Quality Control of Photosystem II during Photoinhibition

  • The reaction center Dl protein of photosystem II is the target of photodamage by excess illumination. The Dl protein is damaged by reactive oxygen species generated by photochemical reactions and then degraded by specific proteolytic enzymes. We found that the Dl protein also cross-links with the surrounding polypeptides, such as D2 and CP43 in isolated thylakoids or photosystem II-enriched membranes from spinach under the illumination with strong visible light. The cross-linking was observed in spinach leaf discs as well when they were illuminated at higher temperature (40°C). It was also shown that the cross-linked products are digested efficiently by a protease(s) in the stroma. Thus the cross-linking/digestion processes of the Dl protein seem to comprise a new pathway in the turnover of the photodamaged Dl protein. It should be noted, however, that the cross-linked products of the Dl protein and CP43 induced by endogenous cationic radicals in the donor-side photoinhibition are resistant to proteolytic digestion. Accumulation of these cross-linked products in the thylakoids may lead to the decay of the function of chloroplasts and finally to the death of plant cells. Thus, we suggest that the quality control of photosystem II, especially removal of the cross-linked products of the Dl protein, is crucial for the survival of chloroplasts under the light stress.

수란우의 혈청화학치의 농도가 수정란이식 후 수태율에 미치는 영향 (The Effect of Serum Chemical Values in Recipients on Pregnancy Rate following Embryo Transfer)

  • This study was done to examine the relationship between serum chemical values (urea nitrogen, glucose, total protein and cholesterol) of recipients and pregnancy rate following embryo transfer. Blood samples were taken from 184 Holstein heifers or cows on Day 6 or 7 (Day 0=day of estrus) to analysis for serum urea nitrogen, glucose, total protein and cholesterol concentrations. After selection of recipients, frozen Holstein embryos were thawed and directly transferred to recipients non-surgically. The average serum concentrations of urea nitrogen, glucose, total protein and cholesterol were 13.8 mg/dl, 56.5 mg/dl, 7.2 mg/dl, 124.8 mg/dl, respectively. The average concentrations of serum urea nitrogen and cholesterol were lower (P<0.05) in pregnant recipients (10.7 mg/dl, 99.2 mg/dl) than in non-pregnant recipients (13.0 mg/dl, 122.2 mg/dl), respectively, although the concentrations of glucose and total protein were not different. These results show serum urea nitrogen and cholesterol concentrations can be used important factors for selection of recipients in Holstein.

한우(韓牛)의 관절활액(關節滑液)에 관한 연구(硏究) 2. 활액단백성상(滑液蛋白性狀) (Studies on Normal Synovial Fluid from Tibiotarsar Joints of Korean Native Cattle II. Synovial Protein Values)

  • The mean values of total protein, albumin, globulins and A/G ratio of synovial fluid from the normal tibiotarsal joints of 55 healthy Korean native cattle were investigated. The results obtained were summarized as followings. 1. The mean values of synovial total protein for the entire group were $0.98{\pm}0.05g/dl$, with a range of 0.43 to 1.83g/dl for individual samples, and $1.00{\pm}0.07g/dl$ in slaughtering cattle and $0.92{\pm}0.06g/dl$ in living group, respectively. Compared with serum, synovial fluid contained far less total protein(p<0.01). 2. The mean values for the group were; albumin, $0.42{\pm}0.02g/dl$, globulins, $0.56{\pm}0.04g/dl$, and A/G ratio, $0.99{\pm}0.10$, with a range of 0.17-0.82g/dl, 0.03-1.32g/dl, and 0.15-3.15 for individual sample, respectively. 3. No statistical significant differences in the mean values of total protein, albumin, globulins, and A/G ratio have been observed between the synovial fluids of slaughtering and living animals. 4. Significant correlations existed between the globulin levels of synovial fluid and serum(r=0.3939), but the other values were not established.

Formation of Cross-Linked Products of The Reaction Center D1 Protein in Photosystem II under Light Stress

  • When illuminated with strong visible light, the reaction center Dl protein of photo system II is photodamage and degraded. Reactive oxygen species and endogenous cationic radicals generated by photochemical reactions are the cause of the damage to the Dl protein. Recently we found that the photodamaged Dl protein cross-links with the surrounding polypeptides such as D2 and CP43 in photosystem II. As the cross-linking reaction is dependent on the presence of oxygen, reactive oxygen species are suggested to be involved. Among the reactive oxygen species examined, ? OH was most effective in the formation of the cross-linked products. These results indicate that the cross-linking is mostly due to ? OH generated at photosystem II. The cross-linking site of the Dl protein is not known. As several tyrosine residues exist at the D­E loop of the Dl protein, there is a possibility that di-Tyr is formed between the D­E loop of the Dl protein and surrounding polypeptides during the strong illumination. Therefore, we examined the formation of di-Tyr using the monoclonal antibody against di-Tyr under excess illumination of the photosystem II membranes. The results obtained here suggest that no di-Tyr is formed during the excess illumination of photosystem II.

임신주령과 진통에 따른 임부 혈청 C-Reactive Protein의 동태 (C-Reactive Protein in Pregnancy and Labor)

  • 임신주령과 진통 및 분만의 진행정도에 따른 정상 임부의 혈청 CRP치의 동태를 파악하기 위해서 1992년 3월 1일 부터 1993년 8월 31일 까지 18개월간 영남대학교 의과대학 부속병원 산부인과에서 임신 20주에서 44주 사이의 건강한 임부 521명을 대상으로 혈청 CRP치를 측정하여 임신 주령과 진통의 유무, 양막의 파열여부 및 분만의 진행정도에 따라 비교 분석한 결과는 다음과 같다. 1. 혈청 CRP치 0.8mg/dl 이상과 2.0mg/dl 이상을 나타낸 임부의 빈도는 각각 12%(61/521)와 4%(22/521)이었다. 2. 임신주령 37주이하의 만삭전 임부군과 38주 이상의 만삭 임부군, 양막 파열 임부군과 비파열 임부군, 자궁경관 개대 3cm이하 임부군과 4cm 이상 임부군의 비교에서 혈청 CRP치 0.8mg/dl 이상과 2.0mg/dl 이상을 나타낸 임부의 빈도는 통계적으로 유의한 차이가 없었다. 3. 진통이 없는 임부군과 진통중인 임부군의 비교에서 혈청 CRP치 0.8mg/dl 이상을 나타낸 임부의 빈도는 각각 5.93%와 13.73%로 통계적으로 유의한 차이(p<0.05)가 있었으나 혈청 CRP치 2.0mg/dl 이상을 나타낸 임부의 빈도는 차이가 없었다. 4. 임신주령 37주 이하의 만삭전 임부군에서 혈청 CRP치 0.8mg/dl 이상을 나타낸 임부의 빈도는 진통중인 임부군이 23.64%로 진통이 없는 임부군의 4.69% 보다 현저히 높았으며(p<0.001), 혈청 CRP치 2.0mg/dl 이상을 나타낸 임부의 빈도도 진통중인 임부군과 진통이 없는 임부군이 각각 12.73%와 3.13%로 통계적으로 유의성 있는 차이(p<0.05)가 있었다. 임신주령 38주 이상의 만삭 임부군에서 진통이 없는 임부군과 진통중인 임부군을 비교하여 혈청 CRP치 0.8mg/dl 이상과 2.0mg/dl 이상을 나타낸 임부의 빈도는 통계적으로 유의성 있는 차이가 없었다. 이상 본 연구의 결과를 종합하면 임부의 감염을 조기 예측하는데 있어 기왕에 조기진통 임부에서 임상적 감염의 지표로 이용되고 있는 혈청 CRP치 2.0mg/dl은 진통중인 만삭임부에서도 양막파열 여부와 분만의 진행정도와 무관하게 유용한 것으로 추정되며, 조기진통 임부의 처치시 혈청 CRP치가 0.8mg/bl 이상인 경우 임부의 불현성 감염을 의심하여야 할 것으로 생각된다.

수유기간에 따른 Preterm Milk의 질소 함량 변화에 관한 연구 (Changes in Contents of Nitrogen of Preterm Milk during Lactation)

  • Nitrogen(N) concentration of preterm(PT) and term (T) milk in various fractions, such as total, protein, nonprotein, whey protein and casein were determined at 2-5 days, 1, 2, 4 and 6 weeks of postpartum. The purpose of this study was to investigate the nitrogen concentration of human milk from mothers delivering at preterm and term, and the propriety of preterm milk for premature infants. The concentration of total N, nonprotein N, protein N, whey protein N and casein N in preterm milk was decreased significantly with time postpartum. Total N was 374mg/이 at colostrum, 232mg/dl at mature milk. Whey protein N was decreased from 42mg/dl at 2-5 days to 32mg/dl at 4-6 weeks. Protein N was 332mg/dl at colostrum, 202mg/dl at mature milk. The proportion of whey protein N and casein N were 39:61 at colostrum, 28:72 at mature milk. No difference were found between T and PT milk for total nitrogen excepted 2 weeks. In this report we show that nitrogen concentration except casetpt casein N is smaller in milk from mothers giving birth prematurely than in milk from mothers giving birth at term, over the first two weeks of lactationl But protein N was higher in preterm milk than term milk, whey protein nitrogen was lower. By comparing predicted nitrogen intakes to estimated requirements of preterm infants fed 150 to 200ml/kg/day of their own mother's milk, we predict that the quantities of protein provided would be adequate to meet the requirements of the prematured infants during the early weeks of life.

  • The inhibitory effect of dl-puerol A as but-2-enolide isolated from Sophora japonica was investigated on copper ion-induced protein oxidative modification in vitro. It inhibited copper-induced protein oxidative modification. However, its inhibitory effect was a little weaker than that of $dl-{\alpha}-tocopherol$ as an antioxidant. The results demonstrated that dl-puerol A, one of but-2-enolides, might be of use in the oxidative stress.

  • Total protein levels of 60 bovine bull sera were treasured with refractometer and the strum proteins were fractionated by cellulose acetate membrane electorphoresis and the relative amount of each fraction was measured with automatic scanning densitometer. The sixty bovine bulls consisted of 17 Charolais bulls, 28 Holstein bulls and 15 Korean bulls. Mean total serum protein level of the total bulls was $7.50{\pm}0.50(Mean{\pm}SD)$, with the mean of Charolais $7.04{\pm}0.37g/dl$, the mean of Holstein $7.62{\pm}0.40g/dl$ and the mean of Korean $7.81{\pm}0.43g/dl$. As barbital-calcium buffer was used in fractionating the bovine serum protein with cellulose acetate membrane, electric current of 0.4 mA per centimeter width of the membrane for an hour resulted in more clear separation between ${\alpha}_1$-globulin and ${\alpha}_2$-globulin than electric current of 0.6 or 1.0 mA for an hour. However ${\alpha}_1$-globulin and ${\alpha}_2$-globulin could not be measured separately with automatic scanning densitometer. Mean serum albumin level of total bulls was $3.54{\pm}0.42g/dl$, with the mean of Charolais $3.42{\pm}0.26g/dl$, the mean of Holstein $3.69{\pm}0.38g/dl$ and the mean of Korean $3.39{\pm}0.55g/dl$. Mean serum ${\alpha}$-globulin level of the total bulls was $0.62{\pm}0.14g/dl$, with the mean of Charolais $0.61{\pm}0.07g/dl$, the mean of Holstein $0.60{\pm}0.11g/dl$ and the mean of Korean $0.67{\pm}0.23g/dl$. Mean ${\beta}$-globulin level of the total bulls was $0.85{\pm}0.21g/dl$, with the mean of Charolais $0.74{\pm}0.11g/dl$, the mean of Holstein $0.83{\pm}0.14g/dl$ and the mean of Korean $1.02{\pm}0.30g/dl$. Mean serum ${\gamma}$-globulin level of the total bulls was $2.48{\pm}0.45g/dl$ with the mean of Charolais $2.27{\pm}0.37g/dl$, the mean of Holstein $2.48{\pm}0.44g/dl$ and the mean of Korean $2.72{\pm}0.47g/dl$.

도시 저소득층 취학전 어린이들의 영양상태에 관한 연구 - II. 단백질영양상태와 면역 기능에 관한 생화학적 지표에 관한 연구 - (Nutritional Status of Preschool Children Residing in Low Income Urban Area - II. Biochemical Parameters of Protein Nutrition and Immunity -)

  • The purpose of this study was to determine biochemical parameters related to protein and immunity. Subjects were 125 preschool children(M:69, F:56) residing in low income area of Seoul. Mean serum total protein of the children aged 6 was 7.3g/dl which was significantly higher than 6.6g/dl of the group aged 3. The mean serum albumin was 4.7g/dl for 3, 4, 5 age group, and 4.9g/dl for 6 age group and there was no significant difference. Serum retinol binding protein(RBP) is used as a sensitive indicatior of protein, because it tends to fall rapidly in response to protein status and respond to quickly dietary treatment. Mean RBP for each group(3, 4, 5 and 6 age group) were $2.5\mu{g}$/dl, $2.9\mu{g}$/dl, $2.7\mu{g}$/dl, $3.0\mu{g}$/dl. The Proportion of children whose RBP was than $2.6\mu{g}$/dl was 15.9%, 19.2%, 24.3% and 16.7%, respectively. The 24-hour urinary excretion of hydroxyproline was 7.9mg, 14.6mg, 11.7mg and 11.8mg for each group and the mean excretion of all children was 12.2mg/day. Children aged 3 were excreting significantly lower amount of hydroxprolinc per day than the children aged 4. The mean hydroxyproline index were 2.18, 2.39, 2.52, 2.80 for each age group and the mean of a group aged 6 was significantly higher than that of the group aged 3. The proportion of children assessed as malnourished and impaired growth(hydroxyproline index <2.0) was 18.8%, 4.9%, 2.5% and 4.3%, respectively. The nutrients which showed significant relationship with protein and immunity parameters were niacin, vitamin C and calcium. Vitamin C showed significant positive relationship(p<0.05-p<0.01) with serum RBP, total protein and globulin. The triceps skinfold thickness was significantly and positively correlated with serum globulin. Serum IgG showed significant positive relationship with height, weight, girth of chest and midarm circumference(p<0.05-p<0.01).

개 혈청화학검사항목의 장기간의 개체 내 변이와 Critical Difference (Long-term Intra-individual Variations and Critical Differences of Clinical Chemical Parameters in Dogs)

  • 이 연구의 목적은 BUN 및 혈청 creatining, total protein, albumin과 glucose의 장기간에 걸친 개체 내 변이를 산정하여, 각각의 critical difference를 구성하는 것이다. 임상적으로 건강한 20마리의 개에서 일주일에 한번씩 총 8주간 채혈하여, 장기간에 걸친 개체 내 변이를 구하고, critical difference를 산출하였다. BUN은 9.01 mg/dl, 혈청 creatining은 0.52 mg/dl, total protein은 0.99 g/dl, albumin은 0.39 g/dl, glucose는 20.64 mg/dl로 critical difference가 산출되었다. 동일한 개에서 연속적으로 측정된 검사항목 치가 critical value보다 차이가 적으면, 이 차이는 생리적인 변화에 기인한다고 할 수 있으며, critical value보다 차이가 크면, 다른 요인, 즉 질병의 진행이나 다른 동반된 질병의 존재를 의미한다고 할 수 있다.
