Characterization of Coolia spp. (Gonyaucales, Dinophyceae) from Southern Tunisia: first record of Coolia malayensis in the Mediterranean Sea

  • This study provides the first report of the presence of Coolia malayensis in the Mediterranean Sea, co-occurring with C. monotis. Isolated strains from the Gulf of Gabès, Tunisia (South-eastern Mediterranean) were identified by morphological characterization and phylogenetic analysis. Examination by light and scanning electron microscopy revealed no significant morphological differences between the Tunisian isolates and other geographically distant strains of C. monotis and C. malayensis. Phylogenetic trees based on ITS1-5.8S-ITS2 and D1-D3/28S rDNA sequences showed that C. monotis strains clustered with others from the Mediterranean and Atlantic whereas the C. malayensis isolate branched with isolates from the Pacific and the Atlantic, therefore revealing no geographical trend among C. monotis and C. malayensis populations. Ultrastructural analyses by transmission electron microscopy revealed the presence of numerous vesicles containing spirally coiled fibers in both C. malayensis and C. monotis cells, which we speculate to be involved in mucus production.

Morphological and molecular characterization of the genus Coolia (Dinophyceae) from Bahía de La Paz, southwest Gulf of California

  • The genus Coolia A. Meunier 1919 has a global distribution and is a common member of epiphytic dinoflagellate assemblages in neritic ecosystems. Coolia monotis is the type species of the genus and was the only known species for 76 years. Over the past few decades, molecular characterization has unveiled two species complexes that group morphologically very similar species, so their limits are often unclear. To provide new knowledge on the biogeography and species composition of the genus Coolia, 16 strains were isolated from Bahía de La Paz, Gulf of California. The species were identified by applying morphological and molecular approaches. The morphometric characteristics of all isolated Coolia species were consistent with the original taxa descriptions. Phylogenetic analyses (large subunit [LSU] rDNA D1 / D2 and internal transcribed spacer [ITS] 1 / 5.8S / ITS2) revealed a species assemblage comprising Coolia malayensis, C. palmyrensis, C. tropicalis, and the C. cf. canariensis lineage. This is the first report of Coolia palmyrensis and C. cf. canariensis in Mexico and C. tropicalis in the Gulf of California. Our results strengthen the biogeographical understanding of these potentially harmful epiphytic dinoflagellate species.

First report of Amphidinium fijiense(Dinophyceae) from the intertidal zone of a sandy beach of Jeju Island, Korea

  • A strain of Amphidinium species was established from samples collected from the intertidal zone of a sandy beach of Jeju Island, Korea. Its cells were 13.0-15.0 ㎛ in length and 10.0-13.0 ㎛ in width. Its cell shape was round or oval and dorsoventrally flat. A pyrenoid was located in the center of the cell and a nucleus was posteriorly located. Its epicone was small and left-deflecting. Its cingulum had V-shape on the ventral side, forming a ventral ridge and extending to the sulcus. Polygonal amphiesmal vesicles and ring-shaped body scales not described previous were observed on the surface of the cell. Its morphological features were consistent with those of previously described Amphidinium fijiense. Phylogeny based on ITS region and LSU rDNA sequences revealed that this Amphidinium isolate was clearly clustered with other A. fijiense strains, but separated from other Amphidinium species. These results indicate that this Amphidinium isolate is A. fijiense. This study reports its presence for the first time in the intertidal zone of a sandy beach of Jeju Island, Korea.

Sequence analysis of partial LSU rDNA of three Alexandrium species (Dinophyceae) hitherto unreported

  • We, for the first time, reported molecular sequences of large subunit ribosomal DNA Dl-D3 region of A. hiranoi, A. leei and A. satoanum hitherto unreported. In addition, this study presented the full-length sequences of A. affine, A. fraterculus, A. catenella and A. tamarense occurring in Korean coastal waters. In total, 17 Alexandrium morphospecies were subjected to the phylogenetic analysis using the Maximum-likelihood (ML) method. The alignment result of sequences of A. hiranoi and A. pseudogonyaulax showed that there were only two substitutions without length heterogeneity implying their genetic affiliation. In ML tree, A. leei formed a deeply diverging branch probably because of the accelerated evolutionary rate, and its phylogenetic position was so ambiguous to resolve the phylogenetic relationship to the residual taxa. An A. satoanum culture showing morphological variation in the sulcal plate formed an independent divergent branch with consistent sister relationship to A. hiranoi/A. pseudogonyaulax clade supported by the high posterior probability (PP) value. Blast search in GenBank showed the sequence data of A. affine, A. fraterculus, A. catenella and A. tamarense corresponded to their morphological species designation. In ML tree, Alexandrium species were commonly split into four main clades. The inter-clade relationships were not clear and usually supported by the week PP values. In general, the sulcal plate of Alexandrium species seemed to reflect the true phylogeny at the main clade level, and the connection between the 1 and the apical pore complex seemed to reflect the phylogeny at the subclade level.

평택호에서 수환경과 식물플랑크톤의 계절적 동태 (Seasonal Dynamics of Aquatic Environment and Phytoplankton in Pyeongtaek Reservoir, Korea)

  • Seasonal investigations were conducted to determine the major aquatic environmental factors and the variation of phytoplankton in Pyeongtaek Reservoir in March, June, September, and December 2000. Heavy rainfall mainly occurs from late June to mid-September, and water quality of reservoir was high in the influent zone of stream and riverine zone of reservoir. The biomass of phytoplankton was related to aquatic environmental factors. In particular, its value increased where nutrient concentration was high. Likewise, the increase of turbidity was found to have anthropogenic effects on the varying quantity of phytoplankton. The phytoplankton composition in quantitative survey identified into 43 genera and 71 species. Species numbers of Bacillariophyceae, Cyanophyceae, and Chlorophyceae accounted for 17%, 15%, and 49%, respectively, with the remainder constituting less than 3-7%. The distribution of such phyla also significantly varied according to seasons, accounting for 25%, 37%, 61%, and 14% in March, June, September, and December, respectively. Bacillariophyceae and Chlorophyceae were observed throughout the year, while Cyanophyceae proliferated in June and September. Euglenophyceae and Dinophyceae were prevalent in March and September, while Cryptophyceae occurred in March and December. The succession trend of phytoplankton showed the maximum cell density was followed by Bacillariophyceae (6.8$\times$$10^3$ cells ${\cdot}$ml)$\rightarrow$ Chlorophyceae (3.7$\times$$10^3$ cells ${\cdot}$ml)$\rightarrow$Cyanophyceae (1.3$\times$$10^4$ cells ${\cdot}$ml)$\rightarrow$Cryptophyceae (1.2$\times$$10^3$ cells ${\cdot}$ml). The cell density was the highest in the upstream. Dominant species were composed of Aulacoseira ambigua, Stephanodiscus hantzschii f. tenuis of Bacillariophyceae, Anabaena spiroides var. crassa, Microcystis aeruginosa, Oscillatoria amphibia of Cyanophyceae, Actinastrum hantzschii var. fluviatile, Pediastrum duplex var. reticulatum of Chlorophyceae, Euglena gracilis, Trachelomonas spp. of Euglenophyceae, and Chroomonas spp., Cryptomonas spp. of Cryptophyceae. As a results, seasonal variation of phytoplankton in Pyeongtaek Reservoir was evident in spite of inflow the high concentration of nutrients from watershed streams, because hydrological control and anthropogenic disturbance in reservoir were found to have major effects on the retention time of water.

Morphological characterization of Symbiodinium minutum and S. psygmophilum belonging to clade B

  • Dinoflagellates in the genus Symbiodinium are known as zooxanthellae and exist as either symbiotic or free-living forms. Among this genus, two species, Symbiodinium minutum and Symbiodinium psygmophilum, were recently established based on genetic characters. However, some critical morphological characters of these species, such as plate formulae and related diagnoses have not been provided yet. In this study, we analyzed the morphology of S. minutum and S. psygmophilum by scanning and transmission electron microscopy. The S. minutum had the Kofoidian plate formula consisting of a small plate (x), elongated amphiesmal vesicle (EAV), 4', 5a, 8", 7s, two cingulum rows, 18-20c, 6''', and 2'''', while S. psygmophilum had x, EAV, 4', 5a, 8", 7-10s, two cingulum rows, 20-22c, 5-6''', and 1''''. These plate formulae are different from any other reported Symbiodinium species. In addition, both species had a pentagonal 1a plate and a hexagonal 2a plate, while other known Symbiodinium species had a hexagonal 1a plate and a pentagonal 2a plate. Thus, we confirm the species status of S. minutum and S. psygmophilum based on morphological and genetic characters and report the detailed morphological characteristics of these two species.

영산강 상류의 담수조류 분포에 관한 연구(1) - 광주지역 상수원을 중심으로 - (Freshwater Algae in the Upstream of the Yongsan River System(1) -on the Drinking Water Supplying Area of Kwangju Districts-)

  • Seasonal occurrences and dynamics on the freshwater algae were examined in the upstream of the Yongsan river system with the Kwangju service water area from 1993 to 1994. Three hundred and three taxi were totally collected and composed of 6 classes, 15 orders, 7 suborders, 31 families, 74 genera, 248 species, 48 varieties and 7 forma. Compositions in each algal class were divided in 136 taxi of Chlorophyceae(44.8% ), 109 taxi of Bacillariophyceae(36.0% ), 30 taxa of Cyanophyceae(10.1% ), 21 taxa of Euglenophyceae(6. 9% ),4 taxi of Chrysophyceae(1.3% ) and 3 taxa of Dinophyceae(1.0% ). The occurrence of taxa was a markedly decrease(-62) between summer and autumn, the critically increase(+43) between spring and summer. And also, it was indicated that the high frequency of taxa in summer, but the high standing biomass In winter in spite of low taxi. In general, it has much algal biomass in station 9 and 10, but low in station 4, 5 and 11, relatively. Through this survey, the major dominant taxa in algal biomass, Microcystis aeruginosa, Melosira varians, Synedra ulna and Navicula cryptocephala, they were all above 2.1×10$^{6}$ce11s/1, 13.2% of totals. There are about 10% of new emigrated taxa by season and they showed a remarkable succession. We may suggested that this system was well sustained for the habitat of freshwater algal community with low dominance and high diversity. Twenty three of nuisance algae including of Microcystis aeruginosa, Peridinium willei and Anabaena flos-aquae were distributed as if had low appearance. Of course, many of them were saprobiontic taxa, extraordinary growttung groups depend on organic loading in water system and also it was not showed severe algal blooming phenomenon.

국내 해양식물플랑크톤의 주요종과 분포에 대한 조사 (A List of Important Species and Distribution of Marine Phytoplankton in Korea)

  • 문헌 32편에서 조사된 출현종수는 최저 43종에서 최고 200종까지 기록되고, 그 중에서 주요종수는 최저 2종에서 최고 27종까지로 종수의 기록에 상당한 차이를 보였다. 본 조사에서 기록한 주요종의 목록은 모두 50속 116종이었고, 규조류 34속 89종(76.6%), 와편모조류 11속 22종(18.9%), 황갈색조류 2속 2종(1.7%), 남조류, 라피도조류, 유글레나조류가 각각 1속 1종(0.9%)이었다. 생태적 특성을 반영하는 주요 종으로는 우점종이 Actinoptychus seranius를 비롯하여 79종, 출현빈번종이 Coscinodiscus centralis를 비롯하여 50종, 적조원인종이 Cochlodinium polykrikoides를 비롯한 36종이었다. 또한 3가지의 주요 특성을 모두 지닌 종은 Prorocentrum micans를 비롯한 11종이었다. 여러 문헌에서 주요종으로 언급했던 종은 규조류인 Skeletonema costatum이었고 단일 속(genus)으로서 18종의 많은 종이 조사된 속은 Chaetoceros속이었다.

경포호의 식물플랑크톤과 환경요인의 계절적 변동 (Seasonal Variation of Phytoplankton and Environmental Factors in Kyungpo Lake)

  • In order to elucidate chatracteristics of water quality and seasonal dynamics of phytoplankton community in Kyungpo lake, the weekly variation of algal population and 11 environmental factors were investigated from March 1998 to February 1999. A total 121 species of phytoplankton belonged to 57 genera, 4 genera 7 species of the Euglenophyceae, 1 genera 2 species of the Chrysophyceae, 2 genera 3 species of the Cryptophyceae, 1 genera 1 species of the Dinophyceae, 6 genera 9 species of the Cyanophyceae, 28 genera 78 species of the Bacillario-phyceae and 15 genera 21 species of the Chlorophyceae, was identified. Cryptomonas sp., Chamydomonas sp., Nitzschia frustulum, Oscillatoria sp., Lyngbya sp, and Dictyosphaerium pulchellum occurred successively as dominant phytoplankton species from March 1998 to February 1999. Density of phytoplankton cell number showed the peak in early April when Chlamydomoas sp. was dominated. The diversity index of phytoplankton community showed low values in the dominating period of Cryptomonas sp. and Chlamydomonas sp. The concentration of chlorophyll a fluctuated between 1.09 and 107.7$\mu\textrm{g}$ㆍ1$^{-1}$. Water temperature, salinity, DO, conduc tivity and SS were ranged 0.5-27.8$^{\circ}C$, 2.8-25.2 ppt, 4.62-19.21 mgO$_2$ㆍ1$^{-1}$, 5.33-34.76 mS/cm and 0.017-0.184 mgㆍ1$^{-1}$, respectively. Annual means of Secchi disc transparency was 36cm and thai of pH value was 8.5. The concentrations of PO$_4$-P was ranged 0.0012-0.077 mgㆍ1$^{-1}$ , and showed high value from August to October. The concentrations of NO$_2$-N, NO$_3$-N and NH$_4$-N was ranged 0.0011-0.1 mgㆍ1$^{-1}$, 0.074-3.33mgㆍ1$^{-1}$ and 0.0024-0.259 mgㆍ1$^{-1}$, respectively. Among environmental factors, water temperature was negatively correlated with DO, salinity and conductivity, DO was positively correlated with salinity and conductivity. Chlorophyll a, one of the indicies of eutropication in lake, was negatively correlated with NH$_4$-N. The cell density of Cryptomonas sp., which was the most dominant species in this lake, was negatively correlated with transparency. 조류생산잠재력조사 방법개발에 의한 육수환경의 부영양화 방지대책에 관한 연구(I) -순수분리종의 형태 및 증식특성- (A Study on Protection Plan of Eutrophication in Fresh Water Environment by Development of Methods for Algal Growth Potential test (I) -Morphology and Growth Characteristics of Isolated algae-)

  • The isolation, morphological study and growth characteristics of the algae were investigated from Lake Chuam. The isolated algae were applied the Agal Growth Potential test. The method of isolation and purification of the algae were used to Agar plating(AP), nutrient enrichment(NE), dilution(DI) and micro capillary technique(MC). Total isolated algae were 21 species. They were composed of Cyanophyceae, Dinophyceae, Bacillariophyceae, Euglenophyceae and Chlorophyceae. The numbers of algal strain by isolation technique were highest in dilution(21 species), and those of the rests were showed in order of NE > MC > AP. The sizes of isolated Selenastrum and Scenedesmus were $1.8\pm 1.4 \mu m$, $3.3\pm 0.9 \mu m$ in diameter and $6.4\pm 2.3 \mu m$, $13.6\pm 1.9 \mu m$ in length respectively. The morphology of isolated algae and NIES-collection strain was very similar each other, but the size was smaller isolated algae than that of NIES-collection. The optimum culture condition of isolated Selenastrum and Scenedesmus was about 30$\circ$C(25$\circ$C-35$\circ$C) in temperature and the maximum growth was appeared between 7,000 lux and 8,000 lux in the light intensity. The comparison of $\mu$(specific growth rate) on the concentration of nutrients such as nitrate and phosphate, isolated Selenastrum was appeared maximum it at 1.0 mg $NO_3-N/l$ but NIES-collection strain was showed 95% of maximum it at same nitrate concentration. Maximum g of isolated algae and NIES-collection strain in Scenedesmus onto nitrate concentration were very similar with the result of selenastrum. The specific growth rates of isolated algae and NIES-collection strain on the gradient concentration of phosphate were showed 0.72/day and 0.70/day at 0.02 mg $PO_4-P/l$ in Selenastrum but those of Scenedesmus were appeared 0.61/day and 0.57/day at same concentration $PO_4-P$.
