A New Salt-Tolerant Thermostable Cellulase from a Marine Bacillus sp. Strain

  • A salt-tolerant cellulase secreted by a marine Bacillus sp. SR22 strain with wide resistance to temperature and pH was purified and characterized. Its approximate mass was 37 kDa. The endoglucanase, named as Bc22Cel, was purified by ammonium sulfate precipitation, gel filtration chromatography, and extraction from the gel after non-reducing sodium dodecyl sufate-polyacrylamide gel electrophoresis. The optimal pH value and temperature of Bc22Cel were 6.5 and $60^{\circ}C$, respectively. The purified Bc22Cel showed a considerable halophilic property, being able to maintain more than 70% of residual activity even when pre-incubated with 1.5 M NaCl for 1 h. Kinetic analysis of the purified enzyme showed the $K_m$ and $V_{max}$ to be 0.704 mg/ml and $29.85{\mu}mol{\cdot}ml^{-1}{\cdot}min^{-1}$, respectively. Taken together, the present data indicate Bc22Cel as a potential and useful candidate for industrial applications, such as the bioconversion of sugarcane bagasse to its derivatives.

The inhibitory effects of 3,4,5-Trimethoxy cinnamate thymol ester(TCTE, Melasolv$\circledR$) on Melanogenesis

  • To date, research on the regulation of melanogenesis has focused on factors which affect tyrosinase, the rate-limiting enzyme in the melanogenic pathway, by searching for chemicals which competitively inhibit tyrosinase function. Many types of tyrosinase inhibitors have been developed, but no satisfactory results have been made clinically until now, To find a new whitening agent, which effectively inhibits melanogenesis, we synthesized several compounds and selected compounds by cell-based assay system. Finally, 3, 4, 5-trimethoxy cinnamaie thymol ester(TCTE, Melasolv) was selected and the effects of TCTE on melanogenesis were investigated. Treatment of mouse-derived melanocyte melan-a cells with TCTE results in a marked down-regulation of tyrosinase activity. 80% decrease of tyrosinase activity occurs with 30uM TCTE treatment for 72 hours without affecting cell growth. The inhibition of tyrosinase activity is dose-dependent and melanin content was also decreased to 40%. From the in vitro tyrosinase assay using cell extract, TCTE does not act as a direct inhibitor of the enzyme. Treatment of melan-a cultures with TCTE blocks the increase in tyrosinase activity by either forskolin, 3-isobutyl-1-methtyl-xanthine. TCTE decreased the expression of tyrosinase, TRP-1 without effects on TRP-2 protein expression through the down regulation of tyrosinase and TRP-1 mRNA. From the results of cAMP immunoassays, intracellular levels of the cyclin nucleotide are unaffected in cells treated with TCTE. The inhibitory effects of melanin synthesis were also shown in reconstitute human epidermis model by topical application. These findings suggest that TCTE can be used for studying the regulation of melanogenesis and depigmenting agent.

Effects of Water Soluble Potassium Silicate by Soil Drenching Application on Watermelon (Citrullus lanatus var. lanatus) (시설수박에 대한 수용성 규산칼륨 토양관주 효과)

  • The objective of this study was to determine the effects of soluble potassium silicate by soil drenching application on watermelon growth, yield, and nutrient uptake. The potassium silicate rates were control (No potassium silicate), 1.63mM, 3.25mM, 6.50mM. The potassium silicate were treated 6 times (twice before fruit forming and 4 times after fruit forming per 7 day. Soil chemical properties, such as soil pH, EC, available phosphorus and silicate, exchangeable K, nitrate-N levels were increased after potassium silicate treatment, while the concentrations of soil organic matter, exchangeable Ca and Mg were similar to control. The growth characteristics of watermelon, such as stem diameter, fresh and dry weight of watermelon at harvest were thicker and heavier for increased potassium silicate treatment than the control, while number of node, and plant length were same for all treatments. With increased potassium silicate treatment, nutrient concentrations, such as P and K in the watermelon leaf at harvest were increased, N concentration in the leaf was decreased, and Ca and Mg concentrations in the leaf were same. Chlorophyll content was increased with increased potassium silicate application. The occurrence of powdery mildew was lower for the potassium silicate treatments than the control. Fresh watermelon weight for the potassium silicate treatments was 0.1 to 0.5kg per watermelon heavier than the control, sugar content was 0.5 to $0.6^{\circ}Brix$ higher than control, and merchantable watermelon was 2 to 4% increased compared to the control. These results suggest that potassium silicate application by soil drenching method in the greenhouse can improve watermelon nutrient uptake, merchantable watermelon and suppress the occurrence of powdery mildew.

Efficient virus elimination for apple dwarfing rootstock M.9 and M.26 via thermotherapy, ribavirin and apical meristem culture (사과 왜성대목 M.9 및 M.26의 고온, ribavirin, 생장점 배양을 통한 바이러스 제거)

  • Apple (Malus pumila) is one of the most economically important fruits in Korea. but virus infection has decreased the sustainable production of apples and caused serious problems such as yield loss and poor fruit quality. Virus or viroid infection including apple chlorotic leaf spot virus (ACLSV), apple stem pitting virus (ASPV), apple stem grooving virus (ASGV), apple mosaic virus (ApMV) and apple scar skin viroid (ASSVd) have been also reported in Korea. In many cases, as apple gets infected with virus and viroid with no specific symptoms, the damage and symptoms caused by the viruses are not detected. In our research, viruses in the rootstock were eliminated for a virus-free apple dwarfing rootstock of M.9 and M.26. The virus elimination methods were apical meristem culture, thermotherapy ($37^{\circ}C$, 6 weeks) and chemotherapy($Ribavirin^{(R)}$). The detection of apple viruses was accomplished by Enzyme-linked Immuno-Sorbent Assay (ELlSA) and reverse transcription-polymerase chain reaction (RT-PCR). RT- PCR method was 10 ~ 30% more sensitive than the ELISA method. The efficiency of virus elimination was enhanced in apical meristem culture method. The acquisition rate of virus-free apple dwarfing rootstocks was 30 ~ 40% higher in apical meristem culture. After the meristem culturing of M.9, the infection ratio of ACLSV, ASPV and ASGV was 45%, 60% and 50%, respectively. In the apple dwarfing rootstock of M.26, the infection ratio of ACLSV, ASPV and ASGV was 40%, 55% and 55%, respectively. Based on this study, the best method for the production of virus-free apple dwarfing rootstocks was the apical meristem culture.

  • To determine the effect of calcium on the preimplantational development of mouse two-cell embryo, the various concentrations of calcium were added into the culture media and the rate of blastocyst formation was observed. Also, to examine the effect of trifluoperazine, an inhibitor of calmodulin which is involved in the several intracellular calcium functions, embryos were cultured for 48 hours at the various concentrations of this inhibitor. An additional 24 hour culture was done to examine the effect of this drug on the transformation from morula to blastocyst. The results are as following ; 1. About 1.71mM of extracellular calcium is adequate for blastocyst formation and the higher concentrations of calcium (3.43mM and 8.55mM) do not affect on the blastocyst formation and the degenerating rate. 2. Trifluoperazine $100{\mu}M$ presents the inhibitory effect on the blastocyst formation while $1{\mu}M$ and $10{\mu}M$ do not so. 3. After an additional 24 hour culture, there is transformation of morula to blastocyst and the degenerating rate of embryo is increased all together.

Four New Species of the Genera Masirana and Longileptoneta (Araneae, Leptonetidae) from Korea

  • Four new species of the genera Masirana Komatsu, 1942 and Longileptoneta n. gen. (Araneae, Leptonetidae) are described from Gyeongsangbuk-do and Chungcheongbuk-do, Korea, under the names Longileptoneta songniensis n. sp., Masirana bonghwaensis n. sp., M. ilweolensis n. sp., and M. flabelli n. sp. A new genus, Longileptoneta n. gen., is also described.

Growth, Flowering, and Ripening Seed Characteristics of Mammillaria goldii, M. theresae, and M. pseudopectinata (선인장 Mammillaria goldii, M. theresae와 M. pseudopectinata의 생육, 개화 및 종자형성 특성)

  • This experiment was conducted to investigate the propagation system by seed to check growth, flowering, and making seed of three-year-old cactus of potted Mammillaria goldii. M. theresae, and M. pseudopeclinata. Plant height and width of them was 2.5~2.8 cm, and 2.8~3.1 cm, and seed formation of Mammillaria goldii and M. theresae had internal body taking130 to 135 days to ripening seeds, however, M. pseudopectinata had externals taking just 26 days to ripening seeds. The mean seeded pod from April to June of M. goldii was 50.0% and the number of seeds in a pod was 25.3. In M. theresae, the mean seeded pod from April to June was 57.3% and the number of seeds in a pod was 35.0. However, the highest rate of seeded pod on May of M. pseudopectinata was 75.0% and the number of seed in a pod was 66.0. The sib crossing was much more ripening seed fertilization than that of self cross ing in the Mammillaria. The self crossing was formed from 12.0% to 20.0% of seeded pod, from 12.0 to 16.0 number of seed in a pod. However, sib crossing obtained 58.0% of seeded pod, 30 seeds in a pod of M. goldii, 65.0% seeded pods and 40 seeds in a pod of M. theresae, and 75.0% seeded pod, 68 seeds in a pod of M. pseudopectinala as the highest of them.

  • 서비스 거부 공격 (Denial of Service: DoS)은 매년 사이버 테러 이슈로 떠오르고 있는 대표적인 공격 양상 중의 하나이다. DoS 공격은 오래된 공격 유형으로 이에 대응하기 위한 연구가 많이 진행되고 있지만, IP(Internet Protocol) 주소를 변조할 수 있는 프로토콜 취약점 때문에 공격 발신지 역추적에 어려움이 있으며 DoS 공격의 다양한 성장으로 이어지고 있다. 현재 대부분의 변형된 DoS 공격들은 IP 주소를 변조하는 IP 스푸핑에 기반하고 있다. 이 때문에 익명성을 이용하여 공격 시도가 지속적으로 발생하고 있으며 정상적인 사용자와 공격자 간의 구분이 불분명하여 방어하기가 어렵다. 그렇기 때문에 본 논문에서는 여러 변형된 DoS 공격 유형 및 IP 스푸핑을 방지하기 위한 연구들을 분석함으로써 향후 공격자의 역추적을 돕고 DoS 공격을 차단하기 위해 필요한 정보를 제공한다.

Numerical simulation of the constructive steps of a cable-stayed bridge using ANSYS

  • This work addresses a three-dimensional nonlinear structural analysis of the constructive phases of a cable-stayed segmental concrete bridge using The Finite Element Method through ANSYS, version 14.5. New subroutines have been added to ANSYS via its UPF customization tool to implement viscoelastoplastic constitutive equations with cracking capability to model concrete's structural behavior. This numerical implementation allowed the use of three-dimensional twenty-node quadratic elements (SOLID186) with the Element-Embedded Rebar model option (REINF264), conducting to a fast and efficient solution. These advantages are of fundamental importance when large structures, such as bridges, are modeled, since an increasing number of finite elements is demanded. After validating the subroutines, the bridge located in Rio de Janeiro, Brazil, and known as "Ponte do Saber" (Bridge of Knowledge, in Portuguese), has been numerically modeled, simulating each of the constructive phases of the bridge. Additionally, the data obtained numerically is compared with the field data collected from monitoring conducted during the construction of the bridge, showing good agreement.

  • The design of segmental bridges, a structure that typically employs precast prestressed concrete elements and the balanced cantilever construction method for the deck, may demand a highly complex structural analysis for increased precision of the results. This work presents a comprehensive numerical analysis of a 3D finite element model using the software ANSYS, version 21.2, to simulate the constructive deck stages of the New Guaiba Bridge, a structure located in Porto Alegre city, southern Brazil. The materials concrete and steel were considered viscoelastic. The concrete used a Generalized Kelvin model, with subroutines written in FORTRAN and added to the main model through the customization tool UPF (User Programmable Features). The steel prestressing tendons used a Generalized Maxwell model available in ANSYS. The balanced cantilever constructive steps of a span of the New Guaiba Bridge were then numerically simulated to follow the actual constructive sequence of the bridge. A comparison between the results obtained with the numerical model and the actual vertical displacement data monitored during the bridge's construction was carried out, showing a good correlation.
