Lyophilized DOTMP kits were prepared using DOTMP, ammonium acetate, and ascorbic acid. The $^{68}Ga$-DOTMP was prepared by incubating the kit dissolved in 0.5 ml of concentrated $^{68}Ga$ using NaCl method and 0.5 ml of DDW, at $100^{\circ}C$ for 7 min. The labeling yield was evaluated by two solvent systems of TLC. 1 MBq of concentrated $^{68}Ga$ was labeled with $0.8{\mu}g$ of DOTMP by high radiolabeling yield (>98%), which was determined by two TLC methods. The composition of the prepared freeze-dried vial is $400{\mu}g$ of DOTMP, 19.27 mg of ammonium acetate and 17.62 mg of ascorbic acid. ~555 MBq of $^{68}Ga$-DOTMP was prepared with excellent radiochemical purity (>98%) and it was stable for 4 hr at room temperature. In conclusion, Freeze-dried DOTMP kits for the convenient preparation of $^{68}Ga$-DOTMP have been developed. Availability of this kit is expected to stimulate the widespread use of $^{68}Ga$-DOTMP in the fields of nuclear medicine.
Microbial community plays important roles in the culture environment of mud crab Scylla serrata. One of the environmental management efforts for the cultivation of S.serrata is by stabilizing microorganisms involved in nitrogen cycle process. The availability of dissolved inorganic nitrogen in its culture environment under a recirculating system closely relates to the nitrogen cycle, which involves both anaerobic and aerobic bacterial activities. Anaerobically, there are two major nitrogen compound degradation processes, i.e., denitrification and dissimilatory nitrate reduction to ammonium (DNRA). This study aimed to identify denitrifying and DNRA bacteria isolated from the recirculating cultivation of S. serrata. The water samples were collected from anaerobic filters called close filter system, which is anaerobically conditioned with the addition of varying physical filter materials in the recirculating mud crab cultures. The results showed that three denitrifying bacterial isolates and seven DNRA bacterial isolates were successfully identified. The phylogenetic analysis based on 16S rRNA gene of the denitrifying bacteria revealed that HIB_7a had the closest similarity to Stenotrophomonas daejeonensis strain MJ03. Meanwhile, DNRA bacterial isolate of HIB_92 showed a 100% similarity to Bacillus sonorensis strain N3, Bacillus vallismortis strain VITS-17, Bacillus tequlensis strain TY5, Geobacillus sp. strain DB24, Bacillus subtilis strain A1, and Bacillus mojavensis strain SSRAI21. This study provides basic information denitrifying and DNRA bacterial isolates identity which might have the potential to be applied as probiotics in aquaculture systems in order to maintain optimal environmental conditions.

In the current study, we examined the quality of surface water in ponds at two golf courses, located in southern (Country Club P) and eastern (Country Club B) areas of Korea respectively. Seasonal measurements were made of following physical parameters;pH, dissolved oxygen, biological oxygen demand, chloride, alkalinity, hardness, and nitrogen compounds [$NH_3$-N, $NO_2$-N, $NO_3$-N] and the concentrations of four heavy metals (Cd, Cu, Pb, and Zn) throughout the courses. The pH values were within the alkaline range (7.3 to 9.0), and the biological oxygen demand was generally between 0.5 and 3.1 mg/L. The alkalinity and hardness ranged from 19.1 to 68.5mg/L and 16.1 to 63.6 mg/L, respectively. Nitrogen as ammonium $(NH_3-N)$ was detected in all samples, and the samples had low concentrations of $NO_2-N$ and $NO_3-N$. Cd concentrations were relatively high (to 22.44 mg/L); the highest Cd concentration was observed in the pond in Country Club P. Other metal concentrations were low compared to the Quebec guidelines.
Using the quarternary ammonium salts as phase transfer catalyst, the nucleophilic substitution reaction of 1-chlorooctane with sodium-cyanide was investigate kinetically with respect to the formation of octanenitrile. The product was analyzed with gas chromatograph, and quantity of octanenitrile was measured. The reaction condition was considered by the effect of the reaction temperature, of the species and the amount of catalyst, of the speed of strirring, and of the concentration of reactants. The reaction was carried out in the first order on the concentration of 1-chlorooctane and sodium cyanide, respectively. The over-all order was 2nd. The activation energies for the nucleophilic substitution reaction of 1-chlorooctane and 1-bromooctane under tetrabutylammonium hydrogen-sulfate were calculated as 2.05 and 10.08kcal/mol, respectively. The effect of various caltalysts was decreased in the order of tetrabutylammonium bromide, terabutylammonium, tetrabutylammonium hydrogensulfate, and tetrabutylammonium iodide. The reaction rate was dependent on the concentration of sodium-cyanide dissolved in the aqueous phase, and the good result was shown when the mol ratio between 1-chlorooctane and sodium cyanide was one per three.
P. spiculispoTum으로부터 생산되는 미생물계변활 성제인 S-acid의 전구체인 O-acid 생산에 영향을 주는 인자들을 검토하고 포도탕 유가배양을 통해 최 척생산조건을 조사하였다. 탄소원으로 oil 등은 별 영향을 마치지 않았고 포도당이 O-acid 생산에 적절 한 탄소원이였으며 무기질소원으로는 $NH_4NO_3$가 높은 O-acid 생산량을 보였다. 미량원소인 $CaCl_2, FeSO_4$등의 첨가에 의해 약 20% 정도의 O-acid가 증가되었으며 용존산소는 포화에 가깡게 유지한 경 우 생산량은 40% 증가되였다. 배양 중 포도당농도는 $50g/\ell$로 유지하는 것이 최적이었으며 Fermentor 에서 포도당의 유가식공급에 의한 경우, 회분식에 비하여 약 60% 의 O-acid 생산증가를 보였다.
In order to estimate the nutrient flux of the Seomjin River into the coastal waters of South Sea, and to understand the estuarine reactions during mixing between river water and seawater, we collected surface water along the salinity gradient in the Seomjin River estuary from Mar. 1999 to Apr. 2001. We found that nitrate and silicate were delivered by fluvial input, while phosphate was, supplied from disposed wastes in the Gwangyang Bay. Mean annual flux of dissolved inorganic nitrogen (DIN), phosphate and silicate into the Gwangyang Bay was estimated 10.9 molesㆍsec$^{-1}$(4,820 tonnesㆍyr$^{-1}$), 0.07 molesㆍsec$^{-1}$(68 tonnesㆍyr$^{-1}$), 13.3 molesㆍsec$^{-1}$(11,747 tonnesㆍy$^{-1}$), respectively. An evident removal of phosphate, silicate and ammonium at the mid-salinity zone during the dry season was attributed to the active uptake of phytoplankton, and consequently nutrient flux into the Gwangyang Bay was low. Whereas, during the flood season in summer, conservative or additional distribution of the nutrients was observed in the estuary. As a rsult nutrient flux into the Gwangyang Bay was maintained high. High concentrations of chlorophyll a and the active removal of nutrient during the dry season at the mid-salinity zone suggest that nutrient distribution in the Seomjin River estuary was mainly controlled by biological processes and nutrient fluxes into the Gwangyang Bay might be significantly modified of by the primary production.

Polyphenol oxidases (PPO) are copper-containing enzymes responsible for tissue browning in fruits and vegetables including potato, apple and pears. Although these enzymes have been studied for many years, their physiological roles in plants are not yet clear. Therefore, these enzymes need to be purified to characterize further from potato tubers. The classical methods used for the purification of PPO involve several steps. So in this study, we developed a one-step chromatography process for the potato tuber PPO purification. After removal of salts from dissolved ammonium sulfate precipitates of potato tuber extracts using Sephadex-G50 gel filtration, affinity chromatography was carried out on NHS-activated Sepharose 4B using p-aminobenzoic acid as a ligand. The purified enzyme was confirmed by silver staining and a zymogram. The optimum temperature and pH for the purified potato tuber PPO were $15^{\circ}C$ and pH 6.0, respectively. The results obtained in the present study will aid to evaluate PPO from various fruits and vegetables.
Sarcodon aspratus (Berk.) S. Ito에서 효소분획을 추출하여 casein 및 여러 식품 단백질기질을 조제하여 그것에 대한 분해작용을 실험하였다. Casein을 크게 분해하고 각 식품단백질기질도 분해하였으므로 protease의 존재를 알게 되었고 여러 기질에 대해 표준 pepsin과 비교할때 거의 동일한 역가를 나타내었으므로 우수한 protease가 함유되어 있음을 규명하였다. 아울러 능이가 육류체증 치료에 민간약으로 사용되는 근거를 제시하였다고 사료된다.
In an attempt to develop a non-aqueous liquid formulation of practically insoluble biphenyl dimethyl dicarboxylate (DDB), dissolution and permeation studies were performed. Various non-aqueous DDB solutions were formulated and filled into empty hard capsules. Dissolution rates of a new formulation were compared with those of commercially available DDB preparations using one and eight dose units. Dissolution rates after 2 hr of DDB tablets (DDB 25 mg), hard capsules (DDB 7.5 mg) and soft capsules (DDB 7.5 mg) on market and new formulation (DDB 7.5 mg) were 6.3, 15.0, 84.5 and 98.0%, respectively. Higher doses (8 units) resulted in a supersaturation within one hr of dissolution, and dissolved amounts were reduced markedly. Due to the saturation and precipitation, a directly proportional dose-dissolution relationship was not observed. The addition of copolyvidone and/or glycyrrhizic acid ammonium salt to DDB solution in polyethylene glycol 300 and 400 inhibited the formation of precipitates during dissolution and markedly enhanced the rabbit duodenal permeation of DDB. From the site-specific gastrointestinal permeation studies, it was found that permeation rates of DDB after mixing of non-aqueous DDB solutions with aqueous buffered solutions were faster in the order of $rectal\;<\;colonic\;{\risingdotseq}\;ileal\;{\risingdotseq}\;duodenal\;<\;jejunal\;<\;gastric$.
