누에품종별 혈당강하물질 축적양상 구명 (Accumulating Pattern of ${\alpha}-glycosidase$ Inhibitor in Various Silkworm Varities)

  • 누에장려품종 중 혈당강하제 개발에 적합한 품종 결정을 위하여 DNJ 함량을 정량한 결과, 봄에 사육한 금옥잠이 5.45 mg/gDW으로 DNJ 함량이 가장 높았다. 또한 누에 유전자원 원종 66품종의 DNJ 함량을 결정한 결과, 품종간 DNJ 함량 차이가 매우 큰 것으로 밝혀져, 혈당 강하제용 누에 육종시 기초자료가 될 수 있으며, 또한 DNJ 함량이 낮은 품종은 DNJ 축적 기작 연구에 많은 도움이 될 것으로 사료된다.

Xylanase를 생산하는 Bacillus sp. AMX-4 균주의 분리와 효소 생산성 (Isolation and Enzyme Production of a Xylanase-producing Strain, Bacillus sp. AMX-4.)

  • 토양으로 부터 xylan 분해능이 우수한 Bacillus sp. AMX-4를 분리하고 동정하였는데 분리균의 화학조성은 B. subtilis와 유사성을 보였다. 분리균이 생산하는 xylanase는 50℃와 pH 6에서 최대활성을 보였다. 배지의 부가 탄소원을 첨가하여 Bacillus sp. AMX-4을 배양한 결과 xylose를 첨가한 배지에서 xylanase 생산성이 가장 증가하는 것으로 나타났으며, xylose를 1.5%(w/v)가 되도록 첨가하였을 때 배양상등액의 xylanase 활성이 29.2 U/ml로 약 16배 정도 생산성이 증가하였다. Xylose를 첨가한 배지와 첨가하지 않은 배지에서 Bacillus sp. AMX-4를 배양하여 균의 성장과 효소 생산성을 비교한 결과 xylose를 첨가한 배지에서 효소 생산뿐만 아니라 분리균의 최대 성장정도도 증가한 것으로 보아 xylose는 균의 성장과 xylanase 생합성을 동시에 유도하는 것으로 판단된다. 또한 xylanase는 균의 성장과 연계되어 생산되는 것으로 확인되었다.

  • The metabolite profile of meju during fermentation was analyzed using mass spectrometry techniques, including GC-MS and LC-MS, and the bacterial diversity was characterized. The relative proportions of bacterial strains indicated that lactic acid bacteria, such as Enterococcus faecium and Leuconostoc lactis, were the dominant species. In partial least-squares discriminate analysis (PLS-DA), the componential changes, which depended on fermentation, proceeded gradually in both the GC-MS and LC-MS data sets. During fermentation, lactic acid, amino acids, monosaccharides, sugar alcohols, and isoflavonoid aglycones (daidzein and genistein) increased, whereas citric acid, glucosides, and disaccharides decreased. MS-based metabolite profiling and bacterial diversity characterization of meju demonstrated the changes in metabolites according to the fermentation period and provided a better understanding of the correlation between metabolites and bacterial diversity.

Effects of Varying Nutritional and Cultural Conditions on Growth of the Ectomycorrhizal Fungus Pisolithus tinctorius SMF

  • The culture conditions and nutritional requirements for enhanced mycelial growth of the ectomycorrhizal fungus P. tinctorius SMF were determined in flask scale experiments. Optimum culture conditions for growth of P. tinctorius SMF in a further modified Melin-Norkrans broth were as follows; temperature 25~$27^{\circ}C$, agitation 120 rpm, and pH 4.0. P. tinctorius SMF utilized various carbon sources including monosaccharides, disaccharides, and polysaccharides. D-Glucose and mannitol were respectively the first and second most suitable carbon sources for mycelial growth. With D-Glucose as the principal carbon source, supplementation of modified Melin-Norkrans (MMN) broth with Lysine (800 mg/l), Glutamic Acid (500 mg/l), or Proline (50 mg/l) enhanced mycelial yields 63%, 34%, and 22% respectively as compared to growth in medium lacking amino acids. ThiaminㆍHCl+biotin+pyridoxine supplementation also enhanced growth. As compared to mycelial growth in the MMN medium, growth of P. tinctorius SMF was enhanced 120% in MMN broth when the carbon/nitrogen ratio was 25/1 in citrate buffer at pH 4.5, and growth was 50% greater in MMN broth of carbon/nitrogen ratio with a 10/1~20/1 without using the buffer. Standard conditions established for growth of P. tinctorius SMF in MMN broth were 25~$27^{\circ}C$, agitation 120 rpm, buffered to pH 4.0 with citrate, in MMN medium containing 10 g/l D-glucose supplemented with 800 mg/l lysine. In this medium the carbon/nitrogen ratio was 20/1~25/1, and the maximal mycelial yield ($Y_{x/s}$ ) was 0.472 (4.72 mg/ml) after 7 days of incubation, as compared to 0.214 (2.14 mg/ml), when the fungus was grown in standard MMN broth.

Effects of a low-FODMAP enteral formula on diarrhea on patients in the intensive care unit

  • BACKGROUND/OBJECTIVES: A dietary restriction on the intake of fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAPs) has been reported to be effective in the treatment of gastrointestinal (GI) tract complications. Enteral nutrition (EN) is widely used for patients who cannot obtain their nutritional requirements orally, but many studies have reported EN complications, especially diarrhea, in up to 50% of patients. SUBJECTS/METHODS: We performed a single-center, non-randomized, controlled trial to determine the effects of a low-FODMAP enteral formula on GI complications in patients in intensive care units (ICUs). Patients in the ICU who needed EN (n = 66) were alternately assigned to the low-FODMAP group (n = 33) or the high-FODMAP group (n = 33). RESULTS: Anthropometric and biochemical parameters were measured, and stool assessment was performed using King's Stool Chart. We excluded patients who received laxatives, GI motility agents, proton pump inhibitors, antifungal agents, and antibiotics other than β-lactams. There were no differences in GI symptoms during 7 days of intervention, including bowel sound, abdominal distension, and vomiting between the 2 groups. However, diarrhea was more frequent in the high-FODMAP group (7/33 patients) than the low-FODMAP group (1/33 patients) (P = 0.044). CONCLUSIONS: Our results suggest that a low-FODMAP enteral formula may be a practical therapeutic approach for patients who exhibit enteral formula complications. Our study warrants further randomized clinical trials and multicenter trials.

한국인의 당류 섭취현황과 만성질환에 미치는 영향 (intake of dietary sugar and its influence on chronic disease in the korean population)

  • As recently many studies on the relationship between excessive sugar intake and chronic disease have been reported across the world, more attention to sugar intake has been paid. Sugars, called simple sugars, is a kind of carbohydrate and sum of monosaccharides and disaccharides. Sugars that are particularly related to health are added sugars or free sugars, which are added to food by processing or cooking. It is generally not to give satiety and increases energy density without other nutrients. Especially, the major food source of added sugar is sugar-sweetened beverages(SSBs), from which sugars are closely linked with health outcome. The total sugar intake in Korea is 61.4 g per day and it is almost half of those in the western countries such as the U.S. or Australia and it is within the recommendation level by the Dietary Reference Intakes for Koreans. However, when it is classified by age groups or food sources, sugar intake in adolescents and young adults are high and the sugar from processed foods is also high that would lead to high intake levels of added sugar. Particularly, the frequency of SSBs, the major food source of added sugar, is the highest among 20's. While the studies of excessive sugar intake with chronic diseases including obesity, type 2 diabetes, metabolic disease, cardiovascular disease reported in the Western populations, those in the Korean populations have just started. Given our dietary practice is very different from those in the western population, more prospective studies would be necessary to evaluate sugar intake by type or food source exactly and examine its relationship with chronic diseases in the Korean population.

Effect of web-based personalized nutrition management on gut microbiota in Korean patients with irritable bowel syndrome aged between 20 and 30 years

  • Purpose: Dietary habits are strongly related to the symptoms of people with irritable bowel syndrome (IBS). Therefore, personalized nutrition management can help reduce symptoms and improve the quality of life of people with IBS. This study assessed the effectiveness of a personalized web-based nutrition management based on the types of food that trigger IBS symptoms. Methods: Sixty Korean adults with IBS according to Rome IV criteria in their 20s and 30s were enrolled in this study. The data from the final 49 patients who completed a three-month personalized nutrition intervention were analyzed. The general information, anthropometry, dietary intake survey, and gut microbiota were examined pre and post-intervention. The gut microbiota analysis included the relative abundance and the Shannon index. The food intake was recorded for two days for personalized nutrition education, followed by three months of personalized nutrition intervention. Statistical analysis was performed using the Wilcoxon signed-rank test in SPSS 26.0, with the significance set to p < 0.05. Results: The relative abundance of the gut microbiota changed after personalized nutrition management, with a significant decrease in the presence of Veillonella (p = 0.048). Furthermore, when the gut microbiota was analyzed according to the type of food that triggers symptoms, the diversity was increased significantly in the high fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAPs) type (p = 0.031) and FODMAPs-containing gluten-type personalized nutrition intervention types (p < 0.001). Conclusions: Gut microbial diversity and gut microbiota distribution changed after using web-based personalized nutrition management. Hence, personalized nutrition management that considers trigger foods may improve IBS symptoms.

상업용 메디컬푸드 및 탄수화물 급원의 발효성 당류 함량에 관한 연구 (Fermentable Sugar Contents of Commercial Medical Foods and Carbohydrate Ingredients)

  • 본 연구에서는 최근 연구 결과들에 기초하여 발효성 당류인 FODMAP(fermentable oligosaccharides, disaccharides, monosaccharides, and polyols)이 설사와 높은 상관성을 가지고 있을 것이라 판단하였다. 따라서 상업용 메디컬푸드와 탄수화물 급원으로부터 FODMAP 함량을 측정하였다. 상업용 메디컬푸드의 단당류(fructose)와 이당류(lactose)의 함량 범위는 각각 ND~0.158, ND~0.304 g/200 mL였다. 삼당류(raffinose)와 사당류(stachyose)의 함량 범위는 각각 0.051~0.738, ND~0.579 g/200 mL였다. 또한 프럭토올리고당의 함량 범위는 1-kestose ND~0.413 g/200 mL, nystose ND~1.239 g/200 mL, 1-fructofuranosylnystose 0.205~0.458 g/200 mL였다. 탄수화물 급원 중 단당류(fructose)는 치커리 식이섬유($18.877{\pm}4.320g/kg$)에서만 검출되었으며, 이당류(lactose)는 모든 급원에서 검출되지 않았다. 삼당류(raffinose)의 함량은 치커리 식이섬유 $61.523{\pm}3.014g/kg$, 대두 식이섬유 $3.273{\pm}0.499g/kg$, 난소화성 말토덱스트린 $5.430{\pm}0.671g/kg$, 말토덱스트린(DE10~15, DE15~20)에서는 검출되지 않았다. 사당류(stachyose)의 함량은 치커리 식이섬유 $78.817{\pm}5.483g/kg$, 대두 식이섬유 $5.547{\pm}0.822g/kg$, 난소화성 말토덱스트린 $13.180{\pm}1.165g/kg$, 말토덱스트린(DE10~15) $16.440{\pm}0.370g/kg$, 말토덱스트린(DE15~20) $22.553{\pm}1.491g/kg$이었다. 프럭토올리고당(1-kestose, nystose, 1-fructofuranosylnystose)의 함량은 치커리 식이섬유 $29.369{\pm}2.553g/kg$, $39.430{\pm}6.740g/kg$, $50.407{\pm}2.642g/kg$, 대두 식이섬유 ND, ND, $18.647{\pm}0.397g/kg$, 난소화성 말토덱스트린 ND, $16.667{\pm}1.719g/kg$, $17.107{\pm}1.814g/kg$, 말토덱스트린 (DE10~15) ND, ND, $16.533{\pm}2.083g/kg$, 말토덱스트린 (DE15~20) ND, ND, $27.490{\pm}1.783g/kg$이었다. FODMAP은 탄수화물의 일종으로 메디컬푸드 제품의 탄수화물과 FODMAP 함량 간에 양의 상관관계를 가질 것으로 예상하였으나 r=0.55로 밀접한 상관성을 나타내지는 않았다. 국내외 메디컬푸드는 모두 유사한 원재료와 영양설계로 구성되어 있으나 사용되는 원재료의 종류에 따라 유래되는 FODMAP 함량이 다르기 때문에 제품 간의 함량 차이가 있는 것으로 생각된다. 본 연구에서 측정된 상업용 메디컬푸드 제품 및 탄수화물 급원의 FODMAP 함량 분석은 상대적으로 부족한 FODMAP 정량 분석 연구에 활용될 것으로 생각되며, 나아가 경관급식의 주요 부적응증인 설사를 최소화하는 메디컬푸드의 제품개발 및 원재료 선정에 기초 자료로 활용될 것이다.

반연속식(半連續式) 배양(培養)에 의(依)한 Rhizobium meliloti M14의 균체생산(菌體生産) (Production of Rhizobium meliloti M14 Inoculum by Semi-continuous Cultivation)

  • 전보(前報)에서 분리(分離)한 Rhizobium meliloti M14를 공시(供試)하여 그 종균생산(種菌生産)을 위한 기초적(基礎的) 연구(硏究)로서, 탄소원(炭素源) 및 질소원(窒素源), chemostat의 반련속식(半連續式) 운용(運用)에 의한 액체배양(液體培養)을 시도(試圖)하여 아래와 같은 결과(結果)를 얻었다. 1. 탄소원(炭素源) 비교(比較)한 단당류(單糖類), 이당류(二糖類) 및 당(糖)알콜류(類) 가운데 당(糖)알콜류(類)에서 가장 생육(生育)이 높았고 단당류중(單糖類中) 오탄당(五炭糖)에서의 생육(生育)은 육탄당(六炭糖)보다 저조(低調)하였다. 2. 천연(天然) 탄소원중(炭素源中) malt extract 및 molasses에서의 생육(生育)은 mannitol에서의 생육수준(生育水準) 이상(以上)이었다. 3. 질소원(窒素源)으로서 yeast water에 대(對)한 편향성(偏向性)이 인정(認定)되었으며 그 10%에서의 생육수준(生育水準)은 yeast extract 0.24%와 상응(相應)하였다. 4. Chemostat (MicroFerm 28L, New Brunswick model CMF-128S) 의 단식배양(單式培養)에 의(依)하여 36시간(時間) 배양(培養)으로 생균수(生菌數) $5{\sim}7{\times}10^9cells/ml$ 수준(水推)의 stationary phase에 도달(到達)하였고, 반련속식(半連續式) 운용(運用)으로 매(每) 24시간(時間)마다 동일수준(同一水準)의 생균(生菌)을 가진 배양액(培養液)을 그 운용량(運用量)에 대(對)하여 5분(分)의 4를 회수(回收)할 수 있었다.

탈지미강의 아임계수 가수분해 생성물을 배지로 이용한 Saccharomyces cerevisiae의 배양 (Cultivation of Saccharomyces cerevisiae using Defatted Rice Bran Hydrolyzed in Near-critical Water as a Culture Medium)

  • 아임계수를 이용하여 탈지미강의 가수분해를 실시하고, 결과로 얻어진 가수분해물이 배지로써 이용할 수 있는지 검토하기 위해 Saccharomyces cerevisiae의 배양을 시도해보았다. 아임계수 가수분해 반응은 회분식 반응을 통해 이루어졌으며, 생성물에 포함된 전당, 이당류, 단당류의 함량, 총유기탄소 (TOC), 총질소 (TN), pH를 측정하였다. Saccharomyces cerevisiae의 성장 속도는 탁도 측정을 통해 확인하였다. 전당 수율, TOC, TN은 $240^{\circ}C$까지 온도에 따라 증가하다가 그 이상의 온도에서는 감소하였다. $240^{\circ}C$ 이상에서의 수율 감소는 유기산 생성에 기인한 것이고 이는 pH 변화를 통해 확인할 수 있었다. 포도당의 수율은 $200^{\circ}C$에서 최대였고 그 이상의 온도에서는 빠르게 감소하였다. 탈지미강 가수분해물에서 배양된 Saccharomyces cerevisiae의 성장 속도는 일부 조건에서 상용 배지와 근접한 수준을 보였고, 성장 속도는 포도당의 함량과 상관 관계가 있었다.
