Development of a Fast Neutron Detector (속중성자 탐지용 반도체 소자 개발)

  • When a Si PIN diode is exposed to fast neutrons, it results in displacement damage to the Si lattice structure of the diode. Defects induced from structural dislocation become effective recombination centers for carriers which pass through the base of a PIN diode. Hence, increasing the resistivity of the diode decreases the current for the applied forward voltage. This paper involves the development of a neutron sensor based on the phenomena of the displacement effect damaged by neutron exposure. The neutron effect on the semiconductor was analyzed. Several PIN diode arrays with various thickness and cross-section area of the intrinsic layer(I layer) were fabricated. Under irradiation tests with a neutron beam, the manufactured diodes have a good linearity to neutron dose and show that the increase of thickness of I layer and the decrease of cross-section of PIN diodes improve the sensitivity. Newly developed PIN diodes with thicker I layer and various cross section, were retested and then showed the best neutron sensitivity at the condition that the I layer thickness was similar to a side length. On the basis of two test results, final discrete PIN diodes with a rectangular shape were manufactured and the characteristics as neutron detectors were analyzed through the neutron beam test using on-line electronic dosimetry system. Developed PIN diode shows a good linearity as dosimetry in the range of 0 to 1,000cGy(Tissue) and its neutron sensitivity is 13mV/cGy at constant current of 5mA, that is three times higher than that of commercially available neutron detectors. And the device shows little dependency on the orientation of the neutron beam and a considerable stability in annealing test for a long period.

Mechanism of Intestinal Transport of an Organic Cation, Tributylmethylammonium in Caco-2 Cell Monolayers

  • Many quaternary ammonium salts are incompletely absorbed after their oral administration and may also be actively secreted into the intestine. However, the underlying mechanism(s) that control the transport of these cations across the intestinal epithelium is not well understood. In this study, the mechanism of absorption of quaternary ammonium salts was investigated using Caco-2 cell monolayers, a human colon carcinoma cell line. Tributylmethylammonium (TBuMA) was used as a model quaternary ammonium salts. When TBuMA was administrated at a dose of 13.3 imole/kg via iv and oral routes, the AUC values were $783.7{\pm}43.6\;and\;249.1{\pm}28.0{\mu}mole\;min/L$ for iv and oral administration, indicating a lower oral bioavailability of TBuMA $(35.6\%)$. The apparent permeability across Caco-2 monolayers from the basal to the apical side was 1.3 times (p<0.05) greater than that from the apical to the basal side, indicating a net secretion of TBuMA in the intestine. This secretion appeared to be responsible for the low oral bioavailability of the compound, probably mediated by p-gp (p-glycoprotein) located in the apical membrane. In addition, the uptake of TBuMA by the apical membrane showed a $Na^+$ dependency. Thus, TBuMA appears to absorbed via a $Na^+$ dependent carrier and is then secreted via p-gp related carriers.

  • In order to evaluate the potential utilization value as a novel probiotic strain, the immunopotentiating activities of the cellular components from Lactobacillus brevis FSB-1 were examined. L. brevis FSB-1 isolated from kimchi were fractionated into the whole cell, cell wall, cytosol and extracellular preparation, and each fraction was examined on intestinal immune system modulating activity in vitro. The cell wall and cytosol preparation showed the relatively high bone marrow cell proliferating activity through Peyer's patch cell in a dose-dependent manner. But these preparations did not directly stimulate the bone marrow cell proliferation. The whole cell, cell wall and cytosol preparation also induced considerable levels of macrophage activation and mitogenicity of murine splenocytes in vitro. The anti-complementary activity (ITCH_(50)) of the cytosol fraction of L. brevis FSB-1 was the most potent in the cellular components, and the activity showed dose dependency. The complement activation by the cytosol fraction of L. brevis FSB-1 occurs via both alternative and classical pathways, which confirmed by the crossed immunoelectrophoresis using anti-human C3.

Measurement of Ir-192 Source Activity for High Dose Rate Brachytherapy (고 선량률 근접치료시 사용되는 Ir-192 선원의 방사능 평가)

  • Ir-192 source activity for high dose rate brachytherapy is measured using Farmertype ionization chamber. The source-to-chamber distance is 10 cm and the measured charge unit is converted to activity unit. The measured values are compared to the values provided from vendor. Because of time dependency of Ir-192 source activity, the activities are regularly checked and compared to calculated values. As the accuracy of Ir-192 source activity is depend on the mechanical measurement setup, we estimated the precision of remote controlled source dwell position using home-made device and film scanner. The difference between measured and predicted dwell position is within 1 mm. As a result, the errors of source activity are 0.7${\pm}$1.5 % for measured and vendor-provided values and 0.l${\pm}$1.2% for measured and time-dependent calculated vlaues. In conclusion, our measured activity has been comparable to the values provided from vendor and our brachytherapy unit has been very accurate until now. Regular quality control of brachytherapy is essential for successful treatment which depends on the accuracy of source position and activity.

Antibacterial and Growth Inhibitory Effects of Liriope Platyphylla Ethanol Extract on Streptococcus Mutnas and Porphyromonas Gingivalis (맥문동 에탄올 추출물(Liriope platyphylla ethanol extract)의 Streptococcus mutnas와 Porphyromonas gingivalis에 대한 항균력과 성장억제 효과)

  • Purpose : This study aimed to analyze the antibacterial activity of Liriope platyphylla ethanol extract (LPEE) against Streptococcus mutans and Porphyromonas gingivalis and to validate its potential for the prevention and treatment of dental caries, gingivitis, and periodontal disease. Methods : To verify the antibacterial effect of L. pulsatilla ethanolic extract (LPEE) against S. mutans and P. gingivalis, the disk diffusion method was used to determine the inhibition zones at concentrations of 50, 100, 200, and 300 mg/㎖. To determine the minimum inhibition concentration (MIC), the final dose of LPEE was .2, .4, .8, 1.6, 2.5, and 5.0 mg/㎖, and the minimum bactericidal concentration (MBC) was determined based on the MIC results. To confirm the growth inhibitory effect of LPEE on both pathogens, the absorbance was measured at 600 nm after each incubation for 0, 3, 6, 12, and 24 hr at concentrations of .8, 1.6, 2.5, and 5.0 mg/㎖. Results : The cytotoxicity of LPEE was evaluated and the cell viability was more than 70 % at 400 mg/㎖. Therefore, concentrations of 50, 100, 200, and 300 mg/㎖ were used in this study. The antimicrobial effect against S. mutans was seen at 100 mg/㎖ and grew in a concentration-dependent manner, while P. gingivalis was effective at 50 mg/㎖ with the dose dependency. The MIC was .8 mg/㎖ for both strains, and the MBC was 1.6 mg/㎖ with the same results. The growth inhibitory effect of LPEE on S. mutans and P. gingivalis was observed, even at low concentrations. Conclusion : The antibacterial effect of LPEE was evaluated through the analysis of MIC, MBC, and growth inhibition effect on S. mutans and P. gingivalis, which suggests LPEE might have the possibility of utilization as a preventive and therapeutic composition for oral diseases.

Radiation-induced Apoptosis in Developing Fetal Rat Cerebral Cortex (발육 중의 백서 태아 대뇌 피질에서 방사선에 의한 아포토시스)

  • Purpose: This study was peformed to Investigate apoptosis by radiation In the developing fetal rat brain. Materials and Methods: Fetal blains were Irradiated In utero between the 17th and 19th days of fetal life (El7-19) by linear accelerator. A dose of Irradiation ranging from 1 Gy to 4 Gy was used to evaluate dose dependency. To test time dependency the ra)s were Irradiated with 2 Gy and then the fetal brain specimens were removed at variable 41me course; 1, 3, 5, 12 and 24 hours after the onset of irradiation. Immunohistochemlcal staining using in situ 707-mediated dUTP nick end labelling (TUNEL) technlfue was used for apoptotic cells. The cerebral cortex, including three zones on coriicai zone (Cf). Intermediate zone (if), and ventricular zone (VZ), was examined. Results : TUNEL positive cells revealed typical features of apoptotic cells under light microscope In the fetal rat cerebral cortex. Apoptotic cells were not found In the cerebral cortex of non-Irradiated fetal rats, but did appear In the entire cerebral cortex after 1 Gy Irradiation, and were more expensive at the ventricular and Intermediate zones than at the cortical zone. The extent of apoptosis was Increased with Increasing doses of radiation. Apoptosis reached the peak at S hours after the onset of 2 Gy Irradiation and persisted until 24 hours. Conclusion: Typical morphological features of apoplosis by irradiation were observed In the developing fetal rat cerebral cortex. It was more extensive at the ventricular and Intermediate zones than at the cortical zone, which suggested that stem cells or early differentiated cells are more radiosensitive than differentiated cells of the cortical zone.

Development of Phantom and Comparison Analysis for Performance Characteristics of MOSFET Dosimeter (MOSFET 선량계 특성분석을 위한 팬톰 개발 및 특성 비교)

  • This study is to develope a phantom for MOSFET (Metal Oxide Semiconductors Field Effect Transistors) dosimetry and compare the dosimetric properties of standard MOSFET and microMOSFET with the phantom. In this study, the developed phantom have two shape: one is the shape of semi-sphere with 10cm diameters and the other one is the flat slab of $30{\times}30cm$with 1 cm thickness. The slab phantom was used for calibration and characterization measurements of reproducibility, linearity and dose rate dependency. The semi-sphere phantom was used for angular and directional dependence on the types of MOSFETs. The measurements were conducted under $10{\times}10cm^2$ fields at 100cm SSD with 6MV photon of Clinac (21EX, Varian, USA). For calibration and reproducibility, five standard MOSFETS and microMOSFETs were repeatedly Irradiated by 200cGy five times. The average calibration factor was a range of $1.09{\pm}0.01{\sim}1.12{\pm}0.02mV/cGy$ for standard MOSFETS and $2.81{\pm}0.03{\sim}2.85{\pm}0.04 mV/cGy$ for microMOSFETs. The response of reproducibility in the two types of MOSFETS was found to be maximum 2% variation. Dose linearity was evaluated In the range of 5 to 600 cGy and showed good linear response with $R^2$ value of 0.997 and 0.999. The dose rate dependence of standard MOSFET and microMOSFET was within 1% for 200 cGy from 100 to 500MU/min. For linearity, reproducibility and calibration factor, two types of MOSFETS showed similar results. On the other hand, the standard MOSFET and microMOSFET were found to be remarkable difference in angular and directional dependence. The measured angular dependence of standard MOSFET and microMOSFET was also found to be the variation of 13%, 10% and standard deviation of ${\pm}4.4%,\;{\pm}2.1%$. The directional dependence was found to be the variation of 5%, 2% and standard deviation of ${\pm}2.1%,\;{\pm}1.5%$. Therefore, dose verification of radiation therapy used multidirectional X-ray beam treatments allows for better the use of microMOSFET which has a reduced angular and directional dependence than that of standard MOSFET.

Clinical Application of Wedge Factor (Wedge Factor의 임상적 응용)

  • Purpose : In general, the wedge factors which are used clinical practices are ignored of dependency on field sizes and depths. In this present, we investigated systematically the depth and field size dependency to determine the absorbed dose more accurately. Methods : The wedge factors for each wedge filter were measured at various depths (depth of Dmax, 5cm, 10cm, and 15cm) and field sizes ($5cm{\times}5cm,\;10cm{\times}10cm,\;15cm{\times}15cm, and 20cm{\times}20cm$) by using 4-, 6-, and 10-MVX rays. By convention, wedge factors are determined by taking the ratio of the central axis ionization readings when the wedge filter is in place to those of the open field in same field size and measurement depth. In this present work, we determined the wedge factors for 4-, 6-, and 10-MV X rays from Clinac 600C and 2100C linear accelerators (manufactured by Varian Associates, Inc., Palo Alto, CA). To confirm that the wedge was centered, measurements were done with the two possible wedge position and various collimator orientations. Results : The standard deviations of measured values are within $0.3\;\%$ and the depth dependence of wedge factor is greater for the lower energies. Especially, the variation of wedge factor is no less than $5\%$ for 4- and 6- MV X rays with more than $45^{\circ}$ wedge filters. But there seems to be a small dependence on field size. Conclusion : The results of this study show a dependence on the point of measurement. There also seems to be a small dependence on field size. And so, we should consider the depth and field size dependence in determining the wedge factors. If one wedge factor were to be used for each wedge filter it seems that the measurement for a 10cm x 10cm field size at a depth of loom would be a reasonable choice.

Inhibitory Potential of Thelephoric Acid on CYP2J2 Activities in Human Liver Microsomes (Thelephoric acid의 CYP2J2 효소 활성 저해제 평가)

  • Cytochrome P450 2J2 (CYP2J2) is an enzyme mainly found in human extrahepatic tissues, with predominant expression in the cardiovascular system. CYP2J2 plays important roles in the metabolism of endogenous metabolites and therapeutic drugs, such as arachidonic acid, astemizole, ebastine, and terfenadine. CYP2J2 is also overexpressed in human cancer tissues and cancer cell lines and may represent a potential target for therapy of human cancers. In this study, 10 natural products obtained from plants and microorganisms were screened as potential CYP2J2 inhibitors. Among them, thelephoric acid showed strong inhibition of astemizole O-demethylation activity ($IC_{50}=3.23{\mu}M$) in a dose-dependent manner. Evaluation of the substrate dependency of the inhibitory activity of thelephoric acid showed that it strongly inhibited CYP2J2-mediated ebastine hydroxylation ($IC_{50}=5.32{\mu}M$) and terfenadine hydroxylation ($IC_{50}=3.27{\mu}M$) in a substrate nondependent manner. The present data suggest that this compound might be a potential candidate for further evaluation for anticancer activity.

A Study on the trabecular change of Femur according to $17{\beta}-Estradiol$ Dosage in Ovariectomized Rat (난소 절제된 백서에서 에스트로젠 투여용량에 따른 대퇴골주 변화에 대한 연구)

  • Osteoporosis is the consequence of an imbalance between osteoclastic and osteoblastic activity, coupled with an increased rate of bone turnover observed with menopause. Estrogen is generally considered to maintain bone mass through suppression of bone resorption. The purpose of this study was to evaluate the rat femoral trabecular change not only in the deficiency of estrogen but also in the administration of estrogen following ovariectomy(OVX). 30 female Sprague-Dawley rats were subjected to bilateral OVX or sham surgery(control). Groups of OVX were divided into 4 groups. The first group was injected daily with vehicle alone for 20 days after 20 weeks following OVX. The additional groups of OVX was injected daily with low, medium, or high doses of $17{\beta}-estradiol$(10, 25 or $50{\mu}g/kg$ BW, respectively). All rats were sacrified 23 weeks after OVX, and their femur were processed for H&E, MT stain and histomorphometry. The results were as follows; 1. In the histomorphometric analysis, the trabecular bone volume/tissue volume, trabecular thickness and trabecular seperation were respectively $31.2{\pm}8.3%$, $54.3{\pm}4.8{\mu}m$ and $280.7{\pm}16.4{\mu}m$ in vehicle treated OVX group and $48.6{\pm}7.3%$, $90.4{\pm}4.5{\mu}m$ and $126.3{\pm}5{\mu}m$ in sham operation group, and they showed statistical significance compare to control group. 2. The trabecular bone volume/tissue volume, trabecular thickness and trabecular separation were respectively $44.4{\pm}4.3%$, $109.5{\pm}12.3{\mu}m$ and $94.9{\pm}8.5{\mu}m$ in low doses of $17{\beta}-estradiol$ injected group and they showed statistical significance compare to OVX group. 3. The trabecular bone volume/tissue volume, trabecular thickness and trabecular separation were respectively $44.4{\pm}4.3%$, $109.5{\pm}12.3{\mu}m$ and $94.9{\pm}8.5{\mu}m$ in medium doses of $17{\beta}-estradiol$ injected group and they showed statistical significance compare to OVX group, but they didn't show statistical significance compare to low doses of $17{\beta}-estradiol$ injected group. 4. The trabecular bone volume/tissue volume, trabecular thickness and trabecular separation were respectively $46.4{\pm}4.5%$, $154.4{\pm}13.2{\mu}m$ and $113.7{\pm}12.8{\mu}m$ in high doses of $17{\beta}-estradiol$ injected group and they also showed statistical significance compare to OVX group, but they didn't show statistical significance compare to other experimental groups. From the above results, metaphyseal bone formation was markedly reduced in OVX rate but treatment of OVX rats with $17{\beta}-estradiol$ resulted in normalization of femur trabecular bone volume. But they didn't show statistical significance the effect of bone formation according to the dose dependency.
