Improvement of Herbicide Use in Crop Production IX. Tank-mix Use of Acifluorfen and Haloxyfop-methyl for Weeding on Soybeans (Glycine max L.)   • For weeding on the after-barley cropping soybean, the tank-mix use of postemergence herbicides acifluorfen and haloxyfop-methyl was tested. Acifluorfen has shown the transient leaf-burn on the newly formed soybean leaves, but no growth inhibitions. Under the treatment of acifluorfen only, grassy weeds occurred successively and explosively. However, under the combinated spraying of both herbicides, the grassy weeds could be controlled in a slight antagonistic, the broadleaved weeds in additive, and the total weed species including sedges in synergic tendencies, respectively. Also, to achieve the full-season weeding efficacies for the prominent crop yields, both herbicides are suggested to be mixed with more dosages than 0.245 kg ai/ha of acifluorfen and 0.15 kg ae/ha of haloxyfop-methyl, respectively.

Effects of Saengjihwangeumja-gami on STZ-induced Diabetic Mice (생지황음자가미(生地黃飮子加味)가 Streptozotocin으로 유발된 당뇨생쥐에 미치는 영향(影響))

  • First, mice were dosed with 50mg/kg of streptozotocin(STZ) twice every 24 hours to cause high blood-sugar. Then, after three days, mice were injected with 100mg/kg of STZ again. Two different dosages of Saengjihwangeumja-gami were given to the experiment groups: SA group, 15mg/kg/day, and SB group, 90mg/kg/day, in order to determine the effects of Saengjihwangeumja-gami, which has been known to be good for DM(Diabetes Mellitus). By observing weight and blood-sugar level changes, blood tolerance, the numerical value of BUN(Blood Urea Nitrogen) and creatinine in blood, and through light-electronicmicroscopic and immunohistologic investigations of pancreas and kidneys, the following results were obtained: 1. The experiment groups showed a high suppressive effect of weight-loss. 2. The experiment groups' blood-sugar and blood tolerance showed an effective lowering of blood-sugar levels. 3. The experiment groups did not show any noticeable change in the numerical value of BUN and creatinine in blood compared with that of the control groups. 4. The experiment groups showed a higher Insulin positive reaction of pancreatic islets ${/beta}-cell$ than the control groups. 5. The experiment groups showed a higher immuno-reaction against IGF- II than the control groups. 6. Observation of apoptosis of the pancreatic islets showed that the cells of experiment groups were less injured compared with those of the control groups, and fewer apoptag-positive reaction cells were seen in experiment groups than in the control groups. 7. Uunder electron-microscopy, the insulin-containing granules in pancreatic islets ${/beta}-cells$ had increased more in the experiment groups than in the control groups. 8. Under light microscopy, the injury on the inner & outer membrane of the glomerulus and epithelial cells of capillaries and cells among vessels were fewer in the experiment groups than in the control groups. 9. More apoptag-positive reaction cells in the kidney were seen in the control groups than in the experiment groups. 10. PAS-positive reaction substances had increased more in the substrate among the vessels of a glomerulus belonging to the control group than those of the experiment group. 11. Uunder electron-microscopy, the nucleonic membrane, nucleoplasm and mitochondria of proximal and distal renal tubular were more injured in the control groups than in the experiment groups. In conclusion, strong evidence for the efficacy of Saengjihwangeumja-gami in lowering blood-sugar, and in recovery and generation of pancreatic tissues injured by DM was observed. Results suggest Saengjihwangeumja-gami is an effective treatment for DM. Further study of the principles of blood-sugar dropping effects of Saengjihwangeumja-gami are needed, as well as further study of recovery and regeneration of pancreatic tissues injured by DM.

Effect of Di-(2-ethylhexyl)phthalate(DBHP) on Spermatogenesis in Rat Testes (흰쥐 정자형성과정에 미치는 Di-(2-ethylhexyl)phthalate의 영향)

  • Di-(2-ethylhexyl) phthalate (DEHP) is a plasticizer known as one of endocrine disruptors. The present study was carried out to investigate the alterations of function and ultrastructure in rat testes after oral intubation of DEHP in dosages of 1g/kg/day, 2 g/kg/day or 3 g/kg/day in 0.5 ml of corn oil for 15 days. DEHP reduced the growth of body and testes, inhibited apermatogenesis and induced structural changes on various cell types of the rat testis. Leydig cells, Sertoli cells and the developing germ cells seemed to be impaired their differentiations in terms of the structural changes of cell organelles. The increase of heterochromatin in amount were common features in all 3 cell types. In addition, the Leydig cells were characterized by the swelling of smooth endoplasmic reticulum and perinuclear space, the increases in number and size of Iysosomes. The Sertoli cells became irregular in nuclear envelope and the number of Iysosomes and vacuoles seemed to be increased. There were some indications of necrosis of the germ cells, such as vacuolized nucleus and segregated nucleolus. And also, DEHP lowered the level of testosterone in experimental rat serum. DEHP suppressed apermatogenesis decreasing developing germ cells and these effects of DEHP on the rat testis were dose dependent. The detrimental effect of DEHP on apermatogenesis and ultrastructure of rat testes seems to be derived from the decreased level of testosterone by Leydig cells, followed by the abnomalities of Sertoli cells and the germ cells.

This study revealed the DEHP inhibited the development of seminiferous tubules and induced structural changes on various cell types of the rat testis. Leydig cells, Sertoli cells and the developing germ cells seemed to be impaired their differentiations in terms of the structural changes of cell organelles. The increase of heterochromatin in amount were common features in all 3 cell types. In addition, the Leydig cells were characterized by the increases in number and size of lysosomes and the scantiness of smooth endoplasmic reticulum. The Sertoli cells became irregular in nuclear envelope and the cytoplasm decreased, but the number of lysosomes and vacuoles seemed to be increased. There were some indications of necrosis of the germ cells, such as vacuolized nucleus and segregated nucleolus. These detrimental effects of DEHP on the rat testis were dose dependent and suppressed spermatogenesis decreasing developing germ cells in number and appearances. The effect of DEHP on ultrastructure of rat testis, as its known physiological functions, seems come from the decreased level of testosterone by Leydig cells, followed by the abnomalities of Sertoli cells and the germ cells.

The Predilution Effect of Al-based Liquid Coagulants for the Optimal Efficacy (최적 응집 효율을 위한 Al계 액상 응집제의 희석 효과)

  • The coagulation efficacy of Al-based coagulants (such as Alum, PACS, and PACC) was investigated to ascertain removal efficiencies of turbidity and phosphate with variation of solution pH, coagulant dosages, and pre-dilution ratios. The efficacy of Al-based coagulants was maximized in the pH range of 6~9. Under the initial condition of pH 8, $10mg/L\;{PO_4}^{3-}$, and 20 NTU, Al-based coagulants exhibited a similar efficacy in the removal of turbidity, whereas the removal efficiency of phosphate was clearly dependent on the basicity of coagulants: Alum (0%) > PACS (45~50%) > PACC (70%). At high initial turbidity of 100 NTU, polymeric coagulants, such as PACS and PACC, exhibited a higher removal efficiency of turbidity compared to Alum. In comparison to direct injection of coagulants at low initial turbidity (20 NTU), 500~2000 times pre-diluted Alum, exhibited reduced coagulation efficacy; however, removal efficiencies of turbidity and phosphate increased with the increase of retention time. Pres-diluted PACC exhibited the enhanced coagulation efficacy followed by silght decrease of the removal efficiencies with increase of the retention time. At high initial turbidity of 100 NTU, pre-diluted Alum and PACC exhibited higher removal efficiencies of turbidity and phosphate.

A COMPARATIVE STUDY ON THE VITAL SIGN AND BEHAVIOR APPEARANCE DEPENDING ON THE ROUTE OF FLUMAZENIL ADMINISTRATION IN CONSCIOUS SEDATION BY MIDAZOLAM (Midazolam을 이용한 의식진정시 flumazenil의 투여경로에 따른 생징후 및 행동양상의 비교 연구)

  • The purpose of this study was to evaluate the efficacy and safety depending on the route of flumazenil, as an antagonist against midazolam. The subjects of this study were 15 volunteers of $22{\sim}24$ years old. They were sedated with midazolam 0.2mg/Kg intranasal spray, and then 40 minutes after midazolam administration, they were given flumazenil 0.2mg intranasal spray for their reversal. For evaluation of the efficacy and safety of intranasal spray for flumazenil, they were monitored with pulse-oxymeter(Nellcor symphony N-3000, Nellcor Puritan CO. USA) and electric sphygmomanometer (Heartcare 200, National CO. Japan), and were assessed themselves using visual analogue scale(VAS) for tranquilization, sleep, fatigue and attitude. All of these subjects were reduced completely without any undesired situations. The results from this study can be summarized as follows ; 1. Nasaly administered flumazenil using spray device produced much more rapid reduction than intravenously administered flumazenil, but soon after fell in more deep sedated state than intravenously administered flumazenil. 2. There were no considerable side effects or bad influence on vital signs of both nasaly administered flumazenil and intravenously administered flumazenil. These results suggested that the flumazenil administered nasaly using spray device for reversal, we could treat patients safely and effectively under conscious sedation using midazolam administration. But, We will have to research about its optimal dosages for flumazenil, used as intranasal spray for reversal agents against the midazolam by evaluating the blood plasma concentration of midazolam and flumazenil.

  • To investigate the possibility of the BPH control by systematic application of insecticides, biological effects of buprofezin and isoprothiolane upon the BPH population was evaluated in the laboratory and pot. Both chemicals did not affect the developmental period of the nymphal stage with the applied dosages. When the buprofezin treated instar nymph was developed to the adult, adult's longevity was fairly reduced and such an effect of buprofezin was greater upon the younger instar nymphs. Residual effect of buprofezin was about 30 days in the pot. And it could suppress the BPH population when the younger instar nymphs were dominant age group in the population at the time of treatment. Isoprothiolane was also more effective for the suppression of the BPH population when the younger instar nymphs were dominant at the time of treatment. And it was expected that by buprofezin treatment in July the control effect of isoprothiolane against the BPH population could be maximized when it was treated to control the rice neck blast in late July or early August, because buprofezin could induce the biased population age distribution in which younger instar nymphs were main group at the time of isoprothiolane treatment.

Acute Toxicity of Lactobacillus plantarum AF1 Isolated from Kimchi in Mice (김치로부터 분리한 Lactobacillus plantarum AF1의 마우스에 대한 급성독성)

  • The $in$ $vivo$ single-dose acute toxicity of $Lactobacillus$ $plantarum$ AF1, a lactic acid bacterium isolated from kimchi, in ICR male and female mice was investigated. The test article was intraperitoneally or orally administered once to both sexes of mice. The motalites, clinical findings, autopsy findings, and body weight changes were monitored daily for 14 days. In the oral acute toxicity test, the male and female mice were gavaged with four doses (5.0, 2.5, 1.25 and 0.625 g/kg) of $Lb.$ $plantarum$ AF1. The oral $LD_{50}$ of the $Lb.$ $plantarum$ AF1 was considered higher than 5.0 g/kg. In the intraperitoneal acute toxicity test, mice were injected intraperitoneally with dosages of 0.7, 0.9, 1.1, 1.3, 1.5, 1.7, 1.9, 2.1, 2.3 and 2.5 g/kg. The intraperitoneal 50% lethal dose ($LD_{50}$) of the $Lb.$ $plantarum$ AF1 was >2.5 g/kg in the male and female mice. No significant changes in the general conditions, body weights, clinical signs, and gross lesions were observed in both sexes of mice to which $Lb.$ $plantarum$ AF1 was administered intraperitoneally or orally. The results suggest that the no-adverse-effect level of $Lb.$ $plantarum$ AF1 is estimated to be more than 5.0 g/kg in the oral route and 2.5 g/kg in the intraperitoneal route.

The study for the mouse cerebellum developments irradiated with γ-ray during embryogenesis (감마선 조사에 의한 발생과정 중인 마우스의 소뇌발달에 관한 연구)

  • In order to study about the lobule and layer formation and cell migration of the mouse cerebellum from at the birth to 15th day effected by 2.5, 5 and 10 Gy r-raddiation at the 19th pregnancy. The routine tissue preparation and staining procedure, Immunohistochemical staining method by the several antibody and western brotting method were utilized from the birth to the15th day. The results were as followings. 1. The body and cerebellum weights were more slowly increase of the the 2.5 Gy, 5 Gy and 10 Gy irradiation group compare to the control group, and the health condition of the 2.5 Gy group was a little bad. but the 10 Gy group was more severe and begun to die from the 12th day after birth. 2. The thickness, proliferation and migration of the 2.5, 5 and 10 Gy irradiated external granular cells from the maginal zone to the medullary area forming the molecular layer from the 6th day to the 15th day after birth were thinner, weaker and more slower according to the radiated dosages than the control group in the cresyl violet staining. 3. The proliteration, migration and lobulation of the 5 Gy radiated groups from the first day to the 15th day after birth were more weak, incomplete and irregular shape in the immunostaining with Dab, Cdk5, P35, calbindin and Zebrin antibody. 4. In the western blotting analysis using the Reelin, Dab, Cdk5 and P35 antibody. The Bands were in the 60 KD, 80 KD, 33 KD and 35 KD, and there were no differences between the control and irradiated groups in the molecular band except the Reelin. 5. As a results, the proliferation and migration of the outer granular and purkinje cells, and lobulation of the cerebellum by the several dosaege of the ${\gamma}$-ray radiation were proportionally incomplete according to dosage.

Identification of Irradiation-induced Volatile Flavor Compounds in Chicken (방사선 조사 닭고기에서의 휘발성 조사물질의 구명)

  • To identify the irradiation-induced volatile flavor compounds, which were not detected in unirradiated sample and had positive correlation with the increment of irradiation dose, the volatile flavor compounds of irradiated (0, 1, 3, 5, 10 kGy) chicken were analyzed by liquid liquid continuous extraction (LLCE) and gas ehromatographyimass spectrometry (GC/MS) methods. One hundred twenty nine compounds were detected in irradiated chicken, and these compounds were composed mainly of hydrocarbons (62 compounds), aromatic compounds (44), aldehydes (9), ketones (5) and miscellaneous compounds (10). Among these, only 3 volatile compounds including 2-methylpentanal (r=0.24), 4-methylcyclohexene (r=0.08) and cyclotetradecene (r=0.92), were detected as irradiation-induced volatile flavor compounds in irradiated chicken. However, only cyclotetradecene was selected as a marker compound for detecting irradiation dosages with high correlation coefficient in irradiated chicken.
