Evaluation of Cultivation Limit Area for Different Types of Barley owing to Climate Change based on Cultivation Status and Area of Certified Seed Request (기후변화에 따른 맥종별 재배실태와 보급종 보급지역에 의한 재배한계지 평가)

  • This study was conducted to determine the extent to which climate change is expanding areas in which barley can be successfully cultivated. In 2019 and 2020, we collected data on areas that had requested certified seeds from the Korea Seed and Variety Service to determine potential cultivation areas. In addition, we surveyed the growth and yield of different types of barley in fields. Certified seeds of hulled and dehulled barley were requested by farmers across Korea from the Korea Seed and Variety Service in both years. Areas that were provided with certified seeds were considered potential barley cultivation areas. The varieties and use rates of certified seeds varied based on the barley type and region. For example, certified seeds of dehulled barley in 2019 and 2020 were not used in some areas, whereas in others, these seeds constituted 100% of the seeds sown for barley crops. In 2019 and 2020, the average sowing days in Korea were from October 17 to November 9 for dehulled barley, October 26 to November 13 for hulled barley, October 19 to November 5 for malting barley, and October 3 to November 1 for naked oats. Thus, the sowing days of the barley types varied depending on the area and year they were used. For example, in the case of hulled barley in Jeonnam, some farmers sowed until December 12. The yield per 10 a of barley cultivation was typically higher in the main production areas than in the cultivation limit areas. In extreme cases, harvest was impossible in some cultivation limited areas, such as Gangwon-do. Based on the current 20-year January minimum average temperature (JMAT) in Korea (2002-2021), climate change scenarios suggest that barley cultivation is feasible, provided that the minimum temperature in January is no lower than -10℃, -8℃, and -4℃ for hulled barley, dehulled barley, and for malting barley and naked oats, respectively. Additionally, cultivation of barley across South Korea seems feasible based on data on certified barley seeds by area. Although both JMAT and certified seed data suggest that barley cultivation across Korea is feasible, our survey results of barley growth and yield showed that harvest was impossible in certain cultivation areas, such as Gangwon-do. Therefore, climate change scenarios related to the cultivation limits of different barley types need to be re-estimated by factoring in survey data on the growth and yield of crops within those cultivation areas.

Simultaneous determination of 11-nor-Δ9-carboxy-tetrahydrocannabinol and 11-nor-Δ9-carboxy-tetrahydrocannabinol-glucuronide in urine samples by LC-MS/MS and its application to forensic science (LC-MS/MS를 이용한 소변 중 11-nor-Δ9-carboxy-tetrahydrocannabinol 및 11-nor-Δ9-carboxy-tetrahydrocannabinol-glucuronide의 동시 분석 및 법과학적 적용)

  • Cannabis (Marijuana) is one of the most widely used drugs in the world, and its distribution has been controlled in South Korea since 1976. Identification of 11-nor-Δ9-carboxy-tetrahydrocannabinol (THCCOOH) in urine can provide important proof of cannabis use, and it is considered scientific evidence in the forensic field. In this study, we describe a simultaneous quantitative method for identifying THCCOOH and THCCOOH-glucuronide in urine, using simple liquid-liquid extraction (LLE), and liquid chromatography-tandem mass spectrometry (LC-MS/MS). THCCOOH-D3 and THCCOOH-glucuronide-D3 were used as internal standards. Validation results of the matrix effect, as well as recovery, linearity, precision, accuracy, process efficiency, and stability were all satisfactory. No carryover, endogenous or exogenous interferences were observed. The limit of detection (LOD) of THCCOOH and THCCOOH-glucuronide were 0.3 and 0.2 ng/mL, respectively. The developed method was applied to 28 authentic human urine samples that tested positive in immunoassay screening and gas chromatography/mass spectrometry (GC/MS) tests. The ranges of concentrations of THCCOOH and THCCOOH-glucuronide in the samples were less than LOQ~266.90 ng/mL and 6.43~2133.03 ng/mL, respectively. The concentrations of THCCOOH-glucuronide were higher than those of THCCOOH in all samples. This method can be effectively and successfully applied for the confirmation of cannabinoid use in human urine samples in the forensic field.

Biological Toxicity Assessment of Sediment at an Ocean Dumping Site in Korea (폐기물 배출해역 퇴적물의 생물학적 독성평가 연구)

  • The effect of sediments in a waste dumping area on marine organisms was evaluated using sediment toxicity tests with a benthic amphipod (Monocorophium acherusicum) and bioluminescent bacterium (Vibrio fischeri) in accordance with the Korean Standard Method for Marine Wastes (KSMMW). Nine sites in the East Sea-Byeong, East Sea-Jeong, and Yellow Sea-Byeong areas were sampled from 2016 to 2019. The test results showed that the relative average survival rate (benthic amphipods) and relative luminescence inhibition rate (luminescent bacteria) were below 30%, which were judged to be "non-toxic." However, in the t-test, a total of 12 benthic amphipod samples (6, 1, 1, and 4 in 2016, 2017, 2018, and 2019, respectively) were significantly different (p<0.05) from the control samples. To identify the source of toxicity on benthic amphipods, a simple linear regression analysis was performed between the levels of eight heavy metals (Cr, As, Ni, Cd, Cu, Pb, Zn, and Hg) in sediments and the relative average survival rate. The results indicated that Cr had the highest contribution to the toxicity of benthic amphipods (p = 0.000, R2 = 0.355). In addition, Cr was detected at the highest concentration at the DB-85 station and exceeded the Marine Environment Standards every year. Although the sediments were determined as "not toxic" according to the ecotoxicity criteria of the KSMMW, the results of the statistical significance tests and toxicity identification evaluation indicated that the toxic effect was not acceptable. Therefore, revising the criteria for determining the toxic effect by deriving a reference value through quantitative risk assessment using species sensitivity distribution curves is necessary in the future.

Evaluation of Temperature and Precipitation over CORDEX-EA Phase 2 Domain using Regional Climate Model HadGEM3-RA (HadGEM3-RA 지역기후모델을 이용한 CORDEX 동아시아 2단계 지역의 기온과 강수 모의 평가)

  • This study evaluates the temperature and precipitation results in East Asia simulated from the Hadley Centre Global Environmental Model version 3 regional climate model (HadGEM3-RA) developed by the UK Met Office. The HadGEM3-RA is conducted in the Coordinated Regional climate Downscaling Experiment-East Asia (CORDEX-EA) Phase II domain for 15 year (2000-2014). The spatial distribution of rainbands produced from the HadGEM3-RA by the summer monsoon is in good agreement with the Asian Precipitation Highly Resolved Observational Data Integration Towards Evaluation of water resources (APRODITE) data over the East Asia. But, precipitation amount is overestimated in Southeast Asia and underestimated over the Korean Peninsula. In particular, the simulated summer rainfall and APRODITE data show the least correlation coefficient and the maximum value of root mean square error in South Korea. Prediction of temperature in Southeast Asia shows underestimation with a maximum error during winter season, while it appears the largest underestimation in South Korea during spring season. In order to evaluate local predictability, the time series of temperature and precipitation compared to the ASOS data of the Seoul Meteorological Station is similar to the spatial average verification results in which the summer precipitation and winter temperature underestimate. Especially, the underestimation of the rainfall increases when the amounts of precipitation increase in summer. The winter temperature tends to underestimate at low temperature, while it overestimates at high temperature. The results of the extreme climate index comparison show that heat wave is overestimated and heavy rainfall is underestimated. The HadGEM3-RA simulated with a horizontal resolution of 25 km shows limitations in the prediction of mesoscale convective system and topographic precipitation. This study indicates that improvement of initial data, horizontal resolution, and physical process are necessary to improve predictability of regional climate model.

Changes in the Linear Compressibility and Bulk Modulus of Natural Stilbite Under Pressure with Varying Pressure-Transmitting Media (천연 스틸바이트의 압력전달매개체에 따른 선형압축률 및 체적탄성률 비교 연구)

  • This study is a preliminary step to understand the reaction between various liquids and zeolite in the subduction zone environment. Stilbite, NaCa4(Al9Si27)O72·28(H2O), was selected and high pressure study was conducted on compressional behavior by the pressure-transmitting medium (PTM). Water and NaHCO3 solution that can exist in the subduction zone was used as PTM, and samples were pressurized from ambient to a maximum of 2.5 GPa. Below 1.0 GPa, both experiments show a low linear compressibility in the range of 0.001 to 0.004 GPa-1 and a high bulk modulus of 220(1) GPa. This is presumably because the structure of the stilbite becomes very dense due to insertion of water molecules or cations into the channel. On the other hand, at 1.0 GPa or higher, the trends of the two experiments are different. In the water run, the linear compressibility of the c-axis is increased to 0.006(1) GPa-1. In the NaHCO3 run, the linear compressibility of the b- and c-axis is increased to 0.006(1) GPa-1. The bulk modulus after 1.0 GPa shows values of 40(1) and 52(7) GPa in water and NaHCO3 run, respectively, confirming that stilbite becomes more compressible than that before 1.0 GPa. It is caused by the migration of cations and water molecules inside the channel, as the water molecules in the PTM start to freeze and stop to insert toward the channel at 1.0 GPa or more. In the NaHCO3 run, it is assumed that the distribution of extra-framework species inside the structure is changed by substitution of the Na+ cation. It can be expected from tendency of the relative intensity ratio of the (001) and (020) peaks which show a different from that of the water run.

Automated Analyses of Ground-Penetrating Radar Images to Determine Spatial Distribution of Buried Cultural Heritage (매장 문화재 공간 분포 결정을 위한 지하투과레이더 영상 분석 자동화 기법 탐색)

  • Geophysical exploration methods are very useful for generating high-resolution images of underground structures, and such methods can be applied to investigation of buried cultural properties and for determining their exact locations. In this study, image feature extraction and image segmentation methods were applied to automatically distinguish the structures of buried relics from the high-resolution ground-penetrating radar (GPR) images obtained at the center of Silla Kingdom, Gyeongju, South Korea. The major purpose for image feature extraction analyses is identifying the circular features from building remains and the linear features from ancient roads and fences. Feature extraction is implemented by applying the Canny edge detection and Hough transform algorithms. We applied the Hough transforms to the edge image resulted from the Canny algorithm in order to determine the locations the target features. However, the Hough transform requires different parameter settings for each survey sector. As for image segmentation, we applied the connected element labeling algorithm and object-based image analysis using Orfeo Toolbox (OTB) in QGIS. The connected components labeled image shows the signals associated with the target buried relics are effectively connected and labeled. However, we often find multiple labels are assigned to a single structure on the given GPR data. Object-based image analysis was conducted by using a Large-Scale Mean-Shift (LSMS) image segmentation. In this analysis, a vector layer containing pixel values for each segmented polygon was estimated first and then used to build a train-validation dataset by assigning the polygons to one class associated with the buried relics and another class for the background field. With the Random Forest Classifier, we find that the polygons on the LSMS image segmentation layer can be successfully classified into the polygons of the buried relics and those of the background. Thus, we propose that these automatic classification methods applied to the GPR images of buried cultural heritage in this study can be useful to obtain consistent analyses results for planning excavation processes.

Effect of Soil Temperatures on Seedling Emergence in Direct Seeding on Dry Paddy (벼 건답직파에서 파종기 지온이 출아에 미치는 영향)

  • Soil temperatures at depths of 1~5cm are important to the germination and emergence of dry seeded-rice. An automated weather station was used to monitor the hourly weather parameters at Experiment Farm, Kyung Hee University from April 21 to May 30 in 1994. The data was analyzed to figure out the 24-hour temporal changes in air 1.5m above ground and soil temperatures under ground of 0, 2.5, 5, 10 and 20cm. The fluctuations of soil temperature were greatest at the soil surface and decreased with increasing depth. Mean soil temperatures at depth of 2.5cm were about 3$^{\circ}C$ higher than mean air temperatures during the observation period. Although mean soil temperatures at depth of 2.5cm during 10 or 15 days after April 21, May 1 and May 11 showed almost same temperatures, the distribution patterns of temperature regime were different from each other. Rice cultivars, Hwasung, Seohae, Nampung, IR60 and CR155, were seeded at depth of 2.5cm on April 21, May 1 and May 11, respectively. The periods of seedling emergence(PSE) varied in accordance with cultivars and seeding dates. PSE was correlated with accumulated daily mean air temperatures and accumulated hours classified by temperature regimes.

Effect of Temperatures during Ripening Period on Morpological Characteristics of Rachis-Grain in Rice (등숙기간중 온도와 벼 이삭 지경간 곡립의 형태 형성차이)

  • To clarify the effect of temperature during ripening on grain quality, rice plant which was grown under normal natural conditions untill heading stage was subjected various temperature regimes during grain filling stage. Three varieties, Odaebyeo, Hwaseongbyeo and Dongjinbyeo were used. Grains on primary and secondary-rachis branches were harvested seperately and measured specific gravity, grain size and weight. The optimum daily temperature for grain filling of rice during ripening period was about 22$^{\circ}C$ and grain filling under alternating temperature(26/18$^{\circ}C$, day /night) was better than constant temperature (22 /22$^{\circ}C$) based on 1, 000-grain weight and yield. Grain size based on volume per grain was smallest under the constant temperature of 22 /22$^{\circ}C$. The size of secondary-rachis branches was smaller than primary-rachis branch. The difference in size between primary and secondary-rachis branches was biggest in Hwaseongbyeo, followed by Odaebyeo and Dongjinbyeo. The temperature regimes treated did not influence the grain size difference between branches so much. The 1000-grain weight was lighter under high temperature than low temperature in Odaebyeo, but reversed tendency was observed in Dongjinbyeo of secondary rachis-branches.

Early Effect of Environment-friendly Harvesting on the Dynamics of Organic Matter in a Japanese Larch (Larix leptolepis) Forest in Central Korea (중부지역 일본잎갈나무림의 친환경벌채가 산림 내 유기물 변화에 미치는 초기 영향)

  • Environment-friendly harvesting is practiced to maintain ecosystem, landscape, and forest protection functions. The present study was conducted at Simgok-ri, Sinbuk-myeon, Pocheon, Gyeonngi-do, where a 41-50-year-old Japanese larch forest was harvested in an environment-friendly manner from 2017 to 2019. The dynamics of organic matter in this forest were investigated at three years after the harvest. Specifically, organic matter content was measured on the forest floor and in overstory biomass, litterfall, and soil up to 30 cm in depth from June 2020 to January 2021. Owing to the harvest, the amount of overstory biomass of the Japanese larch stands decreased from 142.22 to 44.20 t ha-1. On the forest floor, the amount of organic matter was 32.87 t ha-1 in the control plots and 23.34 t ha-1 in the harvest plots. Annual litterfall was 4.43 t ha-1 yr-1 in the control plots and 1.16 t ha-1 yr-1 in the harvest plots. Soil bulk density in the B horizon was 0.97 g cm-3 in the control plots and 1.06 g cm-3 i n the harvest plots. Soil organic matter content was 11.5% in the control plots and 12.8% in the harvest plots. The total amount of soil organic matter did not differ significantly between the control plots (245.21 t ha-1) and harvest plots (263.92 t ha-1), although the amount of soil organic matter tended to be higher in the harvest plots. The total amount of organic matter in the forest was estimated to be 406.48 t ha-1 in the control plots and 338.21 t ha-1 in the harvest plots. In the harvest plots, the ratio of aboveground organic matter decreased to 13.1% and soil organic matter increased to 78.0%, indicating that the distribution of organic matter changed significantly in these plots. Overall, the carbon accumulated in aboveground biomass was substantially reduced by environment-friendly harvesting, whereas the soil carbon level increased, which played a role in mitigating the reduction of system carbon in the forest. These results highlight one possible resolution for forest management in terms of coping with climate change. However, given that only three years of environment-friendly harvesting data were analyzed, further research on the dynamics of organic matter and tree growth is needed.

Fecal Microbiota Profiling of Holstein and Jersey, in South Korea : A Comparative Study (국내에서 사육되는 Holstein 젖소과 Jersey 젖소의 대변 미생물 분석 : 비교연구)

  • In light of the complex interactions between the host animal and its resident gut microbiomes, studies of these microbial communities as a means to improve cattle production are important. This study was conducted to analyze the intestinal microorganisms of Holstein (HT) and Jersey (JS), raised in Korea and to clarify the differences in microbial structures according to cattle species through next-generation sequencing. The alpha-diversity analysis revealed that most species richness and diversity indices were significantly higher in JS than in HT whereas phylogenetic diversity, which is the sum of taxonomic distances, is not significant. Microbial composition analysis showed that the intestinal microbial community structure of the two groups differed. In the both groups, a significant correlation was observed among the distribution of several microbes at the family level. In particular, a highly significant correlation (p<0.0001) among a variety of microbial distributions was found in JS. Beta-diversity analyis was to performed to statistically verify whether a difference exists in the intestinal microbial community structure of the two groups. Principal coordinate analysis and unweighted pair group method with arithmetic mean (UPGMA) clustering analysis showed separation between the HT and JS clusters. Meanwhile, permutational multivariate analysis of variance (PERMANOVA) revealed that their microbial structures are significantly different (p<0.0001). LEfSe biomarker analysis was performed to discover the differenc microbial features between the two groups. We found that several microbes, such as Firmicutes, Bacilli, Moraxellaceae and Pseudomonadales account for most of the difference in intestinal microbial community structure between the two groups.
