Effect of Application Time and Rate of Diquat on Preharvest Field Drying of Rice (작물건조제 Diquat의 벼 수확전처리 건조효과 및 이용성)

  • Most of the currently available rice threshers in Korea are of head-feeding type originally developed for threshing paddy at grain moisture content of 15 to 17% on wet basis. And prevailing rice harvesting system is swathing or cutting and bundling upright rice crop at a maturity stage of grain moisture content of ca. 28 to 20%, followed by transient natural drying in the field and threshing at grain moisture content of about 17%. The system often allows rather high field loss of rice both quantitatively and qualitatively. These necessitate use of a preharvest desiccant, and diquat has been registered recently as a rice desiccant. Aim of the present study is to determine the best time and rate of diquat application and to evaluate its utility as a rice desiccant under its optimal usage conditions in Korea. Diquat was not so effective for japonica rice varieties while it was very effective for the new varieties bred from indica x japonica crosses. The best time of application was the period from 2 days before to 3 days after physiological maturity of grain (moisture content of 28% w.b.) and the optimal rate was about 330g of diquat ion/ha. Applying diquat at the right time and rate dropped grain moisture to 15 to 16% (w.b.) within a week after treatment in contrast to that of 20 to 24% in untreated rice. Desiccation of rice crop with diquat resulted in 10 to 15% increase in grain resistance to shattering and significant reduction in percent green rice. However, there was no reduction in milled rice yield and percent whole kernel and was no change in the percent chalky rice and percent tinted rice. Diquat use appears highly desirable, if there is no residue problem which has not been studied in the present study.

Phytotoxicity and Translocation of Residual Diquat Dibromide from Sandy Loam and Loam Soil to Following Crops Cultivating in the Soils

  • BACKGROUND: Diquat dibromide is a fast-acting nonselective herbicide and plant growth regulator. In this study, in order to understand the possibility of unintentional pesticide contamination in the following crops, the phytotoxicity and transition of diquat dibromide residue in soil into the following crops such as pepper, radish, lettuce and corn have been assessed through phytotoxicity trial and residual evaluation in the unintentional contamination of the higher residual diquat dibromide. METHODS AND RESULTS: The pepper, radish, lettuce and corn were cultivated in the sandy soil and loam soil where the 35 mg/kg and 90 mg/kg diquat dibromide were applied, respectively. Mild growth inhibition symptoms were observed in radish, lettuce and corn crops at the 90 mg/kg- diquat dibromide treatment on the 30 day of cultivation. Diquat dibromide was analyzed using liquid chromatography QTRAP (LC-MS/MS). The recovery rates of diquat dibromide from soil and crop were determined within range from 89.1 to 116.4% with relative standard deviation less than 14.7%. Diquat dibromide residues in soil were found to be 23.90-30.22 and 69.59-82.57 mg/kg from the 35 mg/kg and 90 mg/kg of diquat dibromide-treated soil, respectively after 30 days of crop cultivation. This result implicates that diquat dibromide did not convert to metabolites and remained mostly in the soil, even though it was partially decomposed during crop cultivation. In addition, the diquat dibromide in pepper and radish that were grown for 47 days, and lettuce and corn that were cultivated for 30 days were detected to be 0.01 mg/kg or less in the sandy loam and loam soil where the 90 mg/kg diquat dibromide was applied. CONCLUSION(S): Diquat dibromide did not cause severe phytotoxicity in the following crops as well as it did not uptake and distribute to the following crops, even though it was considered to be residual in the soil.

Glycine alleviated diquat-induced hepatic injury via inhibiting ferroptosis in weaned piglets

  • Objective: The beneficial effects of glycine were tested in piglets with diquat-induced hepatic injury. Methods: Thirty-two piglets were assigned by a 2×2 factorial experimental design including glycine supplementation and diquat challenge. After 3 weeks of feeding with a basic diet or a 1% glycine supplemented diet, piglets were challenged with diquat or saline. After 1 week later, the piglets were slaughtered and samples were collected. Results: Our results indicated that glycine alleviated diquat induced morphological hepatic injury, decreased the activities of plasma alanine aminotransferase, aspartate aminotransferase and glutamyl transpeptidase in the piglets under diquat challenge, and increased total antioxidant capacity and antioxidative enzyme activity significantly. Adding glycine enhanced the concentrations of hepatic adenosine triphosphate and adenosine diphosphate. Transmission electron microscope observation showed that diquat induced clear hepatocytes ferroptosis and its effect could be alleviated by glycine to a certain degree. Moreover, glycine significantly affected mRNA and protein expression of ferroptosis-related signals in the liver. Conclusion: These results demonstrated that glycine attenuated liver damage via inhibiting ferroptosis.

Effects of Oxidative Stress Induced by Diquat on Arginine Metabolism of Postweaning Pigs

  • A total of 16 crossbred post-weaning pigs (10.64${\pm}$0.27 kg BW) were individually penned and assigned to one of two treatments to investigate the influences of diquat-induced oxidative stress on performance and arginine metabolism. Pigs in the oxidative stress group were injected intra-peritoneally with 10 mg/kg BW of diquat, while the control group were injected with isotonic saline. All pigs were fed ad libitum. The experiment lasted for 7 days. The results indicated that compared with control treatment, oxidative stress induced by diquat significantly decreased average daily gain, intake and feed conversion. The treatment decreased activities of antioxidant enzymes, increased concentration of malondialdehyde in plasma, increased cationic amino acid transporter-1 mRNA level and activity of ornithine aminotransferase and concentrations of arginine and citrulline in the jejunum, decreased the concentrations of arginine in plasma and kidney, and decreased induced nitric oxide synthase mRNA level. It is concluded that oxidative stress induced by diquat can influence absorption and metabolism of arginine and consequently modify the requirement of arginine for post-weaning pigs.

Determination of Harvesting Time and Effect of Diquat Treatment in Sesame Cropped After Winter Barley (맥류작 참깨의 수확기 결정과 건조제 처리의 효과)

  • Field experiments were conducted to determine the optimum harvesting time and to evaluate the effect of Diquat spray in late seeded sesame, cultivar 'Suweon 9'. Sesame seed yield reached a plateau from Sept. 18 harvest when seed number was maximum. Thousand seed wt. increased to Sept. 29 harvest. As harvesting was delayed moisture content of capsule decreased and capsule dehiscence increased. Capsule dehiscence did not start until its moisture content dropped below 70%. Optimum harvesting might begin from the time which moisture content of capsule dropped below 70%, leaf senescence reached upper node, and 50% of capsules lost green. About 5% increase in seed weight after defoliation was estimated to be translocation from capsule wall. Diquat spray with 0.3% and 0.5% (v/v) solution of commercial Reglone (20%in A.I.) decreased rapidly capsule moisture content and promoted seed shattering. Dehiscence in 90% capsules was noted at seven days after Diquat spray. Diquat spray as a harvest aid could accelerate sesame desiccation up to 2 wks from normal field condition.

Development of a Simultaneous Analytical Method for Diquat, Paraquat and Chlormequat in Animal Products Using UPLC-MS/MS

  • BACKGROUND: The residual analysis of polar pesticides has remained a challenge. It is even more difficult to simultaneously analyze multiple polar pesticides. Diquat, paraquat, and chlormequat are typical examples of highly polar pesticides. The existing methods for the analysis of diquat, paraquat and chlormequat are complex and time consuming. Therefore, a simple, quick and effective method was developed in the represent study for simultaneous analysis of diquat, paraquat and chlormequat in animal products, meat and fat using UPLC-MS/MS. METHODS AND RESULTS: Sample extraction was carried out using acidified acetonitrile and water and re- extracted with acidified acetonitrile and combine the extracts followed by centrifugation. The extract was then cleaned up with a HLB cartridge after reconstitution with acidic acetonitrile and water. The method was validated in quintuplicate at three different concentrations. The limits of detection (LOD) and quantification (LOQ) were 0.0015 and 0.005 mg/L, respectively. Matrix suppression effect was observed for all of the analytes. A seven point matrix matched calibration curve was constructed for each of the compound resulted excellent linearity with determination coefficients (R2) ≥ 0.991. Accuracy and precision of the method was calculated from the recovery and repeatability and ranged from 62.4 to 119.7% with relative standard deviation less than 18.8%. CONCLUSION: The recovery and repeatability of the developed method were in the acceptable range according to the Codex Alimentarius guideline. The developed method can be applied for the routine monitoring of diquat, paraquat, and chlormequat in animal products, meat and fat.

Effects of Oxidative Stress on Growth Performance, Nutrient Digestibilities and Activities of Antioxidative Enzymes of Weanling Pigs

  • This study was undertaken to investigate the effects of oxidative stress on growth performance, nutrient digestibilities and activities of antioxidant enzymes of weanling pigs. In the experiment, 24 male $Landrance{\times}Yorkshire $weanling pigs were allotted to three groups of 8 animals each. Pigs were fed individually. According to a single factorial arrangement, pigs received diets with 5% of either fresh (group 1 and group 3) or oxidized fish oil (peroxide value was 786.50 meq $O_2/kg$ before inclusion in the diet, group 2). At the beginning of the experiment, pigs in group 3 received an intraperitoneal injection of diquat at 12 mg/kg of body weight. The trial lasted for 26 d. A metabolism test was carried out during the last 4 days of the second week. The results showed that feeding diets containing oxidized fish oil or injection with diquat depressed the growth performance and nutrient digestibilities of weanling pigs, decreased activities of antioxidant enzymes and increased concentration of malondialdehyde in plasma and liver. Intraperitoneal injection of diquat would induce more serious oxidative stress than oral intake of oxidized fish oil in the diet. In conclusion, administration of oxidized fish oil or diquat could induce oxidative stress in weanling pigs, and oxidative stress could depress growth performance and impact anti-oxidative ability of young pigs.

Investigation on the Use of Paraquat and Diquat as a Desiccant for Sesame Harvest-Aid (참깨 수확을(收穫) 위한 건조제(乾燥劑)로서 Paraquat와 Diaquat의 이용(利用)에 관(關)한 연구(硏究))

  • Experiments were carried out to investigate the utilization of paraquat and diquat as desiccants for harvest-aid of sesame. When paraquat and diquat were sprayed at 3days or 3hours before cutting of sesame plants, moisture content in treated plots was evenly rapidly decreased and sesame yeild was not significantly different as compared with that in untreated plot. Percentage of germination of sesame seed in treated plots was similar to that in untreated check. Residual amount in sesame seed was not detectable when these chemicals were treated with 250ppm at 3days before cutting of sesame plant. Percentage of thrashing was high in sesame plants treated with paraquat and diauat with the lapse of time after cutting. Required labor in thrashing of sesame was reduced when applied with these chemicals. Harvest efficiency of sesame after spray of paraquat and diquat was good by comparison with the traditional practice.
