The preventive effects of Sophorae Fructus extracts (I: hot water extract and II: combination product using I) on bone loss in ovariectomized (OVX) rats were investigated. Sophorae Fructus extracts were orally administrated to OVX rats for 9 weeks. Ovariectomy caused the increase of body weight and deoxypyridinoline (Dpd: bone resorption marker) and decrease of calcium (Ca: bone formation marker) level in serum. Dpd level were significantly decreased and Ca levels were elevated at 9 weeks in Sophorae Fructus extracts administered groups after ovariectomy at a dose of 0.556 g/kg/day compared with control group. In administered groups, trabecular bone area (TBA) in the tibia and lumbar were also increased compared with control group in histomorphological analysis. The preventive or treatment effects of Sophorae Fructus extracts on bone loss in OVX rats appears to be due to suppression of bone turnover.
Background : Use of Ma Huang is controversial. Multiple reports of adverse effects led the U.S. Federal Drug Administration (FDA) to announce about prohibiting dietary supplements containing ephedrine alkaloids/Ma Huang. Objective : The objective of the study was to review the evidence from literature and rigorous clinical trials on the dosage, effectiveness, and adverse effects. Methods : Literature searches were conducted in U.S. FDA database after 2003 and digital Uibujeonok(醫部全錄). Hand searches of medical journal and bibliographies of identified articles were conducted. Results : Ma Huang has been used in Korean Oriental Medicine to treatment asthma and other ailments. Recently, Ephedrine/Ma Huang are used for weight loss or to enhance athletic performance in Europe and America, Korea. Ephedrine/Ma Huang are associated with increased risk of psychiatric, autonomic, or gastrointestinal symptoms, and heart palpitations. In addition to that, a risk of serious adverse events, including heart attack, stroke, epilepsy and psychiatric problem are also have been reported. Therefore, max dose of total alkaloids of ephedrine is studied in need of a specialist's part. Conclusion : In most reports, Ma Huang effectively promoted short-term weight loss and enhancement of athletic performance, But, safety with long-term use requires further investigation.
Objectives : This study aimed to investigate the protective effects of an herbal mixture of Atractylodes macrocephala and Taraxacum spp. (ATC) on ulcerative colitis. We have previously screened traditional medicinal herbs to discover the effective candidate by the animal model. A. macrocephala and T. spp were identified as one of the effective herbs in the screening process. Methods : Experimental colitis was induced in male Balb/c mice by administering drinking water containing dextran sulfate sodium, which mimics the clinical and histological features of ulcerative colitis in human. ATC at doses of 30, 100 or 300 mg/kg were orally administered to mice twice per day for 10 consecutive days. To evaluate the damage from experimentla ulcerative colitis, body weight, colon length, disease activity index, myeloperoxidase and histological changes were measured and analyzed. Results : The administration of dextran sulfate sodium with drinking water resulted in markedly reduced colon length, severe body weight loss, increased levels of myeloperoxidase activity and histological damages in mice. ATC treatment significantly ameliorated the colon shortening, histological damage, body weight loss and disease activity index score in a dose-dependent manner. ATC also attenuated the colonic myeloperoxidase activity which reflects the severity and extent of inflammatory damage of colon. Conclusions : ATC exerts protective effects against inflammatory colonic structural damage induced by epithelial barrier integrity impairment. ATC also inhibits weight loss and related symptoms of UC which can be considered as the functional recovery of colon.
Objective : This study was undertaken to determine whether Gamibaegi-eum (BGU) in vitro and in vivo exerts a beneficial effect against cell injury induced by reactive oxygen species (ROS) in the human intestine. Methods : Effects of BGU in vitro on cell injury were examined using Caco-2 cells, cultured human intestinal cell line. Exposure of cells to H₂O₂ induced increases in the loss of cell viability in a time and dose-dependent fashion. Results : BGU prevented H₂O₂-induced cell death and its effect was dose-dependent over a concentration range of 0.051%. H₂O₂-induced cell death was prevented by catalase, the hydrogen peroxide scavenger enzyme, and deferoxamine, the iron chelator. However, the potent antioxidant DPPD did not affect H₂O₂-induced cell death. H₂O₂ increased lipid peroxidation, which was inhibited by BGU and DPPD. H₂O₂ caused DNA damage in a dose-dependent manner, which was prevented by BGU, catalase, and deferoxamine, but not DPPD. BGU restored ATP depletion induced by H₂O₂. BGU inhibited generation of superoxide and H₂O₂ and scavenged directly H₂O₂. Oral administration of mepirizole in vivo at a dose of 200mg/kg resulted in ulcer lesions in the stomach and the proximal duodenum. Pretreatment of BGU(0.1%/kg, orally) and catalase (800Units/kg, i.v.) significantly decreased the size of ulcers. Mepirizole increased lipid peroxidation in the mucosa of the duodenum, suggesting an involvement of ROS. Pretreatment of BGU and catalase significantly inhibited lipid peroxidation induced by mepirizole. Morphological studies showed that mepirizole treatment causes duodenal injury and its effect is prevented by BGU. Conclusion : These results indicate that BGU exerts a protective effect against cell injury in vitro and in vivo through antioxidant action. The present study suggests that BGU may playa therapeutic role in the treatment of human gastrointestinal diseases mediated by ROS.
Physicochemical and microbiological properties of red beans, soybeans, barleys, and corns irradiated by gamma-ray at 1.2 kGy, 10.1 kGy, or 30.5 kGy were investigated. Moisture content and crude lipid content of the irradiated cereals showed no significant difference from the nonirradiated group. TBA values of corns irradiated at 30.5 kGy increased. Gamma irradiation reduced the numbers of mesophilic and psychrotrophic bacteria in the cereals. Yeasts and molds were reduced below detection levels even at 1.2 kGy dose-irradiation. There was no significant difference in reducing sugars between the irradiated cereals and the nonirradiated ones except corns. Stachyose, raffinose, and sucrose of soy beans and sucrose of barleys increased by 30.5 kGy dose-irradiation. Loss of riboflavin content of the irradiated samples was not observed except red beans irradiated at 30.5 kGy. In Hunter's color, b value of the red bean powder increased but L value of the soybean powder decreased by the Irradiation. L, a, and b values of the barley powder slightly increased under 30.5 kGy dose-irradiation and b value of the corn powder decreased under 10.1 or 30.5 kGy-dose irradiation.
In the present study, quantitative and qualitative histology was used to assess the effects of ibuprofen suppositories with various treatments on the rectal mucosa of rats. Two suppositories were prepared with Witepsol W35 and compared with two commercial ibuprofen suppositories Reference I (Showa Pharm.ind., Tokyo, Japan), Reference II (P.Pharm., Seoul, Korea). Single and multiple dose(dosing interval 4 hr, n=4) studies were conducted. All suppositories significantly increased epithelial cell loss, but the extent of rectal irritation was variable. These studies showed that the incorporation of ibuprofen into the suppository bases increases the morphological change in rectal tissue both for the single and multiple administrations of suppositories, but which was significantly recovered within 24 hr although the interanimal variability in scores was very substantial. Multiple administration of ibuprofen suppositories caused significant damage to rectal mucosa, but it must be considered that these were under the severe condition, that is, interval of administration (4 hr) was three times shorter than normal interval of administration and dose was fifteen times larger than usual human dose. Aluminum oxide $(Al_2O_3)$, a dispersing agent, slightly increased the irritation of rectal mucosa in rats at 5 hr and 24 hr after multiple administration, but it was possible to ignore the difference of irritation in the data at 5hr and 24hr after single administration. Finally, it was concluded that Witepsol W35 and ibuprofen had a slight rectal mucosa-irritating effect on the usual human dose, and ibuprofen suppositories prepared with Witepsol W35 or Witepsol W35, $Al_2O_3$ showed almost similar extent of rectal irritation with commercial ibuprofen products.

The Lunar Reconnaissance Orbiter (LRO) launched on June 16, 2009 has six experiments including of the Cosmic Ray Telescope for the Effects of Radiation (CRaTER) onboard. The CRaTER instrument characterizes the radiation environment to be experienced by humans during future lunar missions. The CRaTER instrument measures the effects of ionizing energy loss in matter specifically in silicon solid-state detectors due to penetrating solar energetic protons (SEP) and galactic cosmic rays (GCRs) after interactions with tissue-equivalent plastic (TEP), a synthetic analog of human tissue. The CRaTER instrument houses a compact and highly precise microdosimeter. It measures dose rates below one micro-Rad/sec in silicon in lunar radiation environment. Forbush decrease (FD) event is the sudden decrease of GCR flux. We use the data of cosmic ray and dose rates observed by the CRaTER instrument. We also use the CME list of STEREO SECCHI inner, outer coronagraph and the interplanetary CME data of the ACE/MAG instrument.We examine the origins and the characteristics of the FD-like events in lunar radiation environment. We also compare these events with the FD events on the Earth. We find that whenever the FD events are recorded at ground Neutron Monitor stations, the FD-like events also occur on the lunar environments. The flux variation amplitude of FD-like events on the Moon is approximately two times larger than that of FD events on the Earth. We compare time profiles of GCR flux with of the dose rate of FD-like events in the lunar environment. We figure out that the distinct FD-like events correspond to dose rate events in the CRaTER on lunar environment during the event period.

By applying an ergonomically developed Bismuth shield to the endovascular treatment of cerebral aneurysm the radiation dose of the scalp and lens from the medical radiation exposure was reduced. The enrtance surface dose was analyzed by measuring the occipital parts, bilateral temporal parts, bilateral quadriceps, and nasal tip of the developed bismuth shield using a photostimulable fluorescence dosimeter before (Group A) before use (Group B). Signal to noise ratio (SNR) and contrast to noise ratio (CNR) analysis were used to evaluate the image quality when Bismuth shielding was used. The mean entrance surface dose of A group and B group was 26.92% lower than that of A group. The analysis of CNR and SNR was the same for both Roadmap and DSA. The use of Bismuth shielding is an alternative that can reduce the radiation impairment due to temporary hair loss and other stochastic effects that may occur after cerebrovascular intervention.
Background: Verbenalin is an iridoid glucoside, which is among the active components of some medicinal herbs such as Verbena officinalis Linn, and Cornus officinalis Siebold and Zucc. Previous studies have confirmed the antioxidant activity and neuroprotective potential of verbenalin. To confirm the safety of verbenalin, an approximate lethal dose was determined based on a single oral dose toxicity study. Methods: Institute of Cancer Research mice were randomly assigned to three verbenalin exposure groups (250, 500, and 1,000 mg/kg) and a control group (5% methylcellulose solution). There were (5 male and 5 female mice per group). Mortality, clinical signs, and body weight were monitored for 14 days, and necropsies were conducted. Results: No mortalities were observed in the control group or the verbenalin 250 mg/kg group, whereas mortalities were observed in the 500 mg/kg and 1,000 mg/kg verbenalin groups. During the observation period, stool abnormalities such as mucous stools were observed. Clinical signs such as loss of locomotor activity were observed in the 500 mg/kg and 1,000 mg/kg verbenalin groups. During the study period, significant changes in body weight were observed in the 500 mg/kg and 1,000 mg/kg verbenalin groups; however, no gross abnormalities were observed at necropsy. Overall, no toxicity was found in the 250 mg/kg group. Conclusion: The approximate lethal dose of verbenalin was estimated to be 500 mg/kg. For a more accurate assessment of the safety of verbenalin, other types of studies such as repeated-dose toxicity studies should also be conducted.
The fourth instar larvae of C. riparius were treated with potential endocrine disruption chemicals (EDCs) such as bisphenol A (BPA) and 4-nonylphenol and the effects of morphological abnormalities were observed. The deformities of the mentum following exposure to EDCs showed the smooth tooth, the loss of tooth and deformed tooth. The incidence rates of the mentum deformity were associated with chemicals: BPA $31{\sim}90%$, and 4-nonlyphenol was $40{\sim}80%$. As the concentration of BPA increased, the incidence of deformed mentum was dose dependent. While the incidences of deformed mentum following exposure to 4-nonlyphenol was not associated with their concentrations. The deformed MLT observed smooth or round tooth and the deformity of LT showed loss of one or more than tooth. Also, the MIX type was usually smooth or loss tooth. The abundance of deformity type for the mentum showed MIX (MLT+LT) > LT (lateral teeth) > MLT (median lateral teeth).
