방풍갈근탕(防風葛根湯)의 안전성에 관한 연구 (Study on Safety of Bangpung-galgeun-tang)

  • The purpose of this study was to examine to observe single and four weeks repeated toxicity in mice of Bangpung-galgeun-tang (BGT). We investigated to ascertain safety and toxicity of BGT, we divided into single and four weeks repeated administration test. In single test, three groups were administrated different dosages and routes (2 g/kg/i.p., 4 g/kg/i.p. and 15 g/kg/p.o.) of BGT, and in four weeks repeated test, 0.8 g/kg BGT was administrated. Control groups were administrated with only saline according to on Korean Food and Drug Administration, respectively. We observed attentively motality, abnormal clinical sign, body weight change, organ weight, AST and ALT of mice after BGT administration. During toxicity experiment period, there was no difference in body weight change, organ weight, AST and ALT among different dose groups. Death were found 3 mice from day 2 to day 3 in single test i.p. group. (2 g/kg, 4 g/kg). Several individuals of single test i.p. group were observed that decreased locomotor activity, exophthalmos, bloodshot eyes, loss of eyesight and so on in early period after administration. But there was no difference in clinical signs among p.o. group. These results indicate that BGT have inhibition effects on allergy and suggest that no observable effect level of the test orally administration was considered to be more than 2 g/kg in mice under the conditions employed in this study.

NEUROTOXICITY OF TRIMETHYLTIN IN HIPPOCAMPUS: A HYPEREXCITATORY TOXICITY

  • Trimethyltin (TMT) induced lesions in the rat hippocampal formation was reviewed. Adult rats were treated with a single dose of 6.0 mg TMT/kg b.w. and were sacrificed between 3-60 days following exposure. On the hippocampal formation, the granule cells of fascia dentata showed early changes which subsided considerably at a later time when the destruction of the pyramidal neurons of the Ammon's horn became increasingly pronounced with time, leading to severe destruction of the structure. It is interesting to note that there was an inverse relationship of pathological involvement between the f.d. granule cells and the Ammon's horn neurons; i.e., when there was a large sparing of the granule cells. there was an extensive damage to the Ammon's horn and vice versa. This inverse relationship was also true between the $CA_3$neurons and the $CA_{1,2}$neurons in the Ammon's horn. Progressive zinc loss, as demonstrated by Timm's method, on the Mossy fibers was also observed. Similar Mossy fiber zinc depletion has been demonstrated in electrical stimulatory excitation condition of the perforant path to the hippocampus. Depletion of corticosterone, an inhibitor to the hippocampal neurons, by means of adrenalectomy will exaggerate the TMT induced hippocampal lesion. Neonatal study revealed that a unique degenerative pattern of the Ammon's horn could be established in accordance with exposure to TMT at specific maturation periods of the fippocampal formation: increasing destruction of the Ammon's horn with increasing synaptogenesis between the f.d. granule cells and the Ammon's horn neurons. Thus it is apparent that the damage of the Ammon's horn, upon exposure to TMT, may depend on the integrity and functional state of the f.d. granule cells. A hyperexcitory scheme and mechanism as the toxicity basis of TMT in the hippocampal formation is proposed and discussed.

닭에 있어서 닭와포자충 감염이 파브리시우스낭에 미치는 영향 (Effects of Cryptospoyidium bnileyi infection on the bursa of Fabricius in chickens)

  • 닭에 있어서 닭와포자충 갉염이 면역반응에 미치는 영향을 규명하기 위한 기초적 연구리 일환으로 파브리시우스낭의 병리조직학적 소견을 경시적으로 조사하고자 150마리의 2일령 SPF 병아리 (Drkalb-Warren, Sex-Sal-Link)에 닭와포자충의 오오시스트 $5{\;}{\times}{\;}10^5$를 한 번에 경구투여하였타. 분변 속의 오오시스트 배설양상은 정상적이었으며. 파브리시우스낭 지수는 전 실험기간에 걸쳐 거의 변동이 없얹다 많은 수의 원충체가 접종 후 4-16일에 이 낭상피의 미세융또 가장자리에서 관찰되었으며 많은 수의 비만세포가 출현한 다음 원충체가 급격하게 소실하염다. 원충체의 분포상황과 상피 및 인접점막 고유층의 위호산구 침윤은 일치하였다. 이 병소는 상피. 인접 점막고유층의 위호산구 침율과 점만상피 증식을 동반한 미만성 만성 표재성 화농성 파브리시우스낭영의 병리조직학적 소견이었다 이러한 파브리시우스낭염은 면역억제를 유발할 건으로 생각된다.

A Dunnione Compound MB12662 Improves Cisplatin-Induced Tissue Injury and Emesis

  • The present study was aimed to investigate the effects of MB12662, a synthetic dunnione compound, on cisplatin-induced vomiting reflexes and intestinal, renal, immune system, and hematopoietic toxicities in ferrets and mice, respectively. Male ICR mice were orally administered MB12662 (5, 10, 25 or 50 mg/kg) for 10 days, during which intraperitoneally challenged with cisplatin (3.5 mg/kg) from day 4 to 7, and sacrificed on day 10 for the pathological examination. Male ferrets were orally administered MB12662 (25, 50 or 100 mg/kg) for 7 days, subcutaneously challenged with cisplatin (5 mg/kg), and monitored for vomiting reflexes and survival of the animals. Four-day injection of cisplatin (3.5 mg/kg) to mice caused body weight loss and degeneration and atrophy of intestinal villi, reducing villi/crypt ratio to a half level of control animals. Cisplatin also induced renal and hepatic toxicities, and depletion of splenocytes and bone marrow progenitor cells. The systemic toxicities including decreased villi/crypt ratio, immune system atrophy, splenocyte depletion, and decreased cellularity in bone marrow were improved by MB12662. Cisplatin (5 mg/kg) induced retching and emetic responses of ferrets, which were remarkably attenuated by MB12662 in a dose-dependent manner. All the ferrets pretreated with MB12662 survived the challenge of cisplatin, in comparison with 40% mortality in vehicle-treated animals, and blood parameters of nephrotoxicity and hepatotoxicity were markedly recovered. It is expected that MB12662 could be a candidate for the body protection against burden, including emesis, of chemotherapeutic agents.

The Essential Oil of Artemisia iwayomogi Kitamura Induces Apoptosis on Human Oral Epidermoid Carcinoma Cells

  • The aerial part of Artemisia iwayomogi Kitamura has traditionally been used for inflammation, infectious disease, cancer, pyretic, diuretic, liver protective effect, and choleretic purposes in Korea. We investigated that the essential oil induces apoptosis in KB cell as evidenced by Hoechst-33258 dye staining, flow cytometry (cell cycles), and DNA fragmentation for nuclear condensation and Western blotting for activation of caspases-3, -8, -9, Bax, Bcl-2, cytochrome c, and poly (ADP-ribose) polymerase (PARP) cleavage. In the present study, we found that the essential oil could induce apoptosis in KB cells, as characterized by DNA fragmentation, activation of caspase-3, -8, and -9, and PARP cleavage. The efficacious induction of apoptosis was observed as a dose-dependent. The essential oil-induced apoptotic cell death was accompanied by up-regulation of Bax and down-regulation of Bcl-2. The essential oil also caused the loss of mitochondrial membrane potential and cytochrome c release from mitochondria to cytosol. These findings indicate that mitochondrial pathways might be involved in the essential oil-induced apoptosis and enhance our understanding of the anticancer function of the essential oil in herbal medicine.

자가 처방 감초 다량 복용 후 발생한 저칼륨성 하지마비 1례 (A Case of Self-prescribed Licorice-induced Hypokalemic Paralysis)

  • Glycyrrhetinic acid, which is a component of licorice, can cause hypermineralocorticoidism through the inhibition of $11{\beta}$-hydroxysteroid dehydrogenase. So, a high dose intake of licorice can lead to hypermineralocorticoidism with potassium loss and depression of the renin-angiotensin-aldosterone system. We report a 73-year-old man with muscle weakness of proximal lower limbs with hypokalemia (Serum $K^+$: 1.4 mEq/L) due to taking self-prescribed licorice without OMD's diagnosis. He boiled 60~100g licorice / day in water and drank it intermittently for 1 month due to arthralgia and swelling of both his knees. Patient's serum renin activity and aldosterone level were far beyond the normal range. He also had metabolic alkalosis with pH 7.552. After quitting the licorice, hypokalemia and muscle weakness of proximal lower limbs gradually improved within 1week.

Anti-cancer effects of enzyme-digested fucoidan extract from seaweed Mozuku

  • Fucoidan is a uniquely-structured sulfated fucose-rich polysaccharide derived from brown algae. Recently, the abalone glycosidase-digested fucoidan extract (fucoidan extract) derived from seaweed Cladosiphon novae-caledoniae Kylin (Mozuku) draws much attention because of its clinical anti-cancer effect in Japan. Here, we report the cancer cells-specific apoptosis inducing effects of the fucoidan extract. The fucoidan extract suppressed the growth of various anchorage-dependent and -independent cancer cells. The fucoidan extract contained low molecular weight components, which induced apoptosis of human leukemic HL 60 cells but not of human lymphocytes. It was shown that the fucoidan extract lead caspase 3/7 activation and loss of mitochondrial membrane potential in HL 60 cells. Another function of the fucoidan extract was also observed. It has been known that sugar chain expression on the surface of cancer cell membrane changes dependent on their malignancy. The analysis on sugar chain expression profiling using FITC-labeled lectins revealed that the expression of concanavalin A (Con A) binding sugar chain was enhanced by the treatment of human lung adenocarcinoma A549, human uterine carcinoma HeLa and human fibrosarcoma HT1080 cells with the fucoidan extract. Con A-induced apoptosis of cancer cells was stimulated in a dose-and time-dependent manner by the treatment with the fucoidan extract but not of human normal fibroblast TIG-1 cells.

고리 1호기의 기사용 핵연료 집합체 수송용기 설계에 관한 연구 (Design Study of A Spent Fuel Shipping Cask for Korea Nuclear Unit-1)

  • 본 논문에서는 고리 1호기의 기사용 핵연료 집합체를 수송하기 위한 Cask를 설계하였다. 이를 위하여 고리 1호기의 기사용 핵연료 집합체로부터 방출되는 감마선과 중성자를 계산하여 MORSE 및 ANISN전산 코드로써 차폐 계산을 수행하였다. 그 결과, 9개의 집합체를 동시에 수송할 수 있는 Steel Cask가 가장 적합하다는 것을 밝혔다. 이 Steel Cask에 대한 안전성을 평가하기 위하여 연료봉의 중심 온도와 복재온도를 계산하여 핵연료의 용융점보다 훨씬 낮음을 증명하였다. 또한 KENO와 MORSE전산 코드를 사용하여 임계도 계산을 수행하여 미임계 상태임을 증명하였다. 이로써 9개의 기사용 핵연료 집합체를 동시에 수송할 수 있는 Steel Cask를 간단히 설계하였다.

Fucoidan attenuates 6-hydroxydopamine-induced neurotoxicity by exerting anti-oxidative and anti-apoptotic actions in SH-SY5Y cells

  • Parkinson's disease (PD) is an irreversible neurological disorder with related locomotor dysfunction and is characterized by the selective loss of nigral neurons. PD can be experimentally induced by 6-hydroxydopamine (6-OHDA). It has been reported that reactive oxygen species, which deplete endogenous glutathione (GSH) levels, may play important roles in the dopaminergic cell death characteristic of PD. Fucoidan, a sulfated algal polysaccharide, exhibits anti-inflammatory and anti-oxidant actions. In this study, we investigated whether fucoidan can protect against 6-OHDA-mediated cytotoxicity in SH-SY5Y cells. Cytotoxicity was evaluated by using MTT and LDH assays. Fucoidan alleviated cell damage evoked by 6-OHDA dose-dependently. Fucoidan reduced the number of apoptotic nuclei and the extent of annexin-V-associated apoptosis, as revealed by DAPI staining and flow cytometry. Elevation of lipid peroxidation and caspase-3/7 activities induced by 6-OHDA was attenuated by fucoidan, which also protected against cytotoxicity evoked by buthionine-sulfoximine-mediated GSH depletion. Reduction in the glutathione/glutathione disulfide ratio induced by 6-OHDA was reversed by fucoidan, which also inhibited 6-OHDA-induced disruption of mitochondrial membrane potential. The results indicate that fucoidan may have protective action against 6-OHDA-mediated neurotoxicity by modulating oxidative injury and apoptosis through GSH depletion.

파이로처리 폐기물 처분 시스템 근계 영역 내 핵종 유출 민감도: 제 2 부 결정론적 접근 (A Sensitivity Study on Nuclide Release from the Near-field of a Pyroprocessed Waste Repository System: Part 2. A Deterministic Approach)

  • 당해 연구의 제 1 부 논문에서 확률론적으로 접근한 데에 이어, 파이로처리 방사성 폐기물 처분장에 대하여 폐쇄 후 처분장의 성능에 영향을 줄 수 있는 근계 영역 내 세가지 주요 설계 관련 요소에 대하여 각 핵종별로 최종 피폭 선량에 주는 민감도를 결정론적인 방법을 통하여 조사해 보았다. 농축 피폭 집단에 방사선 피폭을 주는 주요한 핵종들이 처분장에서 유출된 후 처분 시스템 근계 영역 내 다양한 매질을 이동하는 것에 관련되어 이들 요소가 어느 정도의 영향을 주게 되는지 보기 위하여 제 1 부에서 처분 용기의 수명, 선원항으로서의 처분 용기에서의 연간 핵종 유출률, 그리고 처분장 주요 인공 방벽으로서의 완충재의 손실도 등의 변화를 확률론적 접근 방법으로 검토한 데 이어, 제 2 부의 이 연구를 통해서는 통계적인 확률론적 민감도를 검토하는 대신 세가지 인자에 대하여 가장 나쁜 경우와 이상적인 조합을 구성한 후 이를 결정론적으로 평가하여 인지된 3개의 요소들이 제 1 부에서의 결과와 동일하게 처분장 설계에 매우 중요할 수 있다는 결과를 얻을 수 있었다.
