Evaluation of Coarse Spray Vaccination with B$_1$ Strain against Newcastle Disease (뉴캣슬병 B$_1$생독백신의 분무접종 효과)

  • An experiment was conducted to examine the efficacy of spray vaccination against Newcastle disease (ND). Four different coarse sprayers and four different diluents were compared in particle size, immune response and respiratory reaction. The smaller particle size of sprayer was, the better efficacy was and the more respiratory reactions were appeared. Of four diluents, antibody responses and resistance to challenge were higher with skim milk and gelatin, respectively .When day-old broiler chicks with maternal antibodies were vaccinated by coarse. sprayer B with B$_1$strain diluted in 1% skim milk, they showed 100% protection at two weeks of age and 50% from 4 to 8 weeks of age. Another group which boostered at 2 weeks of age by drinking water vaccination showed 100%, 70%, 50%, 40% protection at 2, 4, 6, 8 weeks of age, respectively. Although spray vaccination against ND in this experiments showed similar efficacy, compared to conventional methods, this method may not be appropriate in our field condition because of respiratory vaccinal reaction.

Effect of Electric Frequency on the Partial Discharge Resistance of Epoxy Systems with Two Diluents

  • Partial discharge resistance for the epoxy systems with two diluents was investigated in the rod-plane electrodes arrangement, and the effect of electric frequency on the partial discharge resistance was also studied. Diglycidyl ether of bisphenol A (DGEBA) type epoxy was used as a base resin, and 1,4-butanediol diglycidyl ether (BDGE) or polyglycol (PG) as a reactive diluent was introduced to the DGEBA system, in order to decrease the viscosity of the DGEBA epoxy system. BDGE was acted as a chain extender, and PG acted as a flexibilizer, after the curing reaction. To measure the partial discharge resistance, 5 kV alternating current (ac) with three different frequencies (60, 500 and 1,000 Hz) was applied to the specimen in a rod-plane electrode arrangement, at $30^{\circ}C$. PG had a good effect, while BDGE had a bad effect on the partial discharge resistance of the DGEBA system, regardless of the electric frequency.

  • The enzyme-linked immunosorbent assay using the so-called "double-sandwich"technique was applied to determine Clostridium botulinum type F toxin. Polystyrene tubes were coated with horse anti-type F toxin serum and then toxin sample was added. The tubes were subsequently treated with rabbit anti-type F toxin IgG and sheep anti-rabbit serum IgG-horseradish peroxidase conjugate. By this technique, about 10 mouse intraperitoneal 50% lethal doses (ip LD/50/) of type F toxin could be detected. Low back-ground reading was achieved with the use of phosphate-buffered saline containing 0.05% Tween 20 and 1% bovine serum albumin as diluents of rabbit IgG and conjugate. Addition of EDTA in the diluents of toxin increased ELISA extinction value significantly. No cross-reaction was observed with botulinum type A and B toxin, but type E toxin gave sleight cross-reaction.

  • The effect of diluents on the dissolution rate of indomethacin was studied and the dissolution rate for the marketed products was compared to establish the quality standard for indomethacin preparation. The results are as fellows 1. The effect of test methods was not significant in the case of paddle method but was greatly effected in the case of basket method. 2. The effect of diluents was not significant generally, but in the case of starch dissolution rate was greatly effected. 3. The dissolution rate was remarkably decreased in proprotion to increase of capsule size. 4. The marketed products met the KP dissolution requirement but showed a little difference due to manufacturers.

Conditions for the disinfectant efficacy test under subzero temperatures

  • To establish appropriate conditions for a disinfectant efficacy test at subzero temperatures, this study examined mixtures of frozen foot-and-mouth disease virus or avian influenza virus solutions and disinfectant diluents at $-5^{\circ}C$ and monitored temperature and freezing status of an anti-freezing diluent (AFD, 15% ethanol + 30% propylene glycol + 55% distilled water) over time at various subzero temperatures. Viral solutions and disinfectant diluents froze before the mixtures reached $-5^{\circ}C$, whereas the AFD was not frozen at $-30^{\circ}C$. The times taken for the AFD to reach -10, -20, -30, and $-40^{\circ}C$ from room temperature were 36, 39, 45, and 48 min, respectively.

Effect of Antioxidant Fortification on Preservability of Buffalo Semen

  • During the process of freezing, spermatozoa suffer cold shock which increases their susceptibility to lipid peroxidation which plays an important role in ageing of spermatozoa, shortening their life span and affecting the preservation of semen. An experiment was therefore conducted to study the effect of addition of natural antioxidants into semen diluents on the preservability of buffalo semen. Split semen samples were extended in milk egg yolk diluents fortified with vitamin E (MYE), vitamin C (MYC) and control group (MYO); Tris-egg yolk diluents fortified with vitamin E (TYE), vitamin C (TYC) and control group (TYO) and evaluated for their preservabilities at 4-7$^{\circ}C$ and $37^{\circ}C$. Overall least squares mean of percent motility observed after 0, 24, 48, 72 and 96 h of preservation at 4-7$^{\circ}C$ were 66.70, 54.00, 36.80, 21.90 and 12.50, respectively while the estimates for semen extended in MYE, MYC, MYO, TYE, TYC and TYO were 44.80, 42.70, 38.70, 36.00, 35.20 and 33.00 percent, respectively. The results showed that motility was significantly (p<0.01) affected by extender (extender-antioxidant combination) and preservation interval. Overall least squares mean percent motility observed after 0, 4, 8, 12 and 24 h of preservation at $37^{\circ}C$ were 68.50, 58.90, 45.00, 38.10 and 18.10 percent, respectively, while the estimates for semen extended in MYE, MYC, MYO, TYE, TYC and TYO were 48.20, 49.30, 46.80, 45.30, 42.30 and 42.50 percent, respectively. Extender and storage interval were found to be significantly (p<0.01) affecting spermatozoa motility on room temperature preservation. The results indicated that the incorporation of antioxidants, especially vitamin E, had beneficial effect on preservability of buffalo semen.

Safety and efficacy of modified-live infectious laryngotracheitis vaccines (닭 전염성 후두기관염 생독백신의 안전성과 효능)

  • Modified-live (ML) infectious laryngotracheitis (ILT) vaccines have been widely used as a preventive measure in Korea since the first outbreak of ITL. Recently, it has been observed that chickens vaccinated with the commercially available ML ILT vaccine have sometimes exhibited adverse clinical signs. In this study, we evaluated the quality of the vaccines by comparing titer of each vaccine batch and testing the stability of ILT virus (ILTV) in vaccine diluents and compared the safety and efficacy of vaccines in specific pathogen free (SPF) chickens. The ratio of maximum titer to minimum titer of vaccine produced by most manufacturers was 2 to 15. However, 2 out of 11 manufacturers produced vaccines of which the ratio was 74 to 478. Most vaccines examined were maintained vaccine titers suitable for national regulations within expiry date. However, some vaccines did not keep the titer required for the national regulations. In the test for stability of ILTV in various diluents, ILTV was highly stable in lactose-phosphate-glutamine-gelatin solution, sucrose-phophate-glutamine-albumin solution and some vaccine diluents produced by manufacturers. The safety of ML ILT vaccines was assessed in 10-day-old SPF chicks. Mortality in SPF chicks inoculated intratracheally with one dose of vaccine varied depending on vaccines and some vaccines produced 50-85% mortality. Seven-week-old SPF chickens were vaccinated intraocularly with ML ILT vaccines and then challenged intratracheally with ILT challenge virus 14 days after vaccination. The protection rate was assessed by clinical signs and reisolation of the ILT challenge virus from tracheas taken at day 4 after challenge. There were slight respiratory reactions in some vaccinated chickens after vaccination but these reactions disappeared within 5 days after vaccination. No further clinical signs and death were observed. Protection rate determined by clinical signs and mortality was 100% in all vaccinated groups. However, the challenge virus was isolated from all tracheas of chickens vaccinated with vaccine B or control groups. The challenge virus was also isolated in the trachea of one in five chickens vaccinated with either vaccine F or K, but not in tracheas of chickens vaccinated with other vaccines. In the present study, the stability of vaccine diluents, pathogenicity and protection rate based on reisolation test of the challenge virus were different depending on vaccines produced by eleven manufacturers.

Fabrication of Niobium Powder for Solid-electrolyte Capacitors (고체 전해커패시터용 니오븀 분말제조)

  • The niobium capacitor showed somewhat more unstable characteristics than the commercial tantalum capacitors, but is nonetheless considered applicable as a future substitute for tantalum capacitors. In this study, niobium powder was fabricated by metallothermic reduction process using $K_2NbF_7$ as the raw materials, KCl and KF as the diluents and Na as the reducing agent. The niobium particle size greatly decreased from 0.7um to 0.2 um as the amount of diluent increased. However if a higher surface area of powder is required, more diluents need to be used in the said method in order to produce niobium powder. The niobium powder morphology and particle size are very sensitive to a amount of sodium excess. The particle size of niobium powder increased with a increasing amount of sodium excess. When more diluent and sodium are used, the niobium powder will be contaminated with more impurities such as Fe, Cr, Ni so on.

Effect of Reactive Diluents on the AC Electrical Treeing in Epoxy/Nanosilicate Systems

  • The effect of reactive diluents on the ac electrical treeing in epoxy/nanosilicate systems was studied, in a needle-plate electrode geometry. Diglycidyl ether of bisphenol A (DGEBA) type epoxy was used as a base resin, and layered silicate was used as a nano-sized filler. Polyglycol (PG) or 1,4-butanediol diglycidyl ether (BDGE) was introduced as a reactive diluent to the DGEBA/nanosilicate system, in order to decrease the viscosity of the nanocomposite system. PG acted as a flexibilizer, and BDGE acted as a chain extender, after the curing reaction. To measure the treeing propagation rate, a constant alternating current (ac) of 10 kV/4.2 mm (60 Hz) was applied to the specimen, in a needle-plate electrode arrangement, at $30^{\circ}C$ of insulating oil bath. When 10 kV/4.2 mm (60 Hz) was applied, the treeing propagate rate in the DGEBA system was $1.10{\times}10^{-3}$ mm/min, and that in the DGEBA/PG system was $1.05{\times}10^{-3}$ mm/min. As 1.5 wt% of nanosilicate was added to the DGEGA/PG system, the propagation rate was $0.33{\times}10^{-3}$ mm/min. This meant that the nano-sized layered silicates would act as good barriers to treeing propagation. The effect of chlorine content was also studied, and it was found that chlorine had a bad effect on the electrical insulation property of the epoxy system.

Effect of Vinylic Monomers on Toughness of Unsaturated Polyester (불포화 폴리에스테르 수지의 강인성에 비닐계 단량체가 미치는 효과)

  • We investigated the change of mechanical properties of UP using mixture of SM with VAc or MA that have low glass transition temperature when polymerized instead of SM only for diluents of UP. In case of using mixture of SM/MA, it was elucidated that the toughness of UP was more improved than using SM only. But in case of SM/VAc, it was observed that the content of mixture could not affect on toughness. It was concluded that these results are caused by the effect of each diluents mixtures on stress-relaxation due to phase separation and on decrease glass transition temperature of UP is quite different from each other.
