CY-007 and CY-049 pteridine glycosyltransferases (PGTs) that differ in sugar donor specificity to catalyze either glucose or xylose transfer to tetrahydrobiopterin were studied here to uncover the structural determinants necessary for the specificity. The importance of the C-terminal domain and its residues 218 and 258 that are different between the two PGTs was assessed via structure-guided domain swapping or single and dual amino acid substitutions. Catalytic activity and selectivity were altered in all the mutants (2 chimeric and 6 substitution) to accept both UDP-glucose and UDP-xylose. In addition, the wild type activities were improved 1.6-4.2 fold in 4 substitution mutants and activity was observed towards another substrate UDP-N-acetylglucosamine in all the substitution mutants from CY-007 PGT. The results strongly support essential role of the C-terminal domain and the two residues for catalysis as well as sugar donor specificity, bringing insight into the structural features of the PGTs.
본 연구는 과학 및 시각예술 영역의 창의적 성취사례를 통해 두 영역의 영역보편성과 영역특정성에 대해 살펴보는 데 그 목적을 두었다. 연구를 위해 각각의 영역에서 세계적인 성취를 이룬 한국의 창의적 인물들(과학자: 10인, 시각예술가: 9인)에 대한 인터뷰를 실시하였고, 이로부터 수집한 질적 자료를 토대로 창의적 성취에서 영역보편적으로 드러나는 특성과 영역특정적으로 드러나는 특성을 요약, 기술하였다. 자료분석에는 본 연구의 근간이 되는 총괄적이고 체계적인 개념틀 '지식진화시스템(Knowledge-Evolving Systems: KES)'을 사용하였다. 분석 결과를 보면, 실재시스템에서 두 영역의 성취는 공통적으로 요동, 탐색, 산물을 통해 드러났다. 반면, 과학은 전문지식, 어려운 과제, 세계 최초의 객관적인 지식이 주요한 요인으로, 예술에서는 일상적인 지식, 다양한 주제, 새로운 변화에 대한 인간의 감성 전달이 주된 요인으로 드러났다. 개인시스템에서 두 영역은 모두 분명한 목표를 향한 집중과 독자적인 노력을 보편적으로 요구하였다. 반면 과학은 창조의지, 확산적 및 분석적 사고, 직관 및 통찰, 그리고 도전이 대표적인 요인으로, 시각예술은 즐거움, 민감성, 통합적 사고, 완벽성, 그리고 자유로움이 주요한 요인으로 산출되었다. 마지막으로 사회시스템에서 두 영역의 성취는 전문가의 승인에 의해 결정되었다. 그리고 과학자들의 성취에는 인간네트워크, 기관의 지원, 생존경쟁과 운이 큰 영향을 미쳤고, 시대적 요구와 민족성이 발전의 토대가 되었다. 반면, 시각예술가들은 주변의 반대와 부적인 예술문화 풍토로 인해 고독한 삶을 살아왔다.
This study tries to explain how the relationship-specificity of invested assets affects E-collaboration and consequently on performance of Small and Medium Enterprises (SMEs). We classify the relationship-specificity of invested assets into four types such as business process specificity, physical asset specificity, domain knowledge specificity, and site specificity. We define E-collaboration as composed of Electronic Information Sharing (EIS) and Electronic Cooperation (E-Co). In addition, we articulate firm's performance as operational and strategic one, and investigate the impacts of EIS and E-Co on its performance. The data were collected from 187 SMEs and used for analysis. Based on the survey results, we find the following: (1) EIS is directly influenced by business process specificity and physical asset specificity, (2) E-Co is affected by site specificity and domain knowledge specificity, (3) EIS has a positive and significant impact on E-Co, (4) EIS affects firm's operational performance, (5) E-Co influences on firm's strategic performance. In conclusions, the higher the level of EIS, SMEs seem to get greater operational performance, Respectively, the higher the level of E-Co, they tend to get greater strategic performance.
This study propose a model relating supplier's use of IOIS(Inter-Organizational Information Systems) to strategic benefits through extension of Subramani's research model. In extended model, collaboration serves as a safeguard for relationship-specific intangible asset. Specifically, we evaluate how two patterns of IOIS use by supplier(exploitation and exploration) relate to two specific types of relationship-specific intangible asset(business process specificity and domain knowledge specificity), which in turn are posited to promote collaboration and strategic benefits. To explore the current study, questionnaire survey was conducted on 72 first-tier supplier firms in the manufacturing industry. Based on the survey results, we posits the following : (1) Each pattern of IOIS use directly promotes a specific type of relationship-specific intangible asset. The path of the relationship between IOIS use for exploitation and domain knowledge specificity is positive but not significant. The other paths are positive and significant. (2) Both types of relationship-specific intangible asset have a positive and significant impact on collaboration. (3) Domain knowledge specificity influences on strategic benefits but business process specificity does not have an effect on them. (4) Collaboration affects supplier's strategic benefits. These findings provide a deeper understanding of the mechanism of how the pattern of IOIS use can result in strategic benefits for supplier firms.
Peptide synthetases produced from various microorganisms are multifunctional enzyme complexes and their substrates are recognized and activated by adenylation domains. To identify the substrate specificity of the peptide synthetase isolated from Bacillus subtilis 713, known to produce an antifungal peptide, two adenylation domains containing the minimal functional portion were expressed and purified. ATP-ppi exchange experiments and kinetic studies revealed that the two adenylation enzymes had a substrate specificity to $_{L}-lysine{\;}and{\;}_{L}-tryptophan$, respectively. In addition, based on a signature sequence comparison, the substrate of the third domain was predicted to be L-glutamic acid. These results suggest that this peptide synthetase is novel because there has been no previous report on a peptide synthetase that uses $_{L}-lysine,{\;}_{L}-tryptophan,{\;}and{\;}_{L}-glutamic$ acid as substrates in that order.
Bacillus licheniformis SK-1 naturally produces chitinase 72 (CHI72) with two truncation derivatives at the C-terminus, one with deletion of the chitin binding domain (ChBD), and the other with deletions of both fibronectin type III domain (FnIIID) and ChBD. We constructed deletions mutants of CHI72 with deletion of ChBD (CHI72${\Delta}$ChBD) and deletions of both FnIIID and ChBD (CHI72${\Delta}$FnIIID${\Delta}$ChBD), and studied their activity on soluble, amorphous and crystalline substrates. Interestingly, when equivalent amount of specific activity of each enzyme on soluble substrate was used, the product yield from CHI72-${\Delta}$ChBD and CHI72${\Delta}$FnIIID${\Delta}$ChBD on colloidal chitin was 2.5 and 1.6 fold higher than CHI72, respectively. In contrast, the product yield from CHI72${\Delta}$ChBD and CHI72${\Delta}$FnIIID-${\Delta}$ChBD on ${\beta}$-chitin reduced to 0.7 and 0.5 fold of CHI72, respectively. These results suggest that CHI72 can modulate its substrate specificities through truncations of the functional domains at the C-terminus, producing a mixture of enzymes with elevated efficiency of hydrolysis.
최근 텍스트 분석을 딥러닝에 적용한 연구가 꾸준히 이어지고 있으며, 특히 대용량의 데이터 셋을 학습한 사전학습 언어모델을 통해 단어의 의미를 파악하여 요약, 감정 분류 등의 태스크를 수행하려는 연구가 활발히 이루어지고 있다. 하지만 기존 사전학습 언어모델이 특정 도메인을 잘 이해하지 못한다는 한계를 나타냄에 따라, 최근 특정 도메인에 특화된 언어모델을 만들고자 하는 방향으로 연구의 흐름이 옮겨가고 있는 추세이다. 도메인 특화 추가 사전학습 언어모델은 특정 도메인의 지식을 모델이 더 잘 이해할 수 있게 하여, 해당 분야의 다양한 태스크에서 성능 향상을 가져왔다. 하지만 도메인 특화 추가 사전학습은 해당 도메인의 말뭉치 데이터를 확보하기 위해 많은 비용이 소요될 뿐 아니라, 고성능 컴퓨팅 자원과 개발 인력 등의 측면에서도 많은 비용과 시간이 투입되어야 한다는 부담이 있다. 아울러 일부 도메인에서 추가 사전학습 후의 성능 개선이 미미하다는 사례가 보고됨에 따라, 성능 개선 여부가 확실하지 않은 상태에서 도메인 특화 추가 사전학습 모델의 개발에 막대한 비용을 투입해야 하는지 여부에 대해 판단이 어려운 상황이다. 이러한 상황에도 불구하고 최근 각 도메인의 성능 개선 자체에 초점을 둔 추가 사전학습 연구는 다양한 분야에서 수행되고 있지만, 추가 사전학습을 통한 성능 개선에 영향을 미치는 도메인의 특성을 규명하기 위한 연구는 거의 이루어지지 않고 있다. 본 논문에서는 이러한 한계를 극복하기 위해, 실제로 추가 사전학습을 수행하기 전에 추가 사전학습을 통한 해당 도메인의 성능 개선 정도를 선제적으로 확인할 수 있는 방안을 제시한다. 구체적으로 3개의 도메인을 분석 대상 도메인으로 선정한 후, 각 도메인에서의 추가 사전학습을 통한 분류 정확도 상승 폭을 측정한다. 또한 각 도메인에서 사용된 주요 단어들의 정규화된 빈도를 기반으로 해당 도메인의 특수성을 측정하는 지표를 새롭게 개발하여 제시한다. 사전학습 언어모델과 3개 도메인의 도메인 특화 사전학습 언어모델을 사용한 분류 태스크 실험을 통해, 도메인 특수성 지표가 높을수록 추가 사전학습을 통한 성능 개선 폭이 높음을 확인하였다.
Bacteriophage lambda integrase carries out the site-specific recombination of lambda. Integrase contains two DNA binding domains with distinct sequence specificity, namely arm-type binding and core-type binding domains. The amino-terminal arm-binding domain is structurally related to the three-stranded $\beta-sheet$ family of DNA-binding domains. Integrase binding to the high affinity arm-type site by the amino-terminal domain facilitates Int binding to the low affinity core-type site, where the cleavage and strand exchange occurs. The amino-terminal domain of Int also modulates the core-binding and catalysis through intramolecular domain-domain interaction and/or intermolecular interactions between Int monomers. In addition, the amino-terminal domain interacts cooperatively with excisionase during excision. This indicates that amino-terminal domain of Int plays an important role in formation of proper higher-order nucleoprotein structure required for lambda site-specific recombination.
Receptor binding plays an important role in determining host specificity of the Bacillus thuringiensis Cry $\delta$-endotoxins. Mutations in domains II and III have suggested the participation of certain residues in receptor recognition and insect specificity. In the present study, we expressed the cloned domain II-III fragment of Cry4Ba and examined its binding characteristics to mosquito-larval midgut proteins. The 43-kDa Cry4Ba-domain II-III protein over-expressed in Escherichia coli as inclusion bodies was only soluble when carbonate buffer, pH 10.0 was supplemented with 4M urea. After renaturation via stepwise dialysis and subsequent purification, the refolded domain II-III protein, which specifically reacts with anti Cry4Ba-domain III monoclonal antibody, predominantly exists as a $\beta$-sheet structure determined by circular dichroism spectroscopy. In vitro binding analysis to both histological midgut tissue sections and brush border membrane proteins prepared from susceptible Aedes aegypti mosquito-larvae revealed that the isolated Cry4Ba-domain II-III protein showed binding functionality comparable to the 65-kDa full-length active toxin. Altogether, the data present the 43-kDa Cry4Ba fragment comprising domains II and III that was produced in isolation was able to retain its receptor-binding characteristics to the target larval midgut proteins.
