Synthesis of Galactooligosaccharides in the Cheese Whey-based Medium by a Lactase from Lactobacillus paracasei YSM0308

  • An enzyme ${\beta}$-galactosidase or ${\beta}$-galactohydrolase [EC3.2.1.23], commonly called lactase, mediates galacto-oligosaccharide (GOS) synthesis under conditions of high substrate concentrations. Also, lactase hydrolyzes ${\beta}$($1{\rightarrow}4$) lactose into glucose and galactose, the latter is successively transferred to free lactose to make various oligosaccharides via transgalactosylation. GOS is non-digestible to human digestive enzymes and has been used as a functional prebiotics. Among the 24 lactic acid bacteria (LAB) strains used, Lactobacillus paracasei YSM0308 was selected based on its exhibition of the highest ${\beta}$-galactoside hydrolysis activity, and the crude lactase was prepared for examination of reaction conditions to affect the GOS synthesis. Lactase activity was measured with a spectrophotometer using ONPG (o-nitropheyl ${\beta}$-D-galactopyranoside) method. Lactase activity was not detected in the culture supernatant and was mostly present in the cell pellet after centrifugation. Activity of the crude lactase preparation ranges from102 to 1,053 units/mL, with the highest activity determined for L. paracasei YSM0308. Optimal conditions for GOS synthesis are as follows: concentration of whey powder, pH, temperature, and time were 30%, pH 6.5-7.0, $30^{\circ}C$, and 4 h, respectively. The final GOS concentration was 19.41% (w/v) by the crude YSM0308 lactase, which was obtained from strain YSM0308 grown in the 10% (w/v) reconstituted whey-based medium.

영양강화 Rotifer와 효소활성 향상 Rotifer의 먹이효율 비교 (Comparison of Feed Efficiency Between Rotifers Enriched Lipid-contents to Enrichment and Enhanced Digestive Enzymes Activity to Starch)

  • 본 연구는 rotifer가 가진 소화효소환성과 영양적인 측면에서 자어의 소화력을 평가하기 위한 실험을 하였다. 그래서 rotifer의 소화력을 증가시켜 주기 위해 배양수에 starch를 첨가하여 rotifer (starch-rotifer)의 소화효소 활성을 향상시켰으며 지질 영양강화 rotifer (CE-rotifer)와 비교하여 넙치자어를 이용하여 먹이효율 실험을 하였다. 이들 rotifer의 소화효소 (lipase 제외), 총 단백질, 필수아미노산 총량, 필수아미노산(methionin과 phenylalanine)에서 starch-rotifer가 높게 나타났다. 하지만 총지질, 지질 클래스(sterol 제외)및 DHA, EPA와 같은 지방산은 CE-rotifer에서 높게 나타났다. 그러나 sterol과 ST/TG 비는 starch-rotifer에서 유의적으로 높게 나타났다P<0.05). 이 두 가지 rotifer를 공급받은 넙치자어는 부화 후 6일까지 체장과 체중에서 $3.72{\sim}3.79\;mm$$32.9{\sim}37.8\;mg$/larva의 범위로 유의적인 차이가 없었다(P>0.05). 그러나 부화 후 12일째는 CE-넙치에서 starch-넙치가 보다 높은 $5.94{\pm}0.249\;mm$, $144.0{\pm}23.86\;mg$/larva 및 $26.2{\pm}2.13%$의 생존율을 보였다. 이들 두 rotifer를 공급받은 넙치자어의 가수분해 효소 활성은 부화 후 5일째 acidic -amylase, neutral -amylase, TG-lipase, lysozyme 및 acidic Phosphatase에서 starch-넙치에서 유의적으로 높게 조사되었으나, 부화 후 11일째는 CE-넙치의 neutral $\alpha$-amylase, three pretenses, two phosphatases에서 starch-넙치보다 유의적으로 높은 활성을 보였다. 결과적으로 넙치자어는 부화 후 6일까지 영양강화 rotifer를 공급하는 것보다 소화효소 활성을 향상시킨 먹이를 공급하는 것이 자어의 소화력 측면에서 유익하며, 이후 소화기관의 발달로 영양의 흡수가 원활해져서 영양강화 효과가 자어의 성장으로 나타났다고 판단된다. 따라서 이후 소화효소활성과 영양의 측면에서 부화 후 6일 이후에 대한 상세한 고찰이 필요할 것으로 판단된다.

Biochemical and Genetic Characterization of Arazyme, an Extracellular Metalloprotease Produced from Serratia proteamaculans HY-3

  • Serratia proteamaculans HY-3 isolated from the digestive tract of a spider produces an extracellular protease named arazyme, with an estimated molecular mass of 51.5 kDa. The purified enzyme was characterized as having high activities at wide pH and temperature ranges. We further characterized biochemical features of the enzymatic reactions under various reaction conditions. The protease efficiently hydrolyzed a broad range of protein substrates including albumin, keratin, and collagen. The dependence of enzymatic activities on the presence of metal ions such as calcium and zinc indicated that the enzyme is a metalloprotease, together with the previous observation that the proteolytic activity of the enzyme was not inhibited by aspartate, cysteine, or serine protease inhibitors, but strongly inhibited by 1,10-phenanthroline and EDTA. The araA gene encoding the exoprotease was isolated as a 5.6 kb BamHI fragment after PCR amplification using degenerate primers and subsequent Southern hybridization. The nucleotide sequence revealed that the deduced amino acid sequences shared extensive similarity with those of the serralysin family of metalloproteases from other enteric bacteria. A gene(inh) encoding a putative protease inhibitor was also identified immediately adjacent to the araA structural gene.

Effects of dietary supplementation with different levels and molecular weights of fungal β-glucan on performances, health and meat quality in broilers

  • Objective: To investigate the effects of dietary supplementation with different levels and molecular weights of fungal ${\beta}$-glucan on productive performances, health, carcass traits and meat quality in broilers. Methods: Two hundred and ten of one-day-old chicks with equal sex were assigned to seven experimental groups in $2{\times}4$ factorial arrangement. These groups were supplemented with (0, 10, 30, and 60 ppm) of molecular weight 1-3, 1-6 ${\beta}$-glucan (low or high). High molecular weight ${\beta}$-glucan (H: 943 kDa) was obtained from Ophiocordyceps dipterigena BCC 2073, whereas H with ${\gamma}$-Irradiation treatment was performed to achieve low molecular weight ${\beta}$-glucan (L: 8 kDa). Results: There was no statistical significance in productive performances, apparent digestibility and interaction between fixed factors along 42 days of experiment (p>0.05). A higher caecal amylase activity was present in the group that received L, while there was a dramatic decrease in H and the control groups, respectively (p<0.05). The increase of supplemental dose increased caecal amylase activity (p<0.05). Immunomodulatory effects from L was revealed by the marked increase of phagocytic activity, relative weight of thymus and bursa of fabricius (p<0.05). Similarly, the additive dose at 30 ppm provided the same results, whereas the only significant difference with supplementation at 60 ppm was an increase in phagocytic activity (p<0.05). Interestingly, villi height of broilers fed L was higher than other groups (p<0.05). The treatments did not influence haematology, blood chemistry, antibody production level against vaccination, carcass traits and meat quality (p>0.05). Conclusion: The supplementation of L at 30 ppm was suggested to achieve benefits of immune modulation without adverse effects on other parameters.

아메리카동애등에 장내세균 동정과 생리적 특징 (Identification and Physiological Characters of Intestinal Bacteria of the Black Soldier Fly, Hermetia illucens)

  • 다양한 먹이 조건에서 생활하는 아메리카동애등에(Hermetia illucens)는 장내 세균의 의존성을 가질 수 있다. 이 가설을 증명하기 위해 본 연구는 종령 유충의 소화관에 존재하는 세균을 분리, 동정하고 효소활성 및 항균 능력을 분석하였다. 종령 유충의 소화관은 몸 체장에 약 7 배의 길이를 나타냈다. 한 개체의 소화관 내 존재하는 세균 수는 $5.0{\times}10^6$ cfu로 98% 이상이 후장에 존재했다. 소화관에는 3 종류의 상이한 세균이 존재했고, 미생물 동정 장치는 이들이 각각 Morganella morganii, Providencia rettgeri 및 Bacillus halodurans로 동정하였다. 이들 소화관 세균을 16S rDNA 서열을 분석한 결과 이 외에 Proteus mirabilis, Providencia alcalifaciens, Providencia sp.를 검출하였다. 이들 장내세균은 항생제 내성을 보였고, 타 미생물의 성장을 억제하였다. 또한 섬유소, 지질, 단백질 및 탄수화물의 분해 능력을 보유하였다. 본 연구 결과들은 아메리카동애등에 소화관에 유용성이 높은 세균을 보유하고 있다고 제시하였다.

Effect of ${\beta}$-Glucosidase as a Feed Supplementary on the Growth Performance, Digestive Enzymes and Physiology of Broilers

  • The effects of ${\beta}$-glucosidase on the overall growth performance and a set of physiological parameters of broilers were investigated. 240 male, one-day old Avine broiler chickswere randomly allocated to four treatment groups and fed with a corn-soybean meal supplemented with 0% (control), 0.2%, 0.4% and 0.6% ${\beta}$-glucosidase. The 0.2% ${\beta}$-glucosidase group, but not the 0.4% and 0.6% ${\beta}$-glucosidase groups, showed a significantly increased average daily weight gain (p<0.05) over that of the control. All three ${\beta}$-glucosidase feed groups showed significantly higher feed conversion ratios than the control group (p<0.05). Feed supplementation of 0.2% ${\beta}$-glucosidase significantly raised the contents of serum isoflavone aglycones as shown by decreases of genistin and daizin (p<0.01) and an increase of daidzein (p<0.01). The 0.2% ${\beta}$-glucosidase feeding significantly increased the intestinal amylase activity while it had little effect on lipase and trypsin activities (p>0.05). 0.2% ${\beta}$-glucosidase feeding also significant elevated the levels of highdensity lipoprotein cholesterol and malate dehydrogenase while lowering the level of low-density lipoprotein cholesterol (LDL-C). Finally, ${\beta}$-glucosidase improved the anti-oxidative activities of the animals; the 0.2% ${\beta}$-glucosidase feed group had higher activities of superoxide dismutase (p<0.05), glutathione peroxidase and glutathione reductase in the liver (p<0.05), and malondialdehyde level in the serum (p<0.05).

식이섬유 식이에 적응된 흰쥐에서 고지방식이가 식후 혈장지질농도와 소화효소활성에 미치는 영향 (Postprandial Plasma Lipid Levels and Digestive Enzyme Activities After High Fat Meal in Rats Adapted to Dietary Fiber)

  • 식이섬유에 5주간 적응된 횐쥐 (S.D., male)를 14시간 절식시킨 뒤 총 에너지의 50%가 되는 고지방식이를 급여하고 4시간 뒤의 혈장 지질 농도와 소화효소활성을 조사하여 다음과 같은 결과를 얻었다. 혈장과 지단백의 콜레스테롤 함량은 sodium alginate군에서 가장 낮았다(p<0.05). 혈장 중성지방 함량은 식이군간에 차이가 없었으나 LDL-중성지방은 sodium alginate군에서 가장 낮았다(p<0.05). HDL-중성지방 함량은 식이섬유 군에서 높았다 (p<0.05). 소장 apolipoprotein B는 무섬 유식이군과 cellulose군에 비해 sodium alginate군에서 유의적으로 높았다(p<0.05). 소장 내용물에서의 amylase활성은 각 식이군간에 유의적인 차이는 없었으나 식이 섬유 첨가군에서 다소 높았으며, lipase 활성은 무섬유 식이군과 cellulose군에 비해 sodium alginate군에서 유의적으로 높았다(p<0.05). 소장내용물에서 pretense 활성과 담즙산 함량은 식이군간에 유의적인 차이가 없었으나, 단백질 함량은 무섬유 식이군에 비해 식이섬유 첨가군에서 유의적으로 높았다(p<0.05). 이상의 결과를 종합해 볼 때 장기간의 식이 섬유 섭취는 식이섬유가 배제된 고지방식 후에도 혈장 콜레스테롤 농도를 낮추는 효과가 있으나, 혈장 중성지방 농도는 식이 섬유가 함유된 식이에 비해 영향을 받지 않는 것으로 보인다.

효소 분해에 의한 난황 항체의 항원 결합력 변화 (Changes of Antigen Binding Activities of Hen′s Egg Yolk Immunoglobulins after Proteolysis)

  • 체내 소화효소에 대한 안정성을 검토하기 위해 competitive ELISA를 이용하여 항원 결합력의 변화를 조사하였다. YIgG는 펩신에 대해 상당히 불안정하여 pH 2.0에서 30분간의 반응에 의해 항원 결합력이 소실되었다. 한편 yIgG 용액에 50%(w/v) saccharose를 첨가하여 펩신과 30분 반응시킨 결과 native yIgG에 비해 항원 결합력이 6.7배 정도 저하되었으나 미첨가시에 비해 항원 결합력이 상당히 유지되었으므로, 펩신에 대한 yIgG의 안정성은 saccharose에 의해 상당히 증가되었음을 알 수 있다. YIgG는 트립신 및 키모트립신과 반응 후 항원 결합력의 큰 저하는 나타나지 않아 펩신에 비해 상당히 안정한 것으로 생각된다. 트립신과 8시간 반응 후 yIgG의 항원 결합력은 native yIgG에 비해 2배 감소되었으나 키모트립신과 8시간 반응 후 yIgG의 항원 결합력은 1.5배 감소되었다. 그러므로 yIgG는 키모트립신 대해 가장 안정하였다.

Purification and Characterization of a Protease Produced by a Planomicrobium sp. L-2 from Gut of Octopus vulgaris

  • Protease widely exists in the digestive tract of animals and humans, playing a very important role in protein digestion and absorption. In this study, a high protease-producing strain Planomicrobium sp. L-2 was isolated and identified from the digestive tract of Octopus variabilis. The strain was identified by physiological and biochemical experiments and 16S rDNA sequences analysis. A protease was obtained from the strain Planomicrobium sp. L-2 through ammonium sulfate precipitation, dialysis and enrichment, DEAE-Sephadex A50 anion-exchange chromatography, and Sephadex G-100 gel chromatography. The molecular weight and properties of the protease were characterized, including optimum temperature and pH, thermal stability, protease inhibitions and metal ions. According to our results, the protease from Planomicrobium sp. L-2 strain designated as F1-1 was obtained by three-step separation and purification from crude enzyme. The molecular weight of the protease was 61.4 kDa and its optimum temperature was $40^{\circ}C$. The protease F1-1 showed a broad pH profile for casein hydrolysis between 5.0~11.0. No residual activity was observed after incubation for 40 min at $60^{\circ}C$ and 60 min at $50^{\circ}C$. F1-1 protease was inhibited by $Mn^{2+}$, $Hg^{2+}$, $Pb^{2+}$, $Zn^{2+}$, and $Cu^{2+}$ ions, as well as PMSF, indicating that the protease F1-1 was a serine protease. Additionally, research basis provided by this study could be considered for industrial application of octopus intestinal proteases.

Effect of Graded Levels of Tallow in the Diet on Performance, Digestibility of Fat, Lipogenesis and Body Lipid Deposition of the Weaned Piglet

  • Thirty piglets weaned at 24.5 d of age ($6.9{\pm}0.5kg$) randomly alloted to 3 treatments were used to investigate the effect of dietary tallow on average performance, digestibility of nutrients, metabolic utilization of energy and body composition at 25 kg. Weaned piglets respond to increasing levels of dietary tallow from 0 to 4% and 8% by digestive and metabolic adaptation. Apparent fecal digestibility of fat (AFDf) was highly correlated with the level of dietary tallow (X as % of fat extracted after HCl hydrolysis) by the following curvilinear equation of regression: $AFDf=33.8+6.9X-0.3X^2$. Feed intake expressed as DE was only significantly increased at the higher inclusion level of tallow. But neither average daily gain, nor feed conversion was affected by the addition of fat. On the other hand, body composition at 25 kg was equally affected, by both levels of supplementary fat; dry matter and energy content in the body were significantly higher (p<0.01) in piglets receiving tallow. As a consequence, the energy cost of the live weight gain was also increased from 23 to 24.7 MJ DE/kg (p<0.02) and the efficiency of energy deposition was decreased from 3.2 to 2.8 MJ DE/MJ deposited energy (p<0.01) in the presence of dietary tallow. An increase in the level of fat stimulated the activity of pancreatic lipase up to a constant value of $22{\pm}1.4IU/mg$ protein but conversely depressed the activity of amylase from 300 to 100 IU/mg of protein. The activity of liver acetyl CoA carboxylase and malic enzyme in the perirenal fat were low lind not affected by dietary fat; the activity of glucose-6-phosphate dehydrogenase was high. Opposite to that, the activity of acetyl CoA carboxylase and malic enzyme in the perirenal and backfat were higher than in the liver and both were significantly reduced by the inclusion of fat in the diet. A direct deposition of dietary fat has been demonstrated by increasing the energy and lipid content of the empty body weight gain between 7 and 25 kg of live weight, and decreasing the efficiency of digestible energy utilization.
