Effect of seeding ratio on acidogenic biokinetics in high ammonia concentration

  • Anaerobic digestion is one of the well-known methods for biological treatment handling of concentrated organic matter such as swine $wastewater.^{1)} The anaerobic digestion can reduce organic loading but also hydrolyze non-biodegradable organic $matter.^{2)}$ The feces from the scrapper-type barn are usually collected to make compost and the urine is discarded with swine-slurry wastewater by ocean-dumping or treated by biological methods. The lagoon, aerobic digestion, anaerobic digestion, SBR, $A^{2}/O$, and UCT have been applied for treating swine $wastewater.^{3)} In this study, as a result of the analysis of swine wastewater, the total and soluble chemical oxygen demand was 130g/L and 60g/L, respectively. And the volatile fatty acid as chemical oxygen demand equivalent was 45g/L, which was 75% of soluble chemical oxygen demand. Before everything else, ammonia nitrogen concentration was 6.5 g/L. From biochemical acidogenic potential test, it was concluded that the enhanced acidification process to manage swine waste should be operated in the ammonia nitrogen concentration of less than 1.2 g/L. In the result of seeding ratio experiments with artificial $wastewater^{4)}, the lag period of acidogens was taken the long time because of the inhibition by the $ammonia^{5)}$, however no difference of period by the seeding ratio was not shown. The Haldane-based biokinetics were also evaluated using a method of fourth order Runge-Kutta $approximation.^{6,7)}$ The nonlinear least squares (NLLS) method with a 95% confidence interval was also used. The ranges of maximum microbial growth rate, ${/mu_{max}}$, and half saturation coefficient, $K_{s}$, for acidogenesis of various seeding ratio with artificial wastewater were 6.1 ~ 12.6 $d^{-1}$ and 45,000 ~ 53,500 mg glucose/L, respectively. Also, the methanogenic microbial yield coefficient, Y, and microbial decay rate coefficient, $k_{d}$, and inhibition substrate concentration, $K_{si}$, for the reactors were determined to be 0.32 ~ 0.465 ${/mu}g$/mg glucose; 0.42 ~ 1.01 $d^{-1}$ and 51,500 ~ 55,600 mg glucose/L, respectively.

Effects of Soybean Small Peptides on Rumen Fermentation and on Intestinal and Total Tract Digestion of Luxi Yellow Cattle

  • Four Luxi beef cattle ($400{\pm}10$ kg) fitted with ruminal, duodenal and ileal cannulas were used in a $4{\times}4$ Latin square to assess the effects of soybean small peptide (SSP) infusion on rumen fermentation, diet digestion and flow of nutrient in the gastrointestinal tract. The ruminal infusion of SSP was 0 (control), 100, 200 and 300 g/d. Ruminal SSP infusion linearly (p<0.01) and quadratically (p<0.01) increased microbial protein synthesis and rumen ammonia-N concentration. Concentrations of total volatile fatty acid were linearly increased (p = 0.029) by infusion SSP. Rumen samples were obtained for analysis of microbial ecology by real-time PCR. Populations of rumen Butyrivibrio fibrisolvens, Streptococcus bovis, Ciliate protozoa, Ruminococcus flavefaciens, and Prevotella ruminicola were expressed as a proportion of total Rumen bacterial 16S ribosomal deoxyribonucleic acid (rDNA). Butyrivibrio fibrisolvens populations which related to total bacterial 16S rDNA were increased (p<0.05), while Streptococcus bovis populations were linearly (p = 0.049) and quadratically (p = 0.020) decreased by infusion of SSP. Apparent rumen digestibility of DM and NDF were (Q, p<0.05; L, p<0.05) increased with infusion SSP. Total tract digestion of DM, OM and NDF were linearly (p<0.01) and quadratically (p<0.01) increased by infusing SSP. The flow of total amino acids (AA), essential amino acids (EAA) and individual amino acids were linearly (p<0.01) and quadratically (p<0.01) increased with infusion SSP. The digestibility of Lysine was quadratically (p = 0.033) increased and apparent degradability of Arginine was linearly (p = 0.032) and quadratically (p = 0.042) increased with infusion SSP. The results indicated that infusion SSP could improve nutrient digestion, ruminal fermentation and AA availability.

Evaluation of the Degradation of Carbohydrate-based Material During Anaerobic Digestion for High-efficiency Biogas Production

  • Purpose: In this study, the potential for biogas production, degradation rates, and lag-phase of diauxic growth of carbohydrate-based material, which is one of the proximate compositions, were investigated. Methods: This study was conducted using starch as a carbohydrate-based material. In experimental condition 1, the biogas potential of carbohydrate-based material was measured. In experimental condition 2, the effect of feed to microorganism ratio (F/M ratio) on lag-phase of diauxic growth from carbohydrate-based material was tested. Biochemical methane potential tests were performed at five different feed to microorganism ratios (0.2, 0.4, 0.6, 0.8, and 1.0) under mesophilic conditions. The biogas production patterns, lag-phase, total volatile fatty acids to total alkalinity ratio (TVFA/TA ratio), and time required for 90 percent biogas production were used to evaluate biogas production based on the biochemical methane potential tests. Results: In experimental condition 1, unlike previous studies, biogas was produced in the TVFA/TA ratio ranging from 1.131 to 2.029 (approximately 13-19 days). The methane content in the biogas produced from the digesters was 7% on day 9 and increased rapidly until approximately day 27 (approximately 72%). In experimental condition 2, biogas yield was improved when the feed to microorganism ratio exceeded 0.6, with an initial lag-phase. Conclusions: Even if the TVFA/TA ratio was greater than 1.0, the biogas production was processed continuously, and the $CO_2$ content of the biogas production was as high as 60%. The biogas yield was improved when the F/M ratio was increased more than 0.6, but the lag-phase of carbohydrate-based material digestion became longer starting with high organic loading rate. To clarify the problem of the initial lag-phase, our future study will examine the microbial mechanisms during anaerobic digestion.

Development of the rapid preconcentration method for determination of actinides in large volume seawater sample using Actinide resin

  • A simple and rapid preconcentration method of actinide from seawater using Actinide resin was developed and tested with the seawater spiked with a known U and Th. The developed method of Actinide resin based on column chromatography is less time-consuming and requires less labor compared with a typical co-precipitation technique for preconcentration of actinides. U and Th, which are relatively weak-bonded with Actinide resin among actinides, were used to determine the optimum flow rate of seawater sample and evaluate the capacity of Actinide resin to concentrate actinides from seawater. A flow rate of 50 mL min-1 was available with Actinide resin 2 mL (BV, bed volume). When 5 or 10 L of seawater containing U were loaded on Actinide resin (2 mL, BV) at 50 mL min-1, the recovery of U was 93 % and 86 %, respectively. For extraction of actinides bound with Actinide resin, we compared three methods: solvent extraction, ashing-acid digestion, and ashing-microwave digestion. Ashing-microwave digestion method shows the best performance of which is the recovery of 100 % for U and 81 % for Th. For the preconcentration of actinides in 200 L of seawater, a typical coprecipitation method requires 2-3 days, but the developed method in this study is achieved the high recovery of actinides within 12 h.

Effect of Additives and Fermentation Periods on Chemical Composition and In situ Digestion Kinetics of Mott Grass (Pennisetum purpureum) Silage

  • This study was conducted to see the influence of additives and fermentation periods on Mott grass silage (MGS) characteristics, its chemical composition and to compare the digestion kinetics of Mott grass (MG) and MGS in Nili buffaloes. Mott grass chopped with a locally manufactured chopper was ensiled using two additives, cane molasses and crushed corn grains each at 2, 4 and 6% of forage DM for 30 and 40 days in laboratory silos. The pH, lactic acid concentration, dry matter (DM), crude protein and fiber fractions of MGS were not affected by the type or level of additive and fermentation periods. The non-significant pH lactic acid concentration, and chemical composition of MGS indicated that the both molasses and crushed corn were utilized at similar rate for the growth of lactic acid bacteria and production of organic acids. The MG ensiled with molasses at 2% of fodder DM for 30 days was screened out for in situ digestion kinetics in Nili buffaloes. Ruminal DM and neutral detergent fiber (NDF) degradabilities of MGS were significantly (p<0.05) higher than that of MG. The DM and NDF rate of degradation, lag time and extent of degradation was nonsignificant between MGS and MG. The higher ruminal degradation of DM and NDF of MGS than MG was probably a reflection of fermentation of MG during ensilation that improved its degradability by improving the availability of easily degradable structural polysaccharides to ruminal microbial population. The results in the present study have indicated that MG ensiled with either 2% molasses or 2% crushed corn for 30 days has better nutritive value for buffalo.

IN SITU RUMINAL DEGRADATION KINETICS OF FORAGES AND FEED BYPRODUCTS IN MALE NILI-RAVI BUFFALO CALVES

  • The rate and extent of digestion of dietary carbohydrates has a tremendous impact on ruminal fermentation and the productivity of the animals. The objective of the study was to determine the dry matter (DM) and neutral detergent fiber (NDF) degradabilities and rate and extent of feed byproducts (cotton seed cake, wheat bran), legumes [berseem (Egyptian clover), lucern (Medicago sativa), cowpeas (Vigna sinensis)], grasses [maize (Zea mays), millet (Panicum miliaceum), sorghum (Sorghum vulgare)] and wheat straw in ruminally fistulated male buffalo calves. By using nylon bags, 10 grams sample was exposed to the ruminal fermentation for 0, 1, 2, 4, 6, 10, 16, 24, 36, 48 and 96 hours. Dry matter and NDF degradability was measured at 48 hours. Extent of DM and NDF disappearance was determined at each time point. Rates of disappearance of DM and NDF were determined by regressing the natural logarithm of the percentage of original DM and NDF remaining in the bags between 1 and 96 hours. The dry matter digestibility (DMD) of the feed byproducts (FBP) and legume forages when incubated in the rumen of male buffalo calves were greater (p < 0.05) than grasses. Extent of digestion followed similar pattern as DMD. Rate of DMD was higher in FBP than in legumes and was the lowest in the wheat straw. The NDF degradability (NDFD) of FBP, legumes and grasses did not differ, however, wheat straw had the lowest NDFD from all the feeds tested. The lowest NDFD of wheat straw may have been due to the depressing effect of lignin on fiber digestion. The FBP and legumes had higher (p < 0.05) rates and lower extents of NDF digestion than grasses.

Estimation of Ruminal Degradation and Intestinal Digestion of Tropical Protein Resources Using the Nylon Bag Technique and the Three-step In vitro Procedure in Dairy Cattle on Rice Straw Diets

  • The experiment was carried out using fistulated multiparous Holstein Friesian crossbred (75% Holstein Friesian and 25% Red Sindhi) dairy cows in their dry period fed on untreated rice straw to evaluate the nutritive value of local protein feed resources using the in sacco method and in vitro pepsin-pancreatin digestion. Experimental feeds were cottonseed meal (CSM); soybean meal (SBM); dried brewery's grains (DBG); palm kernel meal (PSM); cassava hay (CH); leucaena leaf meal (LLM). Each feedstuff was weighed into duplicate nylon bags and incubated in each of the two rumen fistulated cows for 0, 2, 4, 8, 16, 24, and 48 h. Rumen feed residues from bags of 16 h incubation were used for estimation of lower gut digestibility by the technique of in vitro pepsin-pancreatin digestion. Ruminal ammonia-nitrogen ($NH_3-N$) concentrations did not differ between treatments or time with a mean of 5.5 mg%. Effective degradability of DM of CSM, SBM, DBG, PSM, CH and LLM were 41.9, 56.1, 30.8, 47.0, 41.1 and 47.5%, respectively. Effective degradabilities of the CP in feedstuffs were 49.6, 59.2, 40.9, 33.5, 47.3 and 65.0% for the respective feedstuffs. The CP in vitro pepsin-pancreatin digestibility as ranked from the highest to the lowest were SBM, CSM, LLM, CH, DBG, PSM, respectively. The intestinal and total tract digestion of feedstuffs in the current study were relatively lower than that obtained from previous literature. The results of this study indicate that SBM and LLM were highly degradable in the rumen, while CH, CSM and DBG were less degradable and, hence resulted in higher rumen undegradable protein. Soybean meal and LLM could be used to improve rumen ecology whilst CH, CSM and DBG could be used as rumen by-pass protein for ruminant feeding in the tropics.

화학적 전처리를 통한 혐기성 슬러지 처리효율의 향상 (Performance Enhancement of Anaerobic Treatment of Waste Sludge by Chemical Pretreatment)

  • Laboratory-scale experiment using anaerobic fluidized bed reactor was carried out to investigate the prehydrolysis step with caustic soda on the treatment efficiency of anaerobic sludge treatment, since the overall rate-limiting step for the complete anaerobic digestion of sludge was the hydrolysis step by extracellular bacterial enzymes of insoluble polymeric molecules. Reactors received a sludge which had not been pretreated, a 50-50 mixture of pretreated and untreated sludge, and the fully pretreated sludge. Hydraulic retention time of 10, 5, 2.5 days and 1 day were applied with an respective equivalent organic loading rate of 1.17, 2.23, 4.17, 11.24 gCOD/L/d. Reactor with the untreated sludge did not archieve adequate digestion even at the HRT of 5 days, and reactor, which received the 50-50 mixture, operated well at the HRT of 5 days, but began to show signs of unstable digestion at the HRT of 2.5 days. While, reactor, which was fed the hydrolyzed sludge, operated reasonably well at the 2.5 days, but was showing somewhat decrease in removal efficiencies. Results, therefore, have substantiated that the limiting reaction in the anaerobic treatment process is hydrolysis. The soluble COD did not significantly accumulate in the reactor as organic acid form, even when they were highly stressed. It was believed that this resistance to a build-up of organic acids and soluble COD behavior was mainly due to the maintenance of the methane bacteria in the fixed-film system which prevents washout as the organic loading increased. The anaerobic fluidized bed reactor was therefore effective for the digestion of waste activated sludge at short HRT.

ICP-AES에 의한 수중의 금속 산화물 직접분석을 위한 연속흐름 Microwave 용해장치 개발 연구 (The Development of Continuous Flow Method Through Microwave Oven for the Analysis of Metal Oxides in Water by ICP-AES)

  • 수중의 금속 산화물을 신속하고 간단하게 용해시킬 수 있는 연속흐름 마이크로파 용해장치를 개발하였다. 이로서 금속 산화물 입자를 용해시킴과 동시에 유도결합 플라즈마 분광기(ICP-AES)를 이용하여 ON-LINE으로 분석하는 완전 자동화 분석 장치의 개발이 가능토록 하였다. 연속흐름 마이크로파 용해장치는 개방형 튜브 장치와 억제형 튜브 장치로 구분하여 제작하고 최적화 시험을 수행하였다. 개방형 튜브 용해장치의 최적조건은 튜브길이가 30m, 마이크로파 출력 20%, 염산 농도 0.6N, 유속은 3.5ml/min였으며, 억제형 튜브 용해장치는 튜브 길이가 7.2m, 마이크로파 출력 30%, 염산 농도 0.6N, 유속은 3.1ml/min였다. 억제형 튜브 용해장치가 개방형 장치에 비하여 용해시간이 3배 정도 단축되었고 용해시킬 수 있는 시료의 농도 범위도 10배 정도 높았다. 제작된 용해장치의 정밀도는 베치식 용기 용해법과 거의 유사한 결과를 나타내어 Fe, Cu는 5% 정도의 상대 표준 편차값을 나타내었고, Zn,Co는 10%내외로 다소 높게 나타났다.

고농도 입자성 유기폐수의 고효율 혐기성 소화 공정 (Efficient Anaerobic Digestion for Highly Concentrated Particulate Organic Wastewater)

  • 고농도 입자성 유기 폐수를 대상으로 기존 혐기성 공정인 CSTR과 UASB공정으로 처리 시 불량한 상징수질, 불안정한 운전 특성과 같은 문제점이 제시되었다. 이에 본 연구는 새로운 개념의 ADEPT(Anaerobic Digestion Elutriated Phased Treatment) 공정을 주정폐수에 적용하여 처리 가능성을 평가하였다. CSTR과 ADEPT의 성능 비교 결과 ADEPT공정에서 gas 발생량은 약 2배였으며, 유기물 제거 효율과 고형물 제거 효율 역시 더 높은 효율을 보여주었다. 또한 ADPET는 비교적 짧은 HRT에서도 휘발성 지방산 생성으로 인한 pH 저하의 영향을 받지 않았으며, 안정적으로 pH를 유지하였다. ADEPT의 recycle ratio를 6Qin과 2Qin으로 변화를 주어 운전 시 6Qin의 운전 기간 동안 높은 처리 효율을 보여주었다. 따라서 ADEPT는 짧은 HRT로 인한 경제성 향상과 메탄 발생을 이용한 에너지 회수, 또한 산 생성조에서 생성되는 유기산을 최대로 생산, BNR 공정의 유기 탄소원으로 활용 가치가 있다고 판단된다.
