transcription factor enhances drought tolerance of rice plants

  • Drought stress is a major constraint of crop development and productivity. Plants have evolutionally developed several mechanisms at the molecular, cellular, and physiological levels to overcome drought stress. The basic Leucine zipper (bZIP) transcription factor (TF) family members are starting to be concerned about their roles in drought stress responses. In this study, we functionally characterized OsbZIP66, a rice group-E bZIP TF, to be associated with rice drought tolerance mechanisms. Expression of OsbZIP66 was significantly induced upon treatments of rice plants with drought, high salinity, and ABA. These observations and the fact that the OsbZIP66 promoter contains ten ABA-responsive elements suggest that OsbZIP66 is up-regulated by drought stress in an ABA-dependent manner. Overexpression of both OsbZIP66 in a whole plant body and specifically in roots enhanced drought tolerance of rice plants, indicating that the rice drought tolerance positively correlates with the expression levels of OsbZIP66. Thus, our results demonstrated that OsbZIP66 has a potential for use in biotechnological development of high-yielding rice plants under drought conditions.

Screening methods for drought and salinity tolerance with transgenic rice seedlings

  • Abiotic stress is one of the major serious limiting factors in rice (Oryza sativa) and caused rice production losses. It is important to precisely screen valuable genetic resources for improving stress tolerance and understanding tolerance mechanism to abiotic stresses. Because there are differences of experiment designs for screening of tolerant plant in several studies related to abiotic stress, this study has performed to provide the rapid and efficiency screening method for selection of tolerance rice to drought and salinity stresses. Two week-old rice seedlings that reached about three leaf stage were treated with drought and salinity stresses and examined tolerant levels with tolerant and susceptible control varieties, and transgenic plants. To determine the optimum concentration for the selection of drought and salinity condition, tolerant, susceptible and wild-type plants were grown under three soil moisture contents (5, 10 and 20% water contents) and three NaCl concentrations (100, 200 and 250 mM) for 10 days at seedling stage. 200 mM NaCl concentration and 5% moisture content soil were determined as the optimum conditions, respectively. The described methodologies in this study are simple and efficiency and might help the selection of drought and salinity tolerance plants at the 3,4-leaf-seedling stage.

P-V 곡선법을 활용한 땃두릅나무의 내건성 평가 (Evaluation of Drought Tolerance of Oplopanax elatus Obtained from Pressure-Volume Curves)

  • This study was carried out to establish a proper cultivation site and to diagnose the drought tolerance of Oplopanax elatus leaves by using pressure-volume curves. As a result of analysing data measured, the leaf of Oplopanax elatus showed the osmotic pressure at full turgor(Ψosat) was -0.77 MPa, and the osmotic pressure at incipient plasmolysis(Ψotlp) was -0.90 MPa. Then, the value of maximum bulk modulus of elasticity Emax was 3.7 MPa, showing that slightly lower drought tolerance of Oplopanax elatus. Furthermore, the values of relative water contents RWCtlp and RWC* were above 80%, showing that the function of osmoregulation is somewhat better. Thus, responses to water relations such as Ψosat, Ψotlp, Emax, RWCtlp and RWC* of Oplopanax elatus showed relatively lower drought-tolerance property indicating that those growth are appropriate in high moisture soil sites.

Overexpression of OsNAC17 enhances drought tolerance in rice

  • Drought conditions during cultivation reduce agricultural production yield less than a theoretical maximum yield under normal condition. Plant specific NAC transcription factors in rice are known to play an essential roles in stress resistance transcriptional regulation. In this study, we report the rice (Oryza sativa L japonica) NAM, AFTF and CUC transcription factor OsNAC17, which is predominantly induced by abiotic stress in leaf, was contribute to the drought tolerance mediated reactive oxygen species (ROS) in transgenic rice plants. Constitutive (PGD1) promoter was introduced to overexpress OsNAC17 and produced the transgenic PDG1:OsNAC17. Overexpression of OsNAC17 throughout the whole plant improved drought resistance phenotype at the vegetative stage. Morphological characteristics such as grain yield, grain filling rate, and total grain weight improved by 22~64% over wild type plants under drought conditions during the reproductive stage. The improved drought tolerance in transgenic rice was involved in reducing stomatal density up to 15% than in wild type plants and in increasing reactive oxygen species-scavenging enzyme. DEG profiling experiment identified 119 up-regulated genes by more than twofold (P<0.01). These genes included UDP-glycosyltransferase family protein, similar to 2-alkenal reductase (NADPH-dependent oxireductase), similar to retinol dehydrogenase 12, Lipoxygenase, and NB-ARC domain containing protein related in cell death. Furthermore, OsNAC17 was act as a transcriptional activator, which has an activation domain in C-terminal region. These result demonstrate that the overexpression of OsNAC17 improve drought tolerance by regulating ROS scavenging enzymes and by reducing stomatal density

Transcriptomic Insights into Abies koreana Drought Tolerance Conferred by Aureobasidium pullulans AK10

  • The conservation of the endangered Korean fir, Abies koreana, is of critical ecological importance. In our previous study, a yeast-like fungus identified as Aureobasidium pullulans AK10, was isolated and shown to enhance drought tolerance in A. koreana seedlings. In this study, the effectiveness of Au. pullulans AK10 treatment in enhancing drought tolerance in A. koreana was confirmed. Furthermore, using transcriptome analysis, we compared A. koreana seedlings treated with Au. pullulans AK10 to untreated controls under drought conditions to elucidate the molecular responses involved in increased drought tolerance. Our findings revealed a predominance of downregulated genes in the treated seedlings, suggesting a strategic reallocation of resources to enhance stress defense. Further exploration of enriched Kyoto Encyclopedia of Genes and Genomes pathways and protein-protein interaction networks revealed significant alterations in functional systems known to fortify drought tolerance, including the terpenoid backbone biosynthesis, calcium signaling pathway, pyruvate metabolism, brassinosteroid biosynthesis, and, crucially, flavonoid biosynthesis, renowned for enhancing plant drought resistance. These findings deepen our comprehension of how AK10 biostimulation enhances the resilience of A. koreana to drought stress, marking a substantial advancement in the effort to conserve this endangered tree species through environmentally sustainable treatment.

qVDT11, a major QTL related to stable tiller formation of rice under drought stress conditions

  • Drought is the most serious abiotic stress limiting rice production. However, little progress has been made in the genetic analysis of drought tolerance, because it is a complex trait controlled by a number of genes and affected by various environmental factors. In here, we screened 218 rice genetic resources for drought tolerance at vegetative stage and selected 32 highly drought tolerant varieties in greenhouse. Under rain-fed conditions, Grain yield of Nagdong was decreased by 53.3% from 517 kg/10a to 241 kg/10a when compare to irrigation condition. By comparison, grain yield of Samgang was decreased by 23.6% from 550 kg/10a to 420 kg/10a. The variety Samgang exhibited strong drought tolerance and stable yield in rain-fed conditions and was selected for further study. To identify QTLs for drought tolerance, we examined visual drought tolerance (VDT) and relative water content (RWC) using a doubled haploid (DH) population consisted of 101 lines derived from a cross between Samgang (a drought tolerance variety) and Nagdong (a drought sensitive variety). Three QTLs for VDT were located on chromosomes 2, 6, and 11, respectively, and explained 41.8% of the total phenotypic variance. qVDT2, flanked by markers RM324 and S2016, explained 8.8% of the phenotypic variance with LOD score of 3.3 and an additive effect of -0.6. qVDT6 was flanked by S6022 and S6023 and explained 12.7% of the phenotypic variance with LOD score of 5.0 and an additive effect of -0.7. qVDT11, flanked by markers RM26765 and RM287, explained 19.9% of the phenotypic variance with LOD score of 7.1 and an additive effect of -1.0. qRWC11 was the only QTL for RWC to be identified; it was in the same locus as qVDT11. qRWC11 explained 19.6% of the phenotypic variance, with a LOD score of 4.0 and an additive effect of 9.7. To determine QTL effects on drought tolerance in rain-fed paddy conditions, seven DH lines were selected according to the number of QTLs they contained. Of the drought tolerance associated QTLs, qVDT2 and qVDT6 did not affect tiller formation, but qVDT11increased tiller number. Tiller formation was most stable when qVDT2 and qVDT11 were combined. DH lines with both of these drought tolerance associated QTLs exhibited the most stable tiller formation. These results suggest that qVDT11 is important for drought tolerance and stable tiller formation under drought stress condition in field.

Over-expression of OsHsfA7 enhanced salt and drought tolerance in transgenic rice

  • Heat shock proteins play an important role in plant stress tolerance and are mainly regulated by heat shock transcription factors (Hsfs). In this study, we generated transgenic rice over-expressing OsHsfA7 and carried out morphological observation and stress tolerance assays. Transgenic plants exhibited less, shorter lateral roots and root hair. Under salt treatment, over-expressing OsHsfA7 rice showed alleviative appearance of damage symptoms and higher survival rate, leaf electrical conductivity and malondialdehyde content of transgenic plants were lower than those of wild type plants. Meanwhile, transgenic rice seedlings restored normal growth but wild type plants could not be rescued after drought and re-watering treatment. These findings indicate that over-expression of OsHsfA7 gene can increase tolerance to salt and drought stresses in rice seedlings.

A Trifloxystrobin Fungicide Induces Systemic Tolerance to Abiotic Stresses

  • Trifloxystrobin is a strobilurin fungicide, which possesses broad spectrum control against fungal plant diseases. We demonstrated that pre-treating red pepper plants with trifloxystrobin resulted in increased plant growth and leaf chlorophyll content compared with those in control plants. Relative water content of the leaves and the survival rate of intact plants indicated that plants acquired systemic tolerance to drought stress following trifloxystrobin pre-treatment. The recovery rate by rehydration in the drought treated plant was better in those pre-treated with trifloxystrobin than that in water treated plants. Induced drought tolerance activity by trifloxystrobin was sustained for 25 days after initial application. The trifloxystrobin treated red pepper plants also had induced systemic tolerance to other abiotic stresses, such as frost, cold, and high temperature stresses. These findings suggest that applying the chemical fungicide trifloxystrobin induced systemic tolerance to certain abiotic stresses in red pepper plants.
