Alcaligenes sp. J-482 로부터 분리한 제한효소 AspJI의 특성 (Characterization of a Restriction Endonuclease AspJI from Alcaligenes sp. J-482)

  • 자연계에서 새로운 제한효소 생산균을 검색하여 한 균주를 선발하고, 형태학적, 생리학적, 생화학적 특성들을 조사하여 Alcaligenes sp.로 동정하고 제한효소의 특성을 조사하였다. Alcaligenes sp. J-482가 생산하는 제한효소를 AspJI으로 명명하였다. AspJI은 pBR322, Adenovirus 2-DNA, ${lambda}$ DNA 등에 대한 절단양식이 AatII와 같아 AatII의 isoschizomer로 추정 되었으며, 효소활성에 12.5mM 이상의 $MgCl_2$를 필요로 하였으며, NaCl에 의하여 저해되었다. AspJI의 반응 최적 온도는 $37^{circ}C$, 최적 pH는 7.5로 확인 되었으며, 내열성을 조사한 결과 $85^{circ}C$이상에서 15분처리 할 때 안전히 실활되는 것으로 관찰되었다.

Genotoxic Effects on Gas Station Attendants in South-southeastern México due to Prolonged and Chronic Exposure to Gasoline

  • Background: Gasoline, a complex mixture of volatile organic compounds is classified as possibly carcinogenic to humans. Gasoline station attendants, consistently exposed to its hazardous components, may face genotoxic effects. This study aimed to assess the influence of varying work shift durations on DNA damage in gasoline station attendants. Methods: Ninety individuals from three locations in southern México were studied. Peripheral blood mononuclear cells (PBMCs) were isolated, and DNA damage was assessed using the comet assay. Demographic, occupational, and lifestyle data were collected. Statistical analyses included t-tests, ANOVA, and Pearson correlation. Results: Significant differences in DNA damage parameters were observed between exposed and unexposed groups. The impact of tobacco, alcohol, and exercise on DNA damage was negligible. Extended work shifts (12 and 24 hours) showed heightened DNA damage compared to 8-hour shifts and the unexposed group. A novel finding revealed a modest but significant correlation between DNA damage and job seniority. Conclusion: The study highlights the intricate relationship between occupational exposure to gasoline components, DNA damage, and work shift lengths. Extended shifts correlate with heightened genotoxic effects, emphasizing the importance of personalized safety measures. The significant correlation between DNA damage and job seniority introduces occupational longevity as a determinant in the genetic health of gasoline station attendants. This discovery has implications for implementing targeted interventions and preventive strategies to safeguard workers' genetic integrity throughout their years of service. The study calls for further exploration of unconsidered factors in understanding the multifactorial nature of DNA damage in this occupational setting.

누에 RAPD-PCR 분석을 위한 기초연구 (Fundamental Study for RAPD-PR Analysis in the Silkworm, Bombyx mori)

  • 누에 RAPD-PCR 최적조건에 대해 실험한 결과, 중합효소 연쇄반응(PCR)의 최적조건은 반응액 25$\mu$l에 대해 주형 DNA 30ng, dNTP mixture 200$\mu$M, primer 300mM, Taq DNA Polymerase 1.0unit 및 Mg2+ 1.5mM로 판단되었으며, 또 상기의 실험조건을 기본으로 하여 annealing 온도를 탐색한 결과 35$^{\circ}C$-42$^{\circ}C$에서 DNA의 증폭이 안정적임이 밝혀졌다. 또한 동일한 품종내에서 개체간 및 암수간의 DNA 다형성은 인정되지 않았고, 품종간 DNA 다형성은 OPM 04 primer를 사용한 결과 5개의 RAPD 마커로 인정 되었다. 양친간 다형성이 인정된 primer를 사용하여 F2 33개체에 대해 DNA를 증폭시킨 결과 800 bp band가 3 :1 로 분리되었으므로 누에 품종간 유전적 유연관계 및 유전자 지도작성의 가능성을 제시했다.

Evidence for a Common Molecular Basis for Sequence Recognition of N3-Guanine and N3-Adenine DNA Adducts Involving the Covalent Bonding Reaction of (+)-CC-1065

  • The antitumor antibiotic (+)-CC-1065 can alkylate N3 of guanine in certain sequences. A previous high-field $^1H$ NMR study on the$(+)-CC-1065d[GCGCAATTG*CGC]_2$ adduct ($^*$ indicates the drug alkylation site) showed that drag modification on N3 of guanine results in protonation of the cross-strand cytosine [Park, H-J.; Hurley, L. H. J. Am. Chem. Soc.1997, 119,629]. In this contribution we describe a further analysis of the NMR data sets together with restrained molecular dynamics. This study provides not only a solution structure of the (+)-CC-1065(N3- guanine) DNA duplex adduct but also new insight into the molecular basis for the sequence- specific interaction between (+)-CC-1065 and N3-guanine in the DNA duplex. On the basis of NOESY data, we propose that the narrow minor groove at the 7T8T step and conformational kinks at the junctions of 16C17A and 18A19T are both related to DNA bending in the drugDNA adduct. Analysis of the one-dimensional $^1H$ NMR (in $H_2O$) data and rMD trajectories strongly suggests that hydrogen bonding linkages between the 8-OH group of the (+)-CC-1065 A-sub-unit and the 9G10C phosphate via a water molecule are present. All the phenomena observed here in the (+)-CC-1065(N3-guanine) adduct at 5'$-AATTG^*$are reminiscent of those obtained from the studies on the (+)-CC-1065(N3-adenine) adduct at $5'-AGTTA^*$, suggesting that (+)-CC-1065 takes advantage of the conformational flexibility of the 5'-TPu step to entrap the bent structure required for the covalent bonding reaction. This study reveals a common molecular basis for (+)-CC-1065 alkylation at both $5'-TTG^*$ and $5'-TTA^*$, which involves a trapping out of sequence-dependent DNA conformational flexibility as well as sequence-dependent general acid and general base catalysis by duplex DNA.

  • DNA-dependent protein kinase catalytic subunit (DNA-PKcs), a member of the phosphatidylinositol 3-kinase-related kinase family is a well-known player in repairing DNA double-strand break through non-homologous end joining pathway. This mechanism has allowed us to understand its critical role in T and B cell development through V(D)J recombination and class switch recombination, respectively. We have also learned that the defects in these mechanisms lead to the severely combined immunodeficiency (SCID). Here we highlight some of the latest evidence where DNA-PKcs has been shown to localize not only in the nucleus but also in the cytoplasm, phosphorylating various proteins involved in cellular metabolism and cytokine production. While it is an exciting time to unveil novel functions of DNA-PKcs, one should carefully choose experimental models to study DNA-PKcs as the experimental evidence has been shown to differ between cells of defective DNA-PKcs and those of DNA-PKcs knockout. Moreover, while there are several DNA-PK inhibitors currently being evaluated in the clinical trials in an attempt to increase the efficacy of radiotherapy or chemotherapy, multiple functions and subcellular localization of DNA-PKcs in various types of cells may further complicate the effects at the cellular and organismal level.

Cloning and Expression of Bovine Polyadenylate Binding Protein 1 cDNA in Mammary Tissues

  • A pregnant-induced clone was identified by differential screening from a cDNA library of bovine mammary gland. The clone was identified as a cDNA encoding a polyadenylate binding protein 1 (PABP). The cDNA clone had a total length of 1,911 nucleotides coding for 636 amino acids. The nucleotide sequence of the bovine PABP was 95% and 94% identical to those of human and mouse species, respectively. Comparison of the deduced amino acid sequences of bovine PABP with those of human species showed 100% identity. Induction of the PABP mRNA was observed in bovine mammary tissues at pregnant 7 and 8 months compared to virgin, lactating and involuted states. Expression of the PABP gene was examined in mammary epithelial HC11 cells at proliferating, differentiated and apoptotic conditions. The mRNA levels of PABP gene were similar between proliferating and differentiated cells, but expression levels were very low in apoptotic cells compared to other conditions. Results demonstrate that the PABP gene is induced during pregnancy at which stage mammary epithelial cells are actively proliferating.

Synthesis and characterization of doxorubicin hydrochloride drug molecule-intercalated DNA nanostructures

  • In this paper, we demonstrate the feasibility of constructing DNA nanostructures (i.e. DNA rings and double-crossover (DX) DNA lattices) with appropriate doxorubicin hydrochloride (DOX) concentration and reveal significant characteristics for specific applications, especially in the fields of biophysics, biochemistry and medicine. DOX-intercalated DNA rings and DX DNA lattices are fabricated on a given substrate using the substrateassisted growth method. For both DNA rings and DX DNA lattices, phase transitions from crystalline to amorphous, observed using atomic force microscopy (AFM) occurred above a certain concentration of DOX (at a critical concentration of DOX, $30{\mu}M$ of $[DOX]_C$) at a fixed DNA concentration. Additionally, the coverage percentage of DNA nanostructures on a given substrate is discussed in order to understand the crystal growth mechanism during the course of annealing. Lastly, we address the significance of optical absorption and photoluminescence characteristics for determining the appropriate DOX binding to DNA molecules and the energy transfer between DOX and DNA, respectively. Both measurements provide evidence of DOX doping and $[DOX]_C$ in DNA nanostructures.

Holstein의 유량과 유지방 생산에 미치는 Mitochondrial DNA D-loop 영역의 염기 서열 변이 효과 (Effect of Sequence Variation in Mitochondrial DNA D-loop Region on Milk and Milk Fat Production in Holstein Cows)

  • 본 연구는 젖소(Holstein종)의 mtDNA D-loop 영역 염기변이 다형성과 경제형질간의 관련성을 분석하기 위하여 수행하였다. 젖소의 mtDNA D-loop 영역에서 단일염기의 치환에 의해 총 35개의 polymorphic site가 확인되었다. 그중 주요 Polymorphic site의 염기변이 빈도는 106, 169, 16057, 16231 및 16255 지역에서 높은 빈도의 염기치환이 검출되었다. 169 지역은 A가 G로 염기치환이 일어났고 그 빈도는 0.555로 높게 나타났으며 염기치환에 의한 산유량 효과는 1259.6 kg(P<0.05)로 나타났다. 유지방과의 관계를 분석한 결과 16118 지역은 A가 G로 치환되는 빈도가 0.02로 검출되었으며, 16135 지역은 T가 C로 치환되는 빈도가 0.02로 검출되었고, 16302 지역은 G가 A로 치환되는 빈도가 0.04로 검출되었다. 이 지역들이 유지방에 미치는 변이 효과는 - 156, - 118.15 및 - 67.77 kg(P<0.1)으로 나타났다. 본 연구에서 검출한 젖소 mtDNA내 D-loop 영역의 염기서열 변이 빈도와 유량, 유지방과의 연관성 분석 결과 등은 젖소집단의 유전적 변이성 추정과 좀 더 다양한 경제형질과의 관련성 분석으로 다양한 유전적 지표인자 발굴에 도움이 될 것이며 이를 통해 분자유전학적 기법을 이용한 젖소의 육종전략을 확립하는데 기초 자료가 되는 것은 물론 분자육종학적인 연구에 기초 자료로서 유용하게 활용할 수 있을 것으로 기대된다.

Identification of Adenosine Deaminase Inhibitor-producing Bacterium Isolated from Soil

  • An adenosine deaminase inhibitor-producing bacterium was isolated from soil. An isolate exhibiting high adenosine deaminase inhibitory activity, was designated J-89, and classified as a strain of Bacillus subtilis on the basis of its morphological, phenotypic characteristics, the menaquinone content and cellular fatty acid composition. To confirm the taxonomic position of the strain we need more information such as DNA-DNA homology and other chemotaxonomic characteristics. In this paper we provisionally named strain J-89 as Bacillus sp. J-89 pending further chemotaxonomic study and analysis of adenosine deaminase inhibitor.

First Record of the Brackish Water Amphipod Jesogammarus (Jesogammarus) hinumensis (Amphipoda: Anisogammaridae) from Korea with DNA Barcode Analysis among Jesogammarus Species

  • Jesogammarus (Jesogammarus) hinumensis Morino, 1993 was discovered firstly from a brackish water region in Jeju Island, Korea. To identification of the specimens we conducted both of morphological and molecular analyses. This species is characterized by having large eyes and a robust seta on the mandibular palp article 1. The morphology of this Korean specimens was well matched with the original description without variation. The mitochondrial cytochrome c oxidase subunit I(COI) sequences of the present specimens were also completely identical to the sequences of J. (J.) hinumensis collected from the type locality of the species. Thus, we concluded that the Jesogammarus species from Jeju Island is J.(J.) hinumensis, based on both morphological and molecular data.
