Effect of Dietary Perilla Seed Oil on Lipid Metabolism in Rats (들깨유가 흰쥐의 체내 지질대사에 미치는 영향에 관한 연구)

  • The effects of various dietary fats on plasma lipids. liver lipids, and Plasma Peroxide levels were studied in rats fed for 6 wk with diets containing 15 wt% fat, as sesame oil. raw perilla seed oil. roasted perilla seed oil, heated perilla seed oil. mackerel oil or beef tallow. TBA values of these lipids during 4 wk storage, and linolenic acid contents of three kinds of perilla seed oil were also measured. Linolenic acid contents of raw perilla seed oil. roasted perilla seed oil and heated perilla seed oil were 62.3%, 61.6% and 53.1% respectively. Raw perilla seed oil showed the lowest rate of lipid peroxidation after 4 wk storage at 4$^{\circ}C$, and mackerel oil showed the highest peroxidation rate. The plasma cholesterol levels of rats consuming diets in which the carbohydrate was rice were not affected by n-3 PUFA. Rather, the degree of peroxidation seems to have a direct effect on cholesterol levels as shown by the hypocholesterolemic effect of raw perilla seed oil and beer tallow. However. the HDL-cholesterol level was greater in rats fed either roasted perilla seed oil or mackerel oil. Rats fed roasted perilla seed oil and raw perilla seed oil had lower levels of plasma triglycerides than rats fed beef tallow. In rats fed roasted perilla seed oil, the total lipid and cholesterol contents of liver were significantly lower than in those fed the other kinds of perilla seed oil. The plasma lipid peroxide levels were lower in rats fed either roasted perilla seed oil or beef tallow.

Improvement of a Rice Seed Pelleting Machine for Direct Seeding in Rice Cultivation(I) - Construction and its performance - (직파용 벼 펠렛종자 제조장치 개선 연구(I) - 장치 제작과 성능분석 -)

  • To enhance the performance of a rice seed pelleting machine and the quality of rice-seed pellets made, improvement of the rice seed pelleting machine developed previously(Park, 2002) was tried and its performance was evaluated. As compared with the previous pelleting machine, a feeding mechanism of pellet materials to the forming rolls was changed from screw conveyor to hydraulic cylinder for proper feeding, rings were installed among rows of semi-spherical forming grooves on the forming rolls for reducing pellet materials loss and seeds damage, and discharging air nozzles were added for complete discharging of the pellets made. Through performance tests, capacity, pelleting ratio, and seed loss ratio of the pelleting machine were investigated at the mixing ratios of soil to rice seed of 6 : 1, 7 : 1, and 8 : 1, and rotating speeds of the forming rolls of 7 rpm, 10 rpm, and 13 rpm. As results of performance evaluation, pelleting ratios were in the range of 77 ∼ 89 %, and maximum pelleting ratio increased by 18 % in comparison with that of the previous machine. Maximum capacity was about 110 kg/h(about 63,000 pellets/h), which was increased by 70 % in comparison with that of the previous machine. But, ratios of seed loss were in the range of 24 - 49 %, which were not improved.

  • Relay intercropping system of direct seeded rice and winter cereal is labor saving cultivation and has high land use efficiency in Japan. In this system, rice seeds are direct seeded into inter-row space of winter cereals (wheat or barley) in March or April. If the direct seeding of rice is conducted before stem elongation phase or using suitable seeder, these are little effect to yield of winter cereals. Though the seeds are generally thiuram treated, it's a matter that seedling establishment rate (SER) of direct seeded rice is low and unstable. The cause of low SER has not been revealed. In present study, with the aim to reveal causes of low SER, we conducted experiments and investigated the SER, and analyzed some factors that might affect SER. Experiment1: In 2015, 2016, we buried rice seeds underground, and investigated the transition of the seed survival rates (SSR). Seeds were thiuram-treated or non-treated. In 2 periods, SSR of thiuram-treated seeds were significantly higher than non-treated seeds. In 2016, thiuram-treated seeds were high in SSR (almost 75%) at April 30th, but low SER (10~27%) after harvest of winter cereals. Therefore, almost all of seed death might have been happen after germination. Analysis 1: We investigated the SER and cultivation conditions in Ibaraki pref. for several years. Meteorological factors were referred from the nearest point of AMeDAS. From mean temperature (MT) among 5days after and before the day, we divided the period of seeding ~June 20 to phase1~4. We defined each phase as below; Phase1: $MT{\leq}10^{\circ}C$, Phase2: $10^{\circ}C<MT{\leq}15^{\circ}C$, Phase3:$15^{\circ}C<MT{\leq}20^{\circ}C$, Phase4: $MT>20^{\circ}C$. We analyzed the correlation of SER and meteorological factors by each phase. Total number of days in phase 1~4 was significantly negative correlated with SER. In phase1, total rain fall and number of soil wetting days were significantly negative correlated with SER. In phase2~4, only MT was significantly positive correlated with SER. This result suggested that rainfalls in phase1 declined seed vigor to emergence from soil surface, by repeated water absorption and re-dry. From these present studies, it was suggested that one of factors of low SER of direct seeded rice in relay intercropping system is changing of water condition by rainfalls in phase1 ($MT{\leq}10^{\circ}C$). To improve SER, it's necessary to consider something seed treatments such as prevent water absorption during phase1. However, 58~60% of seeds seemed to die during May. It's suggested that, if seeds are thiuram treated, almost all of seeds can germinate underground, but the seed vigor to emergence from soil surface are insufficient. Further studies are needed to reveal the rest causes that is happening during emergence from soil surface.

  • The study was performed to evaluate rice seed germination, seedling emergence and seedling establishment under different coating materials such as iron powder, silicate powder and silicate coverage after direct seeding. There were differences among coating materials as follows; 1. In seedling establishment there was the highest in untreated control> silicate-coated seeds and silicate coverage>iron-coated seeds. In case of untreated control this result due to laboratory experiment unlike field conditions where has been constraints in bird damages, seed dry under strong sunlight and buoyance after rainy and/or irrigation. 2. Thus, there was the highest in untreated control>silicate-coated seeds>silicate coverage>iron-coated seeds, respectively. 3. Total fresh weight(shoots and roots) of the seedling was also highest in untreated control>silicate-coated seeds and silicate coverage> iron-coated seeds.

Seedling Stand Influenced by Water Management after Seeding and Seed Soaking with Plant Growth Regulators in Direct Wet Seeding Rice

  • Unstable seedling stand establishment of wet direct seeding culture of rice is one of the major elements preventing the extension of its culture area. In order to develop methods of seedling stand improvement in direct seeded rice on flooded surfaces, three field experiments were conducted on silty loam soil using a cultivar 'Donjinbyeo' for three years, mainly focusing on water management after seeding and seed soaking with plant growth regulators (PGRs). Under the condition of shallow flooding after seeding, seedling stand rate increased and floating seedling rate decreased in both early and normal season seeding compared to deep flooding. With earlier draining time after seeding, there was a tendency towards preferential growth of the seminal root, increase of seedling stand and decrease of the floating seedling rate. Therefore the highest seedling numbers per unit area and the lowest floating seedling numbers were found upon drainage at 1 day after seeding (DAS), while a contrary tendency was shown upon conventional drainage at 7 DAS. Seed soaking with PGRs such as Metalaxyl or mixing of Metalaxyl with gibberellic acid (GA$_3$) significantly increased the seedling stand. In addition the effects of PGR treatment on seedling stand and the early growth of plants were greater under flooded conditions than under drained conditions after seeding, although draining of water after seeding improved the seedling establishment rate more when compared with the PGR treatment. These results suggest that draining management after seeding or maintaining of shallow flooding for a week is the most effective method to improve the seedling stand rate in wet direct seeding.

Rice Plant Growth Characteristics According to Different Seed Germination Length and Damage (볍씨 최아 및 최아 손상정도에 따른 초기생육변화 분석)

  • We checked the emergence rate and early growth characteristics of young rice plants according to the germination length and seed damage ratio in direct seeding. The emergence rate was reduced according to the longer germination length and higher seed damage ratio. The emergence rate was further reduced under high-temperature conditions. The leaf number did not show a significant difference according to germination length and seed damage ratio. However, shoot and root dry weights were reduced according to the germination length and seed damage ratio. The root dry weight was highly correlated with the shoot dry weight. Based on these results, we concluded that a longer germination length and seed damage could induce the loss of nutrients and affect poor root growth, consequently inducing poor emergence and growth in the early growth stage of rice seeds. Therefore, in rice direct seeding, seed conditions of less than 1 mm of germination length should be checked for a sufficiently high standing rate and adequate growth characteristics after seeding.

Seedling Emergence and Initial Growth between Water and Dry Seeding in Forage Millet (사료용 피의 건답과 무논 파종 시 입모 및 초기생육특성)

  • This research was carried out to determine the characteristics of germination in relation to temperature, and seedling emergence and growth affected by soil moisture in forage millet. The seeds of forage millet could germinate at more than $15^{\circ}C$ at the rate of 0, 90.5, 97.3 and 96.8% respectively 10 days after seeding at 10, 15, 20 and $25^{\circ}C$. The result showed that the temperature needs to be $15^{\circ}C$ or more for the seeds to germinate. The effects of seeding depth on the growth characteristics and emergence of millet seed were investigated in waters and direct seeding methods by thoroughly mixing the seed and soil. In water treatment, the emergence rates were 3.7% at $15^{\circ}C$, 7.6% at $20^{\circ}C$, 6.3% at $25^{\circ}C$, while direct seeding treatment showed the emergence rates of 65.6, 75 and 71% at 15, 20 and $25^{\circ}C$, respectively. At 15, 20, and $25^{\circ}C$, seeds in water seeding could germinate at 0.5, 1.8, 1.5 cm of soil depth, while in direct seeding, they could germinate at the depth of 9.5, 10.0, 9.9 cm. When the initial growth characteristics of the seeds were investigated, there was positive correlation between seeding depth and mesocotyl length in both water and direct seed methods. Growth and development in terms of leaf age, root length, coleoptiles and mesocotyl length, and plant height were found maximum at 0.5 cm soil depth for water seeding, and 3-5 cm soil depth for direct seeding. Results showed that there were close relationship among temperature, soil depth and moisture acquiring methods, and the combination of these factors greatly affected the initial growth characteristics and development of millet seeds. It can be concluded that, to get good seedling stand and germination of millet seed, millet should grown in field condition at the depth of 0.5 cm or less for water seeding, and 3-5 cm for direct seeding method.

Physical Seed Treatment Techniques for Germination Enrichment and Seed Sterilization (발아증진 및 소독을 위한 물리적 방법을 이용한 종자처리 기술)

  • Since seeds can be directly used as food resources as well as for crop cultivation or preservation of genetic resources, it is essential to develop high-quality seed processing technology to increase agricultural productivity. Seed treatment means processing technologies of seeds through physical or chemical treatment processes from after harvesting seeds to before sowing of seeds to improve germination and growth rate, durability, and immunity, etc. Since chemical seed treatment technology using pesticides or plant growth regulators has problems of environmental pollution and human toxicity, it is desired to develop an alternative technology. As a physical seed treatment method, various technologies such as ionizing radiation, plasma, microwave, and magnetic field are being developed, and some of them are being used practically. In this paper, I will summarize the mechanism of seed priming and disinfection, and the advantages and disadvantages of application, focusing on these physical seed treatment methods. Low dose or moderate intensity ionizing radiation, microwave, low-temperature plasma, and magnetic field treatments often promoted seed germination and seedling growth. However, effective removal of direct seed pathogens at these treatment intensities appears to be difficult. And it has been shown that relatively high-dose electron beam treatment using low-energy electron beams kills microorganisms on the seed surface and hull layer while not damaging the inner tissue of the seed, and is also effectively used for seed treatment on a commercial scale. In order to put the physical seed treatment technology to practical use in Korea, it is necessary to develop an economical scale treatment device along with the development of individual treatment technology to each crop.

Conservation of Swertia chirata through direct shoot multiplication from leaf explants

  • Swertia chirata is an endangered gentian species that prefers to grow at higher altitudes. This ethnomedicinal herb is known primarily for its bitter taste caused by the presence of important phytochemicals that are directly associated with human health benefits. Due to a continuous loss of habitat and inherent problems of seed viability and seed germination, alternative strategies for propagation and conservation are urgently required to prevent the possible extinction of this species. We have formulated a reproducible protocol for the rapid propagation and conservation of this plant using leaves taken from in vitro shoot cultures. Direct induction of more than seven shoot buds per explant was achieved for the first time when the explants were placed on MS medium supplemented with $2.22{\mu}M$ N-6-benzyladenine, $11.6{\mu}M$ kinetin, and $0.5{\mu}M$ ${\alpha}-naphthalene$ acetic acid. Direct organogenesis was noted exclusively from the adaxial surface of the basal segments of leaves. Leaves closer to the apical meristem were more responsive than those farther away from the meristem. Plants raised through direct organogenesis were evaluated for their clonal fidelity by chromosomal analysis and DNA fingerprinting. Complete plants were successfully transferred to the field condition and produced viable seeds. Given the enormous potential of this age-old medicinal plant in terms of potential health-benefitting drugs, this protocol can be used for commercial propagation purposes and to initiate future genetic improvement studies.

Performance of Heritabilities, Genetic Correlations and Path Coefficients of Some Agronomic Traits at Different Cultural Environment in Sesame

  • This study was conducted to analyze the responses of some agronomic traits to the different cultural environments and relations among the agronomic traits for selecting sesame varieties with higher cultural stabilities. The indexes for stability parameters measured were coefficient of variability, heritabilities, genetic correlations and path coefficients of agronomic traits according to locations and years in Korea. The heritabilities of agronomic traits showed different by locations and years. Number of seeds per capsule and 1000 seeds weight showed higher heritabilities, but stem length and seed weight per plant showed relatively lower heritabilities. Average heritabilities of some agronomic traits in 1998 were comparatively higher than those of 1999. Of six areas, Jinju area showed biggest coefficient of yield variability in 1998-1999. Iksan and Taegu areas showed higher heritabilities in 1998, but Iksan and Jinju areas showed lower heritabilities in 1999. Genetic correlations were slightly higher than corresponding phenotypic correlations. Stem length showed positive genetic correlation with the number of capsules per plant, and seed weight per plant and the number of capsule per plant showed positive genetic correlation with seed weight per plant. On the analysis of path coefficients, stem length and number of capsules effected highly on grain yield. Great regional variations were observed on the effects of agronomic traits on grain yield. Higher direct effects of stem length on grain yield were observed at Suwon, Chungwon, Taegu, Jinju and Naju areas, but in Iksan area was observed higher direct effect of the number of capsules per plant on grain yield in 1998. In 1999, higher direct effect of stem length on grain yield was observed at Chungwon and Suwon areas. Iksan and Taegu areas were also observed higher direct effect of the number of capsule per plant on grain yield.
