Characterization of Bone Marrow Cell Proliferating Arabinogalactan through Peyer`s Patch Cells from Rhizomes of Atractylodes lancea DC

  • Bone marrow cell proliferating arabinogalactan-like polysaccharide (ALR-3IIa-1-1) has been purified from rhizomes of Atractylodes lancea DC. In order to characterize the essential structure of ALR-3IIa-1-1 for expression of the activity, sequential enzymatic digestion using ego-$\alpha$-L-arabinofurasidase (AFase) and ego-$\beta$-D-(1longrightarrow3)-galactanase (GNase) was employed. After ALR-3IIa-1-1 was digested with the AFase, the GNase digestion cleaved only 10% and 23% of 3-linked and 3,6-branched galactose, respectively, from arabinose-trimmed ALR-3IIa-1-1 (AT-ALR-3IIa-1-1), and gave small amounts of intermediate size (AT-G-2) and shorter oligosaccharides (AT-G-3) fractions in addition to a large amount of the GNase resistant fraction (AT-G-1). When AT-G-1 was redigested gradually with the AFase and GNase, it released trace amounts of oligosaccharides in addition to a large amount of the resistant fraction. When the final enzyme-resistant fraction from AT-G-1 was digested simultaneously with both AFase and GNase, the resistant fraction was significantly degraded into two long fragments (3AT-3G-1 and 2). The mixture of digestion products from the first GNase digestion of AT-ALR-3IIa-1-1 showed a significantly decreased bone marrow cell proliferation activity to about 30% of the activity of ALR-3IIa-1-1, but the GNase resistant fraction (AT-7-1) still had significant activity. Although the second gradual enzymatic digestion of AT-G-1 showed a marginal decrease in activity, the resulting fragments (3AT-3G-1 and 2) by the final simultaneous enzymatic digestion lost most of the activity. Component sugar, methylation and FAB-MS analyses indicated that the digestion products (AT-G-21 AT-G-31 2AT-2G-2 and 2AT-2G-3) released from AT-ALR-3IIa-1-1 by the sequential enzymatic digestion contained galactose-containing oligosaccharides mainly comprising 6-linked galactose, that some of which were partially arabinosylated, and these oligosaccharides were attached to $\beta$-D-(1longrightarrow3)-galactan backbone in its non-reducing terminal side as side chains.

Effect of heat treatment of digestion-resistant fraction from soybean on retarding of bile acid transport in vitro

  • In this study, we investigated the heat effect of digestion-resistant fraction (RF) from soybean on retarding bile acid transport in vitro. The RFs from soybean retarded bile acid transport. A raw, unheated RF of soybean (RRF-SOY) was significantly more effective than the heated RF of soybean (HRF-SOY). The RS1 which physically trapped in milled grains and inaccessible to digestive enzyme after 18 hrs incubation level of content in RRF-SOY was found to be as high as 24.1% and after heating the RS1 of HRF-SOY was significantly reduced to 16.8%. The X-ray diffraction pattern of RF from soybean was altered after heat treatment. The RFs from soybean were characterized by peak at diffraction angles of $12.0^{\circ}$ and $20.0^{\circ}$ corresponding to RS content. Cellulose contents of RRF-SOY was 5% higher than that of HRF-SOY and pentosan contents of RRF-SOY was 5% higher than that of HRF-SOY, too. Whereas the hemicellulose content of RRF-SOY was 13% lower than HRF-SOY.

In Vitro Digestibility of Rice and Barley in Forms of Raw Flour and Cooked Kernels

  • Digestion properties of 3 types of cereals, white rice, brown rice, and barley, were measured after cooking or grinding. Regardless of the processing methods, white rice showed the highest rate and the greatest extent of digestion, whereas barley showed the lowest values. During the early digestion period, cooked white rice kernels had a larger k (kinetic constant) value than uncooked white rice flour, indicating that cooking induced faster digestion than grinding. In the case of brown rice and barley, the cell wall in cooked kernels remained intact and resulted in a lower k values than those of uncooked flour. However, after 3 hr of digestion, the total digestion extent was greater for the cooked brown rice and barley than that for uncooked flours. The high content of slowly digestible starch (SDS) in cooked brown rice and barley might be due to the starch fraction which was protected by the cell wall. The resistant starch (RS) content, however, was greater for the uncooked flours than that for cooked kernels. The cooked kernels of 3 cereal samples tested showed higher glycemic index (GI) values than the uncooked flours.

  • 1. Pepsin과 pancreatin 소화물들을 비교한 결과, 순수한 팥 단백질 중에서 소화효소 저항성을 가지는 단백질이 존재하는 것으로 확인되었다. 2. 팥의 주요 단백질을 제거하고 pepsin과 pancreatin으로 소화시켰을 때 더 많은 분해가 일어나는 것으로 보아 팥의 주요단백질들 중에 소화효소 저항성을 가지는 것이 많을 것이라 추측된다. 3. 팥의 주요 단백질들은 장 점막세포와는 크게 작용하지 않는 것을 확인할 수 있었다. 4. 팥 단백질의 데이터베이스 구축과 팥 단백질이 다른 영양소들과의 상호작용을 하는 지에 대한 연구가 진행되어야 할 것이다.

Lactobacillus plantarum 균체 중 항보체 활성물질의 특성과 작용양식 (Characterization and Action Mode of Anti-Complementary Substance Prepared from Lactobacillus plantarum)

  • 김치 및 발효유 제품으로부터 분리한 유산균과 공시균주 12종을 대상으로 보체 용혈 분석법을 이용하여 면역계에서 중요한 역할을 담당하고 있는 보체계 활성화(항보체 활성, $TCH_{50}$) 정도를 측정한 결과, 김치로 부터 분리한 Lactobacillus plantarum이 타 유산균 종에 비해 높은 활성을 나타내었다. 이들 균주로 부터 조제된 세포벽 획분의 경우 세포질 획분보다도 높은 활성을 보였으며 각 획분의 활성은 농도 의존적 경향을 나타내었다. L. plantarum의 세포질 획분과 세포벽 획분의 경우 pronase 소화 후에는 활성의 변화가 없는 반면, 과요오드산 처리에 의해서는 급격한 활성의 감소를 나타내는데 이들 결과로부터 L. plantarum의 세포질과 세포벽 획분에 의한 보체계 활성화가 주로 다당 영역에 기인함을 알수 있었다. 한편 anti-human C3를 이용한 2차원 면역전기영동에 의해, $Ca^{++}$ 이온을 제거한 상태에서도 세포질과 세포벽 획분에 의한 C3 활성화 산물을 동정할 수 있었다. 또한 L. plantarum의 세포벽 획분에 의한 항보체 활성은 동일 조건에서 활성을 유지한 반면, 세포질 획분에 의한 활성화 정도는 동일 조건에서 상당히 감소하였다. 이상의 결과로부터 L. plantarum 세포벽 획분의 보체계 활성화 양식은 주로 alternative pathway의 활성화에 의한 것이며, 세포질 획분에 의한 활성화는 classical pathway와 alternative pathway 양 경로를 경유함을 알 수 있었다.

늙은 호박(Cucurbita moschata D.) 가루 첨가량에 따른 호박편의 품질특성 및 저장성 (Quality Characteristics and Storage Properties of Hobakpyeon with Different Amounts of Pumpkin (Cucurbita moschata D.) Powder)

  • The objective of this study was to investigate the effects of different amounts of pumpkin powder on the quality characteristics of hobakpyeon using physicochemical and sensory properties, as well as on its retrogradation rate during storage. As the amount of pumpkin powder increased, the following effects on the quality characteristics were observed: moisture and amylose contents decreased while protein content increased (p<0.05), and in vitro protein digestibility (IVPD) decreased (p<0.05). Levels of slowly digestible starch and resistant starch fractions increased, while the content of rapidly digestible starch decreased. The starch digestion index and rapidly available glucose content also decreased. Among the physical properties, the L-value decreased while the b-value increased (p<0.05). Texture profile analysis revealed that all textural properties except adhesiveness decreased, and presented significant differences in hardness and chewiness (p<0.05). Sensory properties such as color, flavor, and sweetness increased while others such as wetness and chewiness decreased (p<0.05). Effects on the physicochemical properties during refrigerated storage were also compared between the control and 9% hobakpyeon (9%HP). According to the results, moisture content gradually decreased in the 9%HP compared to the control. Amylose content significantly increased in 9%HP whereas no significant changes were observed in the control. In both types of hobakpyeon, IVSD decreased significantly, showing higher values in 9%HP, whereas mechanical hardness increased, showing lower values in 9%HP. The L-value decreased significantly in the 9%HP whereas no significant changes were observed in the control. In conclusion, the above results suggest that hobakpyeon with low IVSD may be obtained by the addition of 9% pumpkin powder, and also showed that pumpkin powder delayed the rate of retrogradation in 9%HP during storage.

Pleurotus속 균주들의 미토콘드리아 플라스미드 특성 (Characterization of Mitochondrial Plasmids from Pleurotus spp.)

  • 백색 부후균인 plaurotus ostreatus 의 4가지의 균주로부터 각각 10.2 kb 와 7.2 kb (NFFA 2), 두 종류의 10.2 kb (NFFA 4001) 11.2 kb (NFFA 4501), 10.2kb 와 11.2 kb(KFCC 11635) 크기의 미토콘드리아 플라스미드들을 분리해 내었다. NFFA 2 의 변종인 NFFA 2m1 과 NFFA 2m2 에서는 이들 플라스미드가 관찰되지 않았다. 분별 원심분리에 의해 얻은 미토콘드리아에서 핵산을 추출하여 agarose gel 에서 전기영동시키면 플라스미드가 관찰되지 않았으나 proteinase K 를 처리하고 핵산을 추출하여 전기영동한 결과 이들 플라스미드가 관찰되었는에, 이는 플라스미드상에 단백질인 결합되어 있음을 시사한다. Proteinase K 를 처리한 플라스미드 DNA 와 exonuclease 를 반응시킨 결과, 이들 플라스미드들은 5'말단에 단백질이 결합된 성형 이중가닥 DNA의 구조를 가진 것을 확인하였다. 각 플라스미드들의 상호관계를 조사하기 위하여 Southern hybridization 을 수행한 결과 최소 3가지 종류의 플라스미드들로 분류할 수 있었다. 이중 한 그룹 (group I) 은 모든 P. ostreatus 균주들에서 공통적으로 발견되었다. Pleurotus 속의 5가지 다른 종(P. cornucopiae, P. florida, P. pulmonarius, P. sajor-cuja, P. spodoleucus) 의 균주들로부터 미토콘드리아 플라스미드들을 분리하였다. 이들은 한균주당 1-4 개 까지의 플라스미드를 가지며, 플라스미드의 크기는 7.2 kb-14kb 범위에 있었다. P. ostreatus 의 NFFA 2 의 10.2 kb(group I) 플라스미드와 hybridization 을 수행한 결과 P. cornucopiae ASI 2011을 제외한 다른 모든 균주들이 유사한 염기서열의 플라스미드를 갖고 있음을 알았다.
