Air Pollution Risk Prediction System Utilizing Deep Learning Focused on Cardiovascular Disease

  • 이 논문은 대기오염의 심장병에 대한 위험도를 예측하기 위하여 Keras를 활용한 Deep Neural Network Model 시스템을 제안하였다. 연구 데이터로 서울열린데이터광장의 서울시 기간별 시간평균 대기환경 데이터 18,000개의 데이터 셋을 분석하여, 심장병 질병에 미치는 영향에 대한 정보를 얻을 수 있었다. 이 모델은 각각 8개의 노드를 가진 3개의 은닉층, Sigmoid, Binary_crossentropy, Adam과 Accuracy를 사용했을 때 88.92%의 높은 정확도를 얻을 수 있었다. 이 시스템은 각 지역별 대기오염에 따른 심장병 질병 위험도를 예측하여 유용한 질병 예방의 지표로 활용 가능하다고 사료되고, 대기오염과 미세먼지의 각 성분이 유해질환에 미치는 영향에 대한 데이터만 존재한다면 어떠한 호흡기 질환이든 위험도 예측 결과를 알 수 있다는 것에 의미가 있다. 이 시스템을 더욱 발전시킨다면, 마스크 및 공기정화제품 생산기업에게 유용한 정보를 제공하여 기업의 기술개발에 도움이 될 수 있다고 사료된다.

A novel HDAC6 inhibitor, CKD-504, is effective in treating preclinical models of huntington's disease

  • Huntington's disease (HD) is a neurodegenerative disorder, of which pathogenesis is caused by a polyglutamine expansion in the amino-terminus of huntingtin gene that resulted in the aggregation of mutant HTT proteins. HD is characterized by progressive motor dysfunction, cognitive impairment and neuropsychiatric disturbances. Histone deacetylase 6 (HDAC6), a microtubule-associated deacetylase, has been shown to induce transport- and release-defect phenotypes in HD models, whilst treatment with HDAC6 inhibitors ameliorates the phenotypic effects of HD by increasing the levels of α-tubulin acetylation, as well as decreasing the accumulation of mutant huntingtin (mHTT) aggregates, suggesting HDAC6 inhibitor as a HD therapeutics. In this study, we employed in vitro neural stem cell (NSC) model and in vivo YAC128 transgenic (TG) mouse model of HD to test the effect of a novel HDAC6 selective inhibitor, CKD-504, developed by Chong Kun Dang (CKD Pharmaceutical Corp., Korea). We found that treatment of CKD-504 increased tubulin acetylation, microtubule stabilization, axonal transport, and the decrease of mutant huntingtin protein in vitro. From in vivo study, we observed CKD-504 improved the pathology of Huntington's disease: alleviated behavioral deficits, increased axonal transport and number of neurons, restored synaptic function in corticostriatal (CS) circuit, reduced mHTT accumulation, inflammation and tau hyperphosphorylation in YAC128 TG mouse model. These novel results highlight CKD-504 as a potential therapeutic strategy in HD.

Model for end-stage liver disease-3.0 vs. model for end-stage liver disease-sodium: mortality prediction in Korea

  • Background/Aims: The model for end-stage liver disease (MELD) serves as an indicator for short-term mortality among patients diagnosed with liver cirrhosis (LC) and is used to prioritize patients for liver transplantation. In 2021, the updated version of MELD, MELD-3.0, was introduced to improve the accuracy of the mortality prediction of MELD. Therefore, this study aimed to compare the efficacy of MELD 3.0 and MELD-Na in predicting mortality among Korean patients with LC. Methods: A retrospective review was conducted using the medical records of patients diagnosed with LC who were admitted to Konkuk University Hospital From 2011 to 2021. The study calculated the predictive values of MELD-Na and MELD-3.0 for 3- and 6-months mortality using the area under the receiver operating curve (AUROC) and compared the results using the DeLong test. Results: Of the 3,034 patients enrolled in the study, 339 (11.2%) died within 3 months and 421 (14.4%) died within 6 months. The AUROCs values for predicting 3 months mortality were 0.846 for MELD-Na and 0.851 for MELD-3.0. The corresponding AUROC values for predicting 6 months mortality were 0.843 for MELD-Na and 0.848 for MELD-3.0. MELD-3.0 exhibited better discrimination ability than MELD-Na for both 3 (p = 0.03) and 6 months mortality (p = 0.01). Conclusions: Our study found a significant difference between the performance of MELD-3.0 and MELD-Na in Korean patients with LC.

  • Objective: This experiment was designed to investigate the effects of the KBHT hot water extract & ultra-fine powder on Alzheimer's Disease Model Induced by $\beta$A. Method: The effects of the KBHT hot water extract on expression of proinflammatory cytokine mRNA in BV2 microglial celll cell line treated by lipopolysacchaide(LPS). The effects of the KBHT hot water extract & ultra-fine powder on (1) the behavior (2) AChE in serum (3) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with $\beta$A were investigated. Results: 1. The KBHT hot water extract suppressed the expression of proinflammatory cytokine mRNA in BV2 microglial cell line treated with LPS. 2. The KBHT hot water extract & ultra-fine powder a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by $\beta$A in the Morris water maze experiment, which measured stop-through latency and distance movement-through latency 3. The KBHT hot water extract & ultra-fine powder suppressed the over-expression of AChE activity in the serum of the mice with Alzheimer's disease induced by $\alpha$A. 4. The KBHT hot water extract & ultra-fine powder suppressed the expression of TNF-$\alpha$, IL-l$\beta$ protein significantly in the microglial cell of mice with Alzheimer's disease induced by 1$\beta$A. 5. The KBHT ultra-fine powder reduced infarction area of hippocampus significantly in the mice with Alzheimer's disease induced by $\beta$A. Conclusions: These results suggest that the KBHT hot water extract & ultra-fine powder may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the PMCMT hot water extract & ultra-fine powder for Alzheimer's disease is suggested for future research.

Result : The CBD hot water extract suppressed the expression of $IL-1{\beta}$, $TNF-{\alpha}$ mRNA in BV2 microglia cell line treated with LPS. The CBD hot water extract significantly suppressed the production of $IL-1{\beta}$, $TNF-{\alpha}$ in BV2 microglial cell line treated with LPS. 비알콜성 지방간 초음파 영상에 GLCM과 인공신경망을 적용한 비알콜성 지방간 질환 분류 (Non-alcoholic Fatty Liver Disease Classification using Gray Level Co-Ocurrence Matrix and Artificial Neural Network on Non-alcoholic Fatty Liver Ultrasound Images)

  • 비알콜성 지방간은 심혈관계 질환, 당뇨병, 고혈압 및 신장질환의 발생에 있어 독립적인 위험인자에 해당하며, 최근에는 비알콜성 지방간에 대한 임상적 중요성이 증가하고 있다. 본 연구에서는 비알콜성 지방간 환자의 초음파영상에 대하여 질감분석 방법인 GLCM을 적용하여 특징값을 추출하고자 한다. 추출된 특징값들을 이용한 인공신경망 모델의 적용을 통하여 비알콜성 지방간의 지방침착 정도를 정상 간(normal), 경도 지방간(mild), 중등도 지방간(moderate), 중증 지방간(severe)으로 분류를 하고자 한다. GLCM알고리듬 적용 결과 Autocorrelation, Sum of squares, Sum average, Sum variance 파라미터 값들은 경도 지방간, 중등도 지방간을 거쳐 중증 지방간으로 갈수록 특징값의 평균값이 증가하는 경향성을 나타내었다. 인공신경망 모델의 입력은 비알콜성 지방간질환의 초음파영상에 GLCM 알고리듬을 적용하여 추출한 Autocorrelation, Sum of squares, Sum average, Sum variance의 4개의 파라미터들을 인공신경망 모델의 입력값으로 적용하였다. 비알콜성 지방간질환의 초음파영상에 GLCM 알고리듬을 적용하여 추출한 영상을 인공신경망에 적용하여 분류 정확도를 평가한 결과 92.5%의 높은 정확도를 나타내었다. 이러한 결과를 통하여 비알콜성 지방간 환자의 초음파 영상에 대한 질감 분석 GLCM 연구 시 본 연구의 결과를 기초자료로 제시를 하고자 한다.

지역사회 노인의 입원기간에 영향을 주는 개인요인의 분석 (Individual Determinants of Hospital Days m Community-dwelling Elders)

  • This study examines individual determinants of hospital days in community-dwelling elders by estimating a linear structural equation model based upon Andersen's behavioral model. Data were collected through a national survey of elders in Korea in 1994. The subjects for this secondary data analysis were 1687 non-institutionalized elders aged 60 years or older. Except for the effect of age and gender on family help. the predisposing components had direct effects on the enabling components. Of the effects of the enabling components. family help had a direct effect on self-evaluated health; economic status had a direct effect on chronic disease and self-evaluated health; and access had a direct effect on chronic disease. functional health status and self-evaluated health. Of the enabling components. residence and family help directly affected hospital days. Self-evaluated health revealed the greatest direct effect on hospital days followed by functional health. Overall. the effects of the predisposing components and the enabling components on hospital days were not prominent. Since the model explained only $4\%$ of the variance in hospital days. the magnitude of the effect of the need components cannot be judged in terms of equity of distribution of health services. In particular. the effect of family help in the model reveals the importance of family support in health management of elders. The implications of these analyses for improving the proposed model of hospital days were discussed.

공간 자료를 이용한 대기오염이 순환기계 건강에 미치는 영향 분석 (A Study on the effects of air pollution on circulatory health using spatial data)

  • Purpose: In this study, we examine the effects of circulatory diseases mortality in South Korea 2005-2013 using the air pollution index, Methods: We cluster the region of high risk mortality by SaTScan$^{TM}$9.3.1 and compare this result with the regional distribution of air pollution. We use the Geographically Weighted Regression (GWR) to consider the spatial heterogeneity of data collected by administrative district in order to estimate the model. As GWR is spatial analysis techniques utilizing the spatial information, regression model estimated for each region on the assumption that regression coefficients are different by region. Results: As a result of estimating model of the collected air pollution index, circulatory diseases mortality data combined with the spatial information, GWR was found to solve the problem of spatial autocorrelation and increase the fit of the model than OLS regression model. Conclusion: GWR is used to select the air pollution affecting the disease each year, the K-means cluster analysis discover the characteristics of the distribution of air pollution by region.

  • 본 논문에서는 최근 스마트농업의 중요한 이슈로 떠오르고 있는 스마트 축산분야의 가축 유행병 서비스 표준 모델을 제시한다. 네트워크를 이용하여 전 세계적으로 유행하는 가축 전염병 질병 리스크를 파악해서 서비스 유저들에게 관련 모델을 제공하여 실질적으로 가축 소유자들에게 경제적인 이득을 제공하고 궁극적으로 국가 농축산업 경제에도 도움 될 수 있을 것으로 판단된다. ICT와 접목하여 제시되는 가축전염병 서비스 표준모델과 가축 전염병 위기완화 표준모델 공유 방법은 향후 국내 및 국제 농축산업 분야에 표준에 적용하려고 지속적 연구가 진행될 예정이다.

흰쥐 흑질내 수산화도파민 주입으로 유도된 파킨슨병 모델에서 흑질과 선조체의 신경교세포 반응 (Neuroglial Reaction in the Substantia Nigra and Striatum of 6-Hydroxydopamine Induced Parkinson's Disease Rat Model)

  • Objectives : Parkinson's disease is a well-known neurodegenerative disease characterized by dopaminergic cell death in the substantia nigra. The reactive gliosis by activated astrocytes and microglias is no more regarded as a simple sequel of neuronal cell death. Microglial activation takes place in a stereotypic pattern with graded morphologic and functional(resting, activated and phagocytic) changes. In Parkinson's disease animal model, the degree of microglial activation along the nigro-striatal dopaminergic tract has not been studied intensively. The purpose of this study was to elucidate the characteristics of microglial reaction and to grade its degree of activation at substantia nigra and corpus striatum using 6-hydroxydopamine induced rat model of Parkinson's disease. Methods : Using Sprague-Dawley rat, parkinsonian model was made by 6-hydroxydopamine(OHDA) induced destruction of medial and lateral substantia nigra(SN). The rat was sacrificed 3-, 5-, 7-, 14- and 21-day-after operation. For control group, we injected saline with same manner and sacrificed 3-day after operation. With immunohistochemistry, we examined dopaminergic neuronal cells and microglial expression using tyrosine hydroxylase (TH) and OX-42 antibodies, respectively. Also we performed in situ hybridization for osteopontin, a possible marker of subset in activated microglia. Results : 1) In lesioned side of substantia nigra and corpus striatum, the TH immunoreactivity was markedly decreased in whole experimental groups. 2) Using optical densitometry, microglia induced immunoreactivity of OX-42 was counted at SN and corpus striatum. At SN, it was increased significantly on the lesioned side in control and all time-dependent experimental groups. At striatum, it was increased significantly in post lesion 3-day group only(p <0.05). Compared to control group, immunoreactivity of OX-42 on lesioned side was increased in groups, except post lesion 21-day group, at SN. Only post lesion 3-day group showed significance at striatum(p <0.05). Compared to SN region, immunoreactivity of OX-42 was much weaker in striatum. 3) Microscopically, the microglias showed typically different activation pattern. At SN, numerous phagocytic microglias were found at pars compacta and reticularis of lesion side. At striatum, no phagocytic form was found and the intensity of staining was much weaker. 4) At SN, the immunoreactivity of osteopontin showed definite laterality and it was markedly increased at pars compacta of lesion side with relatively short duration time. At striatum, however, it was not detected by in situ hybridization technique. Conclusion : The nigral 6-OHDA induced rat model of Parkinson's disease revealed several characteristic patterns of microglial reaction. At SN, microglias was activated shortly after direct neuronal damage and maintained for about three weeks. In contrast, despite of sufficient dopaminergic insufficiency at striatum, activation of microglias was trivial, and distinguished 3 day later. Antegrade slow neuronal degeneration is major pathophysiology in striatal dopaminergic deficiency. So, the acuteness of neuronal damage and consequential degree of neuronal degeneration may be important factor for microglial activation in neurodegenerative diseases such as Parkinson's disease. Additionally, osteopontin may be a possible marker for several subsets of activated microglia, possibly the phagocytic form.
