Effect of acetazolamide on the diuretic action of furosemide was investigated in rabbits. The rates of urine flow and excretion of salts were significantly reduced when furosemide (0.5 mg/kg) was administered with acetazolamide (10 mg/kg) compared to the diuretic response of the single furosemide (0. 5 mg/kg) administration. Reduction in the fractional excretion rate of urine volume was more pronounced than the fractional excretion rate of salts. The results suggest that reduction of diuretic action on furosemide by combined administration of acetazolamide is probably due elevated urinary pH and interference in the mechanism of inhibition of chloride transport in the ascending Henle's limb.
Buxus leaves(Buxus microphylla var. koreana Nakai) have been used as a folk medicine in treating woman's disease. The chemical constituents, which have teen identified from Buxus sempervirens L. in Europe, are so far known in isolation of buxus alkaloids, cyclobuxine, buxamine, norbuxamine and buxaminol. But But any study on the pharmacological action of Buxus microphylla var. koreana Nakai has not yet teen rallied out, however, active substances have not been identified. As an attempt to isolate the biologically active substances from this plant, the examination of chemical constituents was undertaken. In the experiment, the crystalline component$(C_{10}H_8O_4)$ obtained from Buxus microphylla var. koreana Nakai was identified as a 6-methoxy, 7-hydroxy coumarin by chemical and physical methods. We named this crystal buxuletin. Therefore, we intended to screen the pharmacological action of buxuletin, especially, its diuretic effects in rabbits. Buxuletin was intraveneously injected to the rabbit in the dose of 20 mg/kg, 10 mg/kg and 5 mg/kg. The variations of urine volume and minerals $(Na^+,\;K^+\;and\;Cl^-)$ in urine were measured at an interval of 15 minutes for 4 hours after the treatment of buxuletin. The observed results are as follows: 1) With the administration of buxuletin (20 mg/kg, 10 mg/kg), the remarkable increment of the excreted urine volume was observed during the relatively long period. Excretion rates of urinary minerals $(Na^+,\;K^+\;and\;Cl^-)$ were also increased. 2) In a dose of 5 mg/kg, the slight increment on the diuretic actions of rabbits was observed. From the above results, authors name this crystal buxuletin and buxuletin shows the diuretic action.
Diuretic renal scan, a modified method of conventional radionuclide renography by administration of intravenous furosemide was performed in 54 patients with suspected urinary tract obstruction. Two showed normal responses, 41 showed increased washout patterns, and the remainder 15 revealed no changes in response to the furosemide administration. IVP was done in 34 patients. In comparision to the diuretic renal scan, the following results were obtained. 1) Patients with normal results by the scan, showed normal IVP findings also. 2) 10 patients who showed delayed excretory patterns not influenced by the diuretics administration. 9 patients revealed obstructive lesions and hydronephrotic changes, and CPN was diagnosed for the remainder one and the one patient without obstruction was diagnosed as chronic pyelonephritis. 3) 22 patients showed positive results to the diuretics. Among them 10 patients had positive results in IVP. but, were not related to the speed of decrement of the renal activity after the furosemide administration.
The effects of UK 14,304, an ${\alpha}_2$-adrenergic agonist, on renal function were investigated in dogs. UK 14,304, when given intravenousely($15.0{\mu}g/kg,\;50{\mu}g/kg$), produced the increase of urine flow accompanied with the marked augmentation of free water clearance ($C_{H_2O}$) and reabsorption rates of sodium in renal tubules ($R_{Na}$), and the remarkable decrease of osmolar clearance ($C_{osm}$) and the amounts of sodium excreted in urine ($E_{Na}$). UK 14,304 given into a renal artery($1.5{\mu}g/kg,\;5.0{\mu}g/kg$) elicited the increase of urine flow with the augmentation of $C_{H_2O}$ in both kidney. UK 14,304, when administered into carotid artery($3.0{\mu}g/kg,\;10.0{\mu}g/kg$). exhibited the same aspect as shown in intravenous UK 14.304 at smaller dose than the intravenous dose. Diuretic action of intravenous UK 14,304 were produced together with increase of $C_{H_2O}$ in situation of water diuresis too, changes of renal function in this state were the increase of $C_{osm},\;E_{Na},\;and\;E_K$ (excreted amounts of potassium in urine), and the decrease of $R_{Na}\;and\;R_K$, these were different appearances from situation of saline diuresis. Diuretic action of intravenous UK 14,304 were blocked completely by post or pretreatment of yohim-bine, ${\alpha}_2$-adrenergic blocking agents, and inhibited by pretreatment of vasopressin, antidiuretic hormone. Above results suggest that UK 14,304 produces the diuretic action by the inhibition of vasopressin secretion and suppression of electrolytes reabsorption of electrolytes in renal tubules mediated with central ${\alpha}_2$-adrenoceptor in dog.
The influence of penicillin G infusion(7.5 units/min/kg) on the diuretic action of furosemide$(250{\mu}g/kg)$ in rabbits was studied to investigate the drug interaction between penicillin G and furosemide. The results were as follows: 1) There was no significant change in renal function in the penicillin G infused rabbits compared with the normal ratbit. 2) In comparison with the control group, a significant increase in ${\triangle}U_{flow},\;{\triangle}U_{Na}V,\;{\triangle}U-kV\;and{\triangle}U_{cl}V$ was noted by 30 minutes following the administration of furosemide in the penicillin G infused group. There was no significant difference in ${\triangle}C_{In},\;{\triangle}C_{PAH}$ between the two groups and ${\triangle}Na^+$ reabsorption rate was significantly decreased in the latter. The potentiating effect of penicillin G on the diuretic action of furosemide should be due to the increased luminal concentration of free form of furosemide, and it may be related to the competitive inhibition of plasma protein binding between the two drugs.
The investigation was aimed to study effects of Shirhyung-tang extract and Kamishirhyung-tang extract on the diuretic, antipyretic, anti-inflammatory, analgesic and the injured kidney induced by $HgCl_2$. The extract was administered to the experimental animals. The following results have been obtained. Duiretic effect was noted to be significant in extracts of Shirhyung-tang and Kamishirhyung-tang. Antipyretic effect was noted to be significant according to the density of Shirhyung-tang and Kamishirhyung-tang. Anti-inflammatory effect was noted to be significant in extract of Shirhyung-tang and Kamishirhyung-tang. Analgesic effect was noted to be significant in extracts of Shirhyung-tang and Kamishirhyung-tang.
In order to investigate experimentally the clinical effects of Taeksa-Tang that was prescribed to cure the severe dizziness, the water extract of Taeksa-Tang was experimented about the effects on the diuretic action, the serum lipid, the isolated ileum and heart, the blood vessel, the blood pressure and respiration. The results of this study were obtained as follows: The diuretic action was significantly shown in rabbits. The level of total cholesterol, triglyceride and phopholipid in serum of rabbits given high cholesterol diet were significantly decreased. The contraction of the isolated ileum of mice induced by acetylcholine chloride and barium chloride was remarkably inhibited. The vasodilating and hypotensive actions were noted in rabbits. Negative inotropic action on the isolated frog heart was recognized.
This study was attempted to investigate on renal effect of ($\pm$)6-chloro-7,8-dihydroxy-1-phenol 2,3,4,5-tetrahydro-lH-3 benzazepine (SKF 81297), dopamine $D_1$ receptor agonist, in dog. SKF 81297, when gluten intravenously, produced diuretic action along with the increases of renal plasma flow (RPF), glomerular filtration rate (GFR), amounts of N $a^{+}$ and $K^{+}$ excreted into urine ( $E_{Na}$ , $E_{K}$) and osmolar clearance ( $C_{osm}$). It also decreased the reabsorption rates of N $a^{+}$ and $K^{+}$ in renal tubule ( $R_{Na}$ , $R_{K}$) and free water clearance ( $C_{H2O}$), whereas ratios of $K^{+}$ agonist N $a^{+}$ in urine and filtration fraction (FF) was not changed. SKF 81297, when administered into a renal artery, elicited diuresis both in experimental kidney given the SKF 81297 and control kidney not given, while the effect was more remarkable in experimental kidney than those exhibited in control kidney. SKP-450 which is $K^{+}$ channel opener, When given into duodenum, exhibited the decline of urine flow accompanied with the decrease of glomerular filtration rates (GFR), renal plasma flow (RPF), N $a^{+}$ and $K^{+}$ excreated in the urine ( $E_{Na}$ , $E_{K}$) and the increase of $K^{+}$N $a^{+}$ratios, and then appeared the significant fall of mean arterial pressure (MAP) and unchanged of reabsorption rates of N $a^{+}$, $K^{+}$ in renal tubules ( $R_{Na}$ , $R_{K}$). SKP- 450 injected into the vein elicited the decline of urine flow along with the reduction of $E_{Na}$ , $E_{K}$, and the increase of $R_{Na}$ , $R_{K}$ and $K^{+}$M $a^{+}$ ratios. SKP-450 administered into a renal artery produced diuretic action along with the increase of $E_{Na}$ , $E_{K}$ and the decrease of $R_{Na}$ , $R_{K}$ in experimental kidney, whereas produced the same aspect to intravenous SKP-450 in the control kidney. The crude methanol extract of the roots of Achyranthes aspera Linn. was investigated for its possible antinociceptive, diuretic and neuropharmacological activities in animal models. At the dose of 250 and 500 mg/kg body weight, the extract showed a significant antinociceptive effect in acetic acid induced-writhing in mice comparable to that produced by diclofenac sodium, used as standard drug. The crude extract produced significant diuretic effect at the dose of 500 mg/kg of body weight comparable to that produced by furosemide, used as standard drug. The extract also potentiated significantly the pentobarbital induced sleeping time in mice; decreased the open field score in open field test, decreased the number of hole crossed from one chamber in the hole cross test and decreased the head dip responses. The obtained results provide a support for the use of this plant in traditional medicine and its further investigation.
