Metallo-β-lactamase 생성 Pseudomonas aeruginosa의 시험관내 항균제 병합요법에 대한 연구 (In vitro Antimicrobial Combination Therapy in Metallo-β-lactamase Producing Pseudomonas aeruginosa)

  • Metallo-${\beta}$-lactamase (MBL) can hydrolyze all ${\beta}$-lactams except monobactams and frequently coexists with various antibiotic resistance genes such as aminoglycoside resistance, sulfonamide resistance gene, etc. Therefore, the effective antibiotics against infections by these bacteria are markedly limited or can't even be found. We tried to search in-vitro antimicrobial combinations with synergistic effects for a VIM-2 type MBL producing Pseudomonas aeruginosa, isolated from clinical specimen. On the selection of antibiotic combinations with synergistic effects, we performed a one disk synergy test, modified Pestel's method, in agar without aztreonam (AZT). The bacteriostatic synergistic effects of this tests were scored as $S_1$ (by susceptibility pattern in agar without antibiotics), $S_2$ (by the change of susceptibility in agar with or without antibiotics) and $S_3$ ($S_1$ + $S_2$) and was classified into weak (1 point), moderate (2 points) and strong (3 points) by $S_3$ score. Subsequently, we carried out the time-killing curve for the antibiotic combinations with the strong synergistic bacteriostatic effect. One VIM-2 type MBL producing P. aeruginosa confirmed by the PCR showed all resistance against all ${\beta}$-lactams except AZT, aminoglycoside and ciprofloxacin. In the one disk synergy test, this isolate showed a strong bacteriostatic synergistic effect for the antibiotic combination of AZT and piperacillin-tazobactam (PIP-TZP) or AZT and amikacin (AN). On the time-killing curve after six hours of incubation, the colony forming units (CFUs/mL) of this bacteria in the medium broth with both combination antibiotics were decreased to 1/18.7, 1/17.1 of the least CFUs of each single antibiotics. The triple antibiotic combination therapy including AZT, PIP-TZP and AN was shown to be significantly synergistic after 8 hrs of exposure. In a VIM-2 MBL producing P. aeruginosa with susceptibility for AZT, the triple antibiotic combination therapy including AZT, PIP-TZP and AN may be considered as an alternative antibiotics modality against the infection by some MBL type. But the antimicrobial combination therapy for many more MBL producing isolates is essential to know as soon as possible for the selection of effective treatment against the infection by this bacteria.

  • The sliding wear behavior of $ZrO_2-22wt\%MgO\;(MZ)\;and\;ZrO_2-8wt\%Y_2O_3\;(YZ)$ deposited on a casting aluminum alloy with bond layer (NiCrCoAlY) by plasma spray against an SiC ball was investigated under dry test conditions at room temperature. At all load conditions, the wear mechanisms of the MZ and the YZ coatings were almost the same. The wear mechanisms involved the forming of a smooth film by material transferred on the sliding surface and pullout. The wear rate of the MZ coating was less than that of the YZ coating. With an increase normal load the wear rate of the studied coatings increased. The SEM was used to examine the sliding surfaces and elucidate likely wear mechanisms. The EDX analysis of the worn surface indicated that material transfer was occurred from the SiC ball to the disk. It was suggested that the material transfer played an important role in the wear performance.

  • The hydroformability of aluminum alloy sheets at elevated temperatures have been investigated in this study. It is necessary to analyze the variations of the mechanical properties that depend on the forming temperature and the heat conduction during warm hydroforming. Therefore, in this study a coupled simulation of plastic deformation and temperature distribution in the warm hydroforming process is performed and compared with experimental data. The multi-purpose code DEFORM-2D can handle this type of calculations but it takes high computation time if contact heat transfer between die, tube and pressure medium occurs. Experiments were conducted by high temperature tribometer(pin-on-disk) allowing measuring the friction coefficients of the aluminum alloys at several temperatures and these results are applied to the coupled simulation by which the optimal process parameters such as internal pressure and preset temperature on hydroformability can be determined. The comparison of the FE analysis with the experimental results has shown that hydroformability given by bulge height, and temperature distribution of the tube specimen make a little difference with the FE results but the trend predicted by simulation agrees well with experiments.

Observational Studies of Masers in Star-forming Regions with KVN and KaVA

  • Methanol masers are divided into two classes, I and II. Class II methanol masers trace the disk-outflow systems of massive young stellar objects (YSOs), while class I methanol masers appear to trace the interaction regions of outflows with the ambient molecular gas. Class II masers have been extensively studied by single dishes, connected arrays, and VLBIs. Meanwhile, class I masers have been much less studied. They have not been detected by any VLBI facility. Thus they have been believed to have more extended structures than class II masers. We made fringe surveys of 44GHz class I methanol maser emission towards more than 150 massive YSOs with flux densities >10 Jy using the Korean VLBI Network (KVN), and detected fringes in ~10% of the sources. We performed follow-up imaging observations of the detected maser sources with KVN and KVN+VERA (KaVA). The observations aim to investigate the distribution and kinematics of 44GHz methanol maser features in each source at milli-arcsecond resolutions, and to understand what they trace. In this talk we will present the fringe survey and imaging results and our plans for further studies. Additionally, we will also introduce the preliminary results of single-dish polarization observations of water and class I methanol masers.

Abundant Methanol Ices toward a Massive Young Stellar Object in the Galactic Center

  • Methanol ($CH_3OH$) is a key species in the formation of complex organic molecules. We report the first detection of solid $CH_3OH$ in a line of sight toward the Galactic center (GC) region, based on L-band spectra taken with the Subaru telescope, aided by L'-band imaging data and moderate-resolution spectra from NASA/IRTF. It is found toward a background star, ~8000 AU in projected distance from a newly discovered massive young stellar object (YSO). This YSO also exhibits a strong $CO_2$ ice absorption band at ${\sim}15{\mu}m$ in Spitzer/IRS data, which has a prominent long-wavelength wing. It confirms that a high $CH_3OH$ abundance is responsible for the broad $15{\mu}m$ $CO_2$ ice absorption towards massive YSOs in the GC. Clearly, $CH_3OH$ formation in ices is efficient in the GC region, as it is in star-forming regions in the Galactic disk. We discuss implications of our result on the astrochemical processes in the hostile GC molecular clouds.

Development of a Rapid PCR Test for Identification of Streptococcus agalactiae in Milk Samples Collected on Filter Paper Disks

  • Streptococcus (Strep.) agalactiae is one of the major pathogens of bovine mastitis and is the main cause of subclinical infection. This study attempted to develop a rapid PCR diagnosis procedure using milk samples collected on filter paper disks. Chromatographic filter paper was employed as the preservation media and kept at room temperature for one to four weeks. The revival rate of Strep. agalactiae kept on dried filter paper disks was affected by the pretreatment preservation time. The revival test suggested that not all the bacteria in artificially contaminated milk samples on the filter paper disks could be recovered. After that, a PCR based on the 16-23S intergenic spacer region of Strep agalactiae was performed. The results distinguished the strep. agalactiae from major pathogens of bovine mastitis at a $2{\times}10^2$ colony forming units (CFU)/ml level, which showed similar sensitivity to the results from liquid milk samples. The results also showed that milk samples collected on filter paper disks could be kept at room temperature for one to four weeks with little negative effect on sensitivity and specificity. The field test showed that the diagnostic sensitivity and specificity was 96.15% and 98.60%, respectively. In conclusion, the protocol will provide a rapid and economic procedure for the detection of bovine mastitis.

Sustained Nuclear Star Formation and the Growth of a Nuclear Bulge

  • Hydrodynamic simulations of gas clouds in the central hundred parsecs region of the Milky Way that is modeled with a three-dimensional bar potential are presented. Our simulations consider realistic gas cooling and heating, star formation, and supernova feedback. A ring of dense gas clouds forms as a result of $X_1-X_2$ orbit transfer, and our potential model results in a ring radius of ~200 pc, which coincides with the extraordinary reservoir of dense molecular clouds in the inner bulge, the Central Molecular Zone (CMZ). The gas clouds accumulated in the CMZ can reach high enough densities to form stars, and with an appropriate choice of simulation parameters, we successfully reproduce the observed gas mass and the star formation rate (SFR) in the CMZ, ${\sim}2{\times}10^7\;M_{\odot}$ and ${\sim}0.1\;M_{\odot}/yr$. Star formation in our simulations takes place mostly in the outermost $X_2$ orbits, and the SFR per unit surface area outside the CMZ is much lower. These facts suggest that the inner Galactic bulge may harbor a mild version of the nuclear star-forming rings seen in some external disk galaxies. We also find that the stellar population resulting from sustained star formation in the CMZ would be enlogated perpendicularly to the main bar, and this "inner bar" can migrate the gas in the CMZ further down to the central parsecs region.

  • In order to investigate the cytotoxicity of composite resin in vitro, BALB / C mouse fibroblast were cultured in MEM in which silux, P-50, microrest, clearfil, amalgam and glass - ionomer, in shape of $2{\times}9mm$ circular disk. The experiments were- performed by cell count on 4 hours, 1, 3, 6 days and the composite resin groups, amalgam, glass - ionomer were compared. 1. On the sixth day, the cellular number of resin composite groups were remarkedly reduced, in contrast, the that of amalgam and glass - ionomer group continuously increased. 2. It was only on the 4 hours that the cellular number contained in amalgam were reduced, but increased thereafter, and the cellular number contained in glass - ionomer are greater than other groups. 3. In resin group, especially between self - curing resin and light - curing resin, there is no difference in cellular number statistically (p>0.05). 4. It was amalgam where the round cell without cellular process was found on the 4. hours and on the 6 th day the cell without cellular process was found numeroulsy in resin group whereas in amalgam and glass - ionomer, like control group was contained cell forming monolayer. These result suggested that the toxicity of the self - curing and light - curing resin greater than that of the amalgam and glass - ionomer.

Nature of the Wiggle Instability of Galactic Spiral Shocks

  • Gas in disk galaxies interacts nonlinearly with a underlying stellar spiral potential to form galactic spiral shocks. Numerical simulations typically show that these shocks are unstable to the wiggle instability, forming non-axisymmetric structures with high vorticity. While previous studies suggested that the wiggle instability may arise from the Kelvin-Helmholtz instability or orbit crowding of gas elements near the shock, its physical nature remains uncertain. It was even argued that the wiggle instability is of numerical origin, caused by the inability of a numerical code to resolve a shock that is inclined to numerical grids. In this work, we perform a normal-mode linear stability analysis of galactic spiral shocks as a boundary-value problem. We find that the wiggle instability originates physically from the potential vorticity generation at a distorted shock front. As the gas follows galaxy rotation, it periodically passes through multiple shocks, successively increasing its potential vorticity. This sets up a normal-mode that grows exponentially, with a growth rate comparable to the orbital angular frequency. We show that the results of our linear stability analysis are in good agreement with the those of local hydrodynamic simulations of the wiggle instability.

트윈롤 시스템을 이용한 천연가스 하이드레이트 펠릿의 연속성형 (Study on the Continuous Forming of Natural Gas Hydrate Pellet using Twin Roll System)

  • This study investigates compressive strength of ice pellet strip which is potential medium for Natural Gas Hydrate(NGH) extruded from die holes of Twin-roll Press for Continuous Pelletizing(TPCP). Recently, the prototype of TPCP is newly developed where ice powder is continuously fed and extruded into strip-type pellet between twin rolls. The system is specifically designed for future expansion towards mass-production of ice pellet strips or solid form of natural gas hydrate. It is shown that the compressive strength of pellet strip heavily depends on factors in extrusion process such as disk size, surface smoothness, ring size, taper shape, feeding mechanism, and rotational speed. Here, the mechanism of TPCP, along with compressive strength of pellets is discussed in terms of its feasibility for producing NGH pellets in the future.
