Comparison of gloverin gene expression patterns between domesticated and wild silkworms

  • Bombyx mandarina is widely accepted as ancestor of B. mori. Silkworms are served as well-characterized models for understanding the mechanism for the genetic regulation of development. In this study, we performed RNA-Seq analysis to examine tissue-expression of gloverin isoforms of the silk-gland, mid-gut, and fat body in B. mandarina. BLAST analysis revealed that four gloverin isoform gene sequences of B. mandarina were highly similar to B. mori. To identify the difference between two species, the expression profile of gloverin was measured by semi- RT-PCR analysis. The specific expression of gloverin isoform genes was observed mainly in the fat body from B. mori but not B. mandarina. However, all of tissues in the wild-type silkworm could induce the upregulation of compared with the B. mori. To validate the sudden increase in gloverin gene expression in the mid-gut tissue of B. mandarina, we were using qRT-PCR. Relative mRNA expression rate of gloverin at the wild-type silkworm was much higher than domestic silkworm. Comparative genomics between domesticated and wild silkworms showed different tissue-expression levels in some of immune related genes. These results are suggesting a trend toward decreasing immunity related genes expression during domestication. Further studies are needed to elucidate the silkworm domestication and an invaluable resource for wild silkworm genomics research.

Cloning, Sequencing and Characterization of Mitochondrial Control Region of the Domestic Silkwom, Bombyx mori

  • The nucleotide sequence of the domestic silkworm (Bombyx mori) mitochondrial (mt) control region and its flanking genes was determined from PCR clones. The control region of the silkworm mt genome was located between the small ribosomal RNA gene and transfer RN $A^{Met}$. This 499 bp control region hale 95.4% A+T content. Extensive comparative analysis studies performed with similar control region of other insect genomes could not reveal a highly conserved region containing conserved motifs of animal mito-chondrial genome. The remarkable feature that found in this control region was the presence of tandem motifs containing nine repetitive sequences. The potential usefulness of this motif sequences for Bombyx species or their taxonomically related species is enhanced by its unique localization in the maternally inheritance mitochondrial molecule.e.

Effect of Hydrocortisone on the Economic Parameters of the Domestic Silkworm, Bombyx mori L.

  • The effect of topical application with hydrocortisone on economic parameters was analysed following treatment of last larval stadium. The treated larvae showed a significant increase in larval weight at higher concentrations along with other enhanced larval, cocoon and adult parameters. This suggests that hydrocortisone can be used effectively for commercial silkworm rearing. The larvas period was significantly increased in all the treated groups with increased female cocoon weights, its shell weights and male cocoon shell weight at 20$\mu\textrm{g}$/ml treated group. Filament length, weight and denier were increased significantly in all the treated groups. Moth emergence percentage, length of the ovariole, eggs per ovariole, fecundity and rind hatching percentage were unaffected when compared with that of the carrier control group. This suggests that hydrocortisone in addition to affecting larval growths also affect silk crop.

Construction of the Silkworm, Bombyx mori, with a Green Fluorescence by Autographa californica Nuclear Polyhedrosis Virus

  • We have constructed a recombinant baculovirus, Autographa californica nuclear polyhedrosis virus (AcNPV), containing green fluorescent protein (GFP) gene from the jellyfish, Aequorea victoria, and transferred it into the domestic silkworm Bombyx mori larvae for the production of visible transgenic silkworm of living organism. When one day-old fifth instar female larvae were injected with the recombinant AcNPV of 1x10$^{5}$ plaque forming units, the bright glow of GFP was detected in the recombinant AcNPV-infected larvae and in the newly hatched larvae of the next generation. Our findings demonstrate that the viral replication was detected in the silkworm treated with the recombinant ACNPV and the gfp gene was expressed under the transcriptional control of the polyhedrin gene promoter, Furthermore, the gfp gene was transmitted to the next generation, suggesting that this system can be applied for the development of transgenic silkworms.

누에 품종에 따른 눈꽃동충하초 배양 특성 (Effect of silkworm varieties on Paecilomyces tenuipes culture)

  • 동충하초는 곤충의 몸속에 들어가 버섯이 발생하는 곤충 기생성 약용 버섯으로 세계적으로 인체에 유용한 활성물질이 확인되었으며, 이를 누에를 이용하여 인공재배할 수 있는 기술이 개발되어 활용되고 있다. 그러나 누에품종에 따른 동충하초의 생산성에 대한 연구 보고는 없는 실정이다. 본 연구에서는 장려품종 3품종(백옥잠, 금옥잠, 대성잠)과 특수품종 2품종(연록잠, 골든실크)을 이용하여 눈꽃동충하초의 재배조건과 특징을 조사하였다. 눈꽃동충하초 균주(P. tenuipes)에 대한 감염율은 금옥잠이 가장 높았으며, 백옥잠, 대성잠, 골든실크, 연록잠 순으로 감염율이 높았다. 눈꽃동충하초를 재배하기 위한 최적조건은 어두운 장소에서 번데기 당 약 $4cm^2$의 재배밀도로 $22^{\circ}C{\pm}1$를 유지해 주는 것이었다. 또한 우리는 농가소득 향상을 위해, 동충하초의 수요가 있을 때 마다 수시로 재배할 수 있는 연중재배 기술을 개발하였다. 연중재배 방법은 먼저 동충하초균에 감염된 번데기 체내의 수분함량을 자연건조로 3% 이하로 낮추고, 이를 진공포장 후 $4^{\circ}C$ 이하에서 보관하는 3단계의 과정이 필수적이다. 이렇게 저온에 보관되어 있는 감염번데기는 언제든지 물에 침지하여 번데기 내의 함수율을 원래대로 복원 시킨 다음 최적 재배조건에서 재배하여 동충하초를 생산할 수 있게 함으로 연중생산이 가능하다. 그러므로 우리는 동충하초 재배농가가 누에 품종 중 금옥잠을 이용하고 연중생산 기술을 활용하여 소득증대를 이룰 수 있을 것으로 기대한다.

Isolation of Two cDNAs Encoding a Putative Peptidohlycan Recognition Protein Gene from the Domestic Silkworm, Bombyx mori

  • Peptidohlycan recognition protein (PGRP) is one of the pattern recognition proteins in innate immunity of insect. We isolated differentially expressed two cDNAa, BTL-LPI and BTL-LP2, in the fat body of Bombyx mori larvae injected with bacteria by subtractive hybridization method. These two clones showed amino acid sequence divergence of 30.4%. In the comparison with other insect PGRP genes, BTL-LP2 showed 48.8% and 45.2% of sequence homology to the known PGRP genes of Bombyx mori and Tricoplusia ni, respectively, and BTL-LP2 was 31.8% and 30.9% , respectively. Phylogenetic analysis showed relatively close relationship of the BTL-LP2 to the known insect PGRP, unlike BTL-LPI, which was equidistant both to insect and mammals, suggesting a divergent relationships of the two newly cloned B. mori PGRP genes. Northern blot analyses confirmed an induction of the expression of BTL-LP2 by the bacterial infection in the Int body of B. mori, suggesting the involvement of the gene in the insect immunity.

Collection and Characteristics of the Wild Silkmoth, Samia cynthia pryeri, in Korea

  • The wild silkmoth, Samia cynthia pryeri, was firstly collected at Miryang, Korea. The development and quantitative characters of S. cynthia preyri in the laboratory condition were analysed. The larval body color of dark yellow green or light yellow green of the S. cynthia pryeri was changed into white color during the molting period, and the yellowish bristles in younger larvae were changed into fade green color in advanced stage larvae. Aralia elata and Zanthoxylum piperitum were newly identified as host plants to S. cynthia pryeri. Cocoon weight and cocoon shell percentage were average 2.14 g and 12.3 %, respectively. In addition, the morphology of the silkgland was different from that of the domestic silkworm,Bombyx mori, but was identical with that of the wild silkmoth, Antheraea yamamai.

Gravimetrics of Pupal Weight Loss in the Domestic Silkworm, Bombyx mori Linn. (Lepidoptera : Bombycidae)

  • A comprehensive study on daily pupal weight loss due to histolysis and histgenesis in Bombyx mori Linn., has been undertaken. The percentag of pupal weight loss in the male pupa is higher than the female, conforming that the female pupa require more energy in the form of less pupal weight loss for ovipositional activities. The regression equation clearly shows higher percentage loss of daily pupal weight in male than female with respect to age and, this is also evident from the slope of the regression line. Moreover, analysis of the coefficient of correlation shows that the weight loss of pupa is directly correlated with the age rather than climatic parameters under which the pupa dwells.

Effects of immune-challenged domestic silkworm hemolymph on the regulation of SIRT5 and PRDx1 expression

  • SIRT5 and PRDx1 play crucial roles in cancer and are involved in the basic mechanisms of reactive oxygen species detoxification. In our previous studies, we showed that hemolymph extracts of immune-challenged Bombyx mori have antioxidant properties. Following H2O2 stimulation, immune-challenged B. mori hemolymph extracts elicited SIRT5 downregulation activity, reaching effective activity at the highest concentration of 100 ppm. Additionally, cells treated with immune-challenged B. mori hemolymph extracts demonstrated increased PRDx1 mRNA expression compared to that of PBS-treated cells. Therefore, immune-challenged B. mori hemolymph extracts offer a potential auxiliary means of treating drug-resistant tumors through downregulation of SIRT5 and upregulation of PRDx1 expression. Nevertheless, further studies on the effects of B. mori hemolymph on SIRT5 and PRDx1 regulation are pertinent for using it as a food or pharmaceutical material and understanding its therapeutic effect on tumors, including those that are drug-resistant.
