Reduction of Nitrous Oxide Emission by EGR Method on Diesel Engine (디젤엔진에서 배기가스 재순환 방법을 이용한 아산화질소의 배출률 저감)

  • Nitrous oxide($N_2O$) concentration in the atmosphere has been constantly increased by the human activities with industrial growth after the industrial revolution. One of factors to increase $N_2O$ concentration in the atmosphere is the $N_2O$ emission caused by the combustion of marine fuel. Especially, a sulfur component included in marine fuel oils is known as increasing the $N_2O$ formation in diesel combustion. Form this point of view, $N_2O$ emission from a ship is not negligible. On the other hand, Exhaust gas recirculation(EGR) that have thermal, chemical and dilution effect is effective method for reducing the NOx emission. In this study, an author investigated $N_2O$ reduction by using EGR on a direct injection diesel engine. The test engine was a 4-stroke diesel engine with maximum output of 12 kW at 2600rpm, and operating condition of the engine was a fixed load of 75%. The experimental oil was a blend-fuel that were adjusted with sulfur ratio of 3.5%, and EGR ratio of 0%, 10%, 20% and 30%. In conclusion, diesel fuel that contained 3.5% sulfur component increased $SO_2$ emission in exhaust gas, and increment of EGR ratio reduced NO emission. Moreover, $N_2O$ emission was decreased as over 50% at EGR ratio of 10% and reduced 100% at EGR ratio of 30% compared with $N_2O$ emission of 0% EGR ratio.

  • Measurement of triiodothyronine($T_3$) concentration is useful for the diagnosic and treatment of thyroid gland diseases. Fundermental studies of measurement of $T_3$ concentration by radioimmunoassay were performed and values determined by commercially available kit, Coat-A-Count, Diagnostic Products Corporation. The optimal utilization of the radioimmunoassay in measuring $T_3$ concentrations is dependant not only on high quality performance of the assay, but also on their appropriate application to the clinical situation. These are several aspects that must be considered in every individual case. These include factors such as accurate pippeting in reagent preparation in the assay and through decanting to remove all visible moisture after incubation steps and so forth. In attempt to assess quality control of the radioimmunoassay of serum $T_3$, serum pools with high, medium, low $T_3$ concentrations were assayed for each of 5 samples. The results obtained with this study were as follow: 1. The coefficient of variation(C. V.) for the standard curve ranged between $0.2{\sim}3.5%$. 2. It was necessary that both incubation time and temperature should correctly be maintained all the in the assay performance. 3. The precison with the $T_3$ RIA procedure was good. 4. The measured values of serially diluted serum $T_3$ concentration with Ong/dl standard solution was proportional to the predicted values. However dilution curve of distilled water was not strait. 5. Calculated $T_3$ values of patient serum in normal group was $107.8{\pm}25.90ng/dl$ in male patient and $127.29{\pm}24.08ng/dl$ in female patient.

Current Application of Embryo Cryopreservation for Farm Animals (가축 수정란 동결보존의 최근 이용방법)

  • This paper reviews the most important steps that have generated consistent progress in principles and developmental progress of embryo cryopreservation, and also study on freezing procedure and its application by conventional method and current improved method for freezing procedure and its appilcation of embryo cryopreservation in farm animals. Four were of particular interest: 1.The transport of water across the ccli membrane (zona pellucida) during freezing and thawing accordinglyplays a role in determing whether the celi survives. This movement of water is controlied mainly by extracellular phase changes and by the nature and concentration of any cryoprotective agent present. Therates of cooling, freezing and warming, and the intervals over which they are applied are further decisi've factors in determining whether a cryopreservation procedure allows survival after thawing. 2.The first successful deep freezing experiments with sheep morula and blastocysts during the seventies were based on the early procedures used for mouse embryos.Current research during the eighties is developed with the aim of simplifying and improving current procedures such as one-step dilution and rapid or ultra-rapid cooling by using the model of laboratory animals. 3.The conventional method for the embryo cryopreservation is described. An alternative to this method which may result in high survival and also in reducing of the freezing and thawing time is done by combing a permeable cryoprotectant such as glycerol, DMSO or propanediol and a non-permeable compound such as sucrose, trehalose, raffinose or lactose. 4.Finally a different approach to the preservation of embryos, named vitrification, is introduced. This procedure depends upon the ability of concentrated solutions of cryoprotective agents such as glycerol and propanediol to supercool to very low temperature (-196$^{\circ}C$) during rapid cooling before solidifying without formation of ice. However, more complete data are necessary for successful vitrification of blastocysts.

Role of Oxidative Stress and Mitochondria in Parkinson's Disease

  • Central to developing new treatment strategies for late onset sporadic Parkinson's disease (PD) and early onset familial PD is resolving the enigma of the specific vulnerability exhibited by substantia nigra dopamine (DA) neurons despite multiple risk factors. Neuropathological evidence from both human and experimental models of PD firmly supports a significant role for oxidative stress (OS) and mitochondrial dysfunction in the death of nigral DA neurons. Largely unknown are the genes underlying selective susceptibility of nigral DA neuron to OS and mitochondrial dysfunction and how they effect nigral DA cell death. To overcome the paucity of nigral DA neurons as well as the dilution effect of non-DA cells in brain tissues, we have developed wild type DA cell line model, SN4741 and mutant DJ-1 (-/-) DA cells, appropriate for microarray analysis and differential mitochondrial proteomics. Mutations in the DJ-1 gene (PARK7), localized in cytoplasm and mitochondria, cause autosomal recessive early onset PD. Through microarray analysis using SN4741 cells followed by validation tests, we have identified a novel phylogenically conserved neuroprotective gene, Oxi-a, which is specifically expressed in DA neurons. The knockdown of the gene dramatically increased vulnerability to as. Importantly as down-regulated the expression level of the gene and recovery of its expression via transient transfection exerted significant neuroprotection against as insult. We also have identified altered expression of mitochondrial proteins and other familial PD genes in DJ-1 (-/-) mutant cells by differential mitochondrial proteomics. In DJ-1 (-/-) cells the knockdown of the other familial PD genes (Parkin and PINK1) dramatically increased susceptibility to as. Thus, further functional characterization of the Oxi-$\alpha$ gene family and the mitochondrial alteration in the DJ-1 (-/-) cell model will provide the rationale for the neuroprotective therapy against both sporadic and familial PD.

Ethanol Production Using Alginate Immobilized Cells of Zymomonas rnobilis (고정화 Zymomonas mobilis 균체로부터 에탄올 생산)

  • The fermentation characteristics of ethanol production by the use of immobilized Zymomonas mobilis KCTC 1534 cells were investigated in terms of formation factors such as substrate and product concentration. In batch fermentation, the maximum values of specific ethanol productivity, specific substrate uptake rate, ethanol yield, and glucose conversion rate were $29.14g/{\ell}{\cdot}h$, $60.24g/{\ell}{\cdot}h$, 0.48g/g, and 98.4%, respectively, with 17% glucose medium, and its ethanol productivity was $2.91g/{\ell}{\cdot}h$ in the case of 25 hour fermentation time. Repeated batch fermentation was possible for 30 days with 2.24-$2.94g/{\ell}{\cdot}h$ ethanol productivity. In semicontinuous fermentation, the maximum ethanol productivity was shown to be $15.7g/{\ell}{\cdot}h$ at $0.36h^{-1}$ effective dilution rate with 17% glucose concentration. In this case, ethanol yield coefficient and glucose conversion rate were 0.39 g/g, 64.7%, respectively.

Water Quality Properties of Tributaries of Daechung Lake, Korea (대청호 유입지천의 수질 특성)

  • The tributaries of Daechung Lake play an important role in controlling eutrophication in the lake, which is used for agricultural purposes and as potable water. However, water quality properties were not extensively studied in the tributaries of Daechung Lake. The objectives of this study are to investigate spatial and temporal properties of water quality and to characterize streams which could threaten water quality of Daechung Lake. For this study, water samples were weekly or monthly collected from February 2014 to October 2014 in 9 streams. Water quality parameters analyzed in this study include biochemical oxygen demand (BOD), chemical oxygen demand (COD), total organic carbon (TOC), total nitrogen and phosphorus (TN and TP), suspended solids (SS), and chlorophyll a. Based on temporal distribution and principal component analysis, BOD, COD, TOC, SS, and TP were controlled by not only river discharge that increased during summer due to heavy rain fall, but also due to anthropogenic input (e.g., bridge construction and/or agricultural activity). Dilution is also one of the factors explaining TN and conductivity, both of which decreased with increased discharge. Generally, concentrations of contaminants (BOD, COD, TOC, TN and TP) in the tributaries were higher than those of Daechung Lake. However, pollution load indicated that only the main channel of Geum River and Sook Stream may largely influence lake waters, attributed mostly to their large volumes. This implies that the main channel and Sook Stream are the major influences on the water quality of Daechung Lake.

Isolation and Characterization of 3,4-Dichloroaniline Degrading Bacteria from a Sandbank (갯벌에서 분리한 3,4-Dichloroaniline 분해 미생물의 특성)

  • The compound 3,4-dichloroaniline (DCA) is an aromatic amine used as an intermediate product in the synthesis of herbicides, azo-dyes and harmaceuticals. It is also a degradation product of some herbicides (diuron, propanil, and linuron) and of trichlorocarbanilide, a chemical used as active agent in the cosmetic industry. 3,4-DCA, however, is considered potential pollutants due to their toxic and recalcitrant properties to humans and other species. A bacterium capable of growth on 3,4-DCA was isolated by dilution method from 3,4-DCA-containing enrichment culture. Finally, a strain, YM-14, capable of degrading efficiently 3,4-DCA was isolated from a sandbank. The isolated strain, YM-14 was identified to be Arthrobacter sp.. Fifty ppm 3,4-DCA in 1/10 LB media was completely degraded by the growth of Arthrobacter sp. YM-14 for 12 h at $30^{\circ}C$. The isolated strain is capable of growth on 3,4-DCA as sole carbon source and also able to degrade other chloroaniline compounds. Also, the isolated strain showed high level of catechol 1,2-dioxygenase activity by 3,4-DCA exposure. The catechol 1,2-dioxygenase was supposed to be ones of the important factors for 3,4-DCA degradation.

Evaluation of Organic Matter Sources of Phytoplankton in Paldang Reservoir using Stable Isotope Analysis (팔당호 내 식물플랑크톤 안정동위원소 분석을 통한 유기물 기원 평가)

  • The organic matter sources of phytoplankton and related environmental factors influencing algal bloom in Paldang reservoir were studied using nitrogen and carbon isotope ratio(${\delta}^{15}N$, ${\delta}^{13}C$). Phytoplankton samples for stable isotope analysis were collected from four points in reservoir using a plankton net. Physicochemical water quality, algal taxa and hydrological data were collected from published monitoring material. Phytoplankton samples were analyzed by IRMS. CN ratio of each sample was very similar to that of phytoplankton from literature cited. ${\delta}^{15}N$ of each sample was decreased during July. Mixing and dilution of nitrogen sources due to increment of influx by concentrated rainfall were considered as the main reason for the decline of ${\delta}^{15}N$. Based on analyzed ${\delta}^{15}N$ value of each sample, nitrogen source of Bughan river sample was presumed to come from soil. The nitrogen sources of Namhan river and Kyeongan stream samples seemed to be sewage or animal waste. Low ${\delta}^{15}N$ value in August (2012) seemed to be influenced by isotope fractionation due to the blooming of nitrogen-fixation blue-green algae (Anabaena spp.). Variation in ${\delta}^{15}N$ values particularly by blue-green algal bloom was considered the important factor for estimating the organic matter sources of phytoplankton.

Biological improvement of reclaimed tidal land soil (II) -Changes of soil-microbial populations in reclaimed tidal land- (해안간척지 토양의 생물학적 토성개량에 관한 연구 (제 2 ) -간척지토양에 있어서 생물의 화에 대하여-)

  • The soil of the reclaimed tidal land, located in Chogi-ri, Is. Kanghwa, Korea was used in this experiment. The experimented soil samples were collected from 18 sites with its time elapsed after the shore-protection works, soil-depth and the vegetation of saline plants, and at each site samplings were conducted monthly from March through October, 1968, for the purposes of examining the changes of microbial populations for the microbes such as bacteria, actinomycetes and fungi, by using the dilution plate method. The numbers of the microbes in these soils generally showed lower levels comparing with those of other soils. The more time elapsed after the reclamation, the higher numbers of the microbes inhibited the soils. Higher populations were there in the surface soils than in the lower part of the area. The surface soils included comparatively better conditions in aeration and contents of organic matter than in the lower part, and this fact was. same as in general soils. However, not so was this in the case of March, April and October due to the higher soil temperatures in the lows. At the experimental sites where the halophytes such as Salicorniu were grown vigourously, the more densly the plants grew, the higher populations of actinomycetes and fungi were, but not in the case of bacterial population. This means, in this soil with dense Salicornia, it is difficult to obtain good-natured soils in short time without a higher population of bacteria. For the rapid utilization of the land soil, in this view of point, the methods increasing the number of bacteria in the soil are needed as well as the cultivation and harvesting Salicorniu which indicated in the privious paper(Hong, et al., 1969a). According to the results of this experiment, the changes of soil-microbial populations in the reclaimed tidal land soil containing high salinity depend deeply upon the interrelations of many environmental factors such as soil-salinity, soil-components and contents, concentration of organic matters, pH, aeration, and air and soil temperatures, as in the general soils.

The Skin-Related Biological Activities of Aerially Extract of Oenothera lamarckiana (큰달맞이꽃 지상부 추출물의 피부 관련 생리활성 효과)

  • Skin plays important roles in protecting the internal organs from the chemical-biological risk factors and ultraviolet light. Exposure to the chemical and biological stimuli has a detrimental effect on skin's structure and physiological regulation. Therefore, much attention has been paid to natural products that show biological activities such as anti-oxidation, anti-aging and anti-bacterial activities. In this study, we investigated the skin-related biological activities of Oenothera lamarckiana aerial part extract. The extract contained 229.35 mg TAE (tannic acid equivalents)/g total polyphenolic compounds and the extract showed relative high antioxidant activity ($SC_{50}$ value: $8.52{\mu}g/mL$). The $IC_{50}$ value against tyrosinase and elastase were 307.94 and $181.51{\mu}g/mL$, respectively. This suggested that O. lamarckiana can be applied to whiten skin and slow the aging of skin. O. lamarckiana extract showed a growth inhibitory effect on Staphylococcu epidermidis (minimum inhibitory concentration: $250{\mu}g/mL$). Interestingly, O. lamarckiana extract showed no inhibitory effect on Pseudomonas aeruginosa on the paper disc assay. Yet the extract inhibited the growth of Pseudomonas aeruginosa on the broth dilution assay in a dose-dependent manners. Taken together, O. lamarckiana could have good potential for development as an additive in the cosmetic industry.
